Support bath-assisted additive manufacturing has been extensively employed in constructing 3D complex soft biomaterials by extrusion- or inkjet-based printing. However, the conventional support bath (i.e., slurry of gelatin microparticles used in freeform reversible embedding of suspended hydrogels and nanoclay) can only provide static in-situ support to the deposited constructs, which is not feasible for vat photopolymerization printing since the printed constructs are mobile during the printing process. A dynamic support bath assisted vat photopolymerization printing method using yield stress fluid (YSF) is proposed in this study, which enables the dynamic accompanying supporting for the printing structures. As the moving platform rises continually, the YSF bioink was elevated with the cured construct near the curing region and remaining in a solid-like state to provide support against gravity and surface tension. While the YSF bioink was in a liquid-like state, due to the atmospheric pressure difference during the rising of moving platform, the YSF bioink can flow to fill up the vacuum and ensure continuous forming. Thus, this printing method achieved the vat photopolymerization printing of extremely soft cell-laden bioinks (Young’s modulus of 4.3 KPa) into 3D complex constructs with excellent biological functionality.

Embedded 3D printing is widely adapted for fabricating architected soft and non-self-supporting hydrogels for applications ranging from tissue engineering to soft robotics. Although the matching between hydrogels and supportive baths sets the foundation in embedded 3D printing, the rule-of-thumb for supportive bath selection and creation is not yet established. Herein, the “shapeability” of distinct classes of hydrogel inks (i.e., pH-responsive, photo-crosslinkable, thermal-sensitive, chemically crosslinkable monomeric, and cationic and anionic inks) in diverse representative support baths (i.e., gelatin slurry, agarose fluid gel, Carbopol and oil-based baths) is evaluated. The results show that the dominate mechanisms for interfacial instabilities, including diffusion-driven or charge-driven, can be predicted by evaluating the composition pairing of the pre-crosslinked hydrogel ink and supportive bath. Based on this, a general and simplistic guideline for supportive bath selection to attain hydrogel shapeability is proposed. The approach can widen the spectrum of hydrogel materials that can be structured on-demand for a plethora of functionalities.

Manufacturing biocompatible materials with higher-order structure has great significance because they can mimic the extracellular medium of the human organism and are a novel strategy for tissue regeneration. In this study, a device with stiffness-gradient characteristics based on two biocompatible materials, alginate with presolidification and photocurable acrylamide-containing supporting bath, was designed and constructed by the 3D printing technique. The presolidification can avoid rapid diffusion of alginate in aqueous solutions, improve mechanical properties without the introduction of heterogeneous gel precursor, and endow gradient stiffness by the controlled diffusion of calcium ions. Besides, a photocurable supporting bath was combined to manufacture a device with a dual-gradient structure by a 4-step procedure, including 3D printing, removal of the inner hydrogel, solidification of alginate, and curing of the supporting bath. A cylinder-like container was manufactured as the template, and the wall of the resultant container with two types of gradient structures showed parabola-like stiffness changes (open upward), resulting from calcium ion diffusion-controlled gradient solidification and alginate diffusion-controlled gradient photocuring. Moreover, the resultant device exhibited lower cytotoxicity to both adherent and suspension cells than containers manufactured with alginate. Because of the high water uptake of the photocured supporting bath, the removal of toxic metabolic products together with cell culture medium from the container leads to better cell compatibility. This diffusion-controlled device is also applicable to other additive manufacturers with biomedical significance.

Summary Here, we present a protocol for 3D printing heart tissues using thiol-norbornene photoclick collagen (NorCol). We describe steps for synthesizing NorCol, preparing bioink and the support bath, and cell-laden printing. We then detail procedures for the loading of C2C12 cells into NorCol, ensuring structural integrity and cell viability after printing. This protocol is adaptable to various cell lines and allows for the printing of diverse complex structures, which can be used in drug screening and disease modeling.

Four-dimensional (4D) printing is a promising technology that provides solutions for compelling needs in various fields. Most of the reported 4D printed systems are based on the temporal shape transformation of printed subjects. Induction of temporal heterogenicity in functions in addition to shape may extend the scope of 4D printing. Herein, we report a 4D printing approach using plant protein (zein) gel inspired by the amyloid fibrils formation mechanism. The printing of zein gel in a specialized layered-Carbopol supporting bath with different water concentrations in an ethanol-water mixture modulates hydrophobic and hydrogen bonding that causes temporal changes in functions. The part of the construct printed in a supporting bath with higher water content exhibits higher drug loading, faster drug release and degradation than those printed in the supporting bath with lower water content. Tri-segment conduit and butterfly-shaped construct with two asymmetrical wings are printed using this system to evaluate biomedical function as nerve conduit and drug delivery system. 4D printed conduits are also effective as a drug-eluting urethral stent in the porcine model. Overall, this study extends the concept of 4D printing beyond shape transformation and presents an approach of fabricating specialized baths for 4D printing that can also be extended to other materials to obtain 4D printed medical devices with translational potential.

Vascularized tissue models represent a pivotal approach for enhancing the accuracy of clinical drug prediction by recapitulating the in vivo microenvironment. However, replicating the structural complexity, functional integration, and long-term stability inherent in native vascular tissues remains a significant challenge within the tissue engineering field. Herein, a novel strategy employing aqueous-embedded coaxial bioprinting is proposed. By optimizing the rheological compatibility between alginate-collagen (Alg-Col) bioink and xanthan gum-collagen (XG-Col) support bath, the high-fidelity and efficient fabrication of vascularized tissue models is achieved. Utilizing a triaxial nozzle, synchronous deposition of a bilayered vascular construct containing human umbilical vein endothelial cells (HUVECs) and human lung fibroblasts (HLFs) is performed directly within a tumor cell (human lung adenocarcinoma PC-9 cell line)-laden support bath, resulting in the construction of a perfusable vascularized lung cancer tissue model. Drug assessment revealed significantly lower sensitivity of PC-9 cells to erlotinib within the 3D vascularized model compared to controls. In addition to the physical barrier effect, the vascularized microenvironment activated cancer cell survival pathways primarily through paracrine signaling from endothelial cells and fibroblasts. Gene expression analysis further demonstrated the induction of epithelial-mesenchymal transition (EMT) in the vascularized model. This integrated platform more accurately recapitulates in vivo drug diffusion kinetics and resistance mechanisms, thereby providing an innovative preclinical platform for enhanced anticancer drug screening.
