The pericarp of Sapindus mukorossi Gaertn was used as raw material to extract crude saponin for the preparation of graded samples (fraction a–fraction g). The peak area revealed high total saponin content in fraction g. Based on molecular weights and peak appearance times, we speculated that the main components of total saponins of S. mukorossi Gaertn. include Sapindoside B, Sapindoside A, Mukurozi-saponin E1, Sapindoside L, Mukurozi-saponin G and Sapindoside M. Analysis of the antibacterial and antineoplastic activities of different grade products showed that S. mukorossi Gaertn. and sesquiterpene glycosides display strong antimicrobial activity against dermatophytes, but not bacteria. The ethyl acetate extract displayed strong activity against both bacteria and dermatophytes, presenting blue fluorescence under UV light. In addition, the saponin mixture from S. mukorossi Gaertn. exerted a significant inhibitory effect on growth of the human breast cancer cell line, MCF-7, in vitro.

The evaluation of herbicidal activity and the isolation of plant allelopathic substances can lead to the discovery of new herbicide. This study has confirmed that the ethanol extract of S. mukorossi leaves was inhibitory to growth of Avena fatua L. and Amaranthus retroflexus L. both in petri dish experiment and pot culture assay, with the fresh weight inhibition varied between 47.60% and 62.05% at a concentration of 40 g/L of the leaf extract, being comparable to that of a commercialized natural product herbicide pelargonic acid at a concentration of 4 g/L. Five herbicidal compounds, oleanolic acid (1) , lupeol (2) , d-pinitol (3) , hexadecanoic acid (4) and octadecanoic acid (5) were isolated from the most active fraction, petroleum ether extract. D-pinitol (3) showed the highest growth inhibitory activity against both the shoot and root of A. fatua , with the half maximal inhibitory concentration (IC 50 ) values of 53.61 and 56.43 μg/mL, respectively. Furthermore, oleanolic acid (1) and d-pinitol (3) also inhibited the growth of A. retroflexus with the IC 50 value of 56.05 and 61.89 μg/mL against shoot growth, and 58.07 and 66.70 μg/mL against root growth, respectively. Thus, oleanolic acid (1) and d-pinitol (3) are the main phytotoxin constituents of S. mukorossi , and these compounds have the potential for further development as a botanical herbicide.

Wushanicaritin, a natural polyphenol compound, exerts many biological activities. This study aimed to characterize wushanicaritin glucuronidation by pooled human liver microsomes (HLM), human intestine microsomes and individual uridine diphosphate-glucuronosyltransferase (UGT) enzyme. Glucuronidation rates were determined by incubating wushanicaritin with uridine diphosphoglucuronic acid-supplemented microsomes. Kinetic parameters were derived by appropriate model fitting. Reaction phenotyping, the relative activity factor (RAF) and activity correlation analysis were performed to identify the main UGT isoforms. Wushanicaritin glucuronidation in HLM was efficient with a highCLint(intrinsic clearance) value of 1.25 and 0.69 mL/min/mg for G1 and G2, respectively. UGT1A1 and 1A7 showed the highest activities with the intrinsic clearance (CLint) values of 1.16 and 0.38 mL/min/mg for G1 and G2, respectively. In addition, G1 was significantly correlated with β-estradiol glucuronidation (r= 0.847;p= 0.0005), while G2 was also correlated with chenodeoxycholic acid glucuronidation (r= 0.638,p= 0.026) in a bank of individual HLMs (n= 12). Based on the RAF approach, UGT1A1 contributed 51.2% for G1, and UGT1A3 contributed 26.0% for G2 in HLM. Moreover, glucuronidation of wushanicaritin by liver microsomes showed marked species difference. Taken together, UGT1A1, 1A3, 1A7, 1A8, 1A9 and 2B7 were identified as the main UGT contributors responsible for wushanicaritin glucuronidation.Keywords:wushanicaritin;glucuronidation;UDP-glucuronosyltransferase;activity correlation analysis;relative activity factor;species difference
