Antagonistic yeast and trehalose-enriched gelatin film: A bioactive antifungal packaging film for cherry tomato preservation

Honglian Guo,Yue Xu,Haiyi Chen,Xinyu Si,Menghan Zhou,Enlong Zhu

To extend the shelf-life of cherry tomatoes infected with Botrytis cinerea , this study analyzed the effect of Pichia membranifaciens (PM) on a composite film of gelatin (Gel) and trehalose (Tre), and explored the effect of different concentrations of trehalose on the yeast in the bioactive film and its control effect on cherry tomatoes. Scanning electron microscopy showed that Tre/Gel-based film can be applied as effective carriers to protect the cellular integrity of PM. Fourier-transform infrared spectroscopy, thermogravimetric analysis, and tests of mechanical properties showed that there was interaction between PM and the Tre/Gel base film, which maintained the thermal stability of the base film. The addition of PM reduced the water vapor permeability, hygroscopicity, and moisture content of the Tre/Gel base film, effectively extending the active film's storage time. This study also explored the growth curve of PM and found that 15% trehalose was effective in maintaining PM activity and could result in a 30-day survival rate of 62% for PM in Tre/Gel films, with 72.41% inhibition of B. cinerea . Finally, the PM/Tre/Gel film significantly reduced the incidence of decay in cherry tomatoes and improved the storage quality and fungal resistance of the fruit. Thus, PM/Tre/Gel films may be applied as a new antifungal packaging material for the preservation of fresh fruit.

Fabrication of double crosslinked chitosan/gelatin membranes with Na+ and pH dual-responsive controlled permeability

Jin Liu,Shunyi Wang,Ke Xu,Zaibi Fan,Panpan Wang,Zijun Xu,Xueqin Ren,Shuwen Hu,Zideng Gao

A double crosslinked membrane based on chitosan/gelatin with controlled release properties was synthesized in aqueous system using potassium pyroantimonate and genipin as the ionic crosslinker and covalent crosslinker, respectively. The membranes were characterized using FT-IR, UV–vis spectra, and SEM. In addition, the thermal stability, contact angle, mechanical and controlled release properties were measured. Double-crosslinking significantly improved the hydrophobicity, thermal stability, and tensile strength of the membrane. The permeabilities of various plant nutrients, including urea and plant growth regulators (PGRs), were evaluated. Under the optimized conditions, the lowest flux (J) of urea was 2.89 mg/ (cm 2 ·h). Urea and PGR release were sustained for up to 3 and 10 days, respectively. The release of agrochemicals in this study followed the Weibull model, demonstrating the Na + -responsive and pH-responsive permeability of the films. This dual-responsive film has potential applications in salinized barrier soil, water treatment, separations, and other membrane applications.

The effect of procyanidine crosslinking on the properties of the electrospun gelatin membranes

Zhengjian Chen,Liqun Wang,Hongliang Jiang

In this study, the effect of different crosslinkers including glutaraldehyde (GTA), genipin (GIP) and procyanidine (PA) on the properties of the electrospun gelatin membranes was compared. The water-resistant ability of the membranes could be significantly improved after being crosslinked with PA at T > 40 °C. In contrast with GTA and GIP, the PA-crosslinking process did not apparently affect the fibrous structure, and induced the lowest shrinkage of the membranes. At the concentration of 5% of PA, the ultimate tensile strength and elongation of the hydrated membrane were 0.87 MPa and 148%, respectively, which were higher than those of the GIP-crosslinked counterparts. In addition, the PA-crosslinked membranes displayed the highest resistance to pepsin degradation, and fibroblast cells could migrate deeper into the interior of the membranes due to the good preservation of the fibrous structure during the cell culture process.

Tannic acid cross-linked Poly(lactic-co-caprolactone/Gelatin electrospun nanofibrous membranes for blueberry preservation

Yiting Ye,Jiaqi Liu,Tingzi Yan,Yujia Yan,Jiangkai Fan,Xiao Zhang,Li Gan,Hui Chai,Guoying Zhou

Due to the huge waste caused by fruit mildew during transportation and storage, there is an urgent need to develop novel packing materials with antibacterial and degradable properties. Herein, poly(lactic-co-caprolactone)/Gelatin/Tannic acid (PLCL/GEL/TA) membranes were fabricated by electrospinning of PLCL and GEL to obtain PLCL/GEL membranes, followed by cross-linking with TA. The microstructures of PLCL/GEL/TA membranes were characterized by scanning electron microscope (SEM). The mechanical and wetting properties were evaluated by Instron universal material testing and Drop shape analysis system, respectively. The antibacterial and antioxidant properties were determined. The preservation properties of the membranes were evaluated using blueberry as a model fruit. The results showed that smooth and uniform microstructure was formed on all electrospun membranes. The PLCL/GEL/TA membranes exhibited enhanced mechanical property, wettability, biodegradability, antioxidant and antibacterial properties, when compared to PLCL/GEL membranes without TA treatment. In addition, these increased properties showed positive correlation with TA content. Besides, TA was slowly released from the PLCL/GEL/TA membranes, in a controlled and sustained manner. Moreover, the cytotoxicity assays confirmed the non-toxic nature of these membranes. Finally, the PLCL/GEL/TA membranes effectively inhibited decay degree, weight loss, and extended the shelf life of blueberries. Overall, the antioxidant, antibacterial, biodegradable and biocompatible PLCL/GEL/TA membranes exhibited remarkable preservation performance, demonstrating significant potential as novel packing materials.

