The present study aimed to systematically investigate black tea aroma formation during the fermentation period. In total, 158 volatile compounds were identified. Of these, most amino acid-derived volatiles (AADVs) and carotenoid-derived volatiles (CDVs) showed significant increases, while fatty acid-derived volatiles (FADVs) and volatile terpenoids (VTs) displayed diverse changes during the fermentation period. During this time, fatty acids, amino acids, carotenoids, and glycosidically bound volatiles (GBVs, especially primeverosides) were found to degrade to form aroma components. Further, equivalent quantification of aroma showed that the intensity of green scent was notably decreased, while the intensities of sweet and floral/fruity scents were greatly increased and gradually dominated the aroma of tea leaves. AADVs and CDVs were shown to make greater contributions to the formation of sweet and floral/fruity scents than VTs. Our study provides a detailed characterization of the formation of sweet and floral/fruity aromas in black tea during the fermentation period.

BACKGROUND The diamondback moth, Plutella xylostella , has developed resistance to almost all insecticides used for its control. The ‘push–pull’ method has been shown as an effective control strategy to address this resistance challenge of P. xylostella . The key focus of the strategy is the identification of attractive or repellent volatile components. The aim of this study was to identify attractive volatile compounds released from host plants. Identified compounds were applied in the biological control of this pest. RESULTS Nine active compounds released into the headspace of seven cruciferous plant species were identified using gas chromatography–electroantennographic detection and gas chromatography–mass spectrometry. Electroantennographic detection-active compounds included five green leaf volatiles (hexanal, trans -2-hexen-1-ol, cis -3-hexen-1-ol, cis -3-hexenyl acetate, and 1-penten-3-ol), three isothiocyanates (isopropyl isothiocyanate, allyl isothiocyanate, and butyl isothiocyanate), and nonanal. Except for nonanal, all the identified green leaf volatiles and isothiocyanates elicited strong electrophysiological and behavioral responses in P. xylostella . The strongest attractive compounds, trans -2-hexen-1-ol and isopropyl isothiocyanate, were further evaluated in oviposition and field-trapping assays. Results showed that they both lured female moths to lay eggs, and were highly attractive to P. xylostella adults in field, especially when used in combination with yellow and green sticky boards. However, a blend of the two compounds showed no synergistic effect, but rather an antagonistic effect. CONCLUSIONS Green leaf volatiles and isothiocyanates were identified as key olfactory cues for host selection of P. xylostella . Trans -2- hexen-1-ol and isopropyl isothiocyanate were identified as candidate attractive compounds to serve in a ‘push–pull’ strategy for P. xylostella control. © 2023 Society of Chemical Industry.

This study delved into the aroma characteristics of “Qingxiang” oolong tea, analyzing six different cultivars and their processing modes. The findings showed that both cultivars and processing modes have a significant impact on the oolong tea aroma system. The study identified 18 terpenoid volatiles (VTs), 11 amino-acid-derived volatiles (AADVs), 15 fatty-acid-derived volatiles (FADVs), 3 carotenoid-derived volatiles (CDVs), and 10 other compounds in oolong tea that differentiate it from green and black tea. The turn-over stage was found to be the primary processing stage for oolong tea aroma formation. Molecular sensory analysis revealed that the “fresh” odor attribute is the basis for its aroma, while “floral and fruity” fragrances are its aroma characteristics. The perception of oolong tea as “fresh” and “floral and fruity” is influenced by the interactions of its aroma components. These findings provide a new basis for breed improvement and process enhancement in oolong tea production.

