Burn, trauma, and various medical conditions including bacterial infection, diabetes complication, and surgery could lead to an acute cutaneous wound and scar formation. Application of tissue glues instead of sutures could minimize the additional trauma and scar formation. Despite the countless efforts devoted to the development of high-strength tissue glues, little attention has been paid to their influence on the scar formation. Here, we report the development of a new tissue glue with excellent biocompatibility and tunable degradability for scarless wound closure. A series of catechol-containing poly(amidoamine) (CPAA) polymers were synthesized via the one-step Michael addition of dopamine and bisacrylamide. The tertiary amino group in the polymer backbone was used to introduce a zwitterionic sulfobetaine group by one-step ring-opening polymerization. The introduction of the zwitterionic sulfobetaine group could easily tune the hydrophilicity and the degradability of CPAA without influencing the density of the catechol group in the polymer. Lap-shear tests on the porcine skin demonstrated a high adhesion strength of 7 kPa at 1 h, rising to 24 kPa by 12 h. Addition of silica nanoparticles could further enhance the adhesion strength by 50%. In vivo studies further confirmed that the CPAA tissue glue could effectively accelerate the healing process of incisional wounds on the back of Sprague Dawley rats compared with suture and reduce the scar formation.

Burn wounds are characterized by complex physiological and pathophysiological response and the management of burn wounds is challenging. In situ-forming hydrogels and photothermal therapy (PTT) are promising treatments for wound healing. However, current research on biomaterials for burn treatment mainly focuses on either promoting healing or inhibiting scar formation, few studies have combined the two functions of one material. In this study, we prepared a multifunctional, in-situ cross-linked hydrogel termed CPOD hydrogel, which integrates carboxymethyl chitosan (CMCS), polyvinyl alcohol (PVA), oxidized hyaluronic acid (OHA), and dopamine (DA), coupled with photothermal therapy (PTT). In vitro experiments showed that the CPOD hydrogel had outstanding biocompatibility, suitable tissue adhesion and mechanical properties, excellent antimicrobial and antioxidant properties. This hydrogel showed PTT properties, which could produce mild heat in the early stage and higher temperatures in the later stage under NIR radiation. In the treatment of burn wounds of mice, application of CPOD hydrogel with PTT significantly promoted wound healing in the early stage and inhibited scar formation in the later stage, showing great potential for future innovation and application.

Bacterial cellulose (BC) has been widely used in tissue engineering due to its unique spatial structure and suitable biological properties. In this study, a small biologically active Arginine-Glycine-Aspartic acid-Serine (RGDS) tetrapeptide was incorporated on the porous BC surface followed by a low-energy CO2laser etching operation. As a result, different micropatterns were established on the BC surface with RGDS only anchored on the raised platform surface of the micropatterned BC (MPBC). Material characterization showed that all micropatterned structures exhibited platforms with a width of ~150 μm and grooves with a width of ~100 μm and a depth of ~300 μm, which displayed distinct hydrophilic and hydrophobic properties. The resulting RGDS-MPBC could hold the material integrity, as well as the microstructure morphology under a humid environment. In-vitro and in-vivo assays on cell migration, collagen deposition, and histological analysis revealed that micropatterns led to significant impacts on wound healing progress compared to the BC without surface-engineered micropatterns. Specifically, the basket-woven micropattern etched on the BC surface exhibited the optimal wound healing outcome with the presence of fewer macrophages and the least scar formation. This study further addresses the potential of adopting surface micropatterning strategies to promote skin wounds towards scar-free outcomes. Keywords:bacterial cellulose;lithography;micropatterns optimization;scar-free

