Xingxia Xu,Weihong Lu,Jian Zhu,Xiangqiang Pan,Xiulin Zhu

A new kind of on-demand dissolution hydrogel is successfully synthesized by modification of chitosan using γ-selenobutyrolactone. The chitosan hydrogel with different selenium contents is formed by ring opening of γ-selenobutyrolactone with the amines of D-glucosamine units on the chitosan backbone. The structure of the hydrogel was confirmed by1H NMR, XRD and XPS. Its physical and biological properties were evaluated by rheology characterization, degradation tests and cytotoxicity test. The hydrogel showed excellent biocompatibility and good degradation properties under oxidation or reduction conditions. All the evidence demonstrated that this type of material has good prospects for dressing applications.Keywords:chitosan;hydrogel;diselenide;on-demand dissolution

Construction of aerogels based on N-halamine and chitosan as wound dressing materials with excellent bactericidal and on-demand dissoluble properties

Jie Li,Maoli Yin,Xi Wang,Lei Chen,Yangfeng Wang,Wei Li,Xun Zhang,Zhenzhen Xu,Xiaojuan Li

To address the clinical challenge of wound infection and secondary injury caused by frequent dressing changes during wound healing, this study aims to develop a chitosan-based aerogel dressing with on-demand dissoluble and efficient antibacterial functionality. A low-temperature alkaline urea dissolution strategy enabled the fabrication of chitosan aerogels (CS) through thermally induced gelation, solvent exchange, and freeze-drying. Subsequent ultrasonic-assisted adsorption of N-halamine 1,3-dichloro-5,5-dimethylhydantoin (MC) yielded functionalized N-halamine-loaded chitosan aerogels (CS-MC). Systematic optimization established 2.0 % CS (based on apparent density, porosity, and ethanol adsorption) and 1.5 % MC loading (determined via swelling kinetics, chlorine release profiles, and antibacterial efficacy) as optimal parameters. 2.0 % CS-1.5 % MC had high effective antibacterial effect. The killing rates of E. coli and S. aureus were 78.57 % and 80.30 % at 1 min, respectively, and the killing rates of both bacterial reached 100 % within 5 min. CS-MC dressings were completely dissolved in a dilute acetic acid solution within 20 min. CS-MC aerogels maintain high fibroblast viability, above the 70 % defined by ISO 10993-5 cytotoxicity threshold, achieving an optimal balance between efficient bactericide and histocompatibility. In summary, CS-MC exhibit high antibacterial efficacy and on-demand dissoluble properties, making them suitable for medical dressings.

Ameliorating Myocardial Infarction by Concurrent Pyroptosis Suppression and On-Demand Oxygen Production via Functionalized Nanocarrier

Wenwu Zhu,Yao Lu,Bin Zong,Ziyi Chen,Yong Ma,Haoran Wang,Peng Cao,Bing Han

The ideal treatment for myocardial infarction should address the critical issues of oxygen supply improvement in the infarcted area and prevention of myocardial cell death. Myocardial necrosis is a reversible process, but the challenge lies in mitigating the irreversible pyroptosis of cardiomyocytes, which significantly impedes the recovery of cardiac function. To tackle these complexities, here we have developed MnO2@INN@Alb (MIA) nanoparticles. MIA nanoparticles possess the unique ability to generate oxygen and release glibenclamide (INN) in response to the acidic and H2O2-rich environments present in the infarcted region. MIA plays a dual role by inhibiting the pyroptosis process of cardiomyocytes through the suppression of NLRP3 and by providing vital oxygen to alleviate hypoxia. The results shows that MIA can effectively reverse cardiomyocyte hypoxia, reduce infarct fibrosis, and aid in the reconstruction of myocardial vessels. Moreover, MIA demonstrates high biocompatibility, making it a promising avenue for the future of myocardial infarction improvement. We anticipate that MIA can pave the way for novel solutions and research paradigms in the field.

