Preparation and Properties of Double Sensitive β-Cyclodextrin Hydrogel

Qiuyue Guo,Chen Yuying,Li Xin,Liu Jun,Gu Qintian,Hu Jian,Jiang Yan,Zhang Hongwen,Liu Hongbo

Abstract In this paper, partly esterified β-cyclodextrin derivative had been synthesized by esterification of β-cyclodextrin and maleic anhydride. Then, it is amidated by diethanolamine. β-Cyclodextrin copolymer had been prepared from amidated β-cyclodextrin derivative, cholic acid and epichlorohydrin in sodium hydroxide aqueous solution. β-Cyclodextrin hydrogel had been prepared through radical polymerization of β-cyclodextrin copolymer, N -isopropylacrylamide and acrylic acid. The structures, compositions and morphology of the hydrogel were characterized by infrared spectroscopy, nuclear magnetic resonance, differential scanning calorimetry and field-emission scanning electron microscope. Its properties were tested and analyzed by ultraviolet spectroscopy, thermogravimetric analysis and swelling experiments. The results show that the β-cyclodextrin hydrogel owns sustained release performance, temperature and pH sensitivities.

Novel multi-sensitive pseudo-poly(amino acid) for effective intracellular drug delivery

Yanjuan Wu,Dongfang Zhou,Yanxin Qi,Zhigang Xie,Xuesi Chen,Xiabin Jing,Yubin Huang

Novel intracellular pH, glutathione (GSH) and reactive oxygen species (ROS)-responsive nanoparticles were obtained using mPEG2k-block-redox dual sensitive chain-block-mPEG2k (PRDSP) which was prepared by Cu(I)-catalyzed azide–alkyne cycloaddition (CuAAC) click polymerization. The disulfide bond, peroxalate ester and triazole units were regularly and repeatedly arranged in the hydrophobic blocks. The disulfide bond was GSH-sensitive and the peroxalate ester structure could be disrupted by acid and hydrogen peroxide. In addition, the triazole units are capable of forming pH-responsive hydrogen bonds. Dynamic Light Scattering (DLS) and transmission electron microscopy (TEM) were used to investigate the pH, GSH and ROS sensitivity of the PRDSP nanoparticles (NPs). The results indicated that the average diameter, size distribution and morphology greatly changed upon adding GSH/H2O2 or modulating the pH. As the preloaded model anticancer drug, doxorubicin (DOX) was quickly released from DOX-loaded PRDSP (PRDSP@DOX) NPs by addition of 10 mM glutathione (GSH), or 10 mM H2O2 or under acidic conditions rather than under physiological conditions. Confocal laser scanning microscopy (CLSM) and flow cytometric analyses revealed that PRDSP@DOX could effectively deliver DOX into the cytoplasm and nucleus of cells. Therefore, PRDSP NPs may be a promising redox heterogeneity-sensitive carrier for efficient and controlled anticancer drug release.

Flexible and Multifunctional Composites with Highly Strain Sensing and Impact Resistance Properties

Shu Wang,Jianyu Pu,Shuquan Xu,Yuanhao Tian,Qian Shu,Rui Zou,Tonghua Zhang

The development of smart protective clothing will help detect injuries from contact sports, traffic collisions, and other accidents. The combination of ecoflex, spacer fabric, and graphene-based aerogel provides a multifunctional composite. It shows a strain sensitivity of 17.71 at the strain range of 40~55%, a pressure sensitivity of 0.125 kPa−1at the pressure range of 0~15 kPa, and a temperature sensitivity of −0.648 °C−1. After 50 impact tests, its protection coefficient only dropped from 60% to 55%. Additionally, it shows thermal insulation properties. The compression and impact process results of finite element numerical simulation analysis are in good agreement with the experimental results. The ecoflex/aerogel/spacer fabric sensor exhibits a simple structure, large pressure strain, high sensitivity, flexibility, and ease of fabrication, making it a candidate for smart protective clothing resistant to impact loads.

