Achieving Fast Charge Separation by Ferroelectric Ultrasonic Interfacial Engineering for Rapid Sonotherapy of Bacteria-Infected Osteomyelitis

Jianfang Li,Xiangmei Liu,Yufeng Zheng,Zhenduo Cui,Hui Jiang,Zhaoyang Li,Shengli Zhu,Shuilin Wu

Bacteria-infected osteomyelitis is life-threatening without effective therapeutic methods clinically. Here, a rapid and effective therapeutic strategy to treat osteomyelitis through ferroelectric polarization interfacial engineering of BiFeO 3 /MXene (Ti 3 C 2 ) triggered by ultrasound (US) is reported. Under US, the ferroelectric polarization induces the formation of the piezoelectric field. US cavitation effect induced sonoluminescence stimulates BiFeO 3 /Ti 3 C 2 to produce photogenerated carriers. With synergistic action of the polarization electric field and Schottky junction, BiFeO 3 /Ti 3 C 2 accelerates the separation of electrons and holes and simultaneously inhibits the backflow of electrons, thus improving the utilization of polarized charges and photogenerated charges and consequently enhancing the yield of reactive oxygen species under US. As a result, 99.87 ± 0.05% of Staphylococcus aureus are efficiently killed in 20 min with the assistance of ultrasonic heating. The theory of ferroelectric ultrasonic interfacial engineering is proposed, which brings new insight for developing ferroelectric ultrasonic responsive materials used for the diagnosis and therapy of deep tissue infection and other acoustoelectric devices.

GSH-Triggered Nitric Oxide Release from Polyurethane Nanocarriers for Gas-Photothermal Synergistic Therapy of Bacterial-Infected Wounds

Yuyue Xiong,Shuai Yang,Yaodong Di,Jinglong Yang,Jing Tu,Lesan Yan

Wound bacterial infections pose a significant challenge to global public health. Nitric oxide (NO), an endogenous gaseous molecule, shows great potential in antibacterial therapy. However, efficient NO delivery remains a critical challenge. Photothermal therapy (PTT) offers a noninvasive and highly localized treatment for bacterial infections. Leveraging the advantages of both approaches, this study developed a polymer-based nanocarrier to achieve synergistic NO delivery and PTT. For the first time, an amphiphilic polyurethane PEG–PNU-PEG containing dinitrate NO donors were synthesized, which further self-assembled into polymeric nanomicelles loaded with a NIR emitting conjugated polymer. In the bacterial microenvironment, elevated glutathione (GSH) triggered NO release. Simultaneously, the conjugated polymer enabled effective photothermal therapy, further promoting NO release for gas therapy. Experimental results demonstrated that the synergistic NO and PTT treatment effectively eradicated bacteria, eliminating 69.93% ± 2.79% of S. aureus biofilm, a > 4.5-fold improvement compared to NO treatment alone (14.8% ± 2.92%). The in vitro antibacterial assay showed an inhibition rate exceeding 99.0%, while in vivo studies on infected skin wounds revealed a bacterial clearance rate of 95.0% ± 1.95%. Furthermore, this system suppressed inflammatory cytokines, promoted collagen deposition, and accelerated skin regeneration, providing an effective strategy for broad-spectrum antibacterial therapy and wound healing.

Microneedles loaded with l-arginine-modified puerarin-derived carbon nanoparticles improved treatment of diabetic wound via photothermal and nitric oxide-based gas therapy

Xiangchen Su,Xinrong Geng,Fang Li,Mingzhu Song,Ruizhen Lv,Yifei Zhang,Jiayu Yuan,Jia Dong,Yijie Shi,Liang Zhao

