Optimization of ultrasonic extraction of Lycium barbarum polysaccharides using response surface methodology

Jitian Song,Shi Dongqi,Su Hang,Feng Yongxia,Tian Wei

Ultrasonic extraction was a new development method to achieve high-efficiency extraction of Lycium barbarum polysaccharides instead of hot water extraction. In this paper, the single factor method combined with the box Behnken design of response surface method was used to study the influence of extraction time, extraction temperature, material liquid ratio and ultrasonic power on the extraction rate of L . barbarum polysaccharide. The results indicated that the best extraction rate of L . barbarum polysaccharide was 12.54 ± 0.12% under the conditions of 80 min for extraction time, 73 °C for extraction temperature, 1 g:38 mL for material to liquid ratio, and 185 W for ultrasonic power. Under the same operating conditions, the yield of L . barbarum polysaccharide using ultrasonic extraction was 83.3%, which was higher than that hot water extraction. Moreover, the extraction time of ultrasonic extraction was only 47% of that using hot water extraction. This suggested that there was great potentials of using ultrasonic extraction in the realization of high-efficiency extraction of L. barbarum polysaccharide. The results of this study could also provide a theoretical basis for the coupling of ultrasonic extraction and ultrasonic concentration process to develop the integrated equipment of both ultrasonic extraction and ultrasonic concentration.

Feasibility analysis of extracting and purifying 4-ethylguaiacol using the intermediate product of the reaction between 4-ethylguaiacol and Ca2+ as the extracting agent

Weiyan Luo,Yixin Chen,Ping Hu,Wei Ruan,Yueyuan Ye,Zhifeng Zheng,Shuirong Li,Dechao Wang,Duo Wang

A heterogeneous extracting route of 4-ethylguaiacol is constructed using the intermediate of the reaction between 4-ethylguaiacol and Ca 2+ as the extracting agent. In this work, 4-ethylguaiacol with different concentrations (70, 80, 90, 95%) is prepared using 40 v/v% ethanol aqueous solution as the mother liquor. The extraction parameters optimization, cycle extraction and 4-ethylguaiacol purification are studied, respectively. Extracting parameters investigated are reaction temperature (4–80 ℃), extraction time (5–60 min), and pH value (6.5–13). 4-ethylguaiacol extraction efficiency keeps above 90% at the optimum extracting conditions at the reaction temperature of 4–20 ℃, the extraction time of 30 min, and the pH value of 6.5–10. Further cycle extraction experiments are carried out under the optimal extraction parameters using the intermediate as the extracting agent. After 36 cycle extraction times, the extraction efficiency of 4-ethylguaiacol can still reach 91.7%. Guaiacol has similar physical and chemical properties to 4-ethylguaiacol, thus it is used as an impurity to test the extraction and purification effect of the heterogeneous extracting method. With increasing the cycle extraction times from 1 to 4, 4-ethylguaiacol purity reaches more than 99% for all four mother liquors with different initial concentrations. 4-ethylguaiacol purity of 99.57% is achieved through twice cycle extraction when its initial content in the mother liquor is 95%.

Guanidinium Hydrophobic Magnetic Ionic Liquid-Based Dispersive Droplet Extraction for the Selective Extraction of DNA

Xuelian Wang,Mei Liu,Xueqin Ding

Six hydrophobic magnetic guanidinium ionic liquids (HMILs) were designed and prepared for the extraction of DNA. The physical and thermal properties of the HMILs were characterized using vibrating sample magnetometry, density meter, rotational rheometer, Karl Fischer moisture, Fourier transform infrared spectrometry, and thermogravimetric analysis. Single-stranded DNA and duplex DNA extracted by HMILs can be rapidly collected by a magnet. Three assisted extraction methods, including vortex extraction, mechanical shaking extraction, and ultrasonic extraction, were introduced to extract DNA with HMILs and the extraction efficiencies were evaluated using NanoDrop. Influencing factors of the DNA extraction were comprehensively evaluated, involving the HMIL volume, extraction time, pH, and extraction temperature. The HMIL-based extraction method can well extract DNA from complex matrices and Escherichia coli cell lysates.

