Self-Assembly of Bidirectional-Signal Nanoclusters for Two miRNAs Simultaneously Monitoring in Single Cancer Cells

Yuting Jiao,Mengting Zhu,Xiaomeng Wang,Zhen Zhang,Shusheng Zhang

A novel finding is herein reported that the bidirectional-signal nanoclusters self-assemble simultaneously on the nanoflowers as a result of four-way folding (FWF) nanoprobes and DNA rolling circle replication reactions. The functionalized FWF nanoprobe containing the identification region for two targets monitoring and the trigger region for amplification signals is first used to activate the clustered amplification for two cancer-related microRNAs (miRNAs) assays in single cells. Furthermore, the self-assembled nanoclusters with two-way amplification signals can provide more reliable information in situ in individual cell. Importantly, this new method can significantly distinguish cancer cells from normal cells and identify changes in the expression levels of cancer-related miRNAs for single cells. These findings have exciting potential to provide new opportunities for detection and enhance the accuracy of early disease diagnosis.

3D Biomimetic Hydrangea-Like BiOCl and PtNi Nanocube-Based Electrochemical Immunosensor for Quantitative Detection of CA19–9

Yarui An,Lin Song,Xiaoyan Chen,Chenming Ni,Kuanzheng Mao,Leijing Zhu,Yingying Gu,Yuqing Miao,Bin Song,Hongyun Ma

This work took advantage of 3D biomimetic hydrangea-like bismuth oxychloride (BiOCl) and PtNi nanocubes to fabricate a facile electrochemical immunosensor for the detection of CA19–9. Herein, APTES functionalized BiOCl (APTES-BiOCl) with large specific surface areas can provide eco-friendly microenvironment for biomolecules. The PtNi nanocubes prepared by the classic hydrothermal method have good conductivity, which can facilitate electron transfer highly efficient. Meanwhile, PtNi nanocubes can immobilize the anti-CA19–9 via covalent Pt-N bonds to anchor a great number of biomolecules more firmly. In addition, reduced graphene oxide (rGO) with excellent conductivity was implemented as a matrix, which greatly promoted electron transfer at the sensing interface. Under the optimal conditions, the proposed immunosensor can detect CA19–9 sensitively, from which a linear range of 0.01–150 U·ml–1 was derived accompanied with a low detection limit of 0.0034 U·ml–1. The results illustrated that the designed immunosensor exhibited excellent specificity, good reproducibility, and satisfactory stability. Moreover, it can be applied in the detection of real samples, which indicated that the immunosensor has great potential for early screening and diagnosis of diseases.

One-step and ultrasensitive ATP detection by using positively charged nano-gold@graphene oxide as a versatile nanocomposite

Xue Ning,Wu Shujie,Li Zongbing,Miao Xiangmin

A versatile nanocomposite was simply prepared based upon the electrostatic adsorption of positively charged gold nanoparticles with negatively charged graphene oxide ( nano-gold@GO), and utilized as a novel fluorescence quenching platform for ultrasensitive detection of adenosine triphosphate (ATP). In the designed system, DNA-stabilized Ag nanoclusters (DNA/AgNCs) were used as fluorescent probes, DNA duplex was formed in the presence of ATP, and they can electrostatically adsorb onto the surface of nano-gold@GO to quench the fluorescence signal. Upon the addition of exonuclease III (Exo III), the DNA duplex would be hydrolyzed into DNA fragments and resulted in the recovery of the fluorescence signals due to the diffusion of AgNCs away from nano-gold@GO. Based on these, sensitive detection of ATP was realized with a detection range of 5.0 pM–20 nM. Notably, a good recovery in the range of 94–104% was obtained when detecting ATP in human serum samples, indicating a promising application value in early disease diagnosis. A functional positively charged nano-gold@graphene oxide was fabricated and utilized as an enhanced fluorescence quenching platform for the detection of ATP, coupled with exonuclease III-assisted signal amplification.

