Seiji Samukawa,Masaru Hori,Shahid Rauf,Kunihide Tachibana,Peter Bruggeman,Gerrit Kroesen,J Christopher Whitehead,Anthony B Murphy,Alexander F Gutsol,Svetlana Starikovskaia,Uwe Kortshagen,Jean-Pierre Boeuf,Timothy J Sommerer,Mark J Kushner,Uwe Czarnetzki,Nigel Mason

Low-temperature plasma physics and technology are diverse and interdisciplinary fields. The plasma parameters can span many orders of magnitude and applications are found in quite different areas of daily life and industrial production. As a consequence, the trends in research, science and technology are difficult to follow and it is not easy to identify the major challenges of the field and their many sub-fields. Even for experts the road to the future is sometimes lost in the mist. Journal of Physics D: Applied Physics is addressing this need for clarity and thus providing guidance to the field by this special Review article, The 2012 Plasma Roadmap.Although roadmaps are common in the microelectronic industry and other fields of research and development, constructing a roadmap for the field of low-temperature plasmas is perhaps a unique undertaking. Realizing the difficulty of this task for any individual, the plasma section of the Journal of Physics D Board decided to meet the challenge of developing a roadmap through an unusual and novel concept. The roadmap was divided into 16 formalized short subsections each addressing a particular key topic. For each topic a renowned expert in the sub-field was invited to express his/her individual visions on the status, current and future challenges, and to identify advances in science and technology required to meet these challenges.Together these contributions form a detailed snapshot of the current state of the art which clearly shows the lifelines of the field and the challenges ahead. Novel technologies, fresh ideas and concepts, and new applications discussed by our authors demonstrate that the road to the future is wide and far reaching. We hope that this special plasma science and technology roadmap will provide guidance for colleagues, funding agencies and government institutions. If successful in doing so, the roadmap will be periodically updated to continue to help in guiding the field.

Selective asymmetric polarization of PAANa “diapers”: Solving the water challenge of prussian blue cathode in organic sodium ion battery

Yuhang Du,Xusheng Wang,Yucheng Zhu,Huanrong Zhang,Mengzhen Wei,Huimin Zhang,Hui Ma,Bao Wang,Yanbin Cui,Junhui Ji,Yonglin He,Kai Song,Nabil A. Abdel Ghany,Moustafa M.G. Fouda,Mianqi Xue

Significant research attention has been directed towards the prussian blue analogues (PBAs) cathode for sodium-ion batteries, yet the major obstacle of inferior cycling stability persists. This issue is fundamentally associated with the liberation of their structural and coordinated water into the electrolyte during charge–discharge. In this work, a sodium polyacrylate “diaper” with selective asymmetric polarization is proposed. It’s designed to adsorb water from PBAs during charge–discharge. It ionizes ions to form an asymmetric layer on PBAs, enhancing interfacial kinetics. Density Functional Theory computations validate that PAANa exhibits a more pronounced binding energy with water, efficiently confining moisture within the interface layer and consequently elevating the ionic conductivity of PAANa. Moreover, upon interaction with water, PAANa promptly polarizes, inducing sodium ions to migrate towards the electrolyte side and form a stable charging electrolyte interface. Simultaneously, carboxylate anions engage in robust ion–dipole interactions with the PBA cathode surface, creating a compact coating layer that facilitates sodium ion solvation and desolvation processes, thereby enhancing structural stability. Consequently, PAANa contributes to enhanced rate performance, improve cycle performance (97.9% capacity at 0.15 A g −1 after 300 cycles) and suppresses side reactions at the anode, thereby prolonging the cycling life of the battery.

Multifarious strategies for resolving the deep-curing challenges of PZT piezoceramics slurry in vat photopolymerization

Weigang Ma,Chuanmin Wang,Xinran Wang,Xinyi Zhou,Ya Lu,Xin Guo,Nguyen-Minh-An Tran,Junsheng Yang,Huabin Yang,Danyang Wang,Hua Tan,Haibo Zhang

The vat photopolymerization (VPP) technology offers exceptional flexibility in design and manufaction of high-performance piezoceramic structures. However, due to the high refractive index and strong UV absorption characteristics of Pb-based ceramics, ceramic slurries are required to have a low solid content in for efficient deep-curing. This limitation often results in sintering distortion, cracking, low density, and deteriorated piezoelectric properties. To address these challenges, a heat treatment process was initially employed to spheroidize the PZT powder, which effectively decreased light scattering and absorption. Subsequently, high-refractive-index and highly reactive photosensitive resins were introduced to enhance the penetration of UV light and reduce the critical exposure energy of the slurry. Additionally, a hindered amine light stabilizer is added to capture the peroxy radicals to overcome the oxygen inhibition effect and further increase the curing depth. As a result, a PZT slurry with a high solid content of 45 vol.%, was successfully formulated, facilitating the manufacturing of PZT ceramics with complex structures. The sintered samples achieved a high relative density of 96.5% and a d 33 value of 573 pC/N. These optimized strategies provide a solid foundation for the development of high-performance PZT devices with intricate geometries.

