Tissue-specific regulation of volatile emissions moves predators from flowers to attacked leaves

Asim Munawar,Yi Xu,Amr S. Abou El-Ela,Yadong Zhang,Jian Zhong,Zhiyao Mao,Xuan Chen,Han Guo,Chao Zhang,Yiqiao Sun,Zengrong Zhu,Ian T. Baldwin,Wenwu Zhou

Plant-predator mutualisms have been widely described in nature. 1 , 2 How plants fine-tune their mutualistic interactions with the predators they recruit remains poorly understood. In the wild potato ( Solanum kurtzianum ), predatory mites, Neoseiulus californicus , are recruited to flowers of undamaged plants but rapidly move downward when the herbivorous mites, Tetranychus urticae , damage leaves. This “up-down” movement within the plant corresponds to the shift of N. californicus from palynivory to carnivory, as they change from feeding on pollen to herbivores when moving between different plant organs. This up-down movement of N. californicus is mediated by the organ-specific emissions of volatile organic compounds (VOCs) in flowers and herbivory-elicited leaves. Experiments with exogenous applications, biosynthetic inhibitors, and transient RNAi revealed that salicylic acid and jasmonic acid signaling in flowers and leaves mediates both the changes in VOC emissions and the up-down movement of N. californicus . This alternating communication between flowers and leaves mediated by organ-specific VOC emissions was also found in a cultivated variety of potato, suggesting the agronomic potential of using flowers as reservoirs of natural enemies in the control of potato pests.

Toxic effects of cadmium on the growth and predation capacity of the predator Orius sauteri

Kou Jing,Zhu Zheng-Yang,Wang Su,Zhang Yu,Wang Jie,Jaworski Coline C.,Harwood James D.,Jing Lan,Desneux Nicolas,Di Ning

The heavy metal cadmium (Cd) leads to significant bottom-up effects on food chains of plants, herbivores, and predators in agroecosystems. Through the transfer and accumulation of Cd. In addition to the indirect effects of this cascading effects, predators Orius sauteri fed with artificial diets containing Cd also have a direct effect. We found that O. sauteri fed with sucrose solutions containing Cd at high concentrations of 125 mg/L and 625 mg/L significantly decreased the survival of the 3rd and 5th instar, female, and male adults (except 125 mg/L) of O . sauteri . However, the survival of O. sauteri nymphs was significantly increased when they fed with low concentrations of 1 mg/L, 5 mg/L and 25 mg/L Cd sucrose solutions. Cd stress has differing effects on the LC 50 of each age of O. sauteri , the lowest LC 50 for male adults (153 mg/L), followed by 3rd instar nymphs (192 mg/L), 5th instar nymphs (289 mg/L) and female adults (383 mg/L). It is interesting that 1 mg/L, 5 mg/L, and 25 mg/L of sucrose solutions containing Cd significantly improved the predation capacity of the nymphs of O. sauteri to the B emisia tabaci pupa. The predation ability of the male adults of O . sauteri on B . tabaci pupa were significantly improved, but significantly decreased in the female adults when they fed with 125 mg/L and 625 mg/L sucrose solutions containing Cd. This difference needs to be studied in depth in the future, controlling Cd contamination and protecting natural enemies to enhance the biological control of pests. Graphical Abstract

Visible light-regulated BiVO4-based micromotor with biomimetic ‘predator-bait’ behavior

Chen Zongchen,Jiang Jiwei,Wang Xin,Zhang Hui,Song Bo,Dong Bin

Inspired by the swarming behavior of various bacteria in nature, light-driven micromotor with collective behavior has attracted a great deal of attention, which will be a valuable tool to understand the cooperation and communication between bacterial populations and will contribute to the creation of reconfigurable microrobots and the cooperative transportation of large cargoes. However, it is still challenging to modulate the micromotors’ collective behavior based on local interaction and communication. Herein, we develop a light-powered BiVO 4 -based micromotor with self-propulsion and dynamic ‘predator-bait’ collective behavior, which is based on the photocatalytic reaction of BiVO 4 . The BiVO 4 -based micromotor has great potential in the field of organic pollutant degradation with enhanced apparent rate constant (0.076 min −1 ) which originates from the dynamic ‘predator-bait’ behavior.

