A Targeting Trained Immunity Nanofiber Scaffold for Large Bone Defect Repair

Zhan Jingdi,Chen Zhuolin,Liu Junyan,Pang Qiming,Lei Mingjie,Liu Jiacheng,Song Yang,Huang Wei,Dong Lili

Modulating trained immunity while simultaneously initiating regenerative cues presents a significant challenge in large bone defect therapy. This study introduces a cell-free approach utilizing a 3D microenvironment-responsive scaffold to orchestrate immune reprogramming. To mitigate maladaptive trained immunity and activate regenerative signaling, a composite fibrous scaffold is functionalized with immune-engineered exosomes derived from inflammation-primed mesenchymal stem cells (PSS-iEXO) in a reactive oxygen species (ROS)-responsive manner. The PSS-iEXO scaffolds incorporate boronic ester linkages as ROS-sensitive moieties, enabling rapid, dynamic, and “on-demand” exosome release in response to elevated ROS levels characteristic of the early inflammatory phase post-injury, thereby initiating regeneration. In vitro and in vivo analyses reveal that these scaffolds precisely target and modulate maladaptive trained immunity, reprogramming immune responses by shifting macrophage polarization from a hyperactivated type I phenotype to a balanced state while promoting CD4 + regulatory T cell activation—both critical for coupling angiogenesis and osteogenesis. Mechanistic insights highlight the role of engineered exosomes in enhancing mitochondrial function and oxidative phosphorylation in macrophages, establishing a cell-free immune-regenerative niche for large bone defect therapy. Graphical Schematic diagram of the fabrication, function, and mechanism of ROS-responsive 3D electrospun nanofiber scaffolds loaded with immunoengineered exosomes (PSS-iEXO) for promoting large bone repair.

Enhancing Bone Regeneration and Osteogenic Quality by n-HA Internalized Osteoblasts Synergized with ON Protein: Mechanistic Insights

Wei Yang,Qin Zou,Chenxin Wang,Yuankun Ren,Rui Zhang,Mingyue Lin,Zeyu Huang,Mengxin Huangfu,Lili Lin,Wei Li,Xiyu Li

Bone scaffolds offer hope for oral jawbone repair, yet improving their osteogenic performance remains a clinical challenge. This study investigates a novel approach to enhance early bone formation and osteogenic quality by coloading hydroxyapatite (HA)─internalized osteoblasts (OHA) and osteonectin (ON) onto various scaffolds. Our findings demonstrated that the OHA could effectively facilitate the early bone regeneration by providing rapid calcium and phosphorus ion release via lysosome-mediated HA degradation, while the ON protein helps in ion deposition, cell proliferation, and matrix mineralization. When the PHA (PCL+HA) scaffold was incorporated with both the OHA and ON, the scaffold exhibited superior pro-osteogenic performance, driven by synergistic effects of rapid ion release from the OHA, slow ion release from the PHA, and upregulation of osteogenesis-related genes. The analyses of mechanisms revealed that the OHA activated MAPK and PI3K–Akt pathways, while ON stimulated calcium and Wnt signaling, collectively promoting the osteogenic potential. The strategy presented in this study paves a promising way for the development of advanced bone scaffolds to improve the bone regeneration quality.

Fabrication and characterization of bioactive glass/alginate composite scaffolds by a self-crosslinking processing for bone regeneration