Studies show that plant defenses influence the host-use of herbivores and tend to be evolutionarily more labile than herbivore traits (e.g. feeding preferences). However, all previous studies have focused exclusively on non-volatile plant defenses thereby overlooking the roles of plant volatiles. We hypothesized that volatiles are equally important determinants of herbivore host-use and are evolutionarily more labile than herbivore traits. To test these hypotheses, the following experiments were conducted. We identified the volatiles and non-volatiles of 17 Asteraceae species and measured their relative contents. We also used a highly resolved bipartite trophic network of the 17 host species and 20 herbivorous (pre-dispersal seed predator) tephritid fly species to determine the evolutionary interactions between plants and herbivores. The chemical data showed that interspecific similarity in volatiles—but not non-volatiles and phylogenetic distance—significantly accounted for the herbivore community across the plant species; this implies that plant volatiles—but not non-volatile compounds and species identity—dictate plant-tephritid fly interactions. Moreover, we observed phylogenetic signal for non-volatiles but not for volatiles; therefore closely related herbivores do not necessarily use closely related host species with similar non-volatiles, but do tend to attack plants producing similar volatiles. Thus, plant volatiles are evolutionarily more labile than non-volatiles and herbivore traits associate with host use. These results show that the interactions between plants and herbivores are evolutionary asymmetric, shed light on the role of plant volatiles in plant–herbivore interactions, and highlight the need to include data for both volatiles and non-volatiles when investigating plant–animal interactions.

‘Taro-like’ odor is an important economic trait of pumpkin species. The metabolic and molecular bases of this aromatic trait remain largely unexplored. Therefore, in this study, gas chromatography-mass spectrometry, GC-Olfactometry, and RNA-seq technology were used to illuminate the differential volatile compounds, the key volatile compounds, and differentially expressed genes (DEGs) in leaves from two pumpkin samples. Eight volatile compounds, including (E)-2-nonenal, 3-octanol, 2-ethyl-1-hexanol, 1-nonanol, α-terpineol, 2,3-pentanedione, caryophyllene, and 2-acetyl-1-pyrroline, were only detected in the sample with ‘taro-like’ aroma. Moreover, the variable importance in projection scores of all the above eight volatile compounds were >1.0 using PLS-DA analysis. The compounds 2-acetyl-1-pyrroline, 3-octanol, 1-nonanol, and (E)-3,7-dimethyl-2,6-octadienal were identified as the key contributors using GC-Olfactometry analysis. It was determined that 2-acetyl-1-pyrroline might play a significant role in ‘taro-like’ aroma. Furthermore, most of the differential volatile compounds were derived from fatty acids, and the DEGs were also involved in the pathways related to degradation, metabolism, and biosynthesis of fatty acids. Moreover, five genes involved in the accumulation of 2-acetyl-1-pyrroline showed differential expression, and their expression trends were consistent with 2-acetyl-1-pyrroline. This study offers the basis for further studies on the mechanism of ‘taro-like’ aroma in pumpkins.Keywords:pumpkin;‘taro-like’ aroma;volatiles;transcriptome

Functional and nutritional compounds are increased during foxtail millet germination while bad smell is produced due to the fatty acid oxidation. To eliminate the unpleasant aroma, the origins of the volatiles must be known. A comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry showed forty-nine volatiles containing 8 ketones, 10 aldehydes, 20 alkanes, 4 alcohols, 5 alkenes, and 2 furans were tentatively identified, and they increased during the germination of the foxtail millet. To identify the origin of some volatiles, model experiments by adding 6 fatty acids to the crude enzymes of the foxtail millet was designed, and 17 volatiles could be detected. The saturated fatty acids (palmitic acid and stearic acid) had no contributions to the formation of the volatiles, whereas the unsaturated fatty acid played important roles in the formation of volatiles. Among the unsaturated fatty acids, palmitoleic acid and linoleic acid produced most aldehydes, alcohols, and ketones, while linolenic acid produced the most alkanes and alkenes. This study will be helpful for controlling the smell of germinated seeds from the raw material selection. Keywords:volatile;fatty acid;variation;germination;foxtail millet

