Covalent immobilization of antibacterial peptides (APs) onto silk protein-based materials has always been a challenge due to the lack of green and efficient macromolecular cross-linkers. Here, we proposed a dialdehyde polysaccharide cross-linker oxidized from pullulan for grafting a natural AP protamine (PM) onto silk fiber surface through a simple cold pad-batch process. The oxidized pullulan (OP) was linked to silk fiber surface through Schiff reaction and used for mediated cross-linking of PM also via Schiff base linkages. This modification introduced abundant PM guanidine groups on the fiber leading to much-desired antibacterial activity, and considerable improvement in the moisture transfer properties and shade depth. FTIR, XPS, SEM studies confirmed the presence of PM and the cross-linking structure between the polysaccharide and peptides on the fiber surfaces. The antibacterial activity imparted by this process was retained even after 20 washing and 50 rubbing cycles proving versatility and durability. Further, the process did not affect other critical silk properties such as appearance, tensile strength, biological safety, etc. Immobilization of PM onto silk fibers through this novel green polysaccharide cross-linker makes silk more appealing and usable and could also enlighten the attempts of cross-linking other protein materials.

In the face of mounting global antibiotic resistance, which has become a critical clinical problem, antimicrobial peptides (AMPs) have received considerable interest as new therapeutics with the efficacy for the treatment of multidrug-resistant (MDR) infections due to their novel mechanism. However, certain inherent shortcomings such as instability seriously limit their systemic applications in the clinic. In this study, we intend to clarify the connection between three configurations of IK8 and their stability in plasma and liver S9 of various species by confirming the metabolites. The structural information of these metabolites was scanned and identified using HPLC and Q-TOF, respectively. The results found that IK8-D exhibits superior stability, compared with IK8-2D and IK8-L in plasma and liver S9 incubation, which indicated D- type amino acids could significantly increase the stability of antimicrobial peptides. Keywords:antimicrobial peptide;IK8;metabolite;Q-TOF

Although parasites and microbial pathogens are both detrimental to insects, little information is currently available on the mechanism involved in how parasitized hosts balance their immune responses to defend against microbial infections. We addressed this in the present study by comparing the immune response between unparasitized and parasitized pupae of the chrysomelid beetle, Octodonta nipae (Maulik), to Escherichia coli invasion. In an in vivo survival assay, a markedly reduced number of E. coli colony-forming units per microliter was detected in parasitized pupae at 12 and 24 h post-parasitism, together with decreased phagocytosis and enhanced bactericidal activity at 12 h post-parasitism. The effects that parasitism had on the mRNA expression level of selected antimicrobial peptides (AMPs) of O. nipae pupae showed that nearly all transcripts of AMPs examined were highly upregulated during the early and late parasitism stages except defensin 2B, whose mRNA expression level was downregulated at 24 h post-parasitism. Further elucidation on the main maternal fluids responsible for alteration of the primary immune response against E. coli showed that ovarian fluid increased phagocytosis at 48 h post-injection. These results indicated that the enhanced degradation of E. coli in parasitized pupae resulted mainly from the elevated bactericidal activity without observing the increased transcripts of target AMPs. This study contributes to a better understanding of the mechanisms involved in the immune responses of a parasitized host to bacterial infections.

Osteomyelitis poses substantial therapeutic challenges due to the prevalence of multidrug-resistant bacterial infections and associated inflammation. Current treatment regimens often rely on a combination of corticosteroids and antibiotics, which can lead to complications and impede effective bacterial clearance. In this study, we present CADP-10, a Cecropin A-derived peptide, designed to target methicillin-resistant Staphylococcus aureus (MRSA) and multidrug-resistant Escherichia coli (MRE), while simultaneously addressing inflammatory responses. CADP-10 self-assembles into nanobacterial net (NBacN) that selectively identify and bind to bacterial endotoxins (LPS and LTA), disrupting membrane integrity and depolarizing membrane potential, which culminates in bacterial death. Importantly, these NBacN are bound to LPS and LTA from dead bacteria, preventing their engagement with TLR receptors and effectively blocking downstream inflammatory pathways. Our assessments of CADP-10 demonstrate good biosafety in both in vitro and in vivo models. Notably, in a rabbit osteomyelitis model, CADP-10 eliminated MRSA-induced bone infections, mitigated inflammation, and promoted bone tissue regeneration. This research highlights the potential of CADP-10 as a multifunctional antimicrobial agent for the management of infectious inflammatory diseases.