Programmable CRISPR-Cas9 microneedle patch for long-term capture and real-time monitoring of universal cell-free DNA

Yang Bin,Kong Jilie,Fang Xueen

Recent advances in biointerfaces have led to the development of wearable devices that can provide insights into personal health. As wearable modules, microneedles can extract analytes of interest from interstitial fluid in a minimally invasive fashion. However, some microneedles are limited by their ability to perform highly effective extraction and real-time monitoring for macromolecule biomarkers simultaneously. Here we show the synergetic effect of CRISPR-activated graphene biointerfaces, and report an on-line wearable microneedle patch for extraction and in vivo long-term monitoring of universal cell-free DNA. In this study, this wearable system enables real-time monitoring of Epstein-Barr virus, sepsis, and kidney transplantation cell-free DNA, with anti-interference ability of 60% fetal bovine serum, and has satisfactory stable sensitivity for 10 days in vivo. The experimental results of immunodeficient mouse models shows the feasibility and practicability of this proposed method. This wearable patch holds great promise for long-term in vivo monitoring of cell-free DNA and could potentially be used for early disease screening and prognosis.

Constructing ultrasensitive biosensor for biomarker detection of early diseases through G-quadruplex probe coupled with aggregation-induced electrochemiluminescence platinum(II) complexes

Tingyi Huang,Li Tan,Wenyi Zhang,Ying Ma,Ziqi Lian,Benlin Hong,Jianqiang Hu

Highly sensitive and simple biomarker determination is crucial for diagnosis and treatment of early diseases. However, it remains a tremendous challenge for rapid, accurate and real time detection of biomarkers at the early stage of diseases due to its extremely low content and serious interference. Herein, we have successfully constructed an ultrasensitive electrochemiluminescence (ECL) biosensor based on 2,6-bis(2-benzimidazolyl)pyridine platinum(II) (BZIMPY-Pt) complexes with aggregation-induced electrochemiluminescence (AIECL) phenomenon, which was utilized to detect miRNA-155 at early diseases. The BZIMPY-Pt complexes with AIECL properties and had high affinity with G-quadruplex DNA compared to other square planar pyridyl Pt(II) complexes, which exhibited 13-fold enhancements in ECL intensity in the aggregated state. The constructed biosensor possessed high sensitivity, wide linear range (1 fM-10 nM), low limit of detection (∼0.22 fM), high selectivity and good stability and reproducibility for miRNA-155 detection. The ECL biosensor was successfully applied to determine miRNA-155 in the serum samples of early-stage diagnose. This AIECL-based strategy overcomes traditional limitations in low-abundance biomarker analysis through synergistic signal amplification and background suppression mechanisms, which is potential for clinical diagnosis of early diseases.

Dual-ratiometric magnetic resonance tunable nanoprobe with acidic-microenvironment-responsive property to enhance the visualization of early tumor pathological changes

Cao Rong,Tang Ning,Zhu Yi,Chen An,Li Yumeng,Ge Renbin,Li Yuan,Huang Zhongyi,Guo Jiajing,Deng Jiali,Lu Hongwei,Lu Ziwei,Forgham Helen,Davis Thomas P.,Qiao Ruirui,Wang Zhongling

The development of microenvironment-responsive nanoprobes has shown great promise for use in magnetic resonance imaging (MRI), with the advantage of significantly improved specificity and good biocompatibility. However, the clinical application of responsive probes is hampered by a lack of biological sensitivity for early molecular diagnostics and visualizing microenvionment of metabolism reprogramming in tumor progression. Here, we report on a dual-ratiometric magnetic resonance tunable (DMRT) nanoprobe designed by crosslinking different ratios of transferrin chelating gadolinium and superparamagnetic nanoparticles, complexed to a pH responsive biocompatible polymer. This dually activatable nanoprobe enables pH-dependent tumor microenvironment visualization, providing exceptional quantitative pathophysiological information in vitro and in vivo . When used in combination with dual-contrast enhancement triple subtraction imaging technique (DETSI), this smart nanoprobe guarantees the diagnosis of early-stage diseases. We envisage that this novel integrated nanoplatform will provide a new paradigm for the clinical translation of robust DMRT nanoprobes for early disease detection and staging, as well as microenvironment visualization and disease progression monitoring.