Due to the high-glucose environment of diabetic wounds, a significant proliferation of bacteria can occur, resulting in an inflammatory response that extends the inflammatory phase of the wound, thereby complicating the healing process in diabetic wounds. Eliminating the proliferation of bacteria plays a crucial role in promoting the healing of diabetic wounds. Under near-infrared (NIR) laser irradiation, l -arginine-modified natural product puerarin (Pue)-derived carbon nanoparticles ( l -Arg-CNP) not only exhibited excellent photothermal effects but also produced reactive oxygen species (ROS) to react with l -arginine ( l -Arg) for producing Nitric Oxide (NO), thus contributing to a synergistic antibacterial therapy in diabetic wound. At the same time, l -Arg-CNP retained Pue’s original characteristics to promote cell proliferation and angiogenesis. Following the loading of l -Arg-CNP into microneedle patches ( l -Arg-CNP@MN), it can deliver them into the deeper wound, effectively killing bacteria, reducing inflammatory infiltration, and promoting neovascularization at the wound site. It offers an effective therapeutic strategy for treating diabetic wound healing.

Optimized Antimicrobial Peptide Jelleine-I Derivative Br-J-I Inhibits Fusobacterium Nucleatum to Suppress Colorectal Cancer Progression

Fengjing Jia,Qun Yu,Ruolei Wang,Ling Zhao,Fuwen Yuan,Haidong Guo,Yunhui Shen,Feng He

Colorectal cancer (CRC) is a major health burden worldwide due to its high morbidity, mortality, and complex etiology.Fusobacterium nucleatum(Fn), a Gram-negative anaerobe found in 30% of CRC patients, promotes CRC carcinogenesis, metastasis, and chemoresistance. Effective antimicrobial treatment is an unmet need for the rising CRC burden. Antimicrobial peptides (AMPs) represent a new class of antimicrobial drugs. In our previous study, we did the structure-activity study of Jelleine-I (J-I) and identified several halogenated J-I derivatives Cl-J-I, Br-J-I, and I-J-I. To determine whether those J-I derivatives can be a new therapy for bacterial-associated CRC, here we tested the antibacterial activities of these AMPs againstFnand their effects on CRC development. We found that Br-J-I showed the highest anti-Fnactivity and Br-J-I may target membrane-associated FadA forFnmembrane disruption. More importantly,Fnpromoted the growth of CRC cells-derived xenograft tumors. Br-J-I suppressedFnload, colon inflammation, andFn-induced CRC growth. Of note, Br-J-I induced better anti-CRC effects than common antibiotic metronidazole and Br-J-I sensitized the cancer-killing effect of chemotherapy drug 5-fluorouracil. These results suggest that Br-J-I could be considered as an adjunctive agent for CRC treatment and AMPs-based combination treatment is a new strategy for CRC in the future. Keywords:colorectal cancer;fusobacterium nucleatum;antimicrobial peptides;adjunctive therapy;antimicrobial treatment

Regulating the bacterial oxygen microenvironment via a perfluorocarbon-conjugated bacteriochlorin for enhanced photodynamic antibacterial efficacy

Mengsi Wu,Chao Chen,Zhiyong Liu,Jia Tian,Weian Zhang

Photodynamic therapy (PDT) has attracted considerable attention, since it could effectively kill bacteria and prevent the development of multi-drug resistance. However, PDT currently suffers from oxygen limitation and hypoxia is a prominent feature of pathological states encountered in inflammation, wounds, and bacterial infections. Herein, an oxygen-tunable nanoplatform based on perfluorocarbon-conjugated tetrafluorophenyl bacteriochlorin (FBC-F) was designed for effective antimicrobial therapy . The introduction of fluorine atoms can not only increase the reactive oxygen species (ROS) production capacity of FBC-F by facilitating the intersystem crossing (ISC) process of FBC photosensitizers , but also make FBC-F deliver more oxygen into the treatment sites benefiting from the outstanding oxygen-dissolving capability of perfluorocarbon . As a consequence, the FBC-F nanoplatform was able to efficiently generate singlet oxygens for type II PDT, as well as superoxide anions and hydroxyl radicals for type I PDT, and significantly improve antibacterial efficacy in vitro. In vivo experiments further proved that the FBC-F with a powerful antibacterial capability could well promote wound healing and destroy biofilm . Thus, this FBC-F nanoplatform may open a new path in photodynamic antibacterial therapy . Statement of significance Photodynamic therapy is a promising antibacterial treatment, but its efficacy is severely compromised by hypoxia . To overcome such a limitation, we constructed an oxygen-regulated nanoplatform ( FBC-F ) by attaching perfluorocarbons (PFC) to the NIR photosensitizer ( FBC ). As an analogue of bacteriochlorin , FBC could generate 1 O 2 through energy transfer , as well as O 2 −· and ·OH through electron transfer for synergistic type I and type II photodynamic antibacterial therapy. Benefiting from the oxygen-dissolving capability of PFC, FBC-F could efficiently deliver more oxygen into the treatment site and alleviate the hypoxic environment. As a consequence, FBC-F could effectively generate large amounts of reactive oxygen species to achieve improved antibacterial efficacy and provide a promising approach for eliminating biofilms.