Fujian Zhao,Wen Zhang,Xiaoling Fu,Weihan Xie,Xiaofeng Chen

The objective of this study was to synthesize and characterize self-crosslinked bioactive glass/alginate composite scaffolds, as a kind of potential biomaterial for bone regeneration. The scaffolds, fabricated through a self-crosslinking process of alginate by bioactive glass microspheres provided Ca2+ completely without any organic solvent, crosslinking agent, or binder. The microstructure, mechanical properties, apatite-forming ability, ionic release, adhesion, proliferation, and ALP activity of human bone marrow-derived mesenchymal stem cells (hBMSCs) of the scaffolds were evaluated. The results showed that uniform films could be obtained on the surface as well as in abundant crosslinking bridges in the interior of the scaffolds. The rapidly released Ca2+ from bioactive glass could be temporarily stored in alginate, which achieved the controlling release. The scaffolds, with porosities of 75–57%, had a compressive strength in the range of human trabecular bone. At the same time, they presented an excellent apatite formation ability in vitro. In vitro cellular study confirmed that the alginate films temporarily separated cells from bioactive glass which promoted early cell adhesion and all the groups showed good cell proliferation and ALP activity. All these results demonstrate that self-crosslinked scaffolds represent promising candidates for bone regeneration.

Fabrication of Resveratrol-Loaded Scaffolds and Their Application for Delaying Cell Senescence In Vitro

Rong You,Lu Shi,Surong Chen,Qingpeng Liu,Lingkun Zhang,Liang Yin,Runcai Yang,Yan-Qing Guan

In this research, resveratrol (RSV)-loaded scaffolds have been prepared to control the release of resveratrol and used to delay hepatic stellate cell (HSC) senescence in vitro. The functional carboxyl group–COOH is first introduced to the surface of poly( ε -caprolactone/ d , l -lactide) (P(CL-DLLA)) under the coadministration of ultra-violet (UV) treatment and photo initiator and then resveratrol are conjugated onto the surface of the modified scaffolds through esterification. The characterization of the structure of RSV-AA-P(CL-DLLA) shows that resveratrol has been successfully conjugated onto the modified surface. Cell growth exhibits a higher level of cell viability and much more obvious agglomeration on the surface of the synthetic RSV-AA-P(CL-DLLA). Meanwhile the activity of senescence-associated β -galactosidase (SA- β -gal) and reactive oxygen species (ROS) is downgulated for cells on RSV-AA-P(CL-DLLA), which suggests that cell senescence is delayed on RSV-AA-P(CL-DLLA). And then it is attested that cells have a lower level of p53 but SIRT1 expression is upregulated on RSV-AA-P(CL-DLLA), which might be related to resveratrol release from RSV-AA-P(CL-DLLA). It also suggested cell senescence on RSV-AA-P(CL-DLLA) has been regulated by p53 and the SIRT1 signaling pathway. In all, the present study shows that RSV-AA-P(CL-DLLA) can be successfully prepared to promote cell growth and delay cell senescence and could be used for cell-based therapy in tissue engineering.

Carbon nanotube reinforced polyvinyl alcohol/biphasic calcium phosphate scaffold for bone tissue engineering

Weiwei Lan,Xiumei Zhang,Mengjie Xu,Liqin Zhao,Di Huang,Xiaochun Wei,Weiyi Chen

In this paper, a well-developed porous carbon nanotube (CNT) reinforced polyvinyl alcohol/biphasic calcium phosphate (PVA/BCP) scaffold was fabricated by a freeze-thawing and freeze-drying method. The microstructure, mechanical properties and the composition of the scaffolds were characterized by field emission scanning electron microscopy (FE-SEM), X-ray diffraction (XRD) and Fourier-transform infrared spectroscopy (FTIR). The results illustrate that after the incorporation of CNTs, the compressive strength of the hydrogels (moisture state) reached 81 ± 6 kPa, presenting a significantly higher value than that of pure PVA/BCP hydrogels (48 ± 2 kPa). Meanwhile, CNT reinforced PVA/BCP scaffolds exhibited a porous structure and high interconnectivity (80 ± 0.6%). The degradation analysis indicated that the degradation ratio of scaffolds can be varied by changing the concentrations of BCP powders and CNTs. Cell culture results show that PVA/BCP/CNT porous scaffolds have no negative effects on the survival and proliferation of cells. These results strongly show that the composite scaffolds may possess a potential application in the field of bone tissue engineering and regeneration.