Vaccination with a Human Papillomavirus L2 Multimer Provides Broad Protection against 17 Human Papillomavirus Types in the Mouse Cervicovaginal Challenge Model

Zhenwei Han,Shen Wang,Ting Mu,Ping Zhao,Lingli Song,Ying Zhang,Jin Zhao,Wen Yin,Yue Wu,Huan Wang,Bo Gong,Min Ji,Richard B. S. Roden,Yanping Yang,Michel Klein,Ke Wu

Human papillomavirus (HPV) is a prevalent cause of mucosal and cutaneous infections and underlying conditions ranging from benign warts to anogenital and oropharyngeal cancers affecting both males and females, notably cervical cancer. Cervical cancer is the fourth leading cause of cancer deaths among women globally and is the most impactful in low- and middle-income countries (LMICs), where the costs of screening and licensed L1-based HPV vaccines pose significant barriers to comprehensive administration. Additionally, the licensed L1-based HPV vaccines fail to protect against all oncogenic HPV types. This study generated three independent lots of an L2-based target antigen (LBTA), which was engineered from conserved linear L2-protective epitopes (aa11–88) from five human alphapapillomavirus genotypes inE. coliunder cGMP conditions and adjuvanted with aluminum phosphate. Vaccination of rabbits with LBTA generated high neutralizing antibody titers against all 17 HPV types tested, surpassing the nine types covered by Gardasil®9. Passive transfer of naïve mice with LBTA antiserum revealed its capacity to confer protection against vaginal challenge with all 17 αHPV types tested. LBTA shows stability at room temperature over >1 month. Standardin vitroandin vivotoxicology studies suggest a promising safety profile. These findings suggest LBTA’s promise as a next-generation vaccine with comprehensive coverage aimed at reducing the economic and healthcare burden of cervical and other HPV+ cancers in LMICs, and it has received regulatory approval for a first-in-human clinical study (NCT05672966).

Regulatory T Cells Engineered with Polyphenol-Functionalized Immunosuppressant Nanocomplexes for Rebuilding Periodontal Hard Tissue under Inflammation-Challenged Microenvironment

Bo Zhang,Guidong Gong,Yunxiang He,Jialing Liu,Bo Wang,Yifei Li,Jie Fang,Zhihe Zhao,Junling Guo

Global aging heightens the risk of oral disorders, among which periodontitis is the major cause of tooth loss in the aging population. The regeneration of damaged periodontal hard tissue is highly challenging due to the existence of the refractory local inflammation. Owing to the potent anti-inflammatory capabilities, regulatory T cells hold great promise in immunotherapies for tissue regeneration. However, the transferred regulatory T cells can alter their phenotypes and functions in local inflammatory milieu, significantly impairing their therapeutic efficacy. Herein, we introduce a novel regulatory T cell-based nanobiohybrid system bearing polyphenol-functionalized rapamycin nanocomplexes. The sustained in situ release of immunosuppressant rapamycin from the cell-attached nanocomplexes maintains the anti-inflammatory phenotype of regulatory T cells in the inflammatory milieu. The synergistic actions of the anti-inflammatory cytokines secreted and the immunosuppressant released guide a pro-resolving polarization of macrophages and enhance osteogenic differentiation of bone marrow-derived stromal cells. The stabilized phenotype of the regulatory T cells dramatically promoted the resolution of periodontal inflammation and the repair of the hard tissue (alveolar bone) in vivo . Overall, these studies highlight a potent regulatory T cell-based nanobiohybrid therapy to treat periodontitis by modulating periodontal immune microenvironment.

E. hellem Ser/Thr protein phosphatase PP1 targets the DC MAPK pathway and impairs immune functions

Jialing Bao,Yunlin Tang,Yebo Chen,Jiangyan Jin,Xue Wang,Guozhen An,Lu Cao,Huarui Zhang,Gong Cheng,Guoqing Pan,Zeyang Zhou

Microsporidia are difficult to be completely eliminated once infected, and the persistence disrupts host cell functions. Here in this study, we aimed to elucidate the impairing effects and consequences of microsporidia on host DCs. Enterocytozoon hellem, one of the most commonly diagnosed zoonotic microsporidia species, was applied. In vivo models demonstrated that E. hellem-infected mice were more susceptible to further pathogenic challenges, and DCs were identified as the most affected groups of cells. In vitro assays revealed that E. hellem infection impaired DCs’ immune functions, reflected by down-regulated cytokine expressions, lower extent of maturation, phagocytosis ability, and antigen presentations. E. hellem infection also detained DCs’ potencies to prime and stimulate T cells; therefore, host immunities were disrupted. We found that E. hellem Ser/Thr protein phosphatase PP1 directly interacts with host p38α (MAPK14) to manipulate the p38α(MAPK14)/NFAT5 axis of the MAPK pathway. Our study is the first to elucidate the molecular mechanisms of the impairing effects of microsporidia on host DCs’ immune functions. The emergence of microsporidiosis may be of great threat to public health.