It is important to eliminate lipopolysaccharide (LPS) along with killing bacteria in periprosthetic joint infection (PJI) therapy for promoting bone repair due to its effect to regulate macrophages response. Although natural antimicrobial peptides (AMPs) offer a good solution, the unknown toxicity, high cost and exogenetic immune response hamper their applications in clinic. In this work, we fabricated a nanowire-like composite material, named [email protected] , by combining chitosan and puerarin via solid-phase reaction, which can finely mimic the bio-functions of AMPs. Chitosan, serving as the bacteria membrane puncture agent, and puerarin, serving as the LPS target agent, synergistically destroy the bacterial membrane structure and inhibit its recovery, thus endowing [email protected] with good antibacterial property. In addition, [email protected] possesses good osteoimmunomodulation due to its ability of LPS elimination and macrophage differentiation modulation. The in vivo results show that [email protected] can inhibit the LPS induced bone destruction in the Escherichia coli infected rat. [email protected] exhibits superior bone regeneration in Escherichia coli infected rat due to the comprehensive functions of its superior antibacterial property, and its ability of LPS elimination and immunomodulation. [email protected] can well mimic the functions of AMPs, which provides a novel and effective method for treating the PJI in clinic.

Antimicrobial peptides (AMPs) are a class of peptides consisting of cationic amino acid residues and a hydrophobic segment, which have been used as an alternative to antibiotics in treating multidrug-resistant bacteria. However, the relationship among the molecular design, assembled structures, and resultant efficacy remains elusive. Herein, we report a class of constitutionally isomeric AMPs assembled into filaments with similar dimensions. Spectroscopic characterizations demonstrated that subtle changes in the position of amino acids led to dramatic variations in molecular packing and surface charges, which were verified by molecular dynamics simulations. In vitro antibacterial assays showed that all AMPs exerted antibacterial activity against Gram-positive methicillin-resistant Staphylococcus aureus (MRSA), but the efficacy was dependent on the molecular design. Given the good biocompatibility to eukaryotic cells, these AMPs could be potentially used as antibacterial agents. We believe that this finding provides an avenue to tune the bioactivity of AMPs by rational molecular design.

We present an antibacterial polymersome–hydrogel composite that combines the advantages of both antimicrobial peptides and antibiotics. The polymersomes are self-assembled from poly(ε-caprolactone)-block-poly(lysine-stat-phenylalanine) [PCL-b-P(Lys-stat-Phe)], exhibiting long-acting intrinsic antibacterial capabilities against both Gram-positive and Gram-negative bacteria. In addition, penicillin is encapsulated into the polymersomes, showing quick and enhanced antibacterial activities. Furthermore, a dynamic hydrogel network is prepared based on the Schiff base linkages between the aldehyde groups of dibenzaldehyde-functionalized PEG (DF-PEG) and the amino groups of chitosan. During this process, the penicillin-loaded polymersomes are grafted into the hydrogel networks by Schiff base linkages to afford polymersome–hydrogel composites, which exhibit two-stage antibacterial behavior: (1) penicillin can be released from the hydrogel networks for quick antibacterial function; (2) the polymersomes and the remaining penicillin can be further released for long-term antibacterial effects. Overall, this polymersome–hydrogel composite represents a new type of injectable antibacterial biomaterial for quick and long-acting antibacterial applications.

We show that simply converting the hydrophobic moiety of an antimicrobial peptide (AMP) or synthetic mimic of AMPs (SMAMP) into a hydrophilic one could be a different pathway toward membrane-active antimicrobials preferentially acting against bacteria over host cells. Our biostatistical analysis on natural AMPs indicated that shorter AMPs tend to be more hydrophobic, and the hydrophilic-and-cationic mutants of a long AMP experimentally demonstrated certain membrane activity against bacteria. To isolate the effects of antimicrobials’ hydrophobicity and systematically examine whether hydrophilic-and-cationic mutants could inherit the membrane activity of their parent AMPs/SMAMPs, we constructed a minimal prototypical system based on methacrylate-based polymer SMAMPs and compared the antibacterial membrane activity and hemolytic toxicity of analogues with and without the hydrophobic moiety. Antibacterial assays showed that the hydrophobic moiety of polymer SMAMPs consistently promoted the antibacterial activity but diminished in effectiveness for long polymers, and the resultant long hydrophilic-and-cationic polymers were also membrane active against bacteria. What distinguished these long mutants from their parent SMAMPs were their drastically reduced hemolytic toxicities and, as a result, strikingly enhanced selectivity. Similar toxicity reduction was observed with the hydrophilic-and-cationic mutants of long AMPs. Taken together, our results suggest that long hydrophilic-and-cationic polymers could offer preferential membrane activity against bacteria over host cells, which may have implications in future antimicrobial development.