Impact of Natural Microorganisms on the Removal of COD and the Cells Activity of the Chlorella sp. in Wastewater

Qingnan Sun,Xiaoping Zhang,Xin Zhang

In the treatment of wastewater containing only chemical oxygen demand (COD) byChlorellasp., the cell activity and proliferation ability ofChlorellasp. decreased with the culture time, which affected the removal of COD in wastewater. To solve these problems, theChlorellasp.–natural microorganism symbiosis system was prepared. The system was used to explore how natural microorganisms affect the cell activity and the proliferation ability ofChlorellasp. in wastewater. In the treatment of COD byChlorellasp., the removal rate of COD decreased from 45.47% to 28.88%, with a decrease in the cell activity and proliferation ability ofChlorellasp. In theChlorellasp.–natural microorganism symbiotic system, the removal rate of COD reached 45.75%. With the introduction of natural microorganisms, the circulation of CO2and O2betweenChlorellasp. and natural microorganisms promoted photosynthesis and respiration, which enhanced the cell activity ofChlorellasp. Under the condition that the dosage of natural microorganisms was between 1% and 6%, the concentration ofChlorellasp. was close to the logarithmic growth phase, which maintained the proliferation ability ofChlorellasp. At the same time, the natural microorganisms grew and proliferated in wastewater containing only COD through preying onChlorellasp.Keywords:Chlorellasp.;natural microorganisms;symbiosis system;cell activity;predation

Effects of urea on behavior and functional traits of Asiatic toad (Bufo gargarizans) tadpoles

Zhao Tian,Wang Xiaoyi,Wang Xungang,Wang Sishuo,Chen Youhua,Jiang Jianping

As one of the important contributors of biodiversity, amphibian populations are declining worldwide. Numerous factors are involved in these declines, one of them being the use of fertilizers in agriculture. This is especially true for tadpoles which can live in the fertilizer-polluted farmland water bodies until metamorphosis. The present study aimed to assess the effects of urea (CH 4 N 2 O), as one of the most economical and effective fertilizers, on the anti-predator behavior and intraspecific functional trait variability of Asiatic toad ( Bufo gargarizans ) tadpoles. Based on published literatures and the field observation of urea concentrations in China, glass beakers with a gradient of urea concentrations (0, 200, 400, 600, and 1200 mg/L) were prepared, with 10 tadpoles placed in each glass beaker. Each treatment was replicated three times. Mosquito fish ( Gambusia affini s) cues were used as the predator disturbance, and three main functional traits (body mass, trunk bending shape, and eye position) were selected. Our results revealed that tadpoles activity levels decreased when exposed to urea as well as to mosquito fish cues. However, urea exposure did not alter the anti-predator behaviors of tadpoles. Additionally, we found that increasing urea concentrations might modify some functional traits of tadpoles. Importantly, urea disturbance decreased tadpoles intraspecific functional trait variability. (Functional similarity increased between developmental stages.) Given that functional similarity between developmental stages could potentially increase intraspecific competition, urea could indirectly reduce tadpoles survival by decreasing intraspecific traits variability.

Predation Risk Effects of Lady Beetle Menochilus sexmaculatus (Fabricius) on the Melon Aphid, Aphis gossypii Glover

Xingming Lin,Xiangxin Cui,Jihong Tang,Jiawei Zhu,Jinhua Li

Simple SummaryBiological control is an important method for pest management. Natural enemies not only prey on pests directly but also release predation risk cues to pests, causing physiological and behavioral changes in them, reducing their fitness, and ultimately leading to a decrease in their population.Aphis gossypiiis an important insect pest of melon and many other crops worldwide. This study examined the predation risk effects ofMenochilus sexmaculatus, which is abundant in agricultural areas of southern China, onA. gossypii. The results showed that the presence ofM. sexmaculatussignificantly altered the host preference of the aphids. The longevity of the aphids was shortened, and the reproductive period and offspring production were decreased significantly after exposure toM. sexmaculatusfor 24 h. The enzyme activities of SOD and CAT were increased, and the content of MDA in aphids also showed an increase under the predation risk ofM. sexmaculatus. Through a combined GC-MS and behavioral response experiment, the results showed that alkane compounds may mediate the predation risk. The results will offer an alternative strategy for utilizing these alkanes as repellents to manage aphids.AbstractPredation risk posed by natural enemies can alter pest performance. In our previous study, we foundMenochilus sexmaculatusprovides risk cues to melon aphids, resulting in increased numbers of winged aphids. However, the effects of predation risk on multiple traits including behavior, physiology, growth rate, and reproductive capacity of pests are not clear. This study examined the effects of predation risk on host preference, the activities of two important defense enzymes (CAT and SOD), longevity, and offspring production. The Y-tube trial results showed that the risk ofM. sexmaculatussignificantly altered the host preference of the aphids, leading to avoidance behavior. When exposed toM. sexmaculatusfor a long period (24 h), the reproductive period and offspring production were significantly decreased, and adult longevity was significantly shortened. The defense enzyme activities of SOD and CAT, as well as the MDA content (which is considered a marker of oxidative stress and cellular damage) in the aphids, significantly increased underM. sexmaculatusrisk. The compounds ofM. sexmaculatusextracted with n-hexane and volatile compounds collected with HS-SPME were analyzed by GC-MS, and when combined with the behavior response experiment, the results showed that the alkane compoundsn-henicosane,n-docosane,n-tricosane,n-pentacosane, andn-hentriacontane may contribute to the impact of predation risk. The results will be helpful in the comprehensive evaluation of the ability of lady beetles to affect the aphid population, and provide new ideas for using these compounds in aphid control. Keywords:Menochilus sexmaculatus;Aphis gossypii;predation risk;host preference;defense enzyme activities;fitness

