Plant-inspired TransfOrigami microfluidics

Yi Pan,Zhenyu Yang,Chang Li,Sammer Ul Hassan,Ho Cheung Shum

The healthy functioning of the plants’ vasculature depends on their ability to respond to environmental changes. In contrast, synthetic microfluidic systems have rarely demonstrated this environmental responsiveness. Plants respond to environmental stimuli through nastic movement, which inspires us to introduce transformable microfluidics: By embedding stimuli-responsive materials, the microfluidic device can respond to temperature, humidity, and light irradiance. Furthermore, by designing a foldable geometry, these responsive movements can follow the preset origami transformation. We term this device TransfOrigami microfluidics (TOM) to highlight the close connection between its transformation and the origami structure. TOM can be used as an environmentally adaptive photomicroreactor. It senses the environmental stimuli and feeds them back positively into photosynthetic conversion through morphological transformation. The principle behind this morphable microsystem can potentially be extended to applications that require responsiveness between the environment and the devices, such as dynamic artificial vascular networks and shape-adaptive flexible electronics.

Deformation-Mediated Translocation of DNA Origami Nanoplates through a Narrow Solid-State Nanopore

Libo Zhu,Zhen Zhang,Quanjun Liu

With the development in DNA self-assembly technology, DNA origami nanostructures have been widely applied in biomedical research. Solid-state nanopores represent an emerging single-molecule sensing platform for studying nanostructures with arbitrary dimensions and physical characteristics, including DNA origami. Here, we employed relatively narrow silicon nitride nanopores to detect the deformation and translocation of DNA origami nanoplates with dimensions of approximately 60 × 54 nm. We performed translocation experiments using three nanopore diameters that are all smaller than the plat dimensions. Analysis of current blockade signals and the representative events reveals three types of translocation orientations for the nanoplates. Furthermore, by studying the electrical signal characteristics (current change and dwell time) for the different diameter pores, we obtained information about the translocation behaviors for the DNA nanoplates through different constrictions. Our investigation provides an approach to analyze the deformation and translocation of DNA origami structures.

An Efficient Fog Collection Origami Janus Membrane for Rapid Directional Water Transport and Omnidirectional Fog Harvesting

Qiong Wang,Yihang Zhang,Guangyi Tian,Zhiguang Guo

Existing Janus fog harvesting technologies struggle to achieve efficient and continuous fog collection across various fog flow directions. In this work, we developed a Janus fog harvesting (JHL) system, characterized by a boundary-free Janus system formed by selective modification with octadecanethiol, creating a pair of hydrophilic and hydrophobic domains. The system features a 3D origami macrostructure composed of scallop arrays and shaped like an improved trumpet flower. The wettability gradient Janus system enables rapid droplet capture and ultrafast absorption. The multiple parallel and dual-asymmetric 3D Kirigami superhydrophilic channels facilitate high-speed, directional, and long-range liquid transport. These mechanisms work seamlessly together to keep both the superhydrophilic and hydrophobic surfaces dry at all times. As a result, our JHL achieves a fog harvesting efficiency as high as 0.83 g·cm–2·min–1, which is 3 times that of the original sample. Moreover, our improved trumpet-shaped 3D structure extends the Janus design concept from traditional membrane materials to spatial membrane configurations, making it suitable for omnidirectional fog harvesting in any fog flow environment.

3D Origami-Inspired Electrochemical Paper-Based Analytical Devices with Flow-Through Electrodes and Quasi-Steady Flow