A low-cost bionic interface modification strategy for enhancing the safety performance of energetic powders using PCPA@MXene double coating

Bo-wei Gao,Yan-ze Liu,Xiao-feng Guo,Xu-ran Xu,Hai-feng Yang,Guang-cheng Yang,Jing Lv,Ling-hua Tan

Based on a simple catechol-amine co-precipitation strategy and biomimetic adhesion characteristics, this study prepared a desensitized CL-20@PCPA core-shell energetic powder using the phenolic and amino compound containing low-cost polycatechol/polyamine (PCPA). The results indicate that the CL-20@PCPA composite powder exhibited an elevated characteristic drop height of impact sensitivity ( H 50 ) of 34.9 cm and a reduced friction explosion probability of 56% compared to pure CL-20. PCPA, containing catechol and amine groups, provided a stable platform for secondary modification to enhance the safety of the powder further. Notably, when the CL-20@PCPA@MXene powder was introduced into a propellant system, the resulting propellants displayed a high H 50 value of 20.3 cm while its heat of detonation remained unchanged. This study provides a simple and low-cost biomimetic interface modification strategy for preparing desensitized energetic materials.

Porous Cyclotrimethylenetrinitramine with Reduced Sensitivity Prepared by a Solvation–Desolvation Method

Mi Yan,Yu Liu,Jinjiang Xu,Liyuan Yang,Lin Zhang,Fude Nie,Shiliang Huang

A crystal engineering strategy has been adopted to prepare a porous high explosive, cyclotrimethylenetrinitramine (RDX). Raw RDX was crystallized with TMS, CAM, and HMPT (TMS = tetramethylene sulfone, CAM = ε-caprolactone monomer, and HMPT = hexamethylphosphoramide), forming three solvates, RDX·TMS, RDX·CAM, and RDX·HMPT, respectively. After desolvation by vacuum pyrolysis and antisolvent extraction, porous RDX containing micro- and nanoscale pores was successfully prepared. The porous RDX exhibits remarkably reduced impact, friction, and electrostatic spark sensitivity in comparison to raw RDX. A mechanism of the reduced sensitivity of porous RDX has also been proposed.

Utilization of L-serinyl derivate to preparing triple stimuli-responsive hydrogels for controlled drug delivery

Luo Chun Hui,Sun Xin Xin,Wang Feng,Wei Ning,Luo Fa Liang

Environmentally responsive hydrogels are widely used in various applications. Facile synthesis of multiply stimuli-responsive hydrogels is necessary. Herein, a triple pH-, thermo- and ion-sensitive hydrogel, shorted as HSP, was synthesized by aqueous radical polymerization using L-serinyl acrylate as a monomer and PEG 180 DMA as a crosslinker. The compositions and microstructures of HSP are characterized by FT-IR, 1 H NMR and SEM. The swelling ratio of HSP is the lowest at pH = 3.0 while increases sharply far away from this pH value. Meanwhile, HSP expands with elevated temperatures or ionic strengths, and reaches a plateau when they above 60 °C or 3.0 mol/L, respectively. In addition, these swelling processes are reversible under alternative changing in extern stimuli, and these cycles can be repeated at least 5 times. Furthermore, the release of sodium salicylate can be easily mediated by pH values, temperatures and ion concentrations, all above indicating that HSP is a promising material for controlled drug delivery.

Biomimic pH/reduction dual-sensitive reversibly cross-linked hyaluronic acid prodrug micelles for targeted intracellular drug delivery

Lin Wang,Yin Wang,Qiao Jin,Fan Jia,Haibo Wang,Jian Ji

Biomimic hyaluronic acid (HA) prodrug micelles with pH and reduction responsiveness were prepared by chemical graft of phosphorylcholine and doxorubicin (DOX) to the backbone of HA. DOX was conjugated to HA by pH-sensitive hydrazone bonds which could be hydrolyzed under acidic conditions and thus resulted in controlled release of DOX. Besides, the HA prodrug micelles were cross-linked via reduction-responsive disulfide bonds to improve the stability of the micelles. The HA prodrug can form remarkable core–shell micelles with a quite narrow size distribution which was confirmed by dynamic light scattering (DLS) and transmission electron microscopy (TEM). The in vitro drug release studies showed that there was a dramatic release under endosome pH (pH = 5.0) and reductive environment (10 mM of dithiothreitol) than that at normal physiological environment (pH = 7.4). Greater uptake of DOX from HA prodrug micelles was observed in the CD44 receptor highly expressed MDA-MB-231 cell line, compared to the results from the CD44-negative cell line, NIH3T3, which was demonstrated by the flow cytometry and fluorescence microscopy . The glutathione (GSH)-responsive behaviors were studied as well. The micelles showed higher cellular proliferation inhibition against glutathione monoester (GSH-OEt) pretreated MDA-MB-231 cells than that of the non-pretreated and buthionine sulfoximine (BSO) pretreated ones. These results suggested that such pH/reduction dual-responsive prodrug micelles may enhance the selective inhibition to cancer cells which exhibit higher levels of GSH concentration and provide favorable platforms for cancer therapy.

