Effects of Extraction Methods on Volatile Oil Profiles of Cinnamomi ramulus–Zingiberis rhizoma recens Couplet Medicines

Xiaodong Xin,Xinnong Li,Jiabao Chen,Chuanghui Shen,Xiaohan Shi,Lei Wang,Chunhua Li

Cinnamomi ramulus(CR, Guizhi in Chinese) andZingiberis rhizoma recens(ZRR, Shengjiang in Chinese) are couplet medicines (drug pairs or drug combinations) in traditional Chinese medicine prescriptions. They contain a high amount of volatile oils which endow them with unique flavors and pharmacological activities. Extraction methods have a great influence on the extraction efficiency and composition of volatile oils. Firstly, the volatile oils of CR and ZRR were extracted by steam distillation (SD) and analyzed by GC-MS to obtain their chemical profiles. In total, 35 and 55 compounds were identified in the volatile oils of CR and ZRR, respectively. In order to find a suitable extraction method for the couplet medicine CR-ZRR, subsequently, steam distillation (SD), azeotropic distillation (AD) and supercritical fluid extraction (SFE) were applied to extract the volatile oils from CR-ZRR. The average extraction yields by SD, AD and SFE were 0.573%, 0.62% and 2.135%, respectively. The chemical composition of the volatile oils was then analyzed by GC-MS. In total, 73, 59 and 71 compounds were identified from the extracts obtained by SD, AD and SFE, respectively. Principal component analysis (PCA) and OPLS-DA showed that citral, (E)-cinnamaldehyde dimethyl acetal, zingiberene, cinnamaldehyde and β-sesquiphellandrene were the main contributors to distinguish the volatile oils that were obtained by different processes. Considering the chemical diversity and the total content of the main bioactive components of the volatiles oils, SD was more suitable for CR-ZRR volatile oil extraction. This study provides a basis for elucidating the chemical composition and suitable extraction method for the volatile oils of CR-ZRR.

Ginseng volatile oil prolongs the lifespan and healthspan of Caenorhabditis elegans

Wang Lixin,Qiao Ping,Ouyang Zhuoer,Li Danyang,Zheng Jingtong,Wang Guoqiang,Wang Fang

Ginseng volatile oil (GVO) is one of the main components of ginseng and has antibacterial and anti-inflammatory properties. In this study, gas chromatography–mass spectrometry (GC–MS) was applied to characterize GVO chemical composition, and 73 volatile components were detected from GVO. Caenorhabditis elegans was used as animal model to further elucidate the antioxidant and anti-aging effects of GVO in vivo. The results suggested that GVO significantly prolonged the lifespan of C. elegans and promoted its health without damaging its reproductive capacity. In addition, GVO increased the antioxidant capacity and survival rate of nematodes after heat shock. Transcriptional sequencing showed that autophagy-related genes atg-4.2 , atg-7 , lgg-2 , and cyd-1 were up-regulated, and superoxide dismutase 1 ( sod-1 ) expression was increased after GVO pretreatment. Considering the role of autophagy and antioxidant in aging, the expression of autophagy substrate P62 protein in BC12921 strain was analyzed and found to decrease by more than 50.00% after treatment with GVO. In addition, the lifespan of SOD-1 mutant nematodes was not significantly different from that of the control group. SOD activity and autophagy were activated, which is a clear expression of hormesis. All these results suggest that GVO prolongs the lifespan and healthspan of C. elegans , and its biological functions may be related to hormesis.

Background Frankincense volatile oil (FVO) has long been considered a side product in pharmaceutical industry since frankincense of large molecular weight is the prime target. However, the volatile oil recycled in the extract process might contain a series of functional actives, serving as promising ingredients in the cosmetic field. Methods Gas chromatography-mass spectrometer was utilized to determine the species and amount of active ingredients in FVO. Subsequently, zebrafish models were used to evaluate pigmentation inhibition, ROS elimination and neutrophil activation. In vitro DPPH test was also conducted to consolidate the anti-oxidation efficacy. Based on the test results, network pharmacology was incorporated, where GO and KEGG enrichment analyses were performed to discover the interrelations between active ingredients. Results About 40 actives molecules were identified, including incensole, acetate incensole, and acetate incensole oxide. The FVO demonstrated great depigmentation activity by suppressing melanin synthesis, as well as providing free radical scavenging and anti-inflammation effect. In network pharmacology analysis, 192 intersected targets were identified. By enrichment analysis and network construction, a series of whitening signal pathways, and hub genes, containing STAT3,MAPK3,MAPK1 were identified. Conclusion The current study quantified the components of FVO, evaluated its efficacy in skin depigmentation, and give pioneering insights on the possible mechanism. The results confirmed that the FVO could serve as whitening agent in topical uses.