Flavor, antimicrobial activity, and physical properties of composite film prepared with different surfactants

Xin Li,Zong-Cai Tu,Xiao-Mei Sha,Yun-Hua Ye,Zhong-Ying Li

Different surfactants (lecithin, Tween-20, and Tween-80) were added in composite film during the preparation. Flavor, antimicrobial activity, and physical properties of ginger essential oil -gelatin film were investigated, in order to study the effect of surfactants on the properties of film. The flavor of GEO was not detected in the film prepared with Tween-20 and film prepared with Tween-80, and these two films exhibited stronger antimicrobial activity; film prepared with lecithin possessed higher value in thickness, elongation at break, water solubility, Δ E and opacity, lower value in water vapor property, and tensile strength; attenuated total reflectance-Fourier transform infrared spectrum results suggested, Tween-20 and Tween-80 enhanced the strength of covalent bond, and lecithin weakened the strength of hydrogen bond; and the result of scanning electron microscope showed that Tween-20 and Tween-80 improved the dispersion of oil droplets in film. Therefore, this study suggested that surfactants had an influence on the physical properties and molecular structure of a resulting film; in addition, Tween-20 and Tween-80 could reduce the flavor of GEO in film, improving the antimicrobial activity of film.

The modified electrospun gelatin membrane with in-situ coated silver layer for flexible TENG device

Wen Shen,Fen Ao,Xuemei Ge,Yan Zheng,Xiaomin Luo

Triboelectric nanogenerator (TENG) has shown great potential in real-time monitoring of individual physiological state. Conductive metal nanofibers maintain good stability and mechanical strain as electrodes. Herein, a simple and safe strategy was provided to prepare electrospun gelatin membranes with in-situ coated silver layer for flexible TENG devices. Electrospun gelatin membranes were prepared by a one-pot method of dopamine-grafted gelatinmodification and physical modification through blending with polyvinyl alcohol (PVA). The silver coated electrospun membrane was constructed using the reductive immobilization of silver by dopamine. The content of silver and the output signal of the TENG can be further controlled using bio-crosslinked gelatin membranes. The electrospun gelatin membranes with immobilized silver layers did not show significantly cytotoxic to fibroblasts. The tensile strength of the membranes crosslinked reached 4 MPa, which was more flexible than the membranes uncrosslinked. The membranes showed significant inhibition against Staphylococcus aureus ( S. aureus ) and Pseudomonas aeruginosa ( P. aeruginosa ). The triboelectric output performance of the membrane can be controlled by adjusting the crosslinking method, which providing an effective strategy for implementing TENG in the monitoring of the wound healing process.

Reversible cross-linking of gelatin by a disulphide-containing bis-succinimide for tunable degradation and release

Shengbin He,Jingtong Wang,Zhao Li,Yongqiang Cao,Xueping Ning,Jian Sun,Quanzhi Chen,Min Ling

Generally, gelatin was irreversibly cross-linked by chemical reagents to improve its water-resistance. However, few chemical reagents meet both the requirements of high cross-inking efficiency and tunable degradation. Here a reversible cross-linker, disulphide-containing bis-succinimide, was synthesized and used to control the cross-linking and degradation of edible gelatin film. Mixture of the gelatin and cross-linker for 120 min generated gelatin films that could preserve their morphology in 37 ℃ warm water for above 40 days. The gelatin film changed its microstructure from net to tightness after the cross-linking, thus facilitating the embedding of the targeted molecule into the gelatin material. The degradation of the cross-linked gelatin film and the release of its inclusion could be controlled by biocompatible glutathione. This work provides a good method for preparing modified gelatin with promising water-resistance, good biocompatibility, and tunable degradation for food/biomedical engineering applications.

Chitosan-ginger essential oil nanoemulsions loaded gelatin films: A biodegradable material for food preservation

Xiaohan Wang,Zhaohui Xue,Yijie Sun,Bo Peng,Caie Wu,Xiaohong Kou

The alarming issue of food waste, coupled with the potential risks posed by petroleum-based plastic preservation materials to both the environment and human health necessitate innovative solutions. In this study, we prepared nanoemulsions (NEs) of chitosan (CS) and ginger essential oil (GEO) and systematically evaluated the effects of varying NEs concentrations (0, 10 %, 30 %, 50 %) on the physicochemical properties and biological activities of gelatin films. These films were subsequently applied to blueberry preservation. The scanning electron microscopy confirmed that the NEs were well-integrated with the Gel matrix, significantly enhancing the performance of the Gel films, including improvements of mechanical properties (tensile strength from 7.71 to 19.92 MPa; elongation at break from 38.55 to 113.65 %), thermal, and barrier properties (water vapor permeability from 1.52 × 10 −9 to 6.54 × 10 −10  g·m/Pa·s·m 2 ). The films exhibited notable antibacterial and antioxidant activities due to the gradual release of GEO, thereby extending the storage life of blueberries. Moreover, the prepared composite films demonstrated excellent biodegradability and environmental friendliness, with the majority of the material decomposing within 30 days under soil microbial action. In conclusion, the active films loaded with NEs exhibit superior performance and hold significant potential for developing biodegradable materials for food preservation.