Fabrication of novel dual thermo- and pH-sensitive poly (N-isopropylacrylamide-N-methylolacrylamide-acrylic acid) electrospun ultrafine fibres for controlled drug release

Zhimei Wei,Qingyu Lin,Jie Yang,Shengru Long,Gang Zhang,Xiaojun Wang

Dual temperature- and pH-sensitive materials used as drug carriers have attracted a particularly high level of interest. However, it is limited by low sensitivity to changes in conditions because of large size and low specific surface area. In this work, novel dual temperature- and pH-sensitive poly( N -isopropylacrylamide- N -methylolacrylamide acrylic acid) (PNIPAm-NMA-AAc) ultrafine fibres were successfully prepared by radical copolymerisation, electrospinning and thermal chemical treatment. The PNIPAm-NMA-AAc ultrafine fibres were characterized by Fourier transform infrared (FTIR), proton nuclear magnetic resonance ( 1 H NMR), spectroscopy UV–visible spectroscopy, scanning electron microscopy (SEM) and cell counting kit-8 (CCK-8) assay. The uniformity and small size of PNIPAm-NMA-AAc ultrafine fibres with LCST range from 31 °C to 43 °C were used to drug carrier because of good thermal stability, no cytotoxic. Curcumin as a model drug was well distributed in PNIPAm-NMA-AAc ultrafine fibres. In addition, analysis of curcumin release at different pH values and temperatures confirmed that the PNIPAm-NMA-AAc ultrafine fibres are responsive to both temperature and pH. The maximum amount of curcumin released was 98.47% ± 1.87%. In addition, the release mechanism of curcumin-based PNIPAm-NMA-AAc ultrafine fibres was shown to conform to Fickian diffusion (r 2  ≥ 0.95). These results indicated that dual-responsive PNIPAm-NMA-AAc ultrafine fibres are promising as drug carriers.

One Stone, Two Birds: pH- and Temperature-Sensitive Nitrogen-Doped Carbon Dots for Multiple Anticounterfeiting and Multiple Cell Imaging

Xue-Chun Yang,Qianli Li,Ming Tang,Yun-Ling Yang,Woochul Yang,Jian-Feng Hu,Xiao-Long Pu,Jinliang Liu,Jing-Tai Zhao,Zhi-Jun Zhang

Carbon dots (CDs) as new fluorescent materials with excellent fluorescence properties have shown enormous potential applications, especially in anticounterfeiting and cell imaging. Herein, nitrogen-doped CDs (NCDs) with excellent biocompatibility were prepared by a simple thermal sintering method. An extremely large red shift (∼130 nm) of the emission peak was observed when the excitation wavelength changes from 355 to 550 nm, indicating that NCDs are excellent fluorescent labeling materials for multiple cell imaging. On the other hand, NCDs showed obvious changes of emission intensity and peak position when the temperature increased from 223 to 323 K and the pH values changed from 1 to 13, respectively, which has been demonstrated by the “horse” pattern printed with NCD water-soluble fluorescent inks. The nontoxic NCDs dispersed in a multiple matrix are highly sensitive to excitation wavelength, temperature, and pH, indicating their great potential application in multiple anticounterfeiting and multiple cell imaging.