Simple SummarySince the widespread adoption of Bt cotton in the late 1990s, the green mirid bug,Apolygus lucorum(Hemiptera: Miridae), has become one of the most important pests in cotton fields and some other crops. To manage this destructive pest,Peristenus spretus(Hymenoptera: Braconidae) has been tested in augmentative biological control. In this study, after cotton plants were damaged by different densities ofA. lucorum, the behavioral responses ofA. lucorumandP. spretusto cotton plants volatiles were evaluated, and the quality and quantity of volatiles from cotton plants were analyzed. The results demonstrated that HIPVs emitted by plants in response toA. lucorumcould be influenced by the pest density and could be identified byP. spretusas a signal of the host. Our results would help understand howP. spretusplays a role in biological control againstA. lucorum.AbstractPlants would release herbivore-induced plant volatiles (HIPVs) to repel herbivores and attract natural enemies after being damaged by herbivores. In this study, after cotton plants were damaged by different densities ofApolygus lucorum, the behavioral responses ofA. lucorumandPeristenus spretusto cotton plants volatiles were evaluated, and the quality and quantity of volatiles from cotton plants were analyzed. Only when cotton plants were damaged by four bugs did bothA. lucorumandP. spretusshow an obvious response to damaged cotton plants, which indicates that cotton defense is correlated with pest density. The collection and analysis of volatiles reveals that the increase in pest density results in the emission of new compounds and an increase in the total number of volatiles with an alteration in proportions among the compounds in the blend. These changes in volatile profiles might provide wasps and mirids with specific information on host habitat quality and thus could explain the behavioral responses of parasitoids and pests. Keywords:behavior;pest;natural enemy;HIPVs;density

We determined whether plant volatiles help explain why Cotesia chilonis (a parasitoid of the target pest Chilo suppressalis ) is less abundant in Bt than in non- Bt rice fields. Olfactometer studies revealed that C. chilonis females responded similarly to undamaged Bt and non- Bt rice plants. Parasitoids preferred rice plants damaged by 3 rd -instar larvae of C. suppressalis, but did not differentiate between caterpillar-infested Bt and non- Bt plants. According to GC-MS analyses of rice plant volatiles, undamaged Bt and non- Bt rice plants emitted the same number of volatile compounds and there were no significant differences in the quantity of each volatile compound between the treatments. When plants were infested with and damaged by C. suppressalis larvae, both Bt and non- Bt rice plants emitted higher numbers and larger amounts of volatile compounds than undamaged plants, but there were no significant differences between Bt and non- Bt plants. These results demonstrate that the volatile-mediated interactions of rice plants with the parasitoid C. chilonis were not disrupted by the genetic engineering of the plants. We infer that parasitoid numbers are lower in Bt than in non- Bt fields because damage and volatile induction by C. suppressalis larvae are greatly reduced in Bt fields.

Herbivore-induced plant volatiles (HIPVs) play an important role in insect resistance. As a common HIPV in tea plants (Camellia sinensis), β-ocimene has shown anti-insect function in other plants. However, whether β-ocimene in tea plants also provides insect resistance, and its mechanism of synthesis and emission are unknown. In this study, β-ocimene was confirmed to interfere with tea geometrid growth via signaling. Light was identified as the key factor controlling regular emission of β-ocimene induced by the wounding from tea geometrids. β-Ocimene synthase (CsBOS1) was located in plastids and catalyzed β-ocimene formation in overexpressed tobacco. CsBOS1 expression in tea leaves attacked by tea geometrids showed a day-low and night-high variation pattern, while CsABCG expression involved in volatile emission showed the opposite pattern. These two genes might regulate the regular β-ocimene emission from tea plants induced by tea geometrid attack. This study advances the understanding on HIPV emission and signaling in tea plants.

Tea volatiles’ generation and retention over manufacturing processes are crucial for tea quality. In this study, floral volatile adsorption and retention in green tea scented with Jasminum sambac flowers were examined over the scenting process. Out of 34 enhanced volatiles in the scented tea, β-ionone, β-linalool, indole, and methyl anthranilate were the most potent odorants with 5.1–45.2-fold higher odor activity values than the corresponding controls in the nonscented tea. Scenting efficiencies for the floral volatiles retained in the scented tea (the percentage of volatile abundance over its corresponding amount in jasmine flowers) ranged from 0.22% for α-farnesene to 75.5% for β-myrcene. Moreover, due to additional rounds of heat treatment for scented green tea manufacturing, some volatiles such as carotenoid-derived geraniol and β-ionone and lipid-derived ()-jasmone were heat-enhanced and others such as nonanal were heat-desorbed in the scented green tea. Our study revealed that dynamic volatile absorption and desorption collectively determined tea volatile retention and tea aroma. Our findings may have a great potential for practical improvement of tea aroma.

The diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae) is one of the most destructive pests to cruciferous plants worldwide. The oligophagous moth primarily utilizes its host volatiles for foraging and oviposition. Chemosensory proteins (CSPs) are soluble carrier proteins with low molecular weight, which recognize and transport various semiochemicals in insect chemoreception. At present, there is limited information on the recognition of host volatiles by CSPs of P. xylostella. Here, we investigated expression patterns and binding characteristics of PxylCSP11 in P. xylostella. The open reading frame of PxylCSP11 was 369-bp encoding 122 amino acids. PxylCSP11 possessed four conserved cysteines, which was consistent with the typical characteristic of CSPs. PxylCSP11 was highly expressed in antennae, and the expression level of PxylCSP11 in male antennae was higher than that in female antennae. Fluorescence competitive binding assays showed that PxylCSP11 had strong binding abilities to several ligands, including volatiles of cruciferous plants, and (Z)-11-hexadecenyl acetate (Z11-16:Ac), a major sex pheromone of P. xylostella. Our results suggest that PxylCSP11 may play an important role in host recognition and spouse location in P. xylostella.

Simple SummaryThe Solanaceae crops are the main hosts ofZeugodacus tau, a pest insect that causes serious damage to economically important crops of Solanaceae. In the winter–spring period, female adults ofZ. tauhave been found to oviposit on the pepino melonSolanum muricatum. However, the differences in the ability of the fruits ofS. muricatumand other Solanaceae crops to attract gravidZ. taufemales have not been clarified. In this study, oviposition and trapping bioassays were performed to clarify whether such differences existed. GC-MS was used to identify the compounds and relative contents of the volatile odors ofS. muricatum. The Y-tube olfactometer system was used to determine the compounds that induced behavioral responses in gravidZ. taufemales. The results show thatSolanum muricatumodor was attractive forZ. taufemales. The nine compounds from the volatiles ofS. muricatuminduce behavioral responses in gravidZ. taufemales. These results are beneficial for preventingZ. taufemales from harmingS. muricatum, developing attractants and repellents forZ. taufemales, and establishing a system of ecological control forZ. taufemales.AbstractZeugodacus tau(Walker) (Diptera: Tephritidae) is a pest seriously harmful to Solanaceae crops and was found to oviposit on the pepino melonSolanum muricatum(Aiton). To date, the differences in the ability of the fruits ofS. muricatumand other Solanaceae crops to attract gravidZ. taufemales have seldom been reported. Oviposition and trapping bioassays were performed to clarify whether such differences existed. A combination of GC-MS and the Y-tube olfactometer system was used to identify and determine the compounds inducing behavioral responses in gravidZ. taufemales to the volatile odors ofS. muricatum. The results show thatS. muricatumodors play a role in attracting gravidZ. taufemales. The odors of Solanaceae crops influence their ability to attract these organisms. The nine compounds from the volatiles ofS. muricatuminduce tendency or repellency responses in gravidZ. taufemales. Hexyl acetate, butyl acetate, amyl actate, and isoamyl acetate caused tendency behavior in gravidZ. taufemales, while hexyl hexanoate, butyl isovalerate, butyl valerate, and isoamyl hexanoate caused repellency behavior. Heptyl acetate caused repellency behavior in gravidZ. taufemales at higher concentrations (5 mg/mL) but caused tendency behavior at a low concentration (0.5 mg/mL). These results suggest that vigilance against the harm caused byZ. tauis required during the cultivation ofS. muricatum. The nine compounds of the volatile odors ofS. muricatumcould help to develop attractants and repellents for gravidZ. taufemales. These results are beneficial for preventingZ. taufemales from harmingS. muricatum, developing attractants and repellents forZ. taufemales, and establishing a system of ecological control forZ. taufemales. Keywords:Zeugodacus tau;Solanum muricatum;induced oviposition;attraction rate;volatile
