Bioelectricity plays a pivotal role in human physiology and pathology, particularly within the cardiovascular system. Myocardial ischemia disrupts the normal transmission of electrical signals through the heart, leading to cardiomyocyte death, ventricular remodeling, and ultimately heart failure. Electrical stimulation (ES) has been proven to be an effective strategy for treating cardiac diseases. However, invasive electrode implantation and the unique anatomical position of the heart limit the clinical application of ES therapy. Developing wireless ES systems that effectively enhance bioelectric propagation and electrical activity in the heart can significantly advance therapeutic interventions. In this study, a lead-free, biodegradable and the Food and Drug Administration approved poly-L-lactic acid piezoelectric nanofiber is designed for wireless ES to the myocardium under ultrasonic stimulation, thus promoting structural and functional recovery of cardiac tissue in murine myocardial infarction model. Further study shows that the ultrasound-driven wireless ES system significantly enhanced mitochondria function and angiogenesis in vivo and in vitro experiments. Additionally, the ES system can regulate intracellular calcium ion concentration and cardia tissue contraction rhythm of isolated neonatal rat. Collectively, the findings offer a novel approach for the treatment of heart diseases and potential cardiac pacing applications under the ultrasound-driven wireless ES system.

Nerve guidance conduits capable of wireless stimulation represent a promising approach for addressing peripheral nerve defects. However, traditional electrical stimulation methods are not sufficiently convenient and may cause secondary damage. In this study, a conductive nerve guidance conduit combined with wireless electrical stimulation using alternating magnetic fields is presented. The conduit coated with nanographene and incorporated with Fe 3 O 4 nanoparticles induces currents and creates a supportive microenvironment enhancing nerve regeneration. Finite element analysis confirms that the conduit generates electromotive force under an external alternating magnetic field. The conduit exhibits improved morphology, physicochemical properties, and conductivity by six orders of magnitude. In vitro experiments demonstrate that the conduit promotes Schwann cell proliferation, migration, and intercellular communication through microcurrents, as well as neuronal axon extension. TEM images confirm axon extension and myelin sheath thickness, indicating its high conductivity and efficiency in promoting nerve regeneration across defects. In vivo studies show that the conduit generated microcurrent using wireless electromagnetic stimulation, significantly enhancing myelin restoration, gastrocnemius muscle regeneration, motor function recovery, and nerve tissue growth, achieving results comparable to the gold-standard autograft method. Overall, this work highlights the effectiveness of electromagnetic induction in nerve repair and presents a new, non-invasive stimulation for peripheral nerve regeneration.

Bismuth sulfide (Bi2S3) enabled us to transform light signals into electrical signals via the photoelectric effect, which exhibited tremendous prospects on constructing wireless electrical stimulation for accelerating nerve regeneration. However, too rapid recombination of photogenerated electron–hole pairs weakened its photocurrent. Herein, the Bi2S3/Ti3C2Tx heterojunction was synthesized by in situ growth of Bi2S3 nanoparticles on Ti3C2Tx nanosheets and then mixed with poly-l-lactic acid (PLLA) powder to fabricate the Bi2S3/Ti3C2Tx-PLLA conduit. At the heterojunction interfaces, Ti3C2Tx with a more positive Fermi energy level could form interfacial potential difference with Bi2S3 to promote electron–hole pair separation. Meanwhile, Ti3C2Tx with excellent conductivity could provide channels for photogenerated electron transmission, thus facilitating the generation of the photocurrent. Photoluminescence and electrochemical impedance spectroscopy analysis indicated that electron–hole pair separation and electron transfer were enhanced. As a consequence, under near-infrared light radiation, the output photocurrent of Bi2S3/Ti3C2Tx-PLLA was increased from 0.48 to 1.43 μA compared to that of Bi2S3-PLLA. The enhanced photocurrent effectively promoted the differentiation of rat pheochromocytoma (PC12) into functional neurons by upregulating extracellular Ca2+ influx. Therefore, the above results demonstrated that this work provided a new perspective for wireless electrical stimulated nerve regeneration.