Bojun Chen,Jijun Li,Yuhuan Xie,Xi Ming,Gang Li,Jinjin Wang,Meng Li,Xiaohong Li,Lei Xiong

Ethnopharmacological relevance Cang-ai volatile oil (CAVO) is a traditional Chinese medicine (TCM) inhalational preparation for the treatment of some depressive and emotive disorders. Aim of the study This research aimed to evaluate the efficiency and possible mechanism of intranasal CAVO administration on depression in chronic unpredictable mild stress (CUMS)-induced rats compared to lavender volatile oil (LVO) treatment after CUMS exposure and bilateral olfactory bulb impairment (OBI) in rats. Materials and methods Forty depressive-like model rats induced by CUMS were evaluated by the forced swim test (FST), open field test (OFT), and sucrose preference test (SPT). The model rats were divided into five groups: CUMS (n = 8), CAVOh + CUMS (n = 8), CAVOl + CUMS (n = 8), LVO + CUMS (n = 8), and OBI + CAVO + CUMS (n = 8). The CUMS-induced rats were treated for a period of 4 weeks. The other healthy rats were regarded as the control (CTR, n = 8) subjects. The levels of serotonin (5-HT) and dopamine (DA) and their respective metabolites homovanillic acid (HVA) and 5-hydroxyindol acetic acid (5-HIAA) were measured in brain tissue homogenates of CUMS-induced rats using enzyme-linked immunosorbent assay (ELISA). Results CAVO ameliorated depressive-like behaviors (p < 0.05 ). The levels of DA in the CUMS group were lower than those in the CTR and CAVOh groups (** p < 0.01 and * p < 0.05 ). The levels of HVA were lower in the CUMS group than in the CTR, LVO, OBI + CAVOh and CAVOh groups (** p < 0.01 and * p < 0.05 ) and lower in the OBI + CAVOh group than in the CAVOh group (** p < 0.01 ). The levels of 5-HT in the CUMS group were lower than those in the CTR and CAVOh groups (** p < 0.01 ). The levels of 5-HIAA were lower in the CUMS and OBI + CAVOh groups than in the CTR, LVO and CAVOh groups (** p < 0.01 ). Conclusions CAVO can improve depressive-like behaviors concomitant with the regulation of DA and 5-HT metabolism in the brains of CUMS-induced rats.

Liping Yu,Wang Cheng,Meifen Tian,Zhigang Wu,Xiaoli Wei,Xing Cheng,Mingwei Yang,Xuan Ma

The medicinal value ofP. zhennanhas been documented in traditional Chinese medicine books. The aim of this paper was to study the antioxidant activity of alcoholic extracts ofP. zhennanleavesusing 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2-phenyl-4,4,5,5-tetramethylimidazolineoxyl-1-oxyl-3-oxide (PTIO) radical scavenging and ferric ion reducing antioxidant power (FRAP) assays. The active components of the leaves were identified via headspace solid-phase microextraction and gas chromatography–mass spectrometry (HS-SPME-GC-MS). The results showed that the scavenging rate of DPPH was 94.67%with an EC50 value of 0.674 mg/mL at a concentration of 2 mg/mL. The maximum scavenging rate was 47.40% at a Trolox equivalent of 0.33 mg TE/mL for PTIO radicals. The FRAP reached 84.80% at 0.20 mg/mL concentration. The results confirmed the strong antioxidant activity of the extracts. Furthermore, 44 compounds, mostly terpenoids, obtained from the alcoholic extracts ofP. zhennanleaves were analyzed using HS-SPME-GC-MS and 15 of these compounds had a relative content exceeding 1%. The strong antioxidant activity of the alcoholic extracts ofP. zhennanleaves could be attributed to the presence of copaene (33.97%), β-caryophyllene (4.42%), δ-cadinene (11.04%), γ-muurolene (4.78%), cis-calamenene (2.02%), linalool (1.04%), α-pinene (1.46%), borneol acetate (1.5%), and γ-terpinene (0.66%). This study demonstrates the potential medicinal value of alcoholic extracts ofP. zhennanleaves.

Bao Yu,Dan Zhang,Xue-Wei Yan,Jian-Wei Wang,Ling Yao,Li-Hong Tan,Sheng-Ping Zhao,Na Li,Wei-Guo Cao

In this study, volatile oils of six Hawk tea varieties were studied for their chemical composition, antioxidant and antimicrobial activities to screen the most suitable botanical origins of Hawk tea. A total of 72 components were separated and identified from the six oils. The major constituents of the volatile oils were: α -pinene, camphene, limonene, 1,8-cineole, linalool, cis -nerolidol, and germacrene B. Moreover, the volatile oils were evaluated for antioxidant potential and antimicrobial activities. The results showed that all volatile oils exhibited acceptable antioxidant and antimicrobial activities, which suggested that these volatile oils may serve as natural alternatives to synthetic antioxidants and preservatives to be applied in food and pharmaceutical industries. Principal component analysis results denoted that some major compounds may be closely related to the antioxidant and antimicrobial activities. It also showed that the volatile oils from Litsea coreana var. lanuginosa and Litsea pungens Hemsl . were characterized by positive values of first two principal components, indicating higher active chemical compounds and antioxidant and antimicrobial activities compared with other species. Thus, they were temporarily considered as good sources of Hawk tea. Graphical

