Spodoptera litura larvae are attracted by HvAV-3h-infected S. litura larvae-damaged pepper leaves

Lang Ming,Yuan-Wen Du,Ge-Ge Yuan,Qi Su,Xiao-Bing Shi,Huan Yu,Gong Chen

Background Herbivore-induced plant volatiles (HIPVs) are important self-defense outputs of pepper plants to resist insect pests. Ascoviruses are pathogenic to the larvae of most lepidopteran vegetable pests. However, whether Heliothis virescens ascovirus 3h (HvAV-3h)-infected Spodoptera litura larvae can change pepper leaf HIPVs is not well understood. RESULTS Spodoptera litura larvae preferred S. litura -infested leaves, and this preference was stronger with longer duration of S. litura infestation. In addition, S. litura larvae significantly chose pepper leaves damaged by HvAV-3h-infected S. litura over the healthy pepper leaves. Results also showed that S. litura larvae preferred leaves mechanically damaged and treated with oral secretions from HvAV-3h infected- S. litura larvae in a simulation test. We captured the volatiles emitted by leaves under six treatments. Results showed that the volatile profile changed with the different treatments. Testing of volatile blends, prepared to the proportion released showed that the blend from simulated HvAV-3h-infected S. litura larvae-damaged plants was the most attractive to S. litura larvae. Further, we also found that some of the compounds significantly attracted S. litura larvae at specific concentrations. CONCLUSION HvAV-3h-infected S. litura can alter the release of HIPVs in pepper plants and thus become more attractive to S. litura larvae. We speculate that this may be due to alterations in the concentration of some compounds (such as geranylacetone and prohydrojasmon) affecting the behavior of S. litura larvae. © 2023 Society of Chemical Industry.

Heat stress affects potato's volatile emissions that mediate agronomically important trophic interactions

Asim Munawar,Yadong Zhang,Jian Zhong,Yang Ge,Amr S. Abou El-Ela,Zhiyao Mao,Eric S. Ntiri,Li-Juan Mao,Zengrong Zhu,Wenwu Zhou

Potato, a cool-weather crop, emits volatile organic compounds (VOCs) which attract the specialist herbivore, Phthorimaea operculella , but also this herbivore's parasitic wasp, Trichogramma chilonis , an important biocontrol agent. What happens to this trophic system when heat stress challenges this agro-ecosystem? We studied how high temperature (HT) pre-treatments influence potato's VOC emissions and their subsequent effects on the preferences of insects, as evaluated in oviposition assays and Y-tube olfactometers. HT pre-stressed plants were less attractive to P. operculella adult moths, which were repelled by HT VOCs, but increased the recruitment of the parasitoid, T. chilonis , which were attracted. VOC emissions, including the most abundant constituent, ß-caryophyllene, were enhanced by HT treatments; some constituents elicited stronger behavioural responses than others. Transcripts of many genes in the biosynthetic pathways of these VOCs were significantly enhanced by HT treatment, suggesting increases in de novo biosynthesis. HT increased the plant's stomatal apertures, and exogenous applications of the hormone, ABA, known to suppress stomatal apertures, reduced leaf volatile emissions and affected the HT-altered plant attractions to both insects. From these results, we infer that HT stress affects this plant-insect interaction through its influence on VOC emissions, potentially decreasing herbivore ovipositions while increasing ovipositions of the parasitoid.

Comparative assays revealed distinct toxicity characterizations between pymetrozine and flonicamid

Tengfei Liu,Jianguo Niu,Yuying Gao,Xuan Liu,Jianya Su

Flonicamid is an insecticide with a unique mode of action, primarily inhibiting the feeding behavior of sap-sucking insects. However, its molecular target remains controversial. Reports indicate that both flonicamid and pymetrozine affect insect behavior and mobility. To investigate further, we compared the susceptibility of three insect species ( Myzus persicae , Nilaparvata lugens , and Drosophila melanogaster ) to flonicamid and pymetrozine under two different feeding postures. Results showed that feeding posture did not affect the survival curves or mean lifespans of the three insects. However, insect lifespans were significantly reduced under exposure to either insecticide compared to untreated controls. The effects of the two insecticides on susceptibility and lifespan under different feeding postures were markedly different: feeding posture significantly influenced the sensitivity of aphids, planthoppers, and fruit flies to pymetrozine but had minimal effects on the sensitivity of aphids and planthoppers to flonicamid. Flonicamid had only a minor impact on fruit flies' sensitivity. Survival curve analysis also revealed significant differences under pymetrozine stress, while no such differences were observed under flonicamid stress. These findings indicate that flonicamid exerts different toxicological effects on target insects (aphids and planthoppers) versus non-target insects (fruit flies), depending on feeding posture. This suggests that the toxicological mechanisms of flonicamid differ substantially between target and non-target organisms, with implications that findings in non-target insects may not directly apply to target pests. Our study provides valuable insights and directions for further research into flonicamid's mode of action, aiding in elucidating its underlying mechanisms.