Ultrasensitive detecting of dopamine in complex components by field effect transistor sensor based on the synergistic enhancement effect and overcoming debye length limitations

Meng Tian,Chonghui Li,Renzhong Yu,Congcong Shen,Jihua Wang,Jiajun Lu,Guofeng Liu,Zhenxing Wang,Tiejun Wang,Xiaofei Zhao,Zhen Li,Le Li,Baoyuan Man,Shicai Xu,Chao Zhang

Field effect transistor (FET) has attracted high attention in biomolecules detection. However, the sensitivity of FET-based biosensors is often limited by the charge screening effect. In addition, the facile and ultra-sensitive detection for small biomolecules, which possess weak charge or electroneutral, remains to be studied. Here, the self-assembled monolayer AuNPs/three-dimensional (3D) crumpled graphene FET is structured for a biosensor by shrinking flexible polystyrene (PS) films through heat treatment method and realized label-free and ultra-sensitive detection of dopamine (DA) using DA aptamer as probes immobilized on the AuNPs/3D crumpled graphene surface. The nanoscale deformation caused by thermal expansion effect of flexible graphene/PS can effectively reduce charge screening and the synergistic application of crumpled graphene and AuNPs can promote electron transfer and the graphene carrier concentration, leading to conductivity increase and hydrophilicity enhancement of 3D graphene. In addition, the binding of DA aptamer to DA will cause conformational changes of the aptamer molecule, which affects the charge transport properties of the sensor, thus improving its selectivity and stability. The biosensor can also easily distinguish interfering substances for DA detection in complex components (PBS, human urine, and fetal calf serum) with the detection limits as low as 60, 240 and 316 zM (10 −19 –10 −11  M), respectively. DA released from exocytosis induced by K + stimulation was selectively detected. The results show that the biosensor can be used as an excellent tool for ultrasensitive molecular recognition and detecting the effects of exogenous reagents on living cells, which is promising for clinical diagnosis and early disease prevention.

Dual-Analyte Sensing with a Molecularly Imprinted Polymer Based on Enhancement-Mode Organic Electrochemical Transistors

Panpan Hao,Rufeng Zhu,Yang Tao,Wei Jiang,Xue Liu,Yan Tan,Yuedan Wang,Dong Wang

Novel enhancement-mode organic electrochemical transistors (OECTs) have been prepared by poly(3, 4-ethylenedioxythiophene)-poly(styrenesulfonate) de-doped polyethylenimine on the multi-walled carbon nanotube-modified viscose yarn. The fabricated devices exhibit low power consumption with a high transconductance of 6.7 mS, rapid response time < 2 s, and excellent cyclic stability. In addition, the device has washing durability and bending and long-term stability suitable for wearable applications. Biosensors based on enhancement-mode OECTs for the selective detection of adrenaline and uric acid (UA) are developed by using molecularly imprinted polymer (MIP)-functionalized gate electrodes. The detection limits of adrenaline and UA analysis are as low as 1 pM, with the linear ranges of 0.5 pM to 10 μM and 1 pM to 1 mM, respectively. Moreover, the sensor based on enhancement-mode transistors can efficiently amplify the current signals according to the modulation of the gate voltage. The MIP-modified biosensor has high selectivity in the presence of interferents and desirable reproducibility. Additionally, due to the wearable nature of the developed biosensor, this sensing tool has the capability of being integrated with fabrics. Therefore, it has successfully been applied in textiles for the determination of adrenaline and UA in artificial urine samples. The excellent recoveries and rsds are 90.22–109.05% and 3.97–6.94%, respectively. Ultimately, these sensitive, low-power, wearable, and dual-analyte sensors help to develop non-laboratory tools for early disease diagnosis and clinical research.