Infections associated with biomedical implants and devices pose a serious clinical challenge in hospitals worldwide. Antimicrobial peptides (AMPs) have become a great prospect to inhibit this type of infection due to their broad-spectrum antimicrobial activity and low cytotoxicity. However, it is still a challenge to apply AMPs on the biomaterial surface as the activity of AMPs is sensitive to salt or enzyme. In the present study, we prepared a spacer molecule, poly[2-(methacryloyloxy)ethyl]dimethyl-(3-sulfopropyl)ammonium hydroxide (polySBMA), on a model silicon surface via surface-initiated atom transfer radical polymerization (SI-ATRP). We then modified the antimicrobial peptide HHC36 (KRWWKWWRR) with L-propargylglycine (PraAMP) to improve its salt-tolerant activity and integrated PraAMP onto the spacer molecule using click chemistry. We employed X-ray photoelectron spectroscopy (XPS), contact angle goniometry, and atomic force microscopy (AFM) to confirm the success of the immobilization process. We also characterized the antimicrobial activity and stability of the surface with an antimicrobial assay. The results reveal that the modified surface exhibits good antimicrobial activity to inhibit 98.26% of E. coli, 83.72% of S. aureus, and 81.59% of P. aeruginosa. Furthermore, as compared to the control group without the polySBMA spacer, the modified surface improved its resistance to enzymolysis. An in vitro CCK-8 assay also illustrated that this surface showed negligible cytotoxicity to mouse bone mesenchymal stem cells.

Marine biological fouling has a serious impact on artificial facilities, and traditional antifouling coatings are not beneficial to green development, which lead to the urgent to develop green strategies that can effectively prevent fouling. In this study, an innovative confluence of physical and biological approaches was employed to mitigate the harm caused by biofouling on a material substrate. Firstly, a hierarchical microstructured morphology was prepared on an aluminum surface by reactive ion etching and characterised. The antibacterial peptide, A-2S, was surface modified with dopamine as a coupling agent to produce a physico-biological synergistic antifouling surface. X-ray photoelectron spectroscopy, fourier transform infrared spectroscopy, and electrochemical experiments were then used to analyse the binding effect and corrosion resistance performance. A-2S was successfully modified on the sample surface and exhibited good corrosion resistance with a corrosion inhibition efficiency of 86.57 %. Compared to a bare aluminum plate, the microstructure surface has a certain inhibitory effect on fouling organisms, and the antifouling performance decreases over time. In contrast, the antimicrobial peptide synergistic surfaces demonstrated superior durable antifouling performance. Compared with the 50 % of the antifouling performance on the microstructure surface, the synergistic surface has a unique contact point effect of the microstructure and excellent antibacterial and bactericidal properties of antimicrobial peptides, resulting in a surface antifouling performance of over 80 %. The synergistic modification method enhanced the corrosion resistance of the material and reduced the adhesion of fouling organisms, mitigating the harm caused by biofouling on the marine equipment substrate.

Antimicrobial peptides show promise in enhancing photothermal therapy, but their application is often limited by the challenge of constructing a delivery system that balances efficacy and safety. Our research demonstrated that the bactericidal efficacy of V 2 C MXene-mediated photothermal therapy is enhanced in a concentration-dependent relationship with the introduction and coating of the antimicrobial peptide ε-polylysine (EPL). EPL exhibited a dual role in enhancing bacterial binding and disrupting bacterial membranes, thereby increasing heat transfer efficiency and reducing bacterial resistance to photothermal ablation. The core strategy of this study was to exploit the combined membranolytic-photothermal effect of EPL and V 2 C by extensively applying EPL while regulating V 2 C nanosheets usage to prevent overheating. This approach aims to achieve potent bactericidal efficacy through photothermal therapy below 60 °C. Consequently, we developed dissolving microneedles incorporated with V 2 C nanosheets, where EPL served as the antimicrobial agent and primary matrix, increasing its loading capacity and minimizing the need for inactive excipients. Notably, this microneedle achieved a 99.9 % reduction in the abundance of methicillin-resistant Staphylococcus aureus on infected skin after a single application and resulted in a 92-fold reduction in the bacterial load compared to the group treated with commercial Bactroban ointment, with no apparent toxicity to the mice.

Seed coating technology is vital in agriculture, enhancing seed protection and growth. However, conventional coatings often include chemical fungicides that pose environmental risks, highlighting the need for sustainable alternatives. This study explores silk fibroin (SF), a natural biopolymer with excellent film-forming properties, as a potential seed coating agent, addressing its antimicrobial limitations by combining it with the commercial agent CRUISER®and the antimicrobial peptide Nisin. Experimental methods included solution stability analysis, Fourier-transform infrared spectroscopy (FTIR), scanning electron microscope (SEM), and growth assessments of wheat seeds. Findings reveal that silk fibroin-CRUISER®(SC) composites form stable β-sheet structures, enhancing the coating’s mechanical strength. SF-based coatings improved seedling emergence rates (up to 1.65-fold), plant height (up to 1.05-fold), and root growth (up to 1.2-fold), especially under cold stress. The addition of Nisin further significantly boosted the antibacterial properties, providing sustained pathogen inhibition (p< 0.01). Identifying the optimal concentration of SF was essential for achieving a balance between protection and breathability, a key factor for industrial application. This research provides valuable insights into the development of eco-friendly seed coatings, presenting a viable and sustainable alternative to traditional chemical-based options in agricultural practices.