Protein-based supramolecular adhesive capable of on-demand adhesion and anti-adhesion for preventing undesired epidural tissue adhesion

Tong Li,Guang Wen,He Zhao,Yang Qu,Han Wu,Yingchuan Sun,Jianwu Zhao,Wen Li

The undesired adhesion between the dura mater and surrounding tissues is a great challenge in the clinical treatment of spinal stenosis and disc herniation, and this condition can typically lead to failed back surgery syndrome (FBSS) and persistent or recurrent lumbosacral pain or sciatica. Here, a protein-based supramolecular adhesive featured with wet adhesion and on-demand anti-adhesion via non-covalent interactions between anionic glycyrrhizic acid (GA) nanofibers and N-α-lauroyl-arginine ethyl ester hydrochloride (LAE) dispersed zein colloid (zein-LAE). By controlling the crosslinking density of the zein-LAE/GA supramolecular polymers, the adhesion and anti-adhesion could be conveniently regulated. In detail, suitable amount of GA improved the adhesion strength of the zein-LAE/GA supramolecular adhesives, whereas excessive GA caused the surface curing and anti-adhesion. It was proposed that the anti-adhesion outcome was attributed to the enhanced cross-linking density and Young’s modulus of the supramolecular polymers, and the declined freedom of the binding groups. The excellent biocompatibility and biodegradation enabled the zein-LAE/GA supramolecular polymers to function as easy-to-implement Janus implants for the in vivo prevention of postoperative epidural adhesion in a rat model. In conclusion, the physical barrier of the Janus implant together with the anti-oxidation and anti-inflammatory effects of the GA component played a synergistic role in achieving epidural anti-adhesion.

Precise synchronization of hyperthermia–chemotherapy: photothermally induced on-demand release from injectable hydrogels of gold nanocages

Jiangshan Wan,Shinan Geng,Hao Zhao,Xiaole Peng,Jiabao Xu,Meihe Wei,Junxiong Mao,Yang Zhou,Quan Zhu,Yanbing Zhao,Xiangliang Yang

Though a therapeutic sequence plays a key role in tumor therapy, little attention has been paid to its influence on multimodal combined therapy. Herein, we developed gold nanocages (GNC@PNA-hls) decorated with two kinds of temperature sensitive p(N-isopropyl-acrylamide-acrylic acid) copolymers (PNA-hs and PNA-ls) for precise antitumor coordination of thermo-chemotherapy. Doxorubicin-loaded GNC@PNA-hls (Dox-GNC@PNA-hls) showed a steady photothermally induced on-demand release under multiple near-infrared (NIR) irradiations. In vitro evaluations indicated that concurrent thermo-chemotherapy treatments (Dox − L) showed the best antitumor effect, compared with the sequence of either doxorubicin treatment followed by NIR radiation (Dox + L) or NIR radiation followed by doxorubicin treatment (L + Dox). The in vivo antitumor efficacy also indicated that the tumor volume was totally suppressed (ca. 0.14 cm3) by the treatment of Dox-GNC@PNA-hls with NIR radiation for 14 days. These results indicated that Dox-GNC@PNA-hls could achieve precise synchronization between hyperthermia and chemotherapy, and effectively enhance their antitumor efficacy.

Aptamer-Functionalized Gold Nanostars for on-Demand Delivery of Anticancer Therapeutics

Dalin Wu,Pei Zhao,Lei Wu,Lihua Lin,Guoyi Yu,Linlin Xu,Jun Yue

Gold nanostars (AuNS) are promising carriers for targeted delivery of therapeutic oligonucleotides, but their potential in fabricating an on-demand drug release system in a facile and robust way remains to be explored. In this paper, we used a model aptamer (HApt), acting not only as a target ligand but also as a natural thermal-responsive material, to decorate AuNS. The prepared gold nanoconstruct, HApt@AuNS, displayed stoichiometric loading capacity of the anthracycline drug doxorubicin (Dox). The on-demand drug release was realized by illuminating nanoconstructs with near-infrared (NIR) light. Furthermore, a higher degree of Dox release from the nanoconstructs was achieved in an acidic environment, compared to neutral conditions. The in vitro experiments showed that Dox-intercalation did not affect the cell uptake efficiency of HApt@AuNS, which could enter cells through clathrin-mediated endocytosis and microtubule-dependent active transport to lysosomes. Dox-loaded HApt@AuNS exhibited intracellular on-demand drug release and enhanced toxicity against cancer cells by NIR-irradiation.