Performance of Daphnia simultaneously exposed to nitrite and predation risk: Reduced nitrite tolerance and aggravated predation-induced miniaturization

Lu Zhang,Tian Xia,Qi Liu,Lei Gu,Yunfei Sun,Zhou Yang

Nitrogenous pollutants derived from human activities not only pose direct risk on aquatic organisms but may also indirectly endanger the stability of interspecific relations. To date, the effects of the nitrogen-containing pollutants on the induced defense remain unclear. Here, we aim to investigate the induced defense of an aquatic keystone species , Daphnia pulex, which responds to predation risk under nitrite pollution at environmentally relevant concentrations and simultaneously evaluate the effects of their induced defenses on nitrite tolerance. Results showed that increasing nitrite significantly reduced the survival time of D. pulex and posed severe reproductive toxicity, consequently reducing the offspring and broods. In the morphological defensive responses, early nitrite exposure interfered with the spine elongation, but the relative spine length induced by the predation risk was unaffected by the nitrite concentrations with exposure time prolonged, although high-dose nitrite inhibited the spine elongation and the increase of the body size. The integration of biomarker response index analyses further indicated that the reproductive capacity was more seriously impaired than the morphology and the survival. Moreover, the sensitivity analyses of growth and reproduction indicated that predation risk significantly reduced Daphnia's tolerance to nitrite. Conclusively, these findings highlight that long-term nitrite exposure exacerbates the predator-induced miniaturization of zooplanktons, and predation risk also reduces their tolerance to nitrite, which provides new insights into the performance changes of zooplanktons exposed to pollutants under predation risk and the vulnerability of predator–prey interspecific relationships in polluted environments.

Induction of trap formation in nematode‐trapping fungi by bacteria‐released ammonia

Su H.N.,Xu Y.Y.,Wang X.,Zhang K.Q.,Li G.H.

A total of 11 bacterial strains were assayed for bacteria‐induced trap formation in the nematode‐trapping fungus Arthrobotrys oligospora YMF1·01883 with two‐compartmented Petri dish. These strains were identified on the basis of their 16S rRNA gene sequences. Volatile organic compounds (VOCs) of eight isolates were extracted using solid‐phase micro‐extraction (SPME) and their structures were identified based on gas chromatography‐mass spectrometry (GC‐MS). At the same time, all isolates were used for quantitative measurement of ammonia by the indophenol blue method. The effects of pure commercial compounds on inducement of trap formation in A. oligospora were tested. Taken together, results demonstrated that the predominant bacterial volatile compound inducing trap formation was ammonia. Meanwhile, ammonia also played a role in other nematode‐trapping fungi, including Arthrobotrys guizhouensis YMF1·00014, producing adhesive nets; Dactylellina phymatopaga YMF1·01474, producing adhesive knobs; Dactylellina cionopaga YMF1·01472, producing adhesive columns and Drechslerella brochopaga YMF1·01829, producing constricting rings.

Functionalization of slow-release fertilizers and “passive predation microplastics” mechanism for polylactic acid composites

Haibin Ji,Somia Yassin Hussain Abdalkarim,Ruoxi Nie,Hou-Yong Yu

The large-scale use of nonrenewable plastic mulch has resulted in serious agricultural health pollution. Biobased plastic materials with degradable properties offer attractive sustainable alternatives, but the shortcomings of their properties are slow degradation and extremely monofunctional, making their full-scale promotion still challenging. This work proposes a novel functionalized strategy for the multifunctionality empowerment of bio-based PLA materials for environmental protection and crop yield enhancement. This strategy relies on the effective combination of amine-modified lignin hybrid filler (L-U) and PLA matrix, which generates a hydrogen bonding network that can effectively synergize to improve crystalline ability and mechanical properties of PLA. In addition, L-U can effectively increase the PLA matrix degradation sites and improve its hydrophilicity to enhance its degradation properties further. In agricultural mulch, the functionalized materials (PLU) have high water insulation, biodegradability, and excellent slow release of nutrients, which can effectively improve the utilization of fertilizers and reduce the dependence of industrial fertilizers in agricultural systems. In addition, the “passive predation” mode of plants on bio-based polyester-based micron-sized microplastics was explored, which can better understand the adsorption characteristics of biopolyester-based micron-sized microplastics on plants, thus reducing the potential risk to food yield, quality, and safety.