Yang Wang,Dingding Ye,Yang Xu,Xun Zhu,Yang Yang,Rong Chen,Qiang Liao

Electrochemical paper-based analytical devices (ePADs) are an excellent choice to achieve the goal of global health due to their advantages of low cost, portability, and rapid analysis. However, the low sensitivity limits the application of ePADs. In this study, a 3D origami-inspired ePAD with flow-through electrodes and quasi-steady flow was presented for signal amplification to enhance the sensitivity. A Silhouette Studio plotter/cutter was used to design four schemes with a porous gold electrode as the working electrode. The flow-through electrode with quasi-steady flow exhibited the highest current due to enhanced convective transfer. The peak current of the device combined with flow-through porous electrodes with flow increased by 97.2 and 63.8% compared with the device without flow and the device combined flow-over electrodes with flow, respectively. The finite element simulation results showed that the current increased 10 times compared with the conventional saturated devices with flow-over electrodes and flow-through electrodes. Moreover, the evaluation of the device used copper ions as the detection species. The current of the device that combined flow-through with quasi-steady flow was about twice as large as other schemes and owned higher sensitivity. This strategy provides new insights and a more facile way to amplify signals of ePADs.

QKCMP peptide carrier macromolecule structure and release system based on triangular DNA origami nanostructures: A novel strategy for promoting angiogenesis

Haifeng Wang,Yi Huang,Chenhui Zhou,Fanyong Gong,Jianyong Wang,Hao Liu,Wenhui Zhao,Yuxiang Zhao,Xiang Gao

In recent years, the use of nanocarriers to deliver bioactive peptides has become a new strategy to promote angiogenesis. The aim of this study was to develop a QKCMP peptide delivery system based on triangular DNA origami nanostructure (tDONs) carrier to improve the bioactivity and targeting of the peptide. The main objective of this study was to design and prepare QKCMP peptide-TDOns complexes and to evaluate their angiogenesis promoting effects in vitro and in vivo. tDONs were synthesized and purified, and then QKCMP peptide was loaded into tDONs. A variety of characterization techniques were used to evaluate the physicochemical properties of the complexes, including particle size, morphology, and RNA release efficiency. The effects of QKCMP peptid-TDons on proliferation, apoptosis, migration and angiogenesis of human umbilical vein endothelial cells (HUVEC) were evaluated by CCK8, EdU, Annexin V-FITC/PI, PI staining and Transwell assay. At the same time, its promoting effect on angiogenesis was further verified in zebrafish and mouse models. The successful preparation of QKCMP peptide-TDONS showed good biocompatibility and stability. The results of in vitro experiments showed that QKCMP peptide-TDons significantly promoted HUVEC proliferation, inhibited apoptosis, and activated endothelial cell function. Zebrafish experiments showed that QKCMP peptid-TDons effectively enhanced angiogenesis and promoted rapid repair of carotid vascular endothelium in mouse models.

Programmable, Changeable, Origami Cellulose Films for Magnetically Controllable Soft Robots

Xueqi Li,Ziqiang Shang,Yaoxing Wang,Jiuqing Liu,Yanjun Xie,Jian Li,Yiqi Liu,Wentao Gan

Magnetic soft robots composed of stimuli-responsive materials are promising for biomedical engineering applications; however, typical responsive materials are fabricated with nondegradable polymeric substrates. In this study, we report a flexible, biodegradable, and magnetically sensitive cellulose film (M-film) that can be utilized for magnetically controllable soft robots (M-robots) with programmable locomotion, cargo delivery, and remote wireless operation functions. The M-film with good foldability, origami, and magnetic properties is synthesized by a simple paper-making process using cellulose nanofibers, additive sodium alginate, and BaFe12O19 particles. Through the following origami–magnetization process, the M-robot with multimodal movements is designed: climbing over the obstacles in the walking environment; additionally, this process can complete various cargo transport tasks by clawing, rolling, and flipping. This approach expands the precise controllability and manipulability of environmentally friendly cellulose nanomaterials beyond the known applications and opens the prospects of their implementation in stimuli-responsive robots, wireless control electronics, and intelligent devices.

Transparent origami glass

Xu Yang,Li Ye,Zheng Ning,Zhao Qian,Xie Tao

The art of origami has emerged as an engineering tool with ever increasing potential, but the technique is typically limited to soft and deformable materials. Glass is indispensable in many applications, but its processing options are limited by its brittle nature and the requirement to achieve optical transparency. We report a strategy that allows making three dimensional transparent glass with origami techniques. Our process starts from a dynamic covalent polymer matrix with homogeneously dispersed silica nanoparticles. Particle cavitation and dynamic bond exchange offer two complementary plasticity mechanisms that allow the nanocomposite to be permanently folded into designable geometries. Further pyrolysis and sintering convert it into transparent three dimensional glass. Our method expands the scope of glass shaping and potentially opens up its utilities in unexplored territories.

