Oral delivery of WR-1065 by ROS-responsive PEG-PCL nanoparticles for radioprotection

Xiaona Lin,Xinxin Wang,Hongqi Tian

When a burst of reactive oxygen species (ROS) is induced after radiation exposure, a sufficient radioprotector drug must be quickly released to neutralize the ROS . An oral polymeric carrier of poly(ε-caprolactone)-b-poly(ethylene glycol)-b-poly(ε-caprolactone) was synthesized via the ROS-response linker thioketal (TK-linked PCEC) and used for delivery of the drug WR-1065. The drug-loaded nanoparticles (PCEC/WR-1065 NPs) had high WR-1065 content (19.7 % w/w) in the core and a particle size of 183 nm. The PCEC/WR-1065 NPs could protect the drug from degradation at gastric pH. More than 70 % of the TK-linked PCEC copolymer could be cleaved in 1 h. Therefore, a sufficient amount of WR-1065 could be quickly released from the PCEC/WR-1065 NPs to neutralize the burst of free radicals generated under irradiation. The in vitro cell uptake results of TK-linked PCEC NPs indicated that the NPs could be efficiently endocytosized by Caco-2 cells. Oral administration of the PCEC/WR-1065 led to survival benefit as well as protective effects on hematopoiesis and main organs with respect to the irradiation and oral free WR-1065 groups after 7.2 and 8.0 Gy total body irradiation, respectively. In vivo biodistribution studies indicated that the PCEC/WR-1065 NPs accumulated mainly in intestinal tissue. These results suggest that the design presented here provides a promising oral ROS-sensitive platform for acute radiation syndrome.

Therapeutic effects of G90′-ES2 protein complex from regenerated Eisenia fetida on radiation-induced skin injury

Yuwei Yang,Peng Ni,Lixue Ma,Haikang Tang,Jiaming Yang,Xiaoliang Zhou,Yikun Sun,Wenqing Xu

Ethnopharmacological relevance Radiation-Induced Skin Injury (RISI) is the major cutaneous side effect met by cancer patients who received radiotherapy. Disposed earthworm (Dilong) has been historically employed for treating various ailments in ancient China. Amputated-regeneration and high radiation-tolerant are two distinctive characteristics for earthworms. These features might due to their abundant biologically active substances (65 % composed of proteins), which make them attractive candidates for radioprotector development. Aim of the study This study aimed to investigate therapeutic mechanism of regenerated E. fetida protein extract (G90′-ES2) in mitigating RISI, and reveal therapeutic potential macromolecules by proteomic analysis. Materials and methods Protein salt precipitation method was firstly adopted to obtain G90′-ES2/G90-ES2 protein complex. Their radioprotective activities were evaluated in vitro by cell viability, migration, ROS accumulation, EdU proliferation, cell cycle and apoptosis assays. A combined radiogenic injury mice model was constructed to analyze radiotherapeutic function of G90′-ES2 in vivo . Its mechanism against RISI was illustrated by RNA-Seq analysis, qPCR, and HIF Luciferase Reporter Assay. Finally, a nanoElute UPLC-MS/MS method was used to elucidate therapeutic potential macromolecules in G90′-ES2. Results We discovered that G90′-ES2 displayed higher efficacy than G90-ES2 in mitigating RISI, mainly by activating radio-resistant HIF-1 pathway, redistributing cell cycle phases (S-phase preservation), accelerating re-epithelialization, and enhancing tissue angiogenesis. The significantly up-regulated proteins (including lumbrokinase, aquaporin, RAD51C, TCTP, and SCBP2) in G90′-ES2 might hold particular importance for its therapeutic effect against RISI. Conclusions This investigation will broaden our understanding of the radiation-tolerant and regenerative capacities for earthworm; also provide medical candidates for the development of skin-specific radioprotector.

Renal Clearable Mo-Based Polyoxometalate Nanoclusters: A Promising Radioprotectant against Ionizing Irradiation

Zhenchao Tao,Jingguo Wang,Haitao Wu,Jiaru Hu,Lu Li,Yuhang Zhou,Qi Zheng,Lisha Zha,Zhengbao Zha

In response to diffused ionizing radiation damage throughout the body caused by nuclear leaks and inaccurate radiotherapy, radioprotectants with considerable free radical scavenging capacities, along with negligible adverse effects, are highly regarded. Herein, unlike being performed as toxic chemotherapeutic drug candidates, molybdenum-based polyoxometalate nanoclusters (Mo-POM NCs) were developed as a non-toxic potent radioprotectant with impressive free radical scavenging capacities for ionizing radiation protection. In comparison to the clinically used radioprotectant drug amifostine (AM), the as-prepared Mo-POM NCs exhibited effective shielding capacity by virtue of their antioxidant properties resulting from a valence shift of molybdenum ions, alleviating not only ionizing radiation-induced DNA damage but also disruption of the radiation-sensitive hematopoietic system. More encouragingly, without trouble with long-term retention in the body, ultra-small sized Mo-POM NCs prepared by the mimetic Folin–Ciocalteu assay can be removed from the body through the renal-urinary pathway and the hepato-enteral excretory system after completing the mission of radiation protection. This work broadened the biological applications of metal-based POM chemotherapeutic drugs to act as a neozoic radioprotectant.