Effects of Different Extraction Methods on Physicochemical Characteristics and Bioactivities of Fig (Ficus carica L.) Leaves Polysaccharides

Xin-Yang Jiang,Cai-Wei Wang,Jun Zhang,Pei-Pei Xu,Yi-Ting Xue,Qian Wang

The tea of fig ( Ficus carica L. ) has long been used as a medicine in China due to its excellent effects on hemorrhoids, damp heat diarrhea and other diseases. The present study was designed to evaluate the impacts of different extraction methods, including hot water extraction (HWE), microwave assisted extraction (MAE), ultrasound assisted extraction (UAE), ultrasound assisted enzymatic extraction (UAEE), ultrasound-microwave assisted extraction (UMAE), alkali extraction (AE) and high-speed shearing homogeneous extraction (HSHE), on physicochemical characteristics and bioactivities of polysaccharide from fig leaves (FLPs). Results indicated that there were great differences between the structure and activity of FLPs prepared by seven methods. Among them, FLPs extracted by UAE (FLPs-UAE) showed the strongest antioxidant and hypoglycemic activities due to its higher Mw, total polysaccharide content (60.62±3.06%) and Glucosamine (GlcN) content (7.68%). In addition, FLPs extracted by HWE possessed more total polysaccharide content (62.03±0.70%), total phenolic compounds content (13.55 ± 0.02 %) and GlcN content (5.78%) contributed to its strong antioxidant activity and the inhibition on α-amylase and α-glucosidase, which was second to FLPs-UAE. These finding suggested that UAE and HWE, especially UAE, were good ways to extract FLPs with high antioxidant and anti-diabetic activities, which provide foundations for choosing the suitable extraction strategy to prepare FLPs with desired bioactivities and promoting the application of FLPs in function food industries.

Experiments and simulation on extraction of esmolol enantiomers from single stage to multistage

Weifeng Xu,Shichuan Wang,Wanru Wang,Panliang Zhang,Kewen Tang

Enantioselective extraction of esmolol (ES) enantiomers was performed with tartaric acid derivatives and boric acid (BA) as chiral extractant. The single–stage extraction experiments were carried out to construct the extraction system for ES enantiomers through investigating type of tartaric acid derivatives, type of the organic solvent and temperature. The optimal conditions for extraction system were achieved, which involves 1,2–dichloroethane as organic solvent, iso–butyl–D–tartrate (DT) as chiral selector and 278 K for extraction temperature. A single–stage extraction model was established based on an interfacial reaction mechanism and proved to be reliable by verified experiments. Based on the single–stage extraction model and the law of mass conservation, a fractional extraction model was explored to simulate and optimize the fractional extraction process. The optimal operation conditions at temperature of 278 K were obtained including ES concentration of 0.01 mol/L, DT and BA concentrations of 0.1 mol/L, F/W of 0.25 and O/W of 1.0347, feeding in the middle stage at pH of 5.50. The ee eq of 0.98 and Y eq of 0.99 can be achieved by symmetric separation with the number of stages of 32 under the optimal conditions. As S–ES is the desired product, 10 stages are enough to obtain 0.98 of ee raffinate for S–ES by asymmetric separation under the conditions containing ES concentration of 0.01 mol/L, DT and BA concentrations of 0.1 mol/L, F/W of 0.25 and O/W of 1.64, feeding in the Stage 2 at pH of 5.50.

Optimized High-Pressure Ultrasonic-Microwave-Assisted Extraction of Gingerol from Ginger: Process Design and Performance Evaluation

Yang Zhang,Siyi Yang,Wensi Li,Xiaoyan Li,Xiangqin Lai,Xiang Li,Wuwan Xiong,Bo Zhang

This study employed high-pressure ultrasonic-microwave-assisted extraction (HP-UMAE) to extract gingerols from ginger. The extraction yield and total polyphenol content of the extracts were determined. Their antioxidant activity was assessed by DPPH and ABTS radical scavenging assays, and compared with extracts obtained by leaching extraction, reflux extraction, ultrasonic-assisted extraction (UAE), microwave-assisted extraction (MAE), and ultrasonic-microwave-assisted extraction (UMAE). The results demonstrated that HP-UMAE achieved the highest extraction yield and the strongest ABTS radical scavenging activity among the evaluated methods. Furthermore, HP-UMAE extracts exhibited the highest concentrations of key gingerol constituents: 6-gingerol (14.29 mg/L), 8-gingerol (0.38 mg/L), 10-gingerol (1.95 mg/L), and 6-shogaol (4.32 mg/L). This enhanced efficacy is attributed to the synergistic combination of ultrasonic cavitation and microwave-induced thermal effects under elevated pressure. This synergy creates conditions promoting cellular wall disruption, facilitating the release of intracellular components, while concurrently enhancing solvent penetration and gingerol solubility. Scanning electron microscopy (SEM) analysis confirmed the significant structural damage inflicted on ginger cell walls following HP-UMAE treatment. The process parameters for HP-UMAE were optimized using single-factor experiments. The optimal extraction conditions were determined as follows: microwave power 800 W, ultrasonic power 1000 W, liquid-to-solid ratio 55:1, and temperature 100 °C (corresponding pressure 2 MPa). Under these optimized parameters, the extraction yield and ABTS radical scavenging rate reached their peak performance, yielding values of 4.52% and 43.23%, respectively.