Programmable and flexible wood-based origami electronics

Ma Huashuo,Liu Chaozheng,Yang Zhi,Wu Shuai,Jiao Yue,Feng Xinhao,Xu Bo,Ou Rongxian,Mei Changtong,Xu Zhaoyang,Lyu Jianxiong,Xie Yanjun,Fu Qiliang

Natural polymer substrates are gaining attention as substitutes for plastic substrates in electronics, aiming to combine high performance, intricate shape deformation, and environmental sustainability. Herein, natural wood veneer is converted into a transparent wood film (TWF) substrate. The combination of 3D printing and origami technique is established to create programmable wood-based origami electronics, which exhibit superior flexibility with high tensile strength (393 MPa) due to the highly aligned cellulose fibers and the formation of numerous intermolecular hydrogen bonds between them. Moreover, the flexible TWF electronics exhibit editable multiplexed configurations and maintain stable conductivity. This is attributed to the strong adhesion between the cellulose-based ink and TWF substrate by non-covalent bonds. Benefiting from its anisotropic structure, the programmability of TWF electronics is achieved through sequentially folding into predesigned shapes. This design not only promotes environmental sustainability but also introduces its customizable shapes with potential applications in sensors, microfluidics, and wearable electronics.

A “time-frozen” technique in microchannel used for the thermodynamic studies of DNA origami

Peng Huang,Jingwen Wang,Long Jiao,Dandan Gu,Shusen Jiang,Mingpo Li,Wenlong lv,Hong Chen,Hao Pei

The emergence of DNA origami greatly accelerated the development of DNA nanotechnology. A thorough understanding of origami thermodynamics is very important for both fundamental studies and practical applications. These thermodynamic transitions usually take place in several seconds or even less, and are very difficult to monitor by conventional methods. Numerous tests are required to characterize the origami molecule's behaviors at different temperatures, which is very labor-intensive and time-consuming. In this paper, an axially distributed temperature gradient along a capillary was formed in a spatially varying temperature field. In such a temperature gradient, the origami molecule's thermodynamic processes occur and remain stable at every position along the capillary's microchannel . It looks like the time of the thermodynamic process is frozen along the microchannel. With this method, the origami molecule's thermodynamic characteristics at different temperatures can be obtained in a single experiment, and rapid processes can be monitored with ease by conventional methods for an adequate time period at low cost. In order to show its potential abilities, this method has been demonstrated in applications which the origami's assembly, denaturation and strand displacement are carry out in a flowing or stationary solution.

DNA origami-based composite nanosandwich for iteratively potentiated chemo-immunotherapy

Mengyue Li,Hanxi Zhang,Peizheng Xiong,Yuhan He,Wanyi Zhou,Chunhui Wu,Xiaoling Liao,Wei Zhang,Hong Yang,Yiyao Liu