Creatine-mediated ferroptosis inhibition is involved in the intestinal radioprotection of daytime-restricted feeding

Yingjuan He,Gaomei Zhao,Xue Ouyang,Shaobo Wang,Yin Chen,Chenwenya Li,Yongwu He,Jining Gao,Songling Han,Jinghong Zhao,Junping Wang,Cheng Wang

Ionizing radiation-induced intestinal injury (IRIII) is a catastrophic disease lack of sufficient medical countermeasures currently. Regulation of the gut microbiota through dietary adjustments is a potential strategy to mitigate IRIII. Time-restricted feeding (TRF) is an emerging behavioral nutrition intervention with pleiotropic health benefits. Whether this dietary pattern influences the pathogenesis of IRIII remains vague. We evaluated the impact of TRF on intestinal radiosensitivity in this study and discovered that only daytime TRF (DTRF), not nighttime TRF, could ameliorate intestinal damage in mice that received a high dose of IR. Faecal metagenomic and metabolomic studies revealed that the intestinal creatine level was increased by approximate 9 times by DTRF, to which the Bifidobacterium pseudolongum enrichment contribute. Further investigations showed that creatine could activate the energy sensor AMP-activated protein kinase in irradiated enterocytes and induce phosphorylation of acetyl-CoA carboxylase, resulting in reduced production of polyunsaturated fatty acids and reduced ferroptosis after IR. The administration of creatine mitigated IRIII and reduced bacteremia and proinflammatory responses. Blockade of creatine import compromised the ferroptosis inhibition and mitigation of DTRF on IRIII. Our study demonstrates a radioprotective dietary mode that can reshape the gut microbiota and increase intestinal creatine, which can suppress IR-induced ferroptosis, thereby providing effective countermeasures for IRIII prevention.

Clinically Approved Carbon Nanoparticles with Oral Administration for Intestinal Radioprotection via Protecting the Small Intestinal Crypt Stem Cells and Maintaining the Balance of Intestinal Flora

Chengyan Wang,Jiani Xie,Xinghua Dong,Linqiang Mei,Maoru Zhao,Zhengwei Leng,Houxiang Hu,Lele Li,Zhanjun Gu,Yuliang Zhao

The exploration of an old drug for new biomedical applications has an absolute predominance in shortening the clinical conversion time of drugs for clinical application. In this work, carbon nanoparticles suspension injection (CNSI), the first clinically approved carbon nanoparticles in China, is explored as a new nano-radioprotective agent for potent intestinal radioprotection. CNSI shows powerful radioprotective performance in the intestine under oral administration, including efficient free radical scavenging ability, good biosafety, high chemical stability, and relatively long retention time. For example, CNSI shows high reactive oxygen species (ROS) scavenging activities, which effectively alleviates the mitochondrial dysfunction and DNA double-strand breaks to protect the cells against radiation-induced damage. Most importantly, this efficient ROS scavenging ability greatly helps restrain the apoptosis of the small intestinal epithelial and crypt stem cells, which decreases the damage of the mechanical barrier and thus relieves radiation enteritis. Moreover, CNSI helps remove the free radicals in the intestinal microenvironment and thus maintain the balance of intestinal flora so as to mitigate the radiation enteritis. The finding suggests a new application of clinically approved carbon nanoparticles, which not only promotes the development of new intestinal radioprotector, but also has a great potential for clinical transformation.

Multifunctional melamine-sponge composites with reflection capsules for radiation shielding in nuclear environments

Xue Liu,Jianguo Deng,Yingchun Lu,Xin Li,Fuhan Mai,Lanxiang Ji,Xiaofeng Bai

This study focused on radiation shielding materials for nuclear applications, employing a vacuum-assisted impregnation method to optimise the microstructure and establish a synergistic shielding mechanism combining scattering and absorption. Specifically, melamine sponge (MS) is first modified using KH550 to form –NH 2 active sites on its pore surfaces. Subsequently, it is immersed in a composite solution to construct “reflective capsules”. Finally, gradient encapsulation is performed using polyborosiloxane and polyurethane. The results show that compared with pure MS, the tensile strength of this material doubles, the elongation increases by 179 %, and its shielding capabilities against 59.6 keV γ-rays and 0.025 keV thermal neutrons increase by 1442 % and 1593 % respectively, reaching 27.14 % and 43.35 %. Additionally, it has a photothermal peak temperature of 78.5 °C and stable thermal response. Furthermore, relying on the three-dimensional elastic network of the MS and the conductive pathway of MXene, the material achieves a piezoresistive coefficient of 5.27 kPa −1 , demonstrating potential for application as a pressure-sensitive sensor. Through microstructure design and functional integration, this work makes the material suitable for complex scenarios such as nuclear facility maintenance and emergency rescue.