Mutual boost between free radicals and photothermal effect for synergistic photothermal/thermodynamic antibacterial therapy

Haimeng Liu,Chengyang Ban,Chen Fang,Ru Xia,Jiasheng Qian,Jibin Miao,Ming Cao

Combined treatment is a promising strategy in antibacterial treatment, which could alleviate the shortcomings of monotherapy and achieve better therapeutic effects. In this work, mutual boost between free radical generation and photothermal effect for synergistic photothermal/thermodynamic antibacterial therapy was reported. Mesoporous silica nanoparticles were used as drug carrier for loading dibenzoyl peroxide (BPO). The pores of mesoporous silica nanoparticles were blocked by GSH-responsive polyethylenimine (PEI) layer. 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) was then adsorbed via electrostatic force to prepare the nanosystem. At the site of bacterial infection, the BPO was released and decomposed to aryl radicals. The aryl radicals oxidized ABTS to photothermal reagent of ABTS ·+ , which produced photothermal effect to enhance the antibacterial effect under 808 nm laser irradiation. Moreover, the photothermal effect accelerated the decomposition of BPO to boost the levels of free radicals as a return, further improved the antibacterial efficiency. The in vitro experiments indicated that the photothermal/thermodynamic therapeutic nanosystem have a synergistic antibacterial effect efficiency (˃95 %) for both E. coli and S. aureus , as well as biofilm disruption and inhibition . This mutual boost between free radical ion generation and photothermal effect provides a new and feasible strategy for the synergistic antibacterial therapy.

Liquid Metal Amplified Charge Separation in Photocatalytic Micro/Nanomotors for Antibacterial Therapy

Zichang Guo,Dongdong Jin,Haohui Li,Xinyu Zhu,Tianqi Zheng,Zirong Xu,Yi Chen,Xiaojia Liu,Yinuo Song,Dai Wang,Xiaohui Yan,Xing Ma

Photocatalytic micro/nanomotors (MNMs) driven by electrophoresis have attracted considerable attention by virtue of their active mobility and versatile functionality. However, the rapid recombination of photogenerated electron–hole pairs on light illumination severely compromises the involvement of charge species in the catalytic redox reactions of fuels, thus hindering both the propulsion and the application performance of photocatalytic MNMs. Herein, we report a facile strategy to amplify charge separation by incorporating liquid metal (LM) into the construction of photocatalytic MNMs, thereby strengthening the electrophoretic propulsion of MNMs and promoting the generation of reactive oxygen species (ROS) for antibacterial application. The MNMs are constructed with a gallium (Ga) LM core, coated with abundant graphite-phase carbon nitride (g-C3N4) nanosheets and half covered by a thin platinum layer. These MNMs exhibit self-propulsion in hydrogen peroxide (H2O2) solution, with their motion dynamics further enhanced by light irradiation. Theoretical calculations and simulations reveal that the composition between Ga and g-C3N4 forms an ohmic junction in the electronic energy band structure, which effectively improves the charge separation efficiency of electron–hole pairs. These results align well with the experimental electrochemical tests and consequently intensify the catalytic redox reactions of H2O2, as well as accelerate the charge migration across MNMs, contributing to the enhancement of their propulsion performance. Simultaneously, the amplified separation of electrons facilitates increased ROS generation, empowering the MNMs with motion-enhanced antibacterial activity against Escherichia coli. Finally, an in vivo wound healing experiment is conducted, verifying the superior antibacterial therapeutic performance of photocatalytic MNMs. This work not only provides insights into the role of charge species in phoretic motion of MNMs but also gives inspiration for developing photocatalytic MNMs with advanced biomedical applications.