Cellulose nanocrystal-reinforced thermally responsive dynamic hydrogel

Long Yang,Simei Zhang,Zemei Hu,Jing Li,Guangyin Xie

Hydrogels are regarded as ideal tissue scaffold materials owing to their unique advantages, but problems such as low strength and easy cracking have become bottlenecks that limit their practical applicability. Constructing reinforced hydrogels is a widely studied approach for performance enhancement. Therefore, herein, cellulose nanocrystals were introduced as reinforcement into a thermally responsive hydrogel matrix through a Schiff base, achieving a cellulose nanocrystal-reinforced thermally responsive dynamic hydrogel. The CNC-reinforced hydrogel was characterized in detail, and the influence of CNCs on hydrogel properties was systematically investigated. Moreover, the thermally responsive and dynamic properties of the resulting hydrogels were tested to evaluate their mechanical performance. This work provides a feasible solution for the design of hydrogel-based tissue scaffold materials with long service life.

Rare earth-doped titanium dioxide for enhanced reactive oxygen species generation in photodynamic anti-tumor therapy of scaffolds

Cijun Shuai,Lefei Yuan,Mingli Yang,Xiuwen Gao,Jun Zan,Jin Chen,Xiong Shuai

Photodynamic therapy, as a minimally invasive treatment, selectively and optically damaged skin tumor cells through the generation of reactive oxygen species (ROS). However, its antitumor efficacy is limited by the rapid recombination of electron-hole pair in photosensitizers. In this work, a rare-earth lanthanide-doped titanium dioxide nanosystem was successfully developed via a co-coprecipitation method. In this nanosystem, lanthanide doping induced lattice distortion and generated oxygen vacancy defects, effectively suppressing electron-hole pair recombination. Furthermore, the unique electronic structure of the lanthanide dopant modulated the band positions and Fermi level of titanium dioxide, thereby narrowing the bandgap and broadening its optical absorption spectrum. Subsequently, the nanosystem was incorporated into a poly-L-lactic acid scaffold by selective laser sintering. Photoelectrochemical and ROS analyses revealed significant enhancement in electron–hole pair separation and a marked increase in ROS generation. Density functional theory calculations further confirmed that the reduced bandgap contributed to the improved photocatalytic performance of titanium dioxide. In vitro cell experiments demonstrated that this photodynamic therapy platform achieved an 82.4 % inhibition rate of skin tumour growth. Overall, this work presents a feasible strategy to enhance the therapeutic efficacy of tumour photodynamic therapy.

Tri-layer gradient fibre-aligned electrospun composite: Accelerating skin wound healing

Limin Gong,Hongli Ma,Hui Fang,Xi Yang,Jiacai He,Chenbing Wang

s The structure and topography of the scaffold material play an essential role in the skin wound healing process, and aligned nanofibers can guide cell growth and proliferation to some extent. It has been demonstrated that the optimal scaffold fabrication method for wound healing is to generate three-dimensional scaffolds composed of nanoscale fibers. Inspired by the structure and function of skin, composite aligned electrospun dressings with fiber gradient changes are prepared by continuous spinning through changing the relevant parameters of the electrospinning process, thereby creating a wettability gradient change to mimic the skin's hydrophilic-hydrophobic transition. This design efficiently manages exudates from the wound site and maintains humidity balance. The hydrophilic inner layer (A100) absorbs wound exudates and maintains a moist environment in the wound. The A300 layer, with its appropriate porosity, ensures nutrient and gas exchange at the wound site, providing an ideal microenvironment for cell growth. The outermost layer, A500, is a mechanical barrier to prevent bacterial invasion, wound infection, and over-hydration. In vitro experiments have verified that the dressing exhibits good biocompatibility and mechanical properties, while in vivo experiments have confirmed that the dressing promotes wound healing, collagen deposition, angiogenesis, hair follicle regeneration, and reduces fibrosis at the wound site. Additionally, it improves scar formation to a certain extent.