In this study, an electret-inspired, charge-injected hydrogel called QOSP hydrogel (QCS/OD/SDI/PANI/PS/Plasma) that promotes scar-free healing of bacteria-infected burns through bioelectrical stimulation and immune modulation, is presented. The hydrogel, composed of quaternized chitosan (QCS), oxidized dextran (OD), sulfadiazine (SDI), polystyrene (PS), and polyaniline nanowires (PANI), forms a conductive network capable of storing and releasing electric charges, emulating an electret-like mechanism. This structure delivers bioelectrical signals continuously, enhancing wound healing by regulating immune responses and minimizing fibrosis. In a mouse model of second-degree burns infected with Staphylococcus aureus (SA) and Pseudomonas aeruginosa (PA), the hydrogel accelerates wound healing by 32% and reduces bacterial load by 60%, significantly inhibited scar formation by 40% compared to controls. QOSP hydrogel modulates the Th1/Th2 immune balance toward a Th1-dominant antifibrotic state through quaternized chitosan, thereby reducing collagen deposition by 35%. Electro-dielectric characterization reveals a dielectric constant of 6.2, a 34% improvement in conductivity (3.33 × 10 −5 S/m) and a 30 °C increase in thermal stability. Proteomic analysis highlights a 50% down-regulation of pro-inflammatory and pro-fibrotic pathways, suggesting a controlled immune response conducive to scar-free healing. This study underscores the potential of bioelectrically active hydrogels as a novel approach for treating infected wounds prone to scarring.

Infectious wound healing remains a complex challenge, complicated by bacterial infections, inflammation, and sensory nerve damage, which hinder healing and contribute to excessive scarring. For refractory wound healing, a temperature-programmable deformable microneedle (TPDM) is constructed, which can program at 85 °C through changes in time to maintain the shape for a corresponding period of time at 27 °C before returning to its original shape. In addition, his deformation is not temperature related, but rather caused by the separation of water phases to prevent skin burns from high temperatures and secondary impacts. The microneedles are characterized using scanning electron microscopy, transmission electron microscopy, and Nile red staining. Their antibacterial efficacy is confirmed through co-culture with methicillin-resistant Staphylococcus aureus ( MRSA ). In vitro, it promoted keratinocyte migration and facilitated sensory nerve regeneration. Furthermore, they significantly reduced scar-associated Engrailed-1 (EN-1)-positive fibroblasts and macrophages, which are key contributors to fibrotic responses. In vivo, it accelerated wound healing, reduced the accumulation of EN-1-positive fibroblasts and collagen I, and enhanced sensory nerve density and mitochondrial activity at the wound site. TPDM exhibits strong antibacterial properties against MRSA , promoting sensory nerve regeneration and reduces scarring, offering a promising therapeutic strategy for improving the healing of infectious wounds.

During the process of wound healing, the stimulation of inflammatory factors often leads to abnormal proliferation of blood vessels and collagen, ultimately resulting in scar formation. To address this challenge, we fabricate a novel dermal extracellular matrix (DECM) hydrogel scaffold loaded with ginsenoside Rg3 (Rg3) using 3D printing technology. Mesoporous silica nanoparticles (MSNs) are introduced into the system to encase the Rg3 to control its release rate and enhance its bioavailability. We systematically evaluate the biological, physicochemical, and wound healing properties of this scaffold. In vitro studies demonstrate that the hydrogel exhibits excellent biocompatibility and solid-like rheological properties, ensuring its successful printing. In vivo studies reveal that the composite hydrogel scaffolds effectively accelerate wound healing and achieve scar-free wound healing within three weeks. Histological and immunohistochemical (IHC) analyses show that the composite hydrogel scaffolds reduce the inflammatory response and inhibit excessive collagen accumulation. These combined effects underscore the potential of our approach in effectively inhibiting scar formation.