Diverse nanostructures and gel behaviours contained in a thermo- and dual-pH-sensitive ABC (PNIPAM–PAA–P4VP) terpolymer in an aqueous solution

Ping Yong,Yuejiao Yang,Zhiyong Wang,Liming Yang,Jie Chen

A thermo- and dual-pH-sensitive ABC triblock copolymer composed of poly(N-isopropylacrylamide)-block-poly(acrylic acid)-block-poly(4-vinyl pyridine) (PNIPAM-b-PAA-b-P4VP, NAV) was synthesized via sequential reversible addition–fragmentation chain transfer (RAFT) polymerization and subsequent hydrolysis. NAV, which had the property of multi-stimuli, containing thermo-sensitive PNIPAM block and opposite pH-sensitive PAA and P4VP blocks, formed diverse nanostructures in an aqueous solution by varying the temperature and pH. Nanostructures including large compound micelles (LCMs), vesicles, spherical micelles and 3D topologies were observed using dynamic light scattering (DLS) and transmission electron microscopy (TEM). More significantly, the solation–gelation behaviour and morphology of the NAV system induced by temperature and pH were investigated by rheology and scanning electron microscopy (SEM), respectively. TEM and SEM revealed that the gelation behaviour of NAV resulted from the bridge effect of the ionized PAA chains between the hydrophobic P4VP cores (V-core) and PNIPAM cores (N-core) over the LCST of PNIPAM. The results obtained from the rheology measurements showed that the gelation temperature was independent of alkaline pH, but slightly tuned by the copolymer concentration or pH (only 6 to 7). Repeated sol–gel transition via temperature changes under alkaline conditions showed the good hydrogel recyclability of NAV.

Preparation and characterization of antibacterial magnetic-/pH-sensitive alginate/Ag/Fe3O4 hydrogel beads for controlled drug release

Bahaaldin Rashidzadeh,Ebrahim Shokri,Gholam Reza Mahdavinia,Reza Moradi,Sarvin Mohamadi-Aghdam,Shadi Abdi

This work reports the synthesis of novel antibacterial magnetic-/pH-sensitive hydrogel beads based on ionotropic-gelation of alginate biopolymer . Using pomegranate peels extract, green-Ag nanoparticles were synthesized inside a mixture of alginate and Fe 3 O 4 , via in situ method. The alginate beads were investigated by VSM, TEM , XRD, and FE-SEM techniques. The introducing Ag and Fe 3 O 4 nanoparticles in hydrogel beads caused a reduction in the swelling capacity of hydrogel beads. Besides, a pH-dependent swelling behavior was observed for hydrogel beads with a maximum swelling capacity at pH = 7.4. Diclofenac sodium (DS) as a model drug was loaded in hydrogel beads and its release showed a pH-dependent behavior. Drug release studies exhibited significant behaviors on the subject of physiological simulated pHs with a high release rate at pH = 7.4. The alginate beads have shown a prolonged and successive controlled drug release nearly 83% at pH = 7.4 and time 200 min. In addition to the pH, the release of DS from magnetic beads was affected by the external-magnetic-fields. Also, the Ag-incorporated alginate beads showed strong antibacterial activity against S. aureus and E. coli . These results indicated that the hydrogel beads have potentially applicable in drug delivery systems.

Synthesis of stimuli-responsive copolymeric hydrogels for temperature, reduction and pH-controlled drug delivery

Jun Gao,Miaomiao Li,Haiying Chen,Ze Xu,Junyao Li,Yong Kong,Xiaoming Zuo

Stimuli-responsive copolymeric hydrogels (CH) are synthesized for controlled drug delivery. Oxidized glycyrrhizic acid is connected to acrylamide through Schiff base reaction, which is then copolymerized with N,N-bis (acryloyl) cystamine and N-isopropylacrylamide by free-radical polymerization for the loading of 5-fluorouracil with a loading efficiency of 96.7 %. The lower critical solution temperature of the CH is 39.14 °C. The N-isopropylacrylamide unit in the CH is sensitive to temperature, endowing the CH with temperature sensitivity. The carboxyl groups of oxidized glycyrrhizic acid can be deprotonated in alkaline solutions, and the electrostatic repulsions between the resultant carboxylate can cause swelling of the CH. The disulfide bond (S–S) of the N,N-bis (acryloyl) cystamine unit can be reduced to sulfhydryl (–SH) by glutathione, and thus the CH is also reduction-sensitive. Therefore, temperature, reduction and pH-controlled delivery of 5-fluorouracil from the CH is achieved. Cytotoxicity assay reveals that the drug-free hydrogels have good biocompatibility while the CH shows high cytotoxicity against human hepatoma SMMC-7721 cells, and the cell viability is only 25 % at the CH concentration of 1 mg mL −1 .
