Discovery of a New Microbial Origin Cold-Active Neopullulanase Capable for Effective Conversion of Pullulan to Panose

Meixing Wang,Huizhen Hu,Buyu Zhang,Yang Zheng,Pan Wu,Zhenghui Lu,Guimin Zhang

Panose is a type of functional sugar with diverse bioactivities. The enzymatic conversion bioprocess to produce high purity panose with high efficiency has become increasingly important. Here, a new neopullulanase (NPase), Amy117 fromB. pseudofirmus703, was identified and characterized. Amy117 presented the optimal activity at pH 7.0 and 30 °C, its activity is over 40% at 10 °C and over 80% at 20 °C, which is cold-active. The enzyme cleaved α-1, 4-glycosidic linkages of pullulan to generate panose as the only hydrolysis product, and degraded cyclodextrins (CDs) and starch to glucose and maltose, with an apparent preference for CDs. Furthermore, Amy117 can produce 72.7 mg/mL panose with a conversion yield of 91% (w/w) based on 80 mg/mL pullulan. The sequence and structure analysis showed that the low proportion of Arg, high proportion of Asn and Gln, and high α-helix levels in Amy117 may contribute to its cold-active properties. Root mean square deviation (RMSD) analysis also showed that Amy117 is more flexible than two mesophilic homologues. Hence, we discovered a new high-efficiency panose-producing NPase, which so far achieves the highest panose production and would be an ideal candidate in the food industry. Keywords:neopullulanase (NPase);pullulan;panose production;high-efficiency

Pullulan fermented by Aureobasidium melanogenum TZ-FC3 for the preparation of self-healing, adhesive, injectable and antibacterial pullulan/PVA/borax hydrogel

Guang Yang,Hongjuan Mo,Bingjie Liu,Yiwen Wu,Guiqin Liu,Yadong Hu,Xue Jiao,Kexin Guo,Xin Wei,Yaowei Fang,Mingxuan Pan,Liang Hao

Natural polymer hydrogels, such as pullulan-based hydrogels, offer significant advantages over synthetic materials due to their thermal stability, film-forming capacity, solubility, adhesiveness, and antioxidant properties. In this study, the strain Aureobasidium melanogenum TZ-FC3, which produces a high level of pullulan, was successfully isolated from the mangrove ecosystems of Guangdong Province, China. 66.01 ± 1.10 g/L pullulan without melanin was produced by the TZ-FC3 strain within 120 h at flask level. Pullulan fermented by A. melanogenum TZ-FC3 was added to enhance the hydrogen bond network within the pullulan/PVA/borax hydrogels (P-2, P-3 and P-4 hydrogels) to improve mechanical strength and crosslinking density of PVA/borax hydrogel (P-1 hydrogel). Compared to the P-1 hydrogel, the P-2 hydrogel exhibited a 65.4 % increase in tensile strain, a remarkable 694.03 % increase in tensile strength and improved the degree of internal crosslinking. Additionally, the pullulan/PVA/borax hydrogels demonstrated excellent self-healing properties, adhesion, injectability, and antibacterial activity. The preparation process of pullulan/PVA/borax hydrogels is straightforward and effective, suggesting broad industrial applicability and underscoring their potential as next-generation materials for advanced healthcare solutions.

Application of Pullulan and Chitosan Multilayer Coatings in Fresh Papayas

Linyun Zhang,Chongxing Huang,Hui Zhao

In this work, some multilayer coatings (two-layer, four-layer or six-layer) based on pullulan and chitosan for protecting papayas were prepared by the layer-by-layer technique. The papayas were coated by immersion and stored at 25 °C, 50% relative humidity or up to 14 days. Uncoated and monolayer-coated papayas were used as controls. The pullulan/chitosan coatings decreased the papaya weight loss, softening, color change (b*, ΔE), and pH, retarded the fall of titratable acidity and vitamin C, and maintained respiratory rate and soluble solid contents. Sensory quality evaluation demonstrated that pullulan/chitosan coatings effectively preserved papaya flavor and overall acceptance. In general, the four-layer coatings provided the best fruit preservation. In conclusion, multilayer pullulan/chitosan coatings are efficient in maintaining the post-harvest quality and prolonging the shelf life of fresh papaya. Keywords:papaya;preservation;pullulan;chitosan;multilayer coating

Properties of Guar Gum/Pullulan/Loquat Leaf Extract Green Composite Packaging in Enhancing the Preservation of Chinese Water Chestnut Fresh-Cut Fruit

Kok Bing Tan,Meixia Zheng,Junyan Lin,Yujing Zhu,Guowu Zhan,Jianfu Chen

Loquat leaf extract (LLE) was added to guar gum and pullulan as an environmentally friendly packaging film (GPE) to preserve Chinese water chestnuts (CWCs). The effect of the amount of LLE on the guar gum/pullulan composite film was investigated. The optimal amount of LLE was 4% (GPE4), with lower water vapor permeability (WVP) and greater mechanical strength, antioxidant, and comparable antibacterial performance than many pullulan-based films. Upon packing the CWCs for 4 days, the weight loss rate of GPE4 was only 1.80 ± 0.05%. For GPE4, the POD activity, the soluble solid content, and the vitamin C (Vc) content of the CWCs were 21.61%, 36.16%, and 26.22% higher than those of the control sample, respectively. More importantly, GPE4 was effective in preserving the quality of CWCs after 4 days of storage, better or at least comparable to non-biodegradable plastic wrapping (PE). Therefore, it can be concluded that GPE films hold significant promise as a sustainable alternative packaging material for preserving fruit-based foods like CWCs, potentially replacing PE in the future.

