Cryptochrome 1 activation inhibits melanogenesis and melanosome transport through negative regulation of cAMP/PKA/CREB signaling pathway

Gao Rongyin,Zhang Ximei,Zou Kun,Meng Duo,Lv Jinpeng

Cutaneous pigmentation was recently shown to be an event regulated by clock proteins. Cryptochrome (CRY) is a key protein composing the feedback loop of circadian clock, however, the function of CRY in melanocytes remains unclear. Here, we found that KL001, a synthetic small molecule modulator of CRY1, inhibited melanin synthesis, as well as reduced melanocyte dendrite elongation and melanosome transport. In addition, the dominant role of CRY1 in KL001-induced anti-melanogenesis was revealed by small interfering RNA transfection. Cellular tyrosinase activity and expression level of melanogenic proteins, including tyrosinase, TRP-1, TRP-2, and transport proteins like Rab27a, Cdc42 and Myosin Va induced by α-MSH were remarkably reversed after KL001 treatment. Mechanistically, CRY1 activation inhibited melanogenesis through CREB-dependent downregulation of MITF and CREB phosphorylation was mediated by classical cAMP/PKA pathway. In addition, the other CRY1 activator, KL044 also suppressed cAMP/PKA/CREB pathway and inhibited melanogenesis. Finally, anti-melanogenic efficacy of KL001 was confirmed by determination of melanin contents in UVB-tanning model of brown guinea pigs, which indicated that targeting CRY1 activity, via topical application of small molecule activator, can be utilized therapeutically to manage human pigmentary disorders.

Bioactive Triple-Helical Recombinant Collagen Gels for Improved Healing of Sunburned Skin

Yi Yang,Bei Tian,Yi Xie,Peng Liu,Linyan Yao,Jianxi Xiao

Excessive ultraviolet (UV) exposure can lead to sunburn, characterized by skin barrier damage, inflammation, pain, and an increased risk of skin cancer. Recombinant collagens have gradually attracted attention due to their high purity, low immunogenicity, batch-to-batch consistency, and excellent solubility. Additionally, the type of dressing significantly affects wound repair. Gels are ideal for sunburn treatment because they maintain a moist environment, adhere firmly, and do not need to be removed. Herein, we have created bioactive triple-helical recombinant collagen (THRC) gels for improved healing of sunburned skin. The THRC gels remained stable after a three-month stability test, displaying a rheological behavior characteristic of non-Newtonian pseudoplastic fluids. In vivo skin irritation tests conducted on New Zealand rabbits demonstrated that THRC gels were safe for use. A sunburned mice model was established to study the biological effects of THRC gels. Non-invasive combo evaluations indicated that THRC gels exhibited an exceptional capability of recovering dermis density, erythema index (EI), hydration, and transepidermal water loss (TEWL) of sunburned skin to a healthy state. Histological observations revealed that THRC gels significantly enhanced the repair of damaged skin by accelerating the recovery process, promoting collagen deposition and regeneration. Molecular biological characterizations further demonstrated their remarkable antioxidant properties, including the inhibition of lipoperoxidation and the enhancement in superoxide dismutase (SOD) and glutathione (GSH) activities. These safe and bioactive recombinant collagen gels provide a novel approach for sunburn repair and show potential for long-term cosmetic benefits.

Ionic Hydrogels Based Wearable Sensors to Monitor the Solar Radiation Dose for Vitamin D Production and Sunburn Prevention

Li Lin,Liangen Shen,Junfeng Zhang,Yiyan Xu,Zheng Fang,Peter Müller-Buschbaum,Qi Zhong

Wearable solar radiation sensors based on ionic hydrogels are facilely prepared to simultaneously monitor the radiation dose for the production of vitamin D and the prevention of sunburn. Tetramethylethylenediamine (TEMED) is neutralized with acrylic acid (AA) to obtain tetramethylethylenediamine acrylate (TEMEDA), which is further polymerized with acrylamide by a free radical reaction. By simply adding MB or NR during the polymerization, the final obtained ionic hydrogels can indicate solar radiation. Due to the extent of discoloration, the discoloration speed of MB and NR is correlated to the radiation dose. This wearable sensor can indicate the solar radiation dose required by the human body to synthesize vitamin D through the discoloration of the ionized hydrogel of MB, whereas those with NR are able to illustrate the threshold of radiation dose that causes potential skin hurt. Therefore, the benefit and drawback of solar radiation can be well balanced by optimizing the exposure time to solar irradiation. In addition, polyurethane cross-linked with a thermoresponsive coating is used as band for this wearable sensor. Due to the hydrophilicity below its transition temperature, the cross-linked band possesses the easy cleaning capability of stains after the daily wear. Such type of wearable sensor can be broadly used for monitoring the solar radiation, especially in outdoor activities.