Screening bioactive compounds from Qianghuo (Notopterygium incisum) volatile oil via COX-2 magnetic ligand fishing

Li Zhang,Hui Wu,Xiaoyuan Li,Xiu Zhang,Hua Li,Huaqiao Tang,Daxuan Tang,Shijing Du,Ya’’ou Liu,Yangfan Tang,Xiaoming Bao,Guoqiang Cheng

Qianghuo (the rhizomes and radixes of Notopterygium incisum Ting ex H.T. chang) is a traditional Chinese medicine, and widely used as an anti-inflammatory, anodyne, antifebrile agent. Qianghuo volatile oil also exhibits the same properties, and the effects are probably related to the inhibition of cyclooxygenase (COX), but its bioactive components are not clear yet. To figure out the bioactive components of Qianghuo volatile oil target to cyclooxygenase-2 (COX-2), a ligand fishing method using immobilized COX-2 magnetic beads (COX-2-MB S ) and gas chromatography–mass spectrometry (GC–MS) were developed to identify the compounds that act on COX-2. Nine ligands, including 3-carene, β-caryophyllene, eucalyptol, α-terpinene, Limonene, δ-terpinene, α-pinene, β-pinene and γ-terpinene, were specifically captured by COX-2-MB S . Among them, 3-carene, β-caryophyllene, and eucalyptol in Qianghuo volatile oil were isolated and confirmed by in vitro inhibition and in vivo anti-inflammatory activity assays as selective inhibitor against COX-2. The IC 50 values were 13.5, 10.1, and 7.6 μM, respectively. Compared with the model, 3-carene, β-caryophyllene, and eucalyptol considerably reduced ear swelling and paw edema. Moreover, 3-carene and eucalyptol significantly decreased the changes of COX-2 expression in RAW264.7 cells activated by LPS. Our study validated that the developed ligand-fishing method using COX-2-MBs coupled with GC–MS can be applied to recognize COX-2 inhibitors in Qianghuo volatile oil. In vitro/vivo assay showed 3-carene and eucalyptol in Qianghuo volatile oil had strong inhibition against COX-2. This study may provide novel methods for discovering active components for volatile oil–based drug development or aromatherapy.

Microencapsulation of Ginger Volatile Oil Based on Gelatin/Sodium Alginate Polyelectrolyte Complex

Lixia Wang,Shiwei Yang,Jinli Cao,Shaohua Zhao,Wuwei Wang

The coacervation between gelatin and sodium alginate for ginger volatile oil (GVO) microencapsulation as functions of mass ratio, pH and concentration of wall material and core material load was evaluated. The microencapsulation was characterized by scanning electron microscopy (SEM), Fourier transform infrared (FT-IR), and thermal gravimetric analysis (TGA). SEM and FT-IR studies indicated the formation of polyelectrolyte complexation between gelatin and sodium alginate and successful encapsulation of GVO into the microcapsules. Thermal property study showed that the crosslinked microparticles exhibited higher thermal stability than the neat GVO, gelatin, and sodium alginate. The stability of microencapsulation of GVO in a simulated gastric and an intestinal situation in vitro was also studied. The stability results indicated that the release of GVO from microcapsules was much higher in simulated intestinal fluid, compared with that in simulated-gastric fluid.

Chemical composition and antifungal activity of essential oil from Origanum vulgare against Botrytis cinerea

Yun Zhao,Yun-Hai Yang,Min Ye,Kai-Bo Wang,Li-Ming Fan,Fa-Wu Su

In this study, the chemical composition of the essential oil (EO) of Origanum vulgare was characterized, and the antifungal activity of the EO and some individual components against Botrytis cinerea (Y-BC-1) was determined . Twenty-one components were identified by gas chromatography–mass spectrometry and gas chromatography–flame ionization detection, constituting 95.7% of the EO. The major components were methyleugenol (16.5%), myristicin (15.6%), carvacrol (15.0%), thymol (9.8%), apioline (9.4%), and (Z)-β-farnesene (8.7%). B. cinerea in vitro mycelial growth and spore germination were strongly inhibited by the EO and two of its main components, thymol and carvacrol. In vivo vapor contact assays, the antifungal activity of the EO at 250 mg/L suppressed the decay of cherry tomatoes 96.39%. Moreover, thymol and carvacrol at 125 mg/L completely suppressed the gray mold. Thus, the EO of O. vulgare is a potentially nontoxic and ecofriendly botanical fungicide for postharvest control of gray mold.