Abnormal healing following skin injury, such as slow healing and scar formation, can significantly affect an individual's life. Complex treatment methods and cumbersome instruments have reduced the efficacy of treating such diseases. In this study, a novel biocompatible liquid metal (LM) composite wound dressing (LGPU) is designed by synthesizing polyurea polyurethane (PU) and blending it with LM modified with glutathione (GSH), a bioactive three-peptide compound. The effects of external electrical stimulation (ES) on wound-induced hair follicle neogenesis are explored. The dressings exhibited a few important properties, including conductivity, high stretchability, recyclability, and, most importantly, excellent self-healing capacity, owing to the liquid nature of the LM fillers and the highly dynamic characteristics of hydrogen bonds. Furthermore, the combination therapy with LGPU and ES promoted fibroblast migration and accelerated wound healing. The wounds treated with the combination therapy fully healed in nine days, while the wounds in the blank group are still in a scabbing state. Remarkably, this treatment method can activate the regeneration and healthy growth of hair follicles at the site of injury, which is beneficial for reducing wound scarring. Collectively, this innovative therapy provides a facile strategy to accelerate skin wound healing and achieve scar-free repair.

Rapid and scar-free healing of burn wounds is an urgent clinical issue. Basic fibroblast growth factor (bFGF) has been proven to promote the healing of burn wounds by accelerating ECM remodeling and angiogenesis. However, exudates from burn wounds can accelerate bFGF degradation, thereby affecting its bioactivity. This study proposes an effective protection strategy for bFGF that involves encapsulating bFGF in nanoliposomes (bFGF-NLip) and then incorporating bFGF-NLip into a bovine serum albumin (BSA) hydrogel. This hybrid hydrogel system (bFGF-NLip@B) could maintain the activity of bFGF, achieve sustained release, and allow phospholipids and cholesterol to penetrate the skin, thereby enabling bFGF to function in the dermis. The experimental results showed that the hydrogel was injectable with good mechanical properties and biocompatibility. In a mouse scald wound model, owing to the sustained release of bFGF and skin permeation function of the nanoliposomes, the hydrogel promoted granulation formation, collagen deposition, vascular regeneration, and re-epithelialisation, ultimately accelerating wound healing. In addition, the hydrogel effectively inhibited scar formation. This system provides novel insights into the delivery of bFGF and scar-free healing of burn wounds.

Bacterial cellulose (BC) is a bio-derived polymer, and it has been considered as an excellent candidate material for tissue engineering. In this study, a crossed groove/column micropattern was constructed on spongy, porous BC using low-energy CO 2 laser photolithography. Applying the targeted immobilization of a tetrapeptide consisting of Arginine-Glycine-Aspartic acid-Serine (H-Arg-Gly-Asp-Ser-OH, RGDS) as a fibronectin onto the column platform surface, the resulting micropatterned BC (RGDS-MPBC) exhibited dual affinities to fibroblasts and collagen. Material characterization of RGDS-MPBC revealed that the micropattern was built by the column part with size of ~100 × 100 μm wide and ~100 μm deep, and the groove part with size of ~150 μm wide. Hydrating the MPBC did not result in the collapse of the integrity of the micropattern, suggesting its potential application in a highly hydrated wound environment. Cell culture assays revealed that the RGDS-MPBC exhibited an improved cytotoxicity to mouse fibroblasts L929, as compared to the pristine BC. Meanwhile, it was observed that the RGDS-MPBC was able to guide the ordered aggregation of human skin fibroblast (HSF) cells on the column platform surface, and no HSF cells were found in the groove channels. Over time, it was found that a dense network of collagen was gradually established across the groove channels. Furthermore, the in-vivo animal study preliminarily demonstrated the scar-free healing potential of the micropatterned BC materials. Therefore, this RGDS-MPBC material exhibited its advantages in guiding cell migration and collagen distribution, which could present a prospect in the establishment of “basket-woven” organization of collagen in normal skin tissue against the formation of dense, parallel aggregation of collagen fibers in scar tissue toward scar-free wound healing outcome.

