4D Printing of Overall Radiopaque Customized Bionic Occlusion Devices

Cheng Lin,Zhipeng Huang,Qinglong Wang,Zhichen Zou,Wenbo Wang,Liwu Liu,Yanju Liu,Jinsong Leng

Percutaneous closure of ventricular septal defect (VSD) can effectively occlude abnormal blood flow between ventricles. However, commonly used Nitinol occlusion devices have non-negligible limitations, such as nondegradability leading to life-threatening embolization; limited device size predisposing to displacement and wear; only a few radiopaque markers resulting in inaccurate positioning. Nevertheless, the exploration of customized, biodegradable, and overall radiopaque occluders is still vacant. Here, overall radiopaque, biodegradable, and dynamic reconfigurable 4D printed VSD occluders are developed. Based on wavy bionic structures, various VSD occluders are designed and manufactured to adapt to the position diversity of VSD. The customized configuration, biocompatibility, and biodegradability of the developed 4D printed bionic occluders can eliminate the series of complications caused by traditional occluders. The overall radiopacity of 4D printed VSD occluders is validated ex vivo and in vivo, whereby accurate positioning can be assured. Notably, the preparation strategies for 4D printed occluders are scalable, eliminating the barriers to mass production, and marking a meaningful step in bridging the gap between modeling and clinical application of 4D printed occlusion devices. This work opens attractive perspectives for the rapid manufacturing of customized intelligent medical devices for which overall radiopacity, dynamic reconfigurability, biocompatibility, and biodegradability are sought.

Innovative liquid embolic agents based on deep eutectic solvent: Rapid gelation in situ via solvent exchange with water for endovascular embolization

Yitong Zhou,Menghui Liu,Chuandong He,Jiayuan Lin,Yanlv Chen,Mingyu Yu,Yuhan Jiang,Xin Peng

Current liquid embolic agents face several challenges, including poor biocompatibility and vascular recanalization. Herein, we propose an innovative liquid embolic agent composed of a coenzyme-based polymer (poly lipoic acid, PLA) and a biocompatible solvent (deep eutectic solvent, DES). The agent undergoes phase transformation to form a stable hydrogel in situ through solvent exchange with water, thereby enabling safe and effective embolization. First, DES is obtained by heating a mixture of choline chloride (ChCl) and glycerol (Gly). Subsequently, lipoic acid (LA) is incorporated into the DES and heated to produce the PLA/DES complex. Owing to the strong hydrogen bonding between the DES and PLA, the DES acts as a solvent while also inhibiting PLA depolymerization. Upon contact with blood, most of the DES exchange with water, whereas some amount of ChCl integrates within the PLA via strong hydrogen bonding. This hydrogen bonding not only prevents PLA depolymerization but also reinforces the PLA network, resulting in a stable PLA hydrogel rather than depolymerized LA monomers. Furthermore, liquid-metal (LM) nanoparticles are incorporated to fabricate radiopaque PLA/LM/DES. PLA/LM/DES shows better in vitro hemocompatibility and cytocompatibility, milder inflammatory response in a rat model, and more effective and safer embolization in a rabbit model than a commercial embolic agent (Onyx). Thus, this work provides an innovative liquid embolic agent and broadens the biomedical applications of DES.

A Facile Composite Strategy to Prepare a Biodegradable Polymer Based Radiopaque Raw Material for “Visualizable” Biomedical Implants

Qunsong Wang,Xiaoye Yu,Xianmiao Chen,Jingming Gao,Daokun Shi,Yang Shen,Jingyu Tang,Junhao He,Anning Li,Lin Yu,Jiandong Ding

Enabling a biodegradable polymer radiopaque under X-ray is much desired for many medical devices. Physical blending of a present biodegradable polymer and a commercialized medical contrast agent is convenient yet lacks comprehensive fundamental research. Herein, we prepared a biodegradable polymer-based radiopaque raw material by blending poly(l-lactic acid) (PLLA or simply PLA) and iohexol (IHX), where PLA constituted the continuous phase and IHX particles served as the dispersed phase. The strong X-ray adsorption of IHX enabled the composite radiopaque; the hydrolysis of the polyester and the water solubility of the contrast agent enabled the composite biodegradable in an aqueous medium. The idea was confirmed by in vitro characterizations of the resultant composite, in vivo subcutaneous implantation in rats up to 6 months, and the clear visualization of a part of a biodegradable occluder in a Bama piglet under X-ray. We also found that the crystallization of PLA was significantly enhanced in the presence of the solid particles, which should be taken into consideration in the design of an appropriate biomaterial composite because crystallization degree influences the biodegradation rate and mechanical property of a material to a large extent. We further tried to introduce a small amount of poly(vinylpyrrolidone) into the blend of PLA and IHX. Compared to the bicomponent composite, the tricomponent one exhibited decreased modulus and increased elongation at break and tensile strength. This paves more ways for researchers to select appropriate raw materials according to the regenerated tissue and the application site.

Liquid Metal Microrobots for Magnetically Guided Transvascular Navigation

Xiaohui Ju,Roshan Velluvakandy,Xianghua Wu,Miguel Angel Merlos Rodrigo,Zbyněk Heger,Kamila Bendíčková,Jan Frič,Martin Pumera

Soft microrobots, compared with their rigid counterparts, offer superior adaptability in dynamic and confined biological environments. Here, magnetically-guided liquid metal microrobots composed of gallium-indium alloys embedded with Fe nanoparticles are introduced. The unique combination of magnetic maneuverability, high surface tension, intrinsic radiopacity, and deformability allows liquid metal-based microbots to overcome limitations of both hard microrobots and fragile droplet-based systems. Under magnetic actuation, liquid metal-based magnetic microrobots exhibit controllable rolling and upstream locomotion resembling neutrophil-like navigation, enabling precise maneuvering even against physiological flow. Bridging in vitro with in vivo experiments, quail egg chorioallantoic membrane models are used to demonstrate guided transport of these microrobots through blood vessels, accumulation at tumor xenografts, and migration within subcutaneous tissues. Moreover, their strong X-ray visibility enables real-time fluoroscopic tracking, validated in porcine heart vasculature. Importantly, liquid metal-based magnetic microbots can cross endothelial barriers in a vascular flow-on-a-chip platform, while maintaining endothelial biocompatibility. By integrating deformability, magnetic steerability, and imaging visibility, liquid metal-based microrobots establish a powerful platform for minimally invasive transvascular navigation. This work highlights the potential of liquid metal-based magnetic microrobots for targeted drug delivery, image-guided therapy, and intelligent biomedical interventions.