Thermal-responsive heterogeneous organohydrogels with on-demand oil-release capabilities

Yihui Lv,Qi Shi,Xiaocao Yan,Yudong Cai,Yunxuan Weng,Hainan Gao

The responsive secreting function of artificial materials plays an important role in wettability, self-regulation, self-lubrication, antifouling, and controlled loading and release. Drawing inspiration from the secretory tissues in plant bodies, here we present a facile strategy to prepare a stimuli-responsive organohydrogel with water-driven and thermally-driven oil-release functions, and the responsive wettability is achieved. In essence, the stimuli-responsive secretory function is inextricably linked to the infiltration and exfiltration behavior of different dispersion media between heterogeneous networks. The responsive deswelling of the dispersion medium can be induced by certain environmental factors, including changing the dispersion medium or environmental temperature, which further enables unique on-demand water-driven and thermally-driven oil-release properties. It is anticipated that such smart, self-secreting organohydrogel materials, featured with controllable liquid loading and releasing properties, have great potential in a variety of fields, such as drug delivery systems, self-lubrication and antifouling surfaces, and controlled embedding and release of bioactive molecules.

Underoil superhydrophilic flame-retardant 3D porous composite for efficient on-demand emulsion separation: Interface engineering design on sphagnum moss

Guo Yuyan,Liu Peng,Zhang Wenhao,Guo Zhiguang,Liu Weimin

Oil pollution and the energy crisis make oil-water separation an urgent for human need. The widespread use of materials with a single emulsion separation capability is limited. Multifunctional on-demand separation materials can adapt to a wide range of application scenarios, thus having a wider range of applications. The underoil superhydrophilic surface is of great significance for realizing the on-demand separation of oil/water emulsions through the removal of water in the oil and oil in the water. A 3D porous emulsion separation material based on the superhydrophilic principle of sphagnum moss was designed. The material was prepared in a simple step by taking advantage of the adhesion of polydopamine and the introduction of the as-prepared superhydrophilic BaSO 4 nanoparticles to achieve superhydrophilicity with a water contact angle (WCA) of 0° and an oil contact angle (OCA) of 157.3°, resulting in excellent separation performance for both water-in-oil and oil-in-water emulsions. Underoil superhydrophilic porous composite (OSPC) can complete two kinds of emulsion separations by filtration or adsorption. It adsorbs water from water-in-oil emulsion to achieve separation, with a good adsorption capacity of 74.38 g/g and efficiency up to 99%. It can also filter oil-in-water emulsions with an efficiency of 99.92%. The separation efficiencies are all almost unchanged after ten separation cycles. Furthermore, the material has excellent flame retardancy, which reduces the possibility of secondary disasters. The three-dimensional porous sponge has excellent on-demand separation performance for multiple emulsions. It provides a new preparation strategy for underoil superhydrophilic materials and a new idea for the design direction of special wetting materials for the on-demand separation of oil/water emulsions.

Tough Wet Adhesion of Hydrogen-Bond-Based Hydrogel with On-Demand Debonding and Efficient Hemostasis

Xiaodong Wang,Yaxin Guo,Jiangfeng Li,Min You,Yunlong Yu,Jia Yang,Gang Qin,Qiang Chen

Hydrogels have been widely used in wet tissues. However, the insufficient adhesion of hydrogels for wound hemostasis remains a grand challenge. Herein, a facile yet effective strategy is developed to fabricate tough wet adhesion of hydrogen-bond-based hydrogel (PAAcVI hydrogel) using copolymerization of acrylic acid and 1-vinylimidazole in dimethyl sulfoxide followed by solvent exchange with water. The PAAcVI hydrogel shows equally robust adhesion (>400 J m–2) to both wet and dry tissues. Moreover, the PAAcVI hydrogel also exhibits strong long-term stable adhesion underwater and in various wet environments. Meanwhile, the adhesion of PAAcVI hydrogel can be adjusted through Zn2+-ion-mediated on-demand debonding, which makes it easy to peel off from the tissue reducing pain during dressing removal and avoiding secondary injury. The PAAcVI hydrogel displays efficient hemostasis in the mice-tail docking model and mice-liver bleeding model. This hydrogen-bond-based hydrogel shows tough wet adhesion, and its adhesion is controllable, demonstrating its promising application in moisture-resistant adhesives, medical adhesives, and hemostatic materials.