Comparison of different extraction methods of polysaccharides from cup plant (Silphium perfoliatum L.)

Hongyan Wu,Hongmei Shang,Yang Guo,Hexiang Zhang,Hongxin Wu

The purpose of the present research was to compare the effects of four extraction methods on the extraction yields, physicochemical characteristics, and antioxidant activities of polysaccharides from the cup plant ( Silphium perfoliatum L.) (CPPs). The extraction methods included the hot water extraction method (HEM), ultrasonic-assisted extraction method (UEM), enzyme-assisted extraction method (EEM), and the ultrasonic-enzyme-assisted extraction method (UEEM). Four corresponding CPP extracts, HEM-CPPs, UEM-CPPs, EEM-CPPs, and UEEM-CPPs, were obtained. The results indicated that there were notable differences between the extraction methods regarding the yield, average molecular weight, uronic acid content, monosaccharide proportions, and the antioxidant activities of the CPPs. HEM-CPPs were extracted with the lowest yield (6.44%). EEM-CPPs had the largest extraction yield (9.87%) and the most notable antioxidant abilities of ferric reducing power and free radical scavenging activity. According to correlation analysis, the higher antioxidant abilities of EEM-CPPs might be due to their smaller average molecular weight and a higher content of uronic acid than those of the other extracted CPPs. Therefore, EEM could serve as a promising technique for extracting CPPs.

Comparison of three methods for the sequential extraction of lignite pretreated by ionic liquid

Ning Ding,Mei Li,Jinsong Yue,Sidan Cheng,Caiyun Gao

In order to improve the extraction rate of coal by solvent extraction, Lianhuatang lignite (LHT) was pretreated with 1-butyl-3-methylimidazolium hexafluorophosphate ionic liquid([Bmim]PF 6 ) firstly. Then, the Soxhlet extraction (SE), ultrasound-assisted extraction (UAE), and microwave-assisted extraction (MAE) were carried out to obtain extracts (E S1-3 , E U1-3 , and E M1-3 ) and residues (R S1-3 , R U1-3 , and R M1-3 ) from the Lianhuatang lignite pretreated with ionic liquid (BLHT), respectively, and CS 2 , acetone, and ethanol were used as solvents. The coal samples before and after the ionic liquid pretreatment were characterized by scanning electron microscope (SEM), Fourier-transform infrared spectroscopy (FTIR), and thermogravimetric analyzer (TG). The residues were characterized by FTIR, and the extracts were characterized by a gas chromatograph/mass spectrometer (GC/MS). According to the results, ionic liquid can crack coal particles, increase the contact area of coal surface with solvent, and destroy part of the hydrogen bonds and intermolecular forces in coal, enabling easy extraction of the active small molecules. The macromolecular structure of coal is not destroyed in the three sequential extraction experiments. The types of extracts are similar under different methods, but the specific substances are very different. The total extraction rate by MAE was the highest, which was 48.70%. Microwave radiation can promote the extraction of oxygen-containing substances, especially esters.

Green extraction of carotenoids from apricot flesh by ultrasound assisted corn oil extraction: Optimization, identification, and application

Andi Suo,Gongjian Fan,Caie Wu,Tingting Li,Kaiping Cong

The valorization of waste apricot flesh (WAF) can solve environmental and economic problems, and also meets the demand for natural pigments. Therefore, the aim of this paper was to recover carotenoids from WAF, to determine the type and content of carotenoids in the extracts and to explore the potential of the extracts for food industry applications. The extraction conditions were optimized. The total carotenoids content (TCC) was 42.75 mg/100 g dried weight under the optimized conditions: Time: 60 min, Temperature: 41.53℃, Power: 200 W, Liquid to solid (LS) ratio: 0.10 g/mL. The highest content of carotenoids in the corn oil extracts (COE) was phytoene. Color of COE under high temperature was investigated. Carotenoids in the COE were degraded at high temperatures. The a* of fries fried by COE was 7.31 times higher than that of corn oil. This study provides guidance for the green recovery of carotenoids and valorization of WAF.