Secondary traumatic brain injury (TBI) induces a pro-inflammatory microenvironment that hampers neural stem cells (NSCs) therapy and tissue regeneration. To address this challenge, an immuno-piezoelectric transducer have been developed to create an anti-inflammatory immune microenvironment, deliver wireless electrical stimulation, and facilitate multimodal NSCs therapy. The immuno-piezoelectric transducer drives the polarization of microglia towards the anti-inflammatory M2 phenotype and secretion of anti-inflammatory cytokines. This modulation significantly reduces the inflammatory response, creating an optimal microenvironment for NSCs survival. Furthermore, the wireless electrical stimulation generated by ultrasound facilitates NSCs differentiation into glutamatergic and GABAergic neurons, enhances neurite complexity, upregulates synaptic proteins expression and neural integration in injured regions. The multimodal therapy demonstrates superior outcomes in restoring structural integrity, improving functional, and enhancing behavioral action in TBI rat models. This study integrates piezoelectric with immunomodulation to reprogram the neuroimmune microenvironment, providing novel therapeutic paradigm for brain injury repair.

Magnetic fields play an essential role in material science and biomedical engineering. Magnetic-responsive materials can be arranged orderly in matrix to realize the construction of an aligned scaffold under magnetic induction. However, a single topological cue is insufficient to activate neural tissue regeneration, demanding more cues to promote regeneration synergistically, such as electrical stimulation and a biomimetic matrix. Herein, we propose one-dimensional (1D) magnetoelectric Fe 3 O 4 @BaTiO 3 nanochains with controllable lengths under the regulation of a magnetic field. These nanochains can be oriented in the biomimetic hydrogel under magnetic guidance and induce the hydrogel microfiber to align along the direction of the nanochains, which is beneficial for cell-oriented outgrowth. This aligned hydrogel enabled wireless electrical stimulation mediated by magnetoelectric nanochains under magnetic stimulation, thereby activating the voltage-gated ion channel. Consequently, topological and electrical cues in this multifunctional biomimetic hydrogel synergistically enhanced the expression of neural functional proteins, facilitating synapse remodeling and neural regeneration. Predictably, the construction of multifunctional hydrogels based on low-cost and facile synthesis of magnetoelectric nanochains is an emerging patient-friendly and effective therapeutic strategy for neural or other tissue regeneration. Statement of significance A facile and controllable magnetic strategy is established to manipulate 1D nanomaterial growth, matrix topography, and wireless electrical stimulation of cells. First, the magnetic-assisted interface co-assembly was used to control the length of Fe 3 O 4 @BaTiO 3 nanochains with enhanced magnetoelectric effect. Then, the motion of the magnetic-induced nanochains guided the orientation of nanofibers in a 3D biomimetic hydrogel matrix. Finally, wireless electrical signals and topological cues in the biomimetic matrix synergistically promoted orderly aligned cell outgrowth and membrane depolarization by Ca 2+ influx, thus enhancing nerve cell synaptic plasticity and functional expression. Consequently, this work provides a conceptual strategy from material design to extracellular matrix signal manipulation and synergistic induction of tissue regeneration.

Electrical stimulation is considered an effective way to accelerate peripheral nerve cell growth. However, reliance on external wires and a power supply restricts its further applications. In this work, a wireless magnet-powered electrical stimulation system was constructed based on the electromagnetic induction effect. Specifically, amino-functionalized conductive MXene nanosheets were first wrapped on the PLLA particles’ surface by electrostatic assembly. Then the conductive NH2-MXene/PLLA conduit was fabricated by laser additive manufacturing. Triggered by a rotating magnetic field, the conduit could act as a coil to cut magnetic induction lines, converting magnetic energy into electrical energy and thereby achieving wireless electrical stimulation. The results indicate that the conduit presented an excellent conductivity of 8.44 S m−1, benefiting from its formed conductive network structure. Under excitation of the rotating magnetic field, the conduit generated an electric current of 10 μA, which is in the appropriate range for nerve cell growth. An in vitro cell test confirmed that the generated current effectively enhanced the PC12 cell proliferation, neurite growth and differentiation-related mRNA (Nestin, MAP2 and Tuj1) expression. Moreover, it also promoted PC12 cells to differentiate into mature neurons, evidenced by calcium sparks produced in PC12-derived neurons. Collectively, this wireless electrical stimulation system offers a new perspective on peripheral nerve repair.