Oral supplementation with Bifidobacterium infantis and 2′-fucosyllactose revives gut microbiota perturbation and intestinal and immune developmental delay following early-life antibiotic challenge in BALB/c mice

Yating Luo,Jinxing Li,Simou Wu,Wen Jia,Zhimo Zhou,Meixun Liu,Fengling Jiang,Ting Huang,Xi Shen,Yun Li,Fang He,Ruyue Cheng

ABSTRACT Probiotics and synbiotics can mitigate the negative health consequences of early-life antibiotic exposure. This study aimed to determine whether supplementation with Bifidobacterium longum ssp. infantis 79 (B79) or synbiotics composed of B79 and 2′-fucosyllactose (2′-FL) could mitigate the negative impact of ceftriaxone exposure in early-life. We found that antibiotic-treated mice exhibited lower body weight, crypt depth, short-chain fatty acid content, and α-diversity indices at weaning, while they increased the relative abundance of opportunistic pathogens (such as Enterococcus and Staphylococcus ) and decreased the relative abundance of intestinal commensal bacteria. Supplementation with B79 and 2′-FL revived these antibiotic-induced negative effects and reduced the mRNA expression of IL-6, IL-12p40, and TNF-α in the spleen at weaning. Moreover, B79 and 2′-FL supplementation persistently improved crypt depth, propionic acid synthesis, and IgG and sIgA production and revived the gut microbiota structure and composition in adulthood. In conclusion, our study suggests that early-life supplementation with B79 alone or in combination with 2′-FL can mitigate ceftriaxone-induced negative effects on the gut microbiota and intestinal and immune development of mice, and these improvements can partially last into adulthood.

A xanthan gum and carbomer-codispersed divalent manganese ion-loaded tannic acid nanoparticle adjuvanted inactivated pseudorabies virus vaccine induces balanced humoral and cellular immune responses

Anan Guo,Lvqing Tang,Bing Yang,Niling Xie,Yandong Cui,Wen Sun,Yuan Li,Xiangting Li,Yang Wu,Yingshuai Liu

Adjuvants including aluminum adjuvant (Alum) and oil-water emulsion have been widely used in inactivated pseudorabies virus (PRV) vaccines to improve their performance, however, they are not sufficient to protect from PRV infection because of the weak immune response and poor Th1-type immune response. Divalent manganese ion (Mn 2+ ) has been reported to increase the cellular immune response significantly. In this work, a xanthan gum and carbomer-dispersed Mn 2+ -loaded tannic acid-polyethylene glycol (TPMnXC) nanoparticle colloid is developed and used as an adjuvant to improve the performance of the inactivated PRV vaccine. The good in vitro and in vivo biocompatibility of the developed TPMnXC colloid has been confirmed by the cell viability assay, erythrocyte hemolysis, blood routine analysis, and histological analysis of mouse organs and injection site. The TPMnXC-adjuvanted inactivated PRV vaccine (TPMnXC@PRV) significantly promotes higher and more balanced immune responses indicating with an increased specific total IgG antibody and IgG2a/IgG1 ratio, efficient splenocytes proliferation, and elevated Th1- and Th2-type cytokine secretion than those of control groups. Wild PRV challenge experiment is performed using mice as a model animal, achieving a protection rate of up to 86.67 %, which is much higher than those observed from the commercial Alum. This work not only demonstrates the high potentiality of TPMnXC in practical applications but also provides a new way to develop the Mn 2+ -loaded nanoadjuvant for veterinary vaccines.

Adaptive Design of Nanovesicles Overcoming Immunotherapeutic Limitations of Chemotherapeutic Drugs through Poliovirus Receptor Blockade

Yongkang Yu,Fan Zhang,Wenqing Xiao,Qinzhen Cheng,Tingxuan Li,Jing Tang,Wei Tao,Lin Mei

Chemotherapy is currently a widely used treatment for cancer in clinical settings. Some chemotherapeutic drugs such as oxaliplatin (OXA) can cause tumor immunogenic cell death (ICD), activate immunity, and realize chemoimmunotherapy for tumors. However, the low degree of accumulation and immunosuppressive microenvironment in tumors limit the immunotherapeutic efficacy of these drugs. T cell immunoreceptor with Ig and ITIM domains (TIGIT)/poliovirus receptor (PVR) is an inhibitory immune checkpoint pathway involved in mediating natural killer (NK) cell and T cell exhaustion in tumors. TIGIT expression is up-regulated in NK cells and CD8+ T cells during tumor development. Moreover, we first found that tumors upregulated PVR expression after OXA treatment in previous work. Here, we systematically analyzed the effects of OXA on the TIGIT/PVR pathway, further proving the effectiveness of the combination of OXA and TIGIT/PVR blocking combination. We developed engineered TIGIT-expressing cell membrane nanovesicles loaded with OXA (OXA@TIGIT MVs) for synergistic cancer therapy. OXA@TIGIT showed good efficacy in several cancer models, leading to tumor regression, effectively inhibiting tumor growth and prolonging mouse survival. Furthermore, the OXA@TIGIT MVs activate a strong tumor-specific immune response in the body, providing long-term (more than 2 months) protection from tumor reactivation in the B16F10 melanoma rechallenge mouse model.