Developing effective nanoplatforms for chemo-immunotherapy to achieve enhanced tumor suppression and systemic antitumor immunity has recently received extensive attention. Herein, we formulated a multifunctional DNA sandwich nanodevice, DSWAC/siPD-L1, based on triangular DNA origami, to implement enhanced cancer chemo-immunotherapy. Taking advantage of the tumor-targeting ability of the AS1411 aptamer, DSWAC/siPD-L1 efficiently delivered doxorubicin (DOX), CpG, and siPD-L1 into tumor cells. Moreover, the sandwich cavity spatially protects siPD-L1 from degradation, and the featured design of the DNA/RNA duplex linkers ensures effective intracellular release of siPD-L1. The pH-responsive release of cytotoxic DOX induces apoptosis and initial mild immunogenic cell death of tumor cells, presenting antigens to enhance the maturation of dendritic cells (DCs) with the assistance of the immune adjuvant CpG, thereby activating cytotoxic T lymphocytes to amplify antitumor immunity. Simultaneously, siPD-L1 downregulated the endogenous expression of PD-L1 to inhibit adaptive tumor immune escape. DSWAC/siPD-L1 initiated the iterative revolution of the cancer-immunity cycle, leading to the inhibition of primary and metastatic tumors, as demonstrated by DC maturation and T-cell infiltration in established subcutaneous primary tumor model and metastatic lung tumor model. Furthermore, the superior antitumor effect of DSWAC/siPD-L1 resulted in approximately 91 % inhibition of primary tumor growth and 93 % prevention of lung metastasis. Collectively, this study describes a siPD-L1-based sandwich DNA nanodevice functionalized with AS1411/CpG for enhanced cancer chemo-immunotherapy, inspiring the creation of more innovative drug nanocarriers and the exploitation of novel cancer therapies.

DNA Origami–Anthraquinone Hybrid Nanostructures for In Vivo Quantitative Monitoring of the Progression of Tumor Hypoxia Affected by Chemotherapy

Yun Zeng,Peng Chang,Jingwen Ma,Ke Li,Chunhong Zhang,Yingying Guo,Hanrui Li,Qingxia Zhu,Huifang Liu,Wenjing Wang,Yuwei Chen,Dan Chen,Xu Cao,Yonghua Zhan

Hypoxia is a well-known feature of malignant solid tumors. To explain the misinterpretation of tumor hypoxia variation during chemotherapy, we developed a DNA origami-based theranostic nanoplatform with an intercalated anticancer anthraquinone as both the chemotherapeutic drug and the photoacoustic contrast agent. The size distribution of the DNA origami nanostructure is 44.5 ± 2.3 nm, whereas the encapsulation efficiency of the drug is 90.7 ± 1.0%, and the drug loading content is 92.2 ± 0.1%. The controlled cumulative release rates were measured in vitro, showing an acidic environment induced rapid drug release. The values of free energy of binding between the drugs and the DNA double helix were calculated through molecular simulations. The cell viability assay was used to characterize cytotoxicity, and fluorescence confocal cell imaging illustrates the biodistribution of the probe in vitro. Photoacoustic and fluorescence imaging were used to indicate drug delivery, release, and biodistribution to predict the drug’s chemotherapeutic effect in vivo, whereas the photoacoustic signals were compared with those of deoxygenated/oxygenated hemoglobin to represent the tissue hypoxia/normoxia maps during the chemotherapeutic process and indicate alleviated tumor hypoxia. Staining of tissue sections taken from organs and tumors was used to verify the results of photoacoustic imaging. Our results suggest that photoacoustic imaging can visualize this DNA origami-based theranostic nanoplatform and reveal the mechanisms of chemotherapy on tumor hypoxia.

Fluid-driven hydrogel actuators with an origami structure

Zhexin Huang,Cunyue Wei,Lina Dong,Anyang Wang,Hongyi Yao,Zhongwei Guo,Shengli Mi

Owing to the innate good biocompatibility, tissue-like softness and other unique properties, hydrogels are of particular interest as promising compliant materials for biomimetic soft actuators. However, the actuation diversity of hydrogel actuators is always restricted by their structure design and fabrication methods. Herein, origami structures were introduced to the design of fluid-driven hydrogel actuators to achieve diverse actuation movements, and a facile fabrication strategy based on removable templates and inside-out diffusion-induced in situ hydrogel crosslinking was adopted. As a result, three types of modular cuboid actuator units (CAUs) achieved linear motion, bending, and twisting. Moreover, combinations of multiple CAUs achieved different actuation modes, including actuation decoupling, superposition, and reprogramming. The diverse actuation functionality would enable new possibilities in application fields for hydrogel soft actuators. Several simple application demos, such as grippers for grasping tasks and a multi-way circuit switch, demonstrated their potential for further applications.
