Phytochemical Analysis, Antioxidant and Analgesic Activities of Incarvillea compacta Maxim from the Tibetan Plateau

Jiajia Guo,Dan Zhang,Chao Yu,Ling Yao,Zhuo Chen,Yanduo Tao,Weiguo Cao

Incarvillea compactaMaxim is a traditional Tibetan plant widely used to treat rheumatic pain and bruises. We conducted qualitative analyses by liquid chromatography-mass spectrometry and quantitative analyses of the total phenols, flavonoids, and alkaloids content of different extracts ofI. compactaMaxim. Antioxidant and analgesic activity were analyzed. The results showed that the methanol extract had the highest content of the various ingredients. A total of 25 constituents were identified, of which compounds1–23were found for the first time in this plant. The water extract had the highest capacity to clear free radicals in the antioxidant test. The water extract had dose-dependent analgesic effects in the first and second phase in a formalin test. The latency of pain from a hot-plate test was augmented by the water extract when the dose was greater than or equal to 30 g/kg. The water extract significantly decreased the amount of writhing in a dose-dependent manner compared with the control group in the acetic acid-induced writhing test. These results showed thatI. compactaMaxim is a new antioxidant and analgesic agent, and this study provides information on its ingredients for further study.Keywords:LC-MS;anti-oxidation;analgesia;formalin test;Tibetan plant

A comprehensive study of defatted Idesia polycarpa fruit: Phenolic profile and its relation with anti-inflammatory activity in seed, pulp, and whole fruit

Xiaohua Nie,Mengqing Chen,Chengyu Jin,Xiaoyu Zhou,Ningxiang Yu,Yuanchao Lu,Xianghe Meng

Defatted seed, pulp and whole fruit of Idesia Polycarpa Maxin. ( I. Polycarpa ) are the by-products of the oil extraction process, but have attracted little attention as natural sources of bioactive phenolics. This study aimed to clarify the profiles of phenolic compounds in the extracts from these by-products and their contribution to the anti-inflammatory activity. Twenty-one phenolic compounds were identified, belonging mainly to salicinoid analogues and p -coumaroyl derivatives. Idescarpin was the predominant phenolics in all extracts, especially in seed extract. Nearly half the p -coumaroyl derivatives presented relatively high content in the extracts of pulp and whole fruit. All extracts inhibited the production of nitric oxide (NO) and the mRNA expressions of inducible nitric oxide synthase (iNOS), tumor necrosis factor (TNF-α), and interleukin (IL)-6; and suppressed the accumulation of reactive oxygen species (ROS) along with the restoration of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities. Seed extract exerted better potential than the pulp and whole fruit extracts. Furthermore, correlation analysis demonstrated that idescarpin and idesin were the crucial phenolics against inflammation, whereas some p -coumaroyl derivatives contributed to regulating oxidative stress. These findings suggested that phenolic extracts from defatted I. Polycarpa fruit and its parts, especially seed, could be utilized as inflammation inhibitors in the functional food and cosmetics industries.

Inhibitory effect and mechanism of various fruit extracts on the formation of heterocyclic aromatic amines and flavor changes in roast large yellow croaker (Pseudosciaena crocea)

Meng Li,Songyi Lin,Ruichun Wang,Duo Gao,Zhijie Bao,Dong Chen,Yue Tang,Na Sun,Simin Zhang

The large yellow croaker ( Pseudosciaena crocea ) is one of the most commercially important marine fish in China and is a fixture at Chinese tables. Herein, we determined the formation of heterocyclic aromatic amines (HAAs) in roasted yellow croaker and evaluated the inhibitory effect of natural fruits extracts (blueberry extract, acerola cherry extract and grape seed extract) on the formation of HAAs in roast yellow croaker. All the three fruit extracts treatments minimize the amount of HAAs and the addition of blueberries extracts is the most effective approach on reducing the total HAAs content, which caused a signification reduction of 94.85% and 71.15% of the Norhaman and PhIP content, respectively, led IQ, 8-MeIQX, 4,8-DiMeIQX below the detected limitation, compared to the control group. Furthermore, antioxidant index of fruits extracts, precursors (free amino acids, creatine, creatinine and glucose) of HAAs, lipid and protein oxidation, flavor changes are evaluated to determine the mechanism and physical/chemical changes of fruit extracts on the formation of HAAs. This research develops a new method to minimize the amount of HAAs, which is helpful to food manufacturers for the production of healthier yellow croaker products and may be applicable to various meat systems.