High Fe-Loading Single-Atom Catalyst Boosts ROS Production by Density Effect for Efficient Antibacterial Therapy

Chen Si,Huang Fang,Mao Lijie,Zhang Zhimin,Lin Han,Yan Qixin,Lu Xiangyu,Shi Jianlin

Fe single-atom catalysts (h 3 -FNCs) with high loading, high catalytic activity and high stability were synthesized via a method capable of increasing both the metal loading and mass-specific activity by exchanging zinc with iron. The “density effect,” derived from the sufficiently high density of active sites, has been discovered for the first time, leading to a significant alteration in the intrinsic activity of single-atom metal sites. The superior oxidase-like catalytic performance of h 3 -FNCs ensures highly effective bacterial eradication.

Hollow-structured BaTiO3 nanoparticles with cerium-regulated defect engineering to promote piezocatalytic antibacterial treatment

Junwu Wei,Jing Xia,Xing Liu,Pan Ran,Guiyuan Zhang,Chaoming Wang,Xiaohong Li

The low efficiency of mechanical energy capture and insufficient active sites are the main obstacles of piezocatalytic therapy. Herein, cerium (Ce)-doped hollow BaTiO 3 (hBT Ce ) nanoparticles (NPs) were developed to improve the piezocatalytic effect through simultaneously hollow structure and defect engineering. hBT NPs were prepared through precipitation of TiO 2 layers on SiO 2 NP templates, transformation into BaTiO 3 layer, and simultaneous removal of SiO 2 cores. The hollow structure exhibits a stronger piezoresponse than solid NPs, and the piezoelectric coefficient increases nearly 2.6 times. The Ce doping enhances polarization, oxygen vacancy formation and electron/hole separation, and reduces the band gap of hBT Ce NPs. Both the strong piezoelectric current and abundant generation of reactive oxygen species from hBT Ce under ultrasonication contribute to significant destructions of planktonic bacteria and biofilms. Thus, this study demonstrates the Ce-regulated defect engineering and hollow structure of piezoelectric NPs are effective to promote polarization, piezoelectric potential and piezocatalytic antibacterial therapy.

Engineering of plasmonic metal-semiconductor yolk-shell nanostructures for multi-intensified photodynamic- and immuno- therapy against drug resistant bacteria

Dong Wang,Haojun Li,Lei Ji,Jia Liu,You Li,Meng Xu,Hongzhi Wang,Zeng-ying Qiao,Jiatao Zhang

Photodynamic therapy (PDT) as well as immunotherapy are effective therapies to combat the systemic infection. Nevertheless, the development of the treatments are restrained by the lack of efficient photosensitizers (PS) and the genetic diversity of the bacteria. Here, we report Ag doped Au-CdS 1−y Se y yolk-shell nanostructures (AYSs) which exhibit excellent near-infrared (NIR) light induced PDT and immunotherapy performance. The tailored plasmonic Au nanorods (NRs) are able to gather the NIR light, offering the basis for PDT. Moreover, high yield of reactive oxygen species (ROS) is achieved through the following creativity: i) enhanced light trapping ability by the microcavity, ii) smooth injection process of hot electrons (HEs) by the suitable band gap alignment , iii) inhibiting the combination of electric charges by the doped Ag with appropriate concentration. Therefore, the AYSs exhibit more 30 times of ROS production as well as lower 64 times of MIC than the conventional core-shell nanostructures. Notably, repeated enhanced antibacterial PDT are carried out to obtain versatile antigens from various dead bacteria to awaken the immune cells adequately and boost immune response with the help of Selenium (Se) which is distributed in the shell. As a result, the systemic drug resistant bacterial infection is cured successfully.

