Electrostatic Trapping of Double-Stranded DNA Based on Cd(OH)2 Three-Side Nanobelt Architectures

Di Bao,Peng Gao,Yanbo Wang,Han Zhou,Yujin Chen,Guorui Chen,Piaoping Yang,Xitian Zhang

In this paper, Cd(OH)2 1D three-side nanobelt architectures have been fabricated through a facile two-phase reaction system. Considering their electropositive character proven by zeta potential measurement, the as-obtained Cd(OH)2 nanomaterials have been applied to capture negatively charged DNA. On the basis of the above results, the Cd(OH)2 nanomaterials have been employed for trapping negatively charged double-stranded DNA molecules (salmon sperm DNA). Assisted by the detailed observation of their DNA interaction results as well as through UV–vis spectroscopy, infrared imaging automatic target recognition (IRATR) and inductive coupled plasma atomic emission spectrometer (ICP-AES) measurements, it has been clearly shown that this 1D nanoarchitecture can effectively trap and release DNA molecules without use of any other organic reagents. In addition, a miniaturized electrochemical DNA-releasing device based on the Cd(OH)2 1D three-side nanobelt architectures has been fabricated. Using this device, it is possible to recycle the Cd(OH)2 nanostructures. In addition, the Cd(OH)2 nanomaterials maintain their nanostructures, and no Cd2+ ions were released in repeated DNA separation and transfer processes, which demonstrate their practicality for DNA trapping.

Fabricating upconversion fluorescent nanoparticles modified substrate for dynamical control of cancer cells and pathogenic bacteria

Huanhuan Li,Quansheng Chen,Jiewen Zhao,Khulal Urmila

Lanthanide-doped upconversion nanoparticles (UCNPs) have attracted widespread interests in the field of biomedicine because of their unique upconverting capability by converting near infrared (NIR) excitation to visible or ultraviolet (UV) emission. Here, we developed a novel UCNP-based substrate for dynamic capture and release of cancer cells and pathogenic bacteria under NIR-control. The UCNPs harvest NIR light and convert it to ultraviolet light, which subsequently result in the cleavage of photoresponsive linker (PR linker) from the substrate, and on demand allows the release of a captured cell. The results show that after seeding cells for 5 h, the cells were efficiently captured on the surface of the substrate and ˜89.4% of the originally captured S. aureus was released from the surface after exposure to 2 W/cm 2 NIR light for 30 min, and ˜92.1% of HepG2 cells. These findings provide a unique platform for exploring an entirely new application field for this promising luminescent nanomaterial.

Thermal-responsive PNIPAM/UiO-66 aerogels for controllable water capture and release

Pu Zhou,Han Dong,Wanglin Zhang,Jiachen Liu,Yefei Tian,Yi Li,Jiabei Luo,Song Xiao,Kerui Li,Hongye Yuan

Developing metal–organic framework (MOF)-based sorbents with high water-uptake capacity and mild regeneration conditions remains challenging, but is highly sought after for addressing water shortages. Herein, we report a strategy for fabricating thermal-responsive Poly (N-isopropylacrylamide) (PNIPAM)/UiO-66 composite aerogels with variable PNIPAM loading by leveraging the ordered nanopores of UiO-66 colloids, enabling the confined polymerization of NIPAM monomers within the pores, which allows for controllable water capture and release at above 32 ℃. The thermal-responsive PNIPAM/UiO-66 composite aerogels, with varying PNIPAM loadings, demonstrated an outstanding maxima water capture of over 2100 wt% at room temperature and reversible release at above 32 ℃. This performance sets a new benchmark for the leading water-capture adsorbents reported in the literature, thanks to the extensive specific surface area of the aerogels and the unique hydrophilicity-hydrophobicity transition exhibited by the PNIPAM component. More intriguingly, the aforementioned phase transition can be directly visualized by incorporating 4-(1,2,2-triphenylvinyl) benzoic acid (TPE-COOH) molecules with the well-known aggregation-induced emission (AIE) effect into the aerogels. The remarkable water-uptake ability and mild release conditions showcased in this study open new possibilities for practical water harvesting.

Hyaluronic acid-functionalized redox responsive immunomagnetic nanocarrier for circulating tumor cell capture and release

Yi Zhang,Wenjing Wang,Huiling Guo,Mingxing Liu,Hongda Zhu,Hongmei Sun

Detection of circulating tumor cells (CTCs) in peripheral blood holds significant insights for cancer diagnosis, prognosis evaluation, and precision medicine. To efficiently capture and release CTCs with high viability, we reported the development of hyaluronic acid (HA)-functionalized redox responsive immunomagnetic nanocarrier (Fe3O4@SiO2-SS-HA). First, Fe3O4 nanoparticles were prepared and modified with tetraethyl orthosilicate (TEOS), 3-mercaptopropyltrimethoxysilane (MPTMS) and 2,2'-dithiodipyridine (DDPy) to form the magnetic substrate (Fe3O4@SiO2-SSPy). Modified with targeted segment HA-functionalized L-cysteine ethyl ester hydrochloride (HA-Cys) via disulfide exchange reaction, the Fe3O4@SiO2-SS-HA was formed. The nanocarrier with prominent magnetic property, targeting ligand, and redox-sensitive disulfide linkages was able to specially capture MCF-7 cells with an efficiency of 92% and effectively release captured cells with an efficiency of 81.4%. Furthermore, the Fe3O4@SiO2-SS-HA could successfully be used for the capture of MCF-7 cells, and the captured cells could be diferntiated from the blood cells. Almost all of released tumor cells kept good viability and a robust proliferative capacity after being re-cultured. It is likely that the as-prepared nanocarrier will serve as a new weapon against CD44 receptor-overexpressed cancer cells.