Evaluation of chitosan coatings enriched with turmeric and green tea extracts on postharvest preservation of strawberries

Chun Yang,Jia-Hao Lu,Mei-Ting Xu,Xin-Chi Shi,Zhi-Wei Song,Tang-Min Chen,Daniela D. Herrera-Balandrano,Yun-Jiao Zhang,Pedro Laborda,Mahdi Shahriar,Su-Yan Wang

Strawberries are a widely consumed fruit that contains a variety of antioxidant metabolites. However, postharvest strawberries are highly perishable and susceptible to a variety of fungal pathogens. In this study, a chitosan-based coating containing turmeric (TU) and green tea (GT) extracts was developed and evaluated for the preservation of strawberries. Both TU and GT extracts contained high phenolic contents (>5.8 g kg −1 dry weight), but only TU extract showed strong antifungal activity. SEM observations revealed that high concentrations of extract (3–5% v/v ) caused the formation of agglomerates within the films, reduced elongation at break and increased tensile strength. The postharvest treatment of strawberries with chitosan coating containing TU extract inhibited the proliferation of Botrytis cinerea during 7 d of storage at 20 °C, whereas the coating containing GT extract extended the antioxidant properties of postharvest strawberries from 4 to 8 d at 20 °C. Sensory analysis indicated that the coatings did not significantly affect flavor and taste. Overall, TU extract was used for the first time for food packaging purposes and proved to be a great complement to the antioxidant properties of GT to extend the shelf-life of postharvest strawberries.

Phytochemical profiles, antioxidant activities, and synergistic antiproliferative effects of blueberry and apple peel extracts

Wen Xia,Bingbing Song,Tong Li,Rui Hai Liu

BACKGROUND Blueberries and apples exhibit favorable bioactivity and health benefits as a result of their rich phytochemicals. Natural phytochemicals exist in complex forms, but there are few reports on whether have additive, synergistic or antagonistic effects between different phytochemicals. The present study aimed to elucidate the synergistic effects of blueberry extract (BE) and apple peel extract (APE) together with respect to inhibiting the proliferation of HepG2 liver cancer cells. Meanwhile, phytochemical characterization of BE and APE was conducted by HPLC, and total antioxidant activity was determined via a cellular antioxidant activity assay, oxygen radical absorption capacity assay and peroxy radical scavenging capacity assay. RESULTS The results showed that BE and APE were rich in phytochemicals and had potent antioxidant activities, which synergistically inhibited cell proliferation. In the bilateral combination, the dose reduction index value increased by two-fold, and the combination index value at 95% inhibition was less than 1. Additionally, BE + APE supplementation could promote the expression levels of p53 and c-myc genes. In conclusion, the BE and APE had strong antioxidant activity and exhibited synergistic inhibition against proliferation of HepG2 cells. CONCLUSION The present study can provide a theoretical basis for the synergistic effect of different phytochemicals in health care. © 2023 Society of Chemical Industry.

The effect of Hippophae rhamnoides L and Alkekengi officinarum Moench fruits extracts on the Changes of acrylamide content in roasted chicken breast during storage

Linze Liu,Yawen Deng,Hangqi Ren,Ying Ren,Taizheng Tang,Eerdun bayaer,Shaobo Li,Tungalag Dong,Xueyan Yun

Plant extracts with antioxidant activity are frequently employed to inhibit acrylamide (AM) production, but there has been limited focus on their inhibitory effects on AM during the extended storage of foods. In this study, impregnation solutions were prepared using Hippophae rhamnoides L (Hp) and Alkekengi officinarum Moench (Ak) fruits extracts to investigate their protective effects on roasted chicken breasts (RCB) during processing and storage (vacuum packed at 4°C and stored at 25°C for 90 days). In a 90-day storage experiment with RCB, these extracts also slowed the increase of AM in RCB. However, these extracts exhibit inadequate protective effects on RCB during long-term storage. It is needed to develop new methods for monitoring and inhibiting AM formation in RCB.

