Development of crosslinker-free antibody–enzyme conjugates for immunoassays in pesticide residue detection

Jiayi Chen,Qiyi He,Junkang Pan,El-Sayed A. El-Sheikh,Rubing Zou,Yirong Guo,Bruce D. Hammock,Li Li,Dongyang Li

Immunoassays are biochemical analytical techniques based on the specific binding between antigens and antibodies. Due to their high sensitivity, rapid analysis, high throughput, cost-effectiveness, and broad applicability, immunoassays have been widely utilized across various fields. Antibody–enzyme conjugates are essential components for signal generation in immunoassays, with crosslinking chemical (crosslinker)-based methods traditionally employed for their production. However, these methods often involve hazardous reagents and complex procedures, highlighting the need for more eco-friendly alternatives. In this study, we developed a universal, crosslinker-free approach for preparing antibody–enzyme conjugates. This method eliminates the use of chemical reagents such as glutaraldehyde or periodate, as well as the intricate conjugation and purification steps typically required. Instead, gold nanoparticles (AuNPs) were employed as a versatile nanomaterial to passively adsorb horseradish peroxidase (HRP) and antibodies through a simple procedure completed within 60 minutes. The resulting conjugates were characterized using UV-vis spectroscopy, transmission electron microscopy (TEM), and zeta potential analysis. Using acetamiprid (ACE), a widely-used pesticide, as the target model, an enzyme-linked immunosorbent assay (ELISA) was developed based on the AuNP-mediated conjugates. The assay demonstrated performance comparable to that of traditional crosslinker-based methods, with an IC50 of 0.49 ng mL−1 and a limit of detection (LOD) of 0.11 ng mL−1. Recovery rates for ACE in spiked fruit samples were consistent with those obtained using liquid chromatography-mass spectrometry (LC-MS). The proposed method not only provides a reliable alternative for HRP and antibody conjugation but also advances green analytical chemistry by minimizing the use of hazardous chemicals, reducing derivative waste, and improving energy efficiency.

Crosslinker-free silk/decellularized extracellular matrix porous bioink for 3D bioprinting-based cartilage tissue engineering

Xiao Zhang,Yang Liu,Chunyang Luo,Chenjun Zhai,Zuxi Li,Yi Zhang,Tao Yuan,Shilei Dong,Jiyong Zhang,Weimin Fan

As cartilage tissue lacks the innate ability to mount an adequate regeneration response, damage to it is detrimental to the quality of life of the subject. The emergence of three-dimensional bioprinting (3DBP) technology presents an opportunity to repair articular cartilage defects. However, widespread adoption of this technique has been impeded by difficulty in preparing a suitable bioink and the toxicity inherent in the chemical crosslinking process of most bioinks. Our objective was to develop a crosslinker-free bioink with the same biological activity as the original cartilage extracellular matrix (ECM) and good mechanical strength. We prepared bioinks containing different concentrations of silk fibroin and decellularized extracellular matrix (SF-dECM bioinks) mixed with bone marrow mesenchymal stem cells (BMSCs) for 3D bioprinting. SF and dECM interconnect with each other through physical crosslinking and entanglement. A porous structure was formed by removing the polyethylene glycol from the SF-dECM bioink. The results showed the SF-dECM construct had a suitable mechanical strength and degradation rate, and the expression of chondrogenesis-specific genes was found to be higher than that of the SF control construct group. Finally, we confirmed that a SF-dECM construct that was designed to release TGF-β3 had the ability to promote chondrogenic differentiation of BMSCs and provided a good cartilage repair environment, suggesting it is an ideal scaffold for cartilage tissue engineering.

Multifunctional, Degradable Wearable Sensors Prepared with an Initiator and Crosslinker-Free Method

Jun Hu,Jiangping Guo,Junyan Zhao,Zixun Chen,Mulenga Kalulu,Gaojian Chen,Guodong Fu

The present zwitterionic hydrogel-based wearable sensor exhibits various limitations, such as limited degradation capacity, unavoidable toxicity resulting from initiators, and poor mechanical properties that cannot satisfy practical demands. Herein, we present an initiator and crosslinker-free approach to prepare polyethylene glycol (PEG)@poly[2-(methacryloyloxy)ethyl] dimethyl-(3-sulfopropyl) (PSBMA) interpenetrating polymer network (IPN) hydrogels that are self-polymerized via sunlight-induced and non-covalent crosslinking through electrostatic interaction and hydrogen bonding among polymer chains. The PEG@PSBMA IPN hydrogel possesses tissue-like softness, superior stretchability (∼2344.6% elongation), enhanced fracture strength (∼39.5 kPa), excellent biocompatibility, antibacterial property, reliable adhesion, and ionic conductivity. Furthermore, the sensor based on the IPN hydrogel demonstrates good sensitivity and cyclic stability, enabling effective real-time monitoring of human body activities. Moreover, it is worth noting that the excellent degradability in the saline solution within 8 h makes the prepared hydrogel-based wearable sensor free from the electronic device contamination. We believe that the proposed strategy for preparing physical zwitterionic hydrogels will pave the way for fabricating eco-friendly wearable devices.