Zhao Zhenyu,Liu Pinghuai,Wang Shenglin,Ma Shasha

Ultrasonic-assisted extraction (UAE) and microwave-assisted extraction (MAE) were developed to extract flavonoids antioxidants from the root of Millettia speciosa ( Millettia speciosa Champ.). The optimal experimental parameters for antioxidant extraction from M. speciosa Champ. were measured using single-factor experimentation combined with response surface methodology (RSM). Final extraction rate (FER) of flavonoids assay was used to quantify antioxidant compounds. Then, 2,2ˊ-diphenyl-1-picryhydrazyl (DPPH) scavenging capacity, 2,2ˊ-azino-bis(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) scavenging capacity and ferric reducing antioxidant power (FRAP) were used to determine the antioxidant capacities of extract. Besides, UAE and MAE were compared with conventional Soxhlet extraction (CSE) and M. speciosa Champ. powders were observed by scanning electronic microscopy to analyze the damage of cell structure caused by different extraction methods. The results showed that UAE was the optimal extraction method to extract flavonoids compounds from M. speciosa Champ. and the optimized UAE conditions of flavonoids was as follows: ethanol, 80%, temperature, 40 °C and ultrasonic power, 500 W. This paper provided useful information to utilize the resource of Chinese herbal medicine and manifest the great antioxidant activities of M. speciosa Champ., and the optimized UAE conditions of flavonoids from M. speciosa Champ. could be used in industrial production.

Anti-oxidant and anti-fatigue properties of apple pomace polysaccharides by acid or alkali extraction

Xiangna Chang,Xuefeng Chen,Pin Gong,Wenjuan Yang,Lan Wang,Ning Liu,Yao Su,Yuanyuan Zhao

Summary The anti-fatigue effects and structure-activity relationship of apple polysaccharides (APPs), the active components in apple pomace, remain unknown. We examined the effects of different extraction methods on polysaccharide structures and anti-fatigue activities. Additionally, the structural and biochemical properties of APPs prepared under acidic (ACAPP) or alkaline (ALAPP) conditions, were characterised. ACAPP and ALAPP were acidic heteropolysaccharides primarily comprising arabinose, rhamnose, galactose, glucose, xylose and galacturonic acid. The molecular weights of ACAPP and ALAPP were 110 kDa and 89.6 kDa, respectively. ALAPP had more branched chains than ACAPP and showed stronger free radical-scavenging and anti-fatigue properties. The anti-fatigue properties of APPs correlated with their anti-oxidant activities and were exerted through eliminating free radicals and mediating an oxidative balance. The stronger anti-fatigue activity of ALAPP is probably related to its structural characteristics. Collectively, our study provides a fundamental basis for the development and use of APPs in medicine and functional foods.

UPLC-ESI-MS/MS-based chemometric approach for investigating the effect of conventional versus modern extraction methods on polyphenols recovery from grape seed wastes

Shenxiong Chen,Yi Yuan,Fan Zhang,Shiye Lin,Pengren Zou,Mohamed A. Farag,Jesus Simal-Gandara,Hui Cao,Jianbo Xiao

Grape seed wastes are a rich source of bioactive polyphenols. This study compares the recovery efficiency of traditional maceration extraction (ME) with modern extraction techniques (ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), and accelerated solvent extraction (ASE)), revealing the selective extraction of polyphenolic compounds by different methods. UPLC-ESI-MS/MS analysis was used to identify and quantify 24 polyphenols. ASE showed the highest efficiency for phenolic acids (e.g., gallic acid, 231.75 μg/g) and proanthocyanidins (e.g., procyanidin B1, 126.18 μg/g), while MAE was optimal for flavonoids recovery (e.g., myricetin, 41.52 μg/g). Multivariate chemometric analysis revealed co-extraction patterns among structurally related compounds (e.g., flavan-3-ols with galloylated derivatives), linking extraction parameters to the selectivity of polyphenols. Integrating UPLC-ESI-MS/MS profiling of 24 polyphenols, we suggest that MAE/ASE is most suitable for antioxidant-rich nutraceuticals, while ME is optimal for thermally labile pharmaceuticals. This study provides practical strategies for the sustainable utilization of grape seed by-products, offering industry-specific solutions to reduce waste and improve the recovery of bioactive compounds.