The induction of dental pulp stem cells (DPSCs) into odontogenic differentiation is a cutting-edge method of dental pulp regeneration treatment. However, it remains a challenge to develop biomaterials and therapies that can induce odontogenic differentiation. Here, we propose a wireless electrical stimulation strategy to induce DPSCs odontogenic differentiation via K 0.5 Na 0.5 NbO 3 piezoelectric nanoparticles (KNN) and polarized KNN (PKNN), which can be endocytosed by DPSCs. In vitro, several odontogenic differentiation indexes were also increased in DPSCs treated with KNN and PKNN, and the increase was more obvious in the PKNN group. Intracellular wireless electrical field promoted mitochondrial calcium concentration via mitochondrial calcium unidirectional transporter (MCU), increased the production of adenosine triphosphate (ATP), and induced odontogenic differentiation through the activation of the cAMP-PKA signaling pathway. In vivo, dental pulp-like tissue was induced by electrical stimulation wirelessly with KNN and PKNN, which was more clinically friendly compared with the wired device, and the induction was more obvious in the PKNN group consistent with in vitro experiments. In conclusion, this work demonstrates the potential of PKNN as an effective stimulus that can induce odontogenic differentiation of DPSCs and be applied to dental pulp regeneration treatment.

Electrical stimulation (ES) offers significant advantages in modulating the behavior of stem cells on conductive scaffolds for neural tissue engineering. However, it is necessary to realize wireless ES to avoid the use of external wires in tissues. Thus, herein, a strategy is reported to develop a stem cell scaffold that allows wireless ES. A conductive annular graphene substrate is designed and grown by chemical vapor deposition; this substrate is used as a secondary coil to achieve wireless ES via electromagnetic induction in the presence of a primary coil. The substrate shows excellent biocompatibility for the culture of neural stem cells (NSCs). The results indicate that the applied wireless ES enhances neuronal differentiation, facilitates the formation of neurites, and does not substantially affect the viability and stemness maintenance of NSCs. Collectively, this system provides a strategy for achieving synergy between wireless ES and conductive scaffolds for neural regenerative medicine, which can be further utilized for the regeneration of other tissues.

Electrical stimulation is regarded pivotal to promote repair of nerve injuries, however, failed to get extensive application in vivo due to the challenges in noninvasive electrical loading accompanying with construction of biomimetic cell niche. Herein, a new concept of magneto responsive electric 3D matrix for remote and wireless electrical stimulation is demonstrated. By the preparation of magnetoelectric core/shell structured Fe 3 O 4 @BaTiO 3 NPs-loaded hyaluronan/collagen hydrogels, which recapitulate considerable magneto-electricity and vital features of native neural extracellular matrix, the enhancement of neurogenesis both in cellular level and spinal cord injury in vivo with external pulsed magnetic field applied is proved. The findings pave the way for a novel class of remote controlling and delivering electricity through extracellular niches-mimicked hydrogel network, arising prospects not only in neurogenesis but also in human–computer interaction with higher resolution.