Photoinduced PEG deshielding from ROS-sensitive linkage-bridged block copolymer-based nanocarriers for on-demand drug delivery

Jie Li,Chunyang Sun,Wei Tao,Ziyang Cao,Haisheng Qian,Xianzhu Yang,Jun Wang

Controlling poly(ethylene glycol) (PEG) shielding/deshielding at the desired site of action exhibits great advantages for nanocarrier-based on-demand drug delivery in vivo . However, the current PEG deshielding strategies were mainly designed for anticancer drug delivery; even so, their applications are also limited by tumor heterogeneity. As a proof-of-concept, we explored a photoinduced PEG deshielding nanocarrier TK-NP Ce6&PTX to circumvent the aforementioned challenge. The TK-NP Ce6&PTX encapsulating chlorin e6 (Ce6) and paclitaxel (PTX) was self-assembled from an innovative thioketal (TK) linkage-bridged diblock copolymer of PEG with poly(d, l -lactic acid) (PEG- TK -PLA). We demonstrated that the high PEGylation of TK-NP Ce6&PTX in blood helps the nanocarrier efficiently avoid rapid clearance and consequently prolongs its circulation time . At the desired site (tumor), 660-nm red light irradiation led to ROS generation in situ , which readily cleaved the TK linkage, resulting in PEG deshielding. Such photoinduced PEG deshielding at the desired site significantly enhances the cellular uptake of the nanocarriers, achieving on-demand drug delivery and superior therapeutic efficacy. More importantly, this strategy of photoinducing PEG deshielding of nanocarriers could potentially extend to a variety of therapeutic agents beyond anticancer drugs for on-demand delivery.

ROS-responsive selenium-containing polyphosphoester nanogels for activated anticancer drug release

Yan Zhang,Chuan Ma,Sihang Zhang,Chao Wei,Ying Xu,Wei Lu

Nowadays, the stability and on-demand release of drug carriers are still to be solved. To meet the demand of these issues, we developed a reactive oxygen species (ROS) responsive selenium-containing polyphosphoesters nanogel (PSeP) by a facile one-step ring-opening polymerization of the novel monomer 4-selenoctane-1,8-diyl bis(propylphosphatelane) (Se-COP) with polyethylene glycol (mPEG) employed as the macroinitiator. Their structure was confirmed by NMR, FT-IR and GPC. The crosslinked core-shell structure imparted the nanogels with excellent dimensional stability according to the dynamic light scattering (DLS) and transmission electron microscopy (TEM). Moreover, the selenide groups endowed the nanogels with rich ROS responsiveness when subjected to the stimuli of H 2 O 2 , thus the drug-loaded PSeP nanogels displayed swollen behaviors leading to an activated doxorubicin hydrochloride (DOX · HCl) release. The release mechanisms fitted by the Ritger-Peppas power-law model also proved the swollen release process. MTT assays exhibited that the PSeP nanogels were nontoxic up to a tested concentration of 50 μg mL −1  by A549 and HEK293, and the DOX-loaded PSeP had a high anti-cancer behaviour against A549 cancer cells. Additionally, these nanogels possessed enhanced intracellular drug release by CLSM . Therefore, these results highlighted that the selenium-containing polyphosphoesters nanogels could be a potential platform for the ROS-sensitive drug delivery.

Multifunctional hydrogel wound dressing with rapid on-demand degradation property based on aliphatic polycarbonate and chitosan

Qida Zong,Xinxuan Peng,Yan Ding,Huiying Wu,Chang Lu,Jing Ye,Wei Sun,Jinwei Zhang,Yinglei Zhai

The multifunctional hydrogel dressings are effective strategy to treat chronic wounds of diabetes. In addition, the ability of selective degradation on demand to change dressings could provide better patient compliance. Here, an injectable, self-healing hydrogel with rapid degradability on-demand is designed to promote the healing of diabetes wounds. The block copolymer formed by aldehyde modified aliphatic cyclic carbonate monomer with polyethylene glycol (MBP) and chitosan (CS) were crosslinked through the Schiff base bond to obtain a hydrogel with excellent injectability and self-healing ability. Due to the presence of carbonate bonds in MBP, it showed the rapid on-demand degradation characteristics triggered by N -acetylcysteine (NAC). At the same time, gallic acid (GA) was loaded into the hydrogel, giving the hydrogel dressing antioxidant. In vivo and in vitro experiments showed that the hydrogel wound dressing possesses good natures, such as antibacterial, antioxidant, and friendly cell compatibility, which could promote wound healing. Overall, the multifunctional hydrogel wound dressings with rapid on-demand degradation characteristics are more practical for clinical applications.