Enhanced hydrophobicity, antioxidant activity, and antimicrobial efficacy of edible starch-based films incorporating pullulan and geraniol for fresh salmon preservation

Youning Zou,Jiahao Ren,Yiting Fan,Yifang Cao,Yijia Xie,Xinglong Xiao,Fengsong Liu

The development of sustainable food packaging materials remains a critical challenge due to inherent limitations in hydrophobicity, antioxidant activity, and antimicrobial efficacy. Herein, a novel starch-based film incorporating pullulan and geraniol was designed to overcome these shortcomings through hydrogen bonding and intermolecular entanglement. Structural analyses using Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) confirmed strong molecular interactions, while scanning electron microscopy (SEM) and light transmittance studies demonstrated enhanced compatibility and improved optical properties. Notably, the Starch-Pullulan-Geraniol film exhibited superior hydrophobicity, with a contact angle of 68.11°, reduced water solubility (37.56 %), and minimal water vapor permeability (1.35 ×10⁻ 10 g·m⁻ 1 ·s⁻ 1 ·Pa⁻ 1 ). Mechanical performance was also enhanced, achieving a reduced Young’s modulus of 130.34 MPa, a tensile strength of 22.66 MPa, and a maximum elongation at break of 52.81 %. Furthermore, the incorporation of pullulan and geraniol conferred remarkable antimicrobial and antioxidant properties, significantly improving the film’s effectiveness in food preservation. When applied to salmon storage, the film successfully maintained appearance, lipid stability, and microbial safety, highlighting its potential as an advanced bio-based packaging material. These findings underscore the Starch-Pullulan-Geraniol film as a promising alternative to conventional packaging, offering enhanced functionality for extending food shelf life.

Preparation and performance testing of corn starch/pullulan/gallic acid multicomponent composite films for active food packaging

Min Zhang,Bin Yang,Zimeng Yuan,Qi Sheng,Changchun Jin,Jun Qi,Manman Yu,Yingnan Liu,Guoyuan Xiong

The present study investigated the mechanical characteristics, hydrophobicity, antioxidant and antibacterial properties, FTIR, SEM and XRD of films fabricated with corn starch and pullulan (CS/PUL) by adding different concentrations of Gallic acid (GA) (0%, 0.5%, 1.0%, 1.5% w/v). The mechanical strength and opacity of CS/PUL films were enhanced by the addition of 1.0% GA. The water vapor permeability (WVP) of CS/PUL films was significantly lower in films with GA compared to those without ( P  < 0.05). The addition of GA, especially at concentrations of 1.0% and 1.5%, resulted in considerably better free radical scavenging activities on DPPH than films without GA ( P  < 0.05). Interestingly, the highest water contact angle (WCA) value was observed in films with 0.5% GA, indicating stronger hydrophobicity. Furthermore, the antibacterial capabilities of the films, particularly against E. coli and P. aeruginosa , improved with an increase in GA concentration. The results of FTIR, SEM and XRD analyses showed that GA was well distributed in the CS/PUL matrix.

Preparation and characterization of polyvinyl alcohol/pullulan/ZnO-Nps composite film and its effect on the postharvest quality of Allium mongolicum Regel

Xinhua Zhang,Haowen Sun,Shengzhao Song,Yingying Li,Xiuling Zhang,Wentao Zhang

Allium mongolicum Regel is prone to rapid senescence and quality deterioration during postharvest storage. Herein, polyvinyl alcohol/pullulan/ZnO nanoparticles (PVA/PUL/ZnO-Nps) composite films were prepared via solution casting and studied to analyze the effects of ZnO-Nps on the PVA/PUL film matrix. Results revealed that the incorporation of suitable ZnO-Nps effectively reduced the light transmittance, improved water contact angle, water vapor permeability, and mechanical properties of the composite films, as well as enhanced their antimicrobial activity. The composite films were used for the postharvest preservation of A. mongolicum Regel. Results revealed that the PVA/PUL/ZnO-Nps film effectively reduced malondialdehyde accumulation content, superoxide radical generation rate, hydrogen peroxide content, improve the activity of related enzymes, and extend the storage time compared with that of polyethylene films. Therefore, the PVA/PUL/ZnO-Nps film can be used as a novel packaging material for the postharvest preservation of A. mongolicum Regel.

