Point-of-care (POC) tests increasingly highlight the importance of portable, cost-effective, and visually quantitative detection of biomarkers. Herein, we developed a power-free and visual signal-readout POC sensor based on the target-triggered ultrafast gelation process. In the gelation process, the target triggered the cascade reaction catalyzed by oxidase and ferrous glycinate to produce carbon radicals that immediately initiated the rapid polymerization and cross-linking of acryloylated chondroitin sulfate and dimethylacrylamide. This highly efficient enzymatic polymerization process contributed to the ultrafast generation of chondroitin hydrogel within 1 min at 25 °C. The increase in viscosity of aqueous solution resulted from hydrogel formation was then visually measured according to the distance of solution migration on a tick-labeled pH test strip, which thus realized the quantification of a target. By utilizing glucose oxidase as an oxidase model during the gelation process, this POC sensor was successfully employed in the rapid quantitative detection of glucose without the need for any auxiliary instruments. Benefiting from the specificity and stability of the enzymatic polymerization reaction, the sensor exhibited excellent performance in the detection of glucose in clinical blood samples. Moreover, the sensor was further extended to uric acid detection and enabled accurate assay in clinical urine samples, which indicated the versatility and practicability of this sensor in the POC test.

Electrochemical degradation is a commonly used strategy to remove organic contaminations in water. However, in some remote areas where electricity is insufficient, electrochemical degradation may become ineffective due to the absence of a power supply. Here, a unique self-powered electrochemical system (SPES) is proposed to accomplish the power-free degradation of tetracyclines (TCs) and organic dyes in water with the assistance of flexible magnetoelectric flag generators (MFGs). These MFGs convert wind energy into electricity. Under a wind speed of 6.3 m s −1 , a 22 × 30 cm 2 MFG yields a peak open-circuit voltage of 3.5 V and an ultra-high peak short-circuit current of ≈13 mA. A suchlike MFG charges large capacitors of 5 F to 1 V in 184 min. Powered by a series of MFGs, 5 ppm of tetracycline hydrochloride (TCH) or organic dyes in water are totally degraded with SPES in only 10 min. Moreover, the MFG is fully foldable and portable, which is well-suited for the migrating feature of the nomadic people in remote areas. The study opens up a new solution to the persistent problem of TCs contamination in natural waters in some remote regions where electricity is lacking.

Alpha-fetoprotein (AFP) is an important disease biomarker, relating to cancers such as hepatocarcinomas and gastric cancer. However, traditional methods are time-consuming, relied on bulky instruments and trained professionals, cannot satisfy the demand for low cost and point-of-care testing (POCT). In this study, a power-free POCT device was developed for the rapid and low-cost detection of AFP via one-sampling. Based on the principle of sandwich immunofluorescence, the chip is capable of automatically accomplishing on-chip mixing, labeling and capturing procedures, which only require that operator add 40 μL sample into the chip one time. The proposed device is capable of sensitively detecting human AFP in FBS with a dynamic range of 10 – 1000 ng/mL and LOD (1.88 ng/mL) within a short time of 3 min. Predictably, our method holds a great potential to be applied in the POC diagnostics of proteins, especially for some regions that are resource-limited.

A novel microfluidic chip employing power-free polydimethylsiloxane (PDMS) femtoliter-sized arrays was developed for the detection of low concentrations of protein biomakers by isolating individual paramagnetic beads in single wells. Arrays of femtoliter-sized wells were fabricated with PDMS using well-developed molding techniques. Paramagnetic beads were functionalized with specific antibodies to capture the antigens. These antigens were labeled with enzymes via conventional multistep immunosandwich approach. After suspending in aqueous solutions of enzyme substrate , the solutions were delivered to the arrays using a conventional micropipette . The aqueous solutions were introduced into the microwells by capillarity and the beads were loaded into microwells by gravity. A fluorocarbon oil was then flowed into the chip to remove excess beads from the surface of the array and meanwhile isolated the femtoliter-sized wells. All processes were achieved by conventional micropipette, without external pumping systems and valves. Finally, the arrays were imaged using standard fluorescence imaging after incubation 30 min for digital counting enzyme molecules. It was demonstrated that the chip platform possessed the performance of digital counting with a linear dynamic range from 1 aM to 1 fM for the detection of biotinylated β-galactosidase (BβG), achieving a limit of detection (LOD) of 930 zM. Using this chip, a digital immunoassay to detect Tumor Necrosis Factor α (TNF- α) was developed. Since the chip fabrication is low-cost and circumvents the surface modification, we expect it can become a new chip-based digital immunoassay platform for ultrasensitive diagnostic of biomarkers.