Caffeine is responsible for the bloodglucose-lowering effects of green tea and Puer tea extractsin BALB/c mice

Chong-Ye FANG,Xuan-Jun WANG,Ye-Wei HUANG,Shu-Mei HAO,Jun SHENG

The present study was designed to determine the effects of Puer tea and green tea on blood glucose level. Male BALB/c mice were administered green tea extract (GTE) or Puer tea extract (PTE), either intragastrically or in their drinking water. The major components of these teas are epigallocatechin gallate (EGCG) and caffeine, respectively. Blood glucose measurement results showed that mice fed intragastrically or mice that drank GTE, PTE or caffeine showed significantly lower blood glucose levels compared to the control group. However, EGCG exhibited no influence on the blood glucose levels. When caffeine was eliminated from the GTE and PTE, the effect on the blood glucose levels was abolished, but the effect was recovered when caffeine was re-introduced into the extracts. Evaluation of hematological and biochemical indices at the time of the greatest caffeine-induced decrease in blood glucose levels showed that the effect of caffeine was specific. Microarray analyses were performed in 3T3-L1 preadipocytes and mature adipocytes treated with 0.1 mg·mL −1 caffeine to identify factors that might be involved in the mechanisms underlying these effects. The results showed that few genes were changed after caffeine treatment in adipocytes, and of them only phospholipid transfer protein (PLTP) may be ralated to blood glucose. In conclusion, this study indicates that caffeine may be the key constituent of tea that decreases blood glucose levels, and it may be used to treat type 2 diabetes.

Determination of 1-aminocyclopropane-1-carboxylic Acid in Apple and Peach Extracts by High Performance Liquid Chromatography Coupled to a Fluorescence Detector

Meirong Liu,Shuhua Zhu,Jie Zhou

A new, sensitive, and simple HPLC method was described for the determination of 1-aminocyclopropane-1-carboxylic acid (ACC) in apple and peach extracts. The method was based on the derivatization of ACC with fluorescamine in borate buffer systems of pH 8.0 to yield a highly fluorescent product. The experimental parameters affecting the derivatization reaction efficiency were optimized by fluorimetric analysis. Under optimum derivatization conditions, the derivative product of ACC in apple and peach extracts without extra purification was successfully chromatographed on a C-18 column by HPLC coupled to fluorescence detection. The derivative product of ACC with fluorescamine could be well separated from other concomitant substances or their derivatives that might interfere with the determination of ACC. The linearity of ACC was measured in the range of 23.82–238.82 µg · L−1 with a good correlation coefficient of 0.9997. Based on signal-to-noise ratio of 3, a low detection limit of 5.0 µg · L−1 could be reached. The proposed method was applied successfully to the determination of ACC in the crude apple and peach extracts without extra purification with low RSDs of 0.19–1.9% and good recoveries of 90.89–104.4%. The sensitive HPLC quantitative method is of great significance for the investigations of ACC metabolism in plants.

Jasmine (Jasminum grandiflorum) Flower Extracts Ameliorate Tetradecanoylphorbol Acetate Induced Ear Edema in Mice

Dongli Li,Xiaodan Tang,Chang Liu,Huifang Li,Shuzhen Li,Shili Sun,Xi Zheng,Panpan Wu,Xuetao Xu,Kun Zhang,Hang Ma

Published data from in vitro assays support the anti-inflammatory effects of jasmine (Jasminum grandiflorum Linn.) but limited studies are reported in animal models. Herein, the anti-inflammatory effects of jasmine flower extracts (JFEs) including ethanol extract (JF-EE), petroleum ether extract (JF-PEE), ethyl acetate extract (JF-EAE), and n-butanol extract (JF-BE) were evaluated in a mouse ear edema model. Acute mouse ear skin inflammation was induced by tetradecanoylphorbol acetate (TPA; 125 µg/mL) and then treated with JFEs (100 mg/mL) or dexamethasone (DEX; 6.25 mg/mL; as a positive control). Jasmine flower extracts alleviated ear edema by reducing TPA-increased ear thickness and ear weight by 30.8% to 64.1% and 24.0% to 47.1%, respectively, whereas DEX showed comparable activity (by 71.8% and 49.1%, respectively). Their anti-inflammatory effects were supported by data from the immunohistochemical assays. Jasmine flower extracts reduced the inflammatory cells (from 5.5- to 9.5-fold) and the expressions of inflammation related enzymes including cyclooxygenase-2 and inhibitor of kappa-B kinase (from 1.9- to 2.8-fold and from 7.1- to 11.0-fold, respectively). Findings from this study showed that JFEs were able to ameliorate TPA-induced mouse skin inflammation. However, future studies on the underlying mechanisms of jasmine flower’s anti-inflammatory effects are warranted.