Construction of perfluorohexane/IR780@liposome coating on Ti for rapid bacteria killing under permeable near infrared light

Xiuhua Wang,Lei Tan,Xiangmei Liu,Zhenduo Cui,Xianjin Yang,Kelvin W. K. Yeung,Paul K. Chu,Shuilin Wu

Near infrared (NIR) light induced photodynamic antibacterial therapy (PDAT) is a promising antibacterial technique in rapid in situ disinfection of bacterially infected artificial implants due to its penetration ability into tissues. However, the lower oxygen content in vivo may restrict the yields of reactive oxygen species (ROS), thus reducing the antibacterial efficacy of PADT significantly. Herein, liposome encapsulated photosensitizers (PS), IR780 and perfluorohexane (PFH), have been constructed on the surface of Ti implants via a covalent linkage to overcome this issue. Thanks to the high oxygen capacity of PFH, more ROS can be generated during NIR irradiation regardless of the low content of oxygen in vivo. As a result, in vitro tests demonstrated that 15 minutes of 808 nm near-infrared irradiation could achieve a high antibacterial efficacy of 99.62% and 99.63% on the implant surface against Escherichia coli and Staphylococcus aureus, respectively. By contrast, the PDAT system without PFH modification shows a lower antibacterial efficacy (only 66.54% and 48.04%, respectively). In addition, this enhanced PDAT system also possesses great biocompatibility based on the in vitro and in vivo subcutaneous assays. This surface system makes it possible for rapid bacteria-killing in artificial implants that have been implanted in vivo under local conditions with lower oxygen content.

Roxarsone Delivery Nanocomposite Based on Nitrite-Functionalized Mesoporous Polydopamine for Multidrug-Resistant Bacterial Infections via Enhanced Chemo-Photothermal Therapy

Zhecheng Kang,Qian Zhou,Kun Wang,An Wang,Tianyi Chen,Hongbo Wei,Tengjiao Wang

Graphical An anti-infection nanocomposite was constructed by loading bactericide Roxarsone onto nitrosylated mesoporous polydopamine. The designed nanocomposite achieves NO-enhanced chemo-photothermal antibacterial therapy under near-infrared laser irradiation, and in vitro antibacterial evaluation has confirmed the effective elimination against the Gram-negative tetracycline-resistant Escherichia coil and Gram-positive methicillin-resistant Staphylococcus aureus of the nanocomposite. Current treatments for infections caused by multidrug-resistant bacteria still remain challenging and therapeutic materials with high efficacy are of demand. Herein, a bactericidal nanocomposite was constructed by loading Roxarsone (ROX) onto nitrosylated mesoporous polydopamine (named mPDA@NO-ROX). The designed nanocomposite exhibited considerable photothermal effect and controlled NO and ROX co-delivery under the irradiation of near-infrared laser (NIR) to achieve enhanced chemo-photothermal antibacterial therapy. The in vitro antibacterial evaluation of the mPDA@NO-ROX demonstrated the effective elimination of the Gram-negative tetracycline-resistant Escherichia coil and Gram-positive methicillin-resistant Staphylococcus aureus under mild NIR irradiation compared to merely ROX loaded unmodified mPDA, indicating the NO enhanced chemo-photothermal therapy. In addition, the cytotoxicity experiments indicated that mPDA@NO-ROX exhibited only 5 % of hemolysis rate and high cell viability at 1 mg mL −1 against mammalian fibroblasts, suggesting the excellent biocompatibility. In conclusion, the mPDA@NO-ROX could be a promising candidate for anti-infection therapy of multidrug-resistant bacteria.
