Optimization of the Ethanol Recycling Reflux Extraction Process for Saponins Using a Design Space Approach

Xingchu Gong,Ying Zhang,Jianyang Pan,Haibin Qu

A solvent recycling reflux extraction process for Panax notoginseng was optimized using a design space approach to improve the batch-to-batch consistency of the extract. Saponin yields, total saponin purity, and pigment yield were defined as the process critical quality attributes (CQAs). Ethanol content, extraction time, and the ratio of the recycling ethanol flow rate and initial solvent volume in the extraction tank (RES) were identified as the critical process parameters (CPPs) via quantitative risk assessment. Box-Behnken design experiments were performed. Quadratic models between CPPs and process CQAs were developed, with determination coefficients higher than 0.88. As the ethanol concentration decreases, saponin yields first increase and then decrease. A longer extraction time leads to higher yields of the ginsenosides Rb 1 and Rd. The total saponin purity increases as the ethanol concentration increases. The pigment yield increases as the ethanol concentration decreases or extraction time increases. The design space was calculated using a Monte-Carlo simulation method with an acceptable probability of 0.90. Normal operation ranges to attain process CQA criteria with a probability of more than 0.914 are recommended as follows: ethanol content of 79–82%, extraction time of 6.1–7.1 h, and RES of 0.039–0.040 min −1 . Most of the results of the verification experiments agreed well with the predictions. The verification experiment results showed that the selection of proper operating ethanol content, extraction time, and RES within the design space can ensure that the CQA criteria are met.

Development of Pretreatment Approaches for Authentic Representation of Tea Infusion Aroma

Mingming Zhang,Zhihui Feng,Fang Wang,Jianxin Chen,Yifan Li,Yuqiong Chen,Junfeng Yin

Appropriate aroma extraction methods are crucial prerequisites for accurately and objectively characterizing the authentic aroma profile of samples. Purified water and ionized water were used as brewing water, and the effects of different tea-to-water ratios, extraction temperatures, and extraction times on the aroma authenticity and component enrichment of tea infusions were compared. The conditions of a tea-to-water ratio of 1 g:10 mL, extraction at 30 °C for 30 or 45 min were identified as the optimal parameter range, which could maximize the enrichment of aroma while maintaining fidelity. The cosine value of the aroma attribute scores between the optimal parameter set and the control group (tea brewed at 1 g:10 mL ratio for 4 min) exceeded 0.979, and the correlation coefficient surpassed 0.828. Test evaluation results indicate the method had good reproducibility and effectively highlighted the differential impacts of ionic content in brewing water on tea aroma constituents. This approach effectively solved the problem of sensory distortion caused by conventional high-temperature and long-duration extraction, enabling precise analysis of how water quality authentically influences tea infusion aroma characteristics.

Optimization of the ultrasound-assisted extraction for phenolic compounds content and antioxidant activity of Cortex fraxini using response surface methodology

Huang Dan,Wang Jing,Li Fanghua,Xie Min,Qu Qingli,Wang Yimin,Sun Wenjuan,Wu Caie,Xu Wenhui,Xiong Ranhua,Ding Yichun,Yang Anquan,Huang Chaobo

In this study, for the first time, optimized phenolic compounds and antioxidant properties were derived from the extraction process of Cortex fraxini (CFR) using response surface methodology. Four factors and three levels of Box-Behnken (BBD) were used to study the extraction time (X 1 ), extraction temperature (X 2 ), ethanol concentration (X 3 ), and solvent-material ratio (X 4 ). The analysis of variance (ANOVA) revealed that temperature and solvent-material ratio had a relatively large effect on the response. The optimal combination of phenolic compounds and antioxidant activity was obtained with the optimal conditions of X 1  = 1.629 h, X 2  = 53.367 ℃, X 3  = 60.971% (v/v), and X 4  = 45.582 mL/g under ultrasonic assisted extraction (UAE). The experimental parameters were consistent with the predicted values. Caffeic acid, erucic acid, ferulic acid and p-hydroxycinnamic acid and other 19 phenolic acids were characterized by high performance liquid chromatography-mass spectrometry (HPLC–MS). The results of this study indicate that optimization of the extraction process is essential for the accurately determined measurement of phenolic components and antioxidants in CFR for its industrial applications.

Extraction of DNA from complex biological sample matrices using guanidinium ionic liquid modified magnetic nanocomposites

Mei Liu,Xueqin Ding,Xuelian Wang,Jianzhong Li,Huansheng Yang,Yulong Yin

A series of guanidinium ionic liquid modified magnetic chitosan/graphene oxide (GIL-MCGO) nanocomposites have been prepared for DNA extraction via magnetic solid-phase extraction technology. These nanocomposites are of only 20 nanometers in diameter. Single stranded DNA or DNA sodium salts that were absorbed by GIL-MCGO could be quickly collected by an external magnet and extracted. The DNA extraction efficiency of 11 GIL-MCGO nanocomposites was evaluated using NanoDrop. Factors that could impact the DNA extraction process, such as pH, temperature, extraction time, and ionic strength were systematically investigated via single-factor experimental analysis. Under the optimum extraction conditions, a maximum DNA extraction capacity of 233.0 ± 0.4 mg g−1 of GIL-MCGO nanocomposite was achieved. The solid phase extraction method based on GIL-MCGO nanocomposites has been demonstrated with the extraction of DNA from a series of complex sample matrices, including single stranded DNA samples, salmon sperm DNA sodium salt, human whole blood and E. coli cell lysate. The DNA extracted by using the GIL-MCGO nanocomposites are well suitable for PCR amplifications. In addition, an initial study on the interaction between GIL-MCGO and DNA was conducted: the preference of GIL-MCGO on DNA absorption with varying base composition was tested. Only a slight loss in the DNA extraction efficiency of GIL-MCGO was observed after four extraction–desorption cycles, proving excellent regeneration performance and recyclability of the GIL-MCGO nanocomposites in the DNA extraction process.

Highly Selective and Considerable Subcritical Butane Extraction to Separate Abamectin in Green Tea

Yating Zhang,Lingbiao Gu,Fei Wang,Lingjun Kong,Huili Pang,Guangyong Qin

We specially carried out the subcritical butane extraction to separate abamectin from tea leaves. Four parameters, such as extraction temperature, extraction time, number of extraction cycles, and solid-liquid ratio were studied and optimized through the response surface methodology with design matrix developed by Box–Behnken. Seventeen experiments with three various factors and three variable levels were employed to investigate the effect of these parameters on the extraction of abamectin. Besides, catechins, theanine, caffeine, and aroma components were determined by both high-performance liquid chromatography and gas chromatography–mass spectrometry to evaluate the tea quality before and after the extraction. The results showed that the extraction temperature was the uppermost parameter compared with others. The optimal extraction conditions selected as follows: extraction temperature, 42°C; number of extraction cycles and extraction time, 1 and 30 min, respectively; and solid–liquid ratio, 1:10. Based on the above study, the separation efficiency of abamectin was up to 93.95%. It is notable that there has a quite low loss rate, including the negligible damage of aroma components, the bits reduce of catechins within the range of 0.7%–13.1%, and a handful lessen of caffeine and theanine of 1.81% and 2.6%, respectively. The proposed method suggested subcritical butane possesses solubility for lipid–soluble pesticides, and since most of the pesticides are attached to the surfaces of tea, thus the as-applied method was successfully effective to separate abamectin because of the so practical and promising method.