Impact of atrazine on the dynamic response of Daphnia pulex populations to fish predation risk

Qin Shanshan,Xia Tian,Li Gongzheng,Gu Lei,Sun Yunfei,Yang Zhou

Herbicide pollution is persistent, which not only has a negative impact on individual organisms, but also may alter population dynamics and stability of interspecific relationships. Cladocerans, an important part of zooplankton, are often simultaneously exposed to environmental pollutants and predation risk in the aquatic environment. To evaluate the combined effects of atrazine and fish predation risk on the population traits of cladocerans, we exposed Daphnia pulex to different concentrations of atrazine (0, 0.05, 0.10, and 1.0 mg L−1) with or without fish (Rhodeus ocellatus) kairomone, recorded the key population traits, and fitted Gaussian model to population dynamics. Results showed that fish kairomone increased the population density at the end of the experiment and resting eggs production, and tended to decrease the total biomass and the average dry weight per individual of D. pulex. Atrazine reduced the total biomass, the average dry weight per individual, and resting eggs production of D. pulex populations. Atrazine also decreased the population density at the end of the experiment of D. pulex in fish kairomone treatment, and attenuated the promoting effect of fish kairomone on resting eggs production and the reduction of the total biomass. The findings highlighted the importance of considering the combined impact of environmental pollutants and predation risks on zooplankton populations.

Safety of Bacillus thuringiensis Cry1Ah and Vip3Aa toxins for the predatory stink bug Arma custos (Hemiptera: Pentatomidae)

Limei Cai,Xiaoming Liu,Zhiqiang Tian,J.P. Michaud,Zhongjian Shen,Zhen Li,Songdou Zhang,Xiaoxia Liu

The widespread adoption of Bt crops expressing insecticidal proteins derived from Bacillus thuringiensis has created a need to assess the potential effects of these toxins on non-target organisms, especially species such as Arma custos , a generalist predator that provides important biological control services in many field crops in Asia. Direct dietary exposure of A. custos to Cry1Ah and Vip3Aa proteins produced no adverse effects on life history traits, despite continuous exposure throughout development and early adult life to concentrations significantly higher than the Bt protein concentration likely encountered by A.custos in the field, even when feeding directly on Bt plants. Enzyme-linked immunosorbent assay confirmed the presence of Bt proteins in A. custos midguts, but quantitative real-time PCR analysis of 12 genes associated with detoxification, antioxidative responses, immune responses, and metabolism revealed no significant changes in expression in adult bugs. Indirect exposure to these toxins via consumption of intoxicated prey, larvae of Helicoverpa armigera (Hübner), likewise produced no negative impacts on survival, development, adult weight, or female fecundity in either the F0 (exposed) or F1 (unexposed) generation, but female fresh weight was reduced in the F0 generation by the Cry1Ah (50 μg/g) treatment. Finally, a competitive binding assay with labelled protein and a ligand blotting assay both demonstrated that the Cry1Ah protein could not bind to receptors on the midgut brush border membrane vesicles (BBMVs) of A. custos adults. Therefore, we conclude that Cry1Ah and Vip3Aa proteins are unlikely to have significant negative effects on A. custos populations if employed as plant-incorporated protectants in field crops.

Mandelonitrile produced by commensal bacteria protects the Colorado potato beetle against predation

Kang Wei-Nan,Pan Yang,Liao Lan-Lan,Wu Yi-Kuan,Zhang Xiao-Qing,Jin Lin,Fu Kai-Yun,Guo Wen-Chao,Li Guo-Qing

Larvae and adults of the Colorado potato beetle ( Leptinotarsa decemlineata ), a major pest of potato crops, display conspicuous coloration to advertise their toxicity to predators. However, the identity of the toxic compounds remains unclear. Here, we show that larvae and adults release toxic hydrogen cyanide (HCN) from the degradation of mandelonitrile and other cyano-compounds, which are produced by commensal bacteria. We isolate the bacterium Proteus vulgaris Ld01 from the insect’s gut, and show that it produces HCN and a mandelonitrile-producing cyanoglucoside, amygdalin. Knockout of a gene ( hcnB ) encoding putative hydrogen cyanide synthase impairs HCN production in P. vulgaris Ld01. Antibiotic treatment of larvae, to eliminate their commensal bacteria, leads to a substantial reduction of HCN emission in larvae and adults. HCN release by bacteria-deprived beetles can be restored by addition of mandelonitrile or by re-infection with P. vulgari s Ld01 (but not with its ∆ hcnB1 or ∆ hcnB2  mutants). Finally, we use dual-choice experiments to show that domestic chicks prefer to eat bacteria-deprived larvae over control larvae, larvae re-colonized with P. vulgaris Ld01, or mandelonitrile-injected larvae. Our work highlights the role of the beetle’s intestinal bacteria in the production of the cyanoglucoside amygdalin and its derived metabolites, including mandelonitrile and HCN, which protect the insect from predation.