Comparison of the protective effect of black and white mulberry against ethyl carbamate-induced cytotoxicity and oxidative damage

Yuting Li,Tao Bao,Wei Chen

Increasing evidence indicates that crude extracts derived from mulberry confer protection against oxidative stress. However, the antioxidant capacity of mulberry extract among different cultivars remains elusive. The main objective of the present study was therefore to investigate the effect of black mulberry extract (BMB) and white mulberry extract (WMB) on ethyl carbamate (EC)-induced cytotoxicity and oxidative stress. This study showed that the contents of total phenolics, total flavonoids, total procyanidins, cyanidin- 3 - O -glucoside, cyanidin- 3 - O -rutinoside and pelargonidin- 3 - O -glucoside of BMB were higher than that of WMB. Moreover, our results showed that phenolics-abundant BMB was stronger than WMB in scavenging ABTS and DPPH free radicals. BMB was more effective in ameliorating EC-induced cytotoxicity by inhibiting excessive ROS generation, suppressing mitochondrial dysfunction and increasing GSH concentration in HepG2 cells than WMB. Taken together, our study revealed that BMB afforded better protection against EC-caused cytotoxicity than WMB.

Molecular Characterization of Lignite Extracts of Methanol and Carbon Disulfide/N-Methyl-2-pyrrolidone by High-Resolution Mass Spectrometry

Lulu Yan,Yahe Zhang,Hongyu Yan,Yu Fan,Xuxia Liu,Chao Ma,Quan Shi

CS2/N-methyl-2-pyrrolidone (NMP) could extract much more substance from coals than any other solvents. Investigation on the molecular composition of CS2/NMP extracts from lignite is significant for the understanding of high extraction yield and the clean utilization of coal. The methanol-soluble portions from lignites and CS2/NMP extracts of lignites were characterized by electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry. The yield of CS2/NMP extracts from lignite was quite higher than that of methanol extracts. Furthermore, the yield of methanol-soluble portions from CS2/NMP extracts was far more than that of methanol extracts from lignite. At the level of molecular composition, the relative content of heteroatom compounds with more oxygen atoms, longer side chain, and higher condensation in the CS2/NMP extract was also higher than that in the methanol extract. Despite great difference in the yield and the relative content of components, the distributions of species, molecular weight, carbon number, and double-bond equivalent were similar to those of most organic molecules for the methanol extract and methanol-soluble portions from the CS2/NMP extract. These phenomena suggested that organic molecules with similar structure but different composition, nonuniformly distributed in the coal matrix, were released more in the CS2/NMP extract compared to the single methanol extract.

Protective Effects of Different Molecular Weights of Purslane (Portulaca oleracea L.) Aqueous Extract on DSS-Induced Ulcerative Colitis in Mice

Ke Ning,Yameng Duan,Weiwei Tong,Yue Chen,Qinghui Zhang,Qiuhong Xie,Hongyu Xiang

Purslane, a common wild vegetable, contains active substances with various biological functions. However, its effects have been under-investigated in ulcerative colitis (UC). Therefore, this study investigated the therapeutic effects of purslane macromolecular (POEM) and small molecular extracts (POES) on dextran sulfate sodium (DSS)-induced UC in mice. Membrane separation was used to obtain extracts of different molecular weights, and their compositional differences were compared using liquid chromatography–mass spectrometry (LC/MS). POEM contained more proteins and polysaccharides, whereas POES contained more organic acids and alkaloids. These differences in composition were directly responsible for the different degrees of remission of the alleviated UC in model mice. POEM may alleviate UC by regulating the antioxidant capacity and the gut microbiota, whereas the major alleviatory effect of POES was primarily related to the regulation of antioxidant capacity. The POEM and POES effects identified in this study provide a theoretical basis for the development of purslane as a functional food. Keywords:ulcerative colitis;macromolecular extract;purslane;oxidative stress;small molecular extract;gut microbiota;Portulaca oleraceaL.