Pathological angiogenesis frequently occurs in tumor tissue, limiting the efficiency of chemotherapeutic drug delivery and accelerating tumor progression. However, traditional vascular normalization strategies are not fully effective and limited by the development of resistance. Herein, inspired by the intervention of endogenous bioelectricity in vessel formation, we propose a wireless electrical stimulation therapeutic strategy, capable of breaking bioelectric homeostasis within cells, to achieve tumor vascular normalization. Polarized barium titanate nanoparticles with high mechano-electrical conversion performance were developed, which could generate pulsed open-circuit voltage under low-intensity pulsed ultrasound. We demonstrated that wireless electrical stimulation significantly inhibited endothelial cell migration and differentiation in vitro . Interestingly, we found that the angiogenesis-related eNOS/NO pathway was inhibited, which could be attributed to the destruction of the intracellular calcium ion gradient by wireless electrical stimulation. In vivo tumor-bearing mouse model indicated that wireless electrical stimulation normalized tumor vasculature by optimizing vascular structure, enhancing blood perfusion, reducing vascular leakage, and restoring local oxygenation. Ultimately, the anti-tumor efficacy of combination treatment was 1.8 times that of the single chemotherapeutic drug doxorubicin group. This work provides a wireless electrical stimulation strategy based on the mechano-electrical conversion performance of piezoelectric nanoparticles, which is expected to achieve safe and effective clinical adjuvant treatment of malignant tumors.

Electrical stimulation (ES) can restore motor function after spinal cord injury (SCI). However, traditional intraspinal ES has many disadvantages, such as the need for a complex circuitry device, an external power source, and a second surgery to remove the implant. Piezoelectric materials have received increasing attention due to their potential to convert ambient mechanical energy into ES without an external power source or implantation of electrodes. Herein, a novel polydopamine (PDA) coated barium titanate (BaTiO 3 ) nanoparticles (BaTiO 3 @PDA) incorporated silk fibroin (SF) hydrogel (SFBT) was established. The SFBT hydrogel was crosslinked by horseradish peroxidase (HRP)/hydrogel peroxide (H 2 O 2 ) to integrate the piezoelectric potency of BaTiO 3 with the thermal sensitivity of HRP/H 2 O 2 for SCI recovery. The hydrogel containing 5% (w/v) BaTiO 3 @PDA nanoparticles (SFBT-5), which was selected for in vivo study, exhibited a short gelation time (5 min), suitable storage modulus (925 ± 35 Pa), and wireless ES (average current of 124 nA). Moreover, Basso-Beattie-Bresnahan (BBB) scores test and footprint analysis demonstrated that the SFBT-5 hydrogel successfully enhanced motor functional recovery of SCI. In histopathological assessments, the SFBT-5 hydrogel significantly accelerated spinal cord healing, as indicated by smaller lesion cavities (∼16.7% of the SCI group). Meanwhile, the SFBT-5 hydrogel accelerated neurogenesis, facilitated axon regrowth and synapse formation, and promoted remyelination. Overall, this study highlights the potential of piezoelectric hydrogels for SCI regeneration.

Nerve guidance conduits (NGCs) are considered as promising treatment strategy and frontier trend for peripheral nerve regeneration, while their therapeutic outcomes are limited by the lack of controllable drug delivery and available physicochemical cues. Herein, novel aligned piezoelectric nanofibers derived hydrogel NGCs with ultrasound (US)-triggered electrical stimulation (ES) and controllable drug release for repairing peripheral nerve injury are proposed. The inner layer of the NGCs is the barium titanate piezoelectric nanoparticles (BTNPs)-doped polyvinylidene fluoride-trifluoroethylene [BTNPs/P(VDF-TrFE)] electrospinning nanofibers with improved piezoelectricity and aligned orientation. The outer side of the NGCs is the thermoresponsive poly(N-isopropylacrylamide) hybrid hydrogel with bioactive drug encapsulation. Such NGCs can not only induce neuronal-oriented extension and promote neurite outgrowth with US-triggered wireless ES, but also realize the controllable nerve growth factor release with the hydrogel shrinkage under US-triggered heating. Thus, the NGC can positively accelerate the functional recovery and nerve axonal regeneration of rat models with long sciatic nerve defects. It is believed that the proposed US-responsive aligned piezoelectric nanofibers derived hydrogel NGCs will find important applications in clinic neural tissue engineering.