Folic Acid-Functionalized Hybrid Photonic Barcodes for Capture and Release of Circulating Tumor Cells

Chengxin Luan,Huan Wang,Qi Han,Xiaoyan Ma,Dagan Zhang,Yueshuang Xu,Baoan Chen,Minli Li,Yuanjin Zhao

Recovery of circulating tumor cells (CTCs) from cancer patients by an efficient CTCs capture and release method can greatly increase their application in diagnostics and treatment of cancers. In this paper, we presented a folic acid (FA)-functionalized hybrid photonic barcode for capture and release of CTCs. The hybrid photonic barcodes were formed by two nano-ordered parts, poly(ethylene glycol) diacrylate (PEGDA) inverse opal structure for sustaining integrity and methacrylated gelatin (GelMA) gel filler for conjugating FA molecules to mediate cell capture. The nano-ordered structures of the hybrid photonic barcodes not only increased contact area, but also decreased steric hindrance among FA molecules. Thus, the topographic interaction between the barcodes and CTCs was greatly enhanced. In addition, GelMA gel was soft and extracellular matrix (ECM) alike, which was beneficial to decrease impairment to CTCs during the CTCs-barcode interaction as well as preserving their viability. Demonstrated by four CTCs types, Hela (epithelial tissue, folate receptor positive, FR+), A02 (bone marrow, FR+), Raji (lymphoid tissue, FR+), and A549 (epithelial tissue, folate receptor negative, FR-), FR+ CTCs could be captured efficiently with reliability and specificity. The captured cells could be controllably released with high viability just by quick trypsinization. The whole processes were simple and efficient. These features indicated that the FA-functionalized hybrid photonic barcodes were promising for full recovery of CTCs from cancer patients, which was important for diagnosis and treatment of cancer.

Reversible capture and release of circulating tumor cells on a three-dimensional conductive interface to improve cell purity for gene mutation analysis

Yi-Ke Wang,Ming Wang,Shi-Bo Cheng,Yi-Jing Chen,Cui-Wen Li,Min Xie,Wei-Hua Huang

Rare circulating tumor cells (CTCs) are always harvested with large numbers of white blood cells (WBCs) by current CTC isolation techniques, which influences the accuracy of CTC-related gene and protein analysis. Therefore, it is very urgent to develop a method for efficient isolation of CTCs with high purity. In this work, we fabricated a reversibly assembled interface by layer-by-layer assembly of biotinylated poly-(L-lysine-graft-ethylene glycol) (PLL-g-PEG-biotin), streptavidin, and CTC-specific antibody on a three-dimensional conductive scaffold, and further embedded it into a customized microchip for CTC capture. The assembled multilayers increased the roughness of the scaffold, which improved the capture efficiency. Importantly, the PLL-g-PEG-biotin covalently coupled with CTCs could be detached from the scaffold by electrostatic repulsion, and the released CTCs could be recaptured on the conductive scaffold via electrostatic attraction while the WBCs were continuously drifted away. This reversible capture and release strategy significantly improved the purity of CTCs, and pure CTCs were obtained from cancer patients’ blood samples for gene mutation assay, which will be of great significance for assisting cancer diagnosis and treatment in a noninvasive way.

Bioinspired three-dimensional hierarchical micro/nano-structured microdevice for enhanced capture and effective release of circulating tumor cells

Le Wang,Song Huang,Qin-Ying Li,Li-Yun Ma,Cong Zhang,Feng Liu,Ming Jiang,Xu Yu,Li Xu

The efficient capture and release of the circulating tumor cells (CTCs) are promising for the CTC related cancer diagnosis and subsequent analysis. However, it is a great challenge to isolate and enrich the rare CTCs from the complex peripheral blood sample, which inhibits its broad translating to the clinical applications. Although various papers reported that the nanostructure substrates can improve the capture efficiency of CTCs, few of researches constructed 3D hierarchical micro/nano-structured microdevice to enhance the efficiency of CTC capture and release with a simple manner. Herein, a bio-inspirited 3D epithelial cell adhesion molecule (EpCAM) aptamer modified rose petal derived ZnO microchip (EpCAM-RPD-ZnO-Chip) was designed to realize efficient capture and release of the CTCs. The 3D rough spherical concave surfaces on the EpCAM-RPD-ZnO-Chip guaranteed the maximum contact with the CTCs, thus improving the capture efficiency. Moreover, simple release of CTCs by dissolving ZnO under moderate acidic condition was achieved. The topographical and target molecular recognition cooperatively contributed to highly efficient capture of CTCs. The scanning electron microscope (SEM) image clearly showed that the filopodias and lamellipodias of CTCs stretched and attached to the ZnO nanomaterials. In sum, the proposed 3D EpCAM-RPD-ZnO-Chip is expected to be a promising tool for capture and release of CTCs in clinical trials, which might lay a foundation for downstream analysis and precise drug susceptibility test for the cancer treatment.