Vocal fold (VF) scarring results from injury to the unique layered structure and is one of the main reasons for long-lasting dysphonia. A minimally invasive procedure with injectable hydrogels is a promising method for therapy. However, current surgical techniques or standard injectable fillers do not yield satisfactory outcomes. In this work, an injectable hybrid hydrogel consisting of oxide hyaluronic acid and hydrazide-modified waterborne polyurethane emulsion was injected precisely into the injury site and cross-linked in situ by a dynamic hydrazone bond. The prepared hydrogel displays excellent injectability and self-healing ability, showing favorable biocompatibility and biodegradability to facilitate endogenous newborn cell migration and growth for tissue regeneration. With the aim of evaluating the antifibrosis and regeneration capacity of the hybrid hydrogel in the VF scarring model, the morphology and vibration characteristics of VFs, inflammatory response, and healing status were collected. The hybrid hydrogel can decrease the inflammation and increase the ratio of collagen III/collagen I to heal damaged scar-free tissue. Fascinatingly, the mucosal wave oscillations of healing VF by injecting the hybrid hydrogel were vibrated like the normal VF, achieving functional restoration. This work highlights the utility of hybrid hydrogels consisting of synthetic biodegradable waterborne polyurethane emulsions and natural hyaluronic acid as promising biomaterials for scarless healing of damaged VFs.

Traumatic multidrug-resistant (MDR) bacterial infections are deadly threat to the public. To combat MDR bacteria, we developed a dual functional pH-sensitive hydrogel based on peptide DP7 (VQWRIRVAVIRK) and oxidized dextran (DP7-ODEX hydrogel). As an antimicrobial peptide , DP7 can synergize with many antibiotics; thus, we loaded ceftazidime into DP7-ODEX hydrogel, which showed an obvious advantage in MDR P. aeruginosa inhibition. Additionally, due to the interaction between aldehyde groups in oxidized dextran and amine groups from wound tissue, the hydrogel could extend on the irregular surface of skin defects and promote epithelial cells adhesion . DP7 could also be used as a wound-healing peptide and accelerate the healing process. We confirmed that the DP7-ODEX hydrogel exerted formidable therapeutic effects in normal or diabetic wound infection model. According to histomorphology analysis we found that DP7 hydrogel also have a scarless wound healing ability. In summary, we developed a hydrogel fabricated by the dual functional peptide DP7 that can kill multidrug-resistant bacteria colonizing the wound bed and boost scarless wound healing.

Abstract As one of the most common forms of skin injuries, skin burns are often accompanied by edema pain, suppuration of infection, slow tissue regeneration, and severe scar formation, which significantly delay wound healing as well as affect the quality of life. We prepared multifunctional electrospun poly(L-lactide-co-glycolide)/gelatin (P/G)-based dressings to synergistically harness the therapeutic benefits of peppermint essential oil (T), burn ointment (B), and Oregano essential oil (O) (P/G@TBO) for skin regeneration in punch and burn injury models. The P/G@TBO can afford the sustained release of bioactive cues for up to 72 h as well as remarkably promote cell migration ( ca. P/G@TBO, 89% vs. control group, 51%) at 24 h. The P/G@TBO membranes also showed significant angiogenic effect as well as antibacterial and anti-inflammatory properties than that of the control group in vitro. Moreover, P/G@TBO dressings enabled fast wound healing ( ca. P/G@TBO, 100% wound closure vs. control group, 95%) in a full-thickness excisional defect model up to 14 days in rats. Further evaluation of membranes in different animal models, including tail wagging model, facial itch model, and hot burn injury model showed significant pain relieve effect as well as itching and swelling relief functions during earlier stages of wound healing. Membranes were next transplanted into a scalded wound model in rats and an ear punch wound model in rabbits, which manifested antibacterial and anti-inflammatory properties and promoted re-epithelialization to achieve scarless wound healing percentage wound closure at day 28: P/G@TBO, 96% vs. control group 66%. Taken together our approach of simultaneously harnessing T, B, and O to enable multifunctionality to fibrous dressings may hold great promise for burn wound healing applications and other related disciplines.