Preparation of pullulan-shellac edible films with improved water-resistance and UV barrier properties for Chinese cherries preservation

Baoshan Tang,Jinju Ma,Lanxiang Liu,Juan Xu,Hong Zhang,Kun Li,Xinghao Tu,Qingfang Guan

In order to improve the water-resistance and ultraviolet (UV) barrier properties of pullulan (PL) film, edible composite films were prepared by solution casting based on PL and shellac (SH). The physical properties, thermal stability, mechanical properties, water vapor permeability and UV barrier of the composite films were characterized. The analysis of the functional properties revealed that the composite films had good compatibility, water-resistance (33.50˚–60.15˚), and strong UV-blocking properties (84.60%–0.00%). The addition of SH improved the water-resistance and mechanical properties of the films, but the thermal stability was slightly reduced. These films can be used as an edible coating on Chinese cherries. According to the characteristics of weight loss, decay incidence and other indicators of cherries, the PL-SH composite films can keep Chinese cherries fresh at room temperature at least for 7 days. Therefore, PL-SH composite films may be an environmentally friendly packaging material with application prospects in the fruit preservation industry.

Soft and flexible polyvinyl alcohol/pullulan aerogels with fast and high water absorption capacity for facial mask substrates

Man Xiao,Mo Tan,Chun Peng,Fatang Jiang,Kao Wu,Ning Liu,Dan Li,Xiaolin Yao

Facial mask substrates commonly used in skincare are often considered unhealthy and environmentally unfriendly due to their composition of premoistened nonwovens containing various preservatives. This study aims to address this issue by developing a preservative-free degradable aerogel made from polyvinyl alcohol (PVA)/pullulan (PUL) using a unidirectional freeze-drying method. The aerogels had ordered three-dimensional porous structures and exhibited desirable mechanical properties. They were soft and flexible in both dry and wet states, and their Young's moduli were comparable to that of human skin. The aerogels had high porosity, ranging from 93.0 % to 95.1 %, and exhibited a high water absorption rate and water absorption capacity (ranging from 7.5 g/g to 10.1 g/g). After 30 min of water evaporation, the aerogels showed excellent moisture retention, ranging from 88 % to 93 %. Additionally, the PVA/PUL aerogel efficiently loaded and released active ingredients, such as rapidly releasing ascorbic acid (> 90 % within 30 min). These findings suggest that the PVA/PUL aerogel has potential as a material for facial mask substrates.

Enhancement of a recombinase-aided amplification assay using betaine and pullulan

Jinrong Wang,Guowei Song,Yue Ming,Jing Pan,Ruiqing Zhang,Guohao Fan,Xinxin Shen,Xuejun Ma,Lixin Li

Background Nucleic acid amplification enhancers suitable for use in a recombinase-aided amplification (RAA) assay were studied for the first time, and amplification of a long-fragment (509 bp) was initially explored. Methods Using recombinant plasmids and clinical samples, RAA fluorescence and basic methods were used to evaluate the efficacy. The fluorescence method was evaluated by threshold time and fluorescence value, and the basic method was characterized by 2% agarose gel electrophoresis. Results Taking a previously established RAA assay for HPV18 as an example, we demonstrated that the addition of 0.2 M, 0.4 M, and 0.6 M betaine and 10% pullulan could enhance the RAA. The new RAA assays with betaine and pullulan were named B-RAA and P-RAA, respectively. Using the B-RAA and P-RAA fluorescence methods, the threshold time values could be shortened by 1.72–2.32 minutes and 2.60 minutes, respectively, and the fluorescence values could be enhanced by 8847.25–9094.37 mv and 5250 mv, respectively. Using the basic method, the sensitivity could be increased 10-fold. We successfully amplified a long-fragment of 509 bp using a P-RAA assay with a sensitivity of 10 2 copies/µL (compared with 10 3 copies/µL in the RAA assay). Conclusions Thus, we concluded that betaine and pullulan are effective additives to enhance the sensitivity of RAA assays.

Conductive polyacrylamide/pullulan/ammonium sulfate hydrogels with high toughness, low-hysteresis and tissue-like modulus as flexible strain sensors

Hongyao Ding,Bingyao Wang,Xu Yang,Jie Liu,Wei Sang,Xuewen Li,Yuefang Wen,Hui Li,Xiaodong Shen

Conductive hydrogels have great potential for applications in flexible wearable sensors due to the combination of biocompatibility, mechanical flexibility and electrical conductivity. However, constructing conductive hydrogels with high toughness, low hysteresis and skin-like modulus simultaneously remains challenging. In the present study, we prepared a tough and conductive polyacrylamide/pullulan/ammonium sulfate hydrogel with a semi-interpenetrating network. Ammonium sulfate promoted the formation of low-energy-dissipating motifs between polymer chains, reinforcing the gel matrix and resulting in excellent mechanical properties, including a high stretchability of 2063 %, a high strength of 890 kPa, and a high toughness of 4268 kJ/m 3 . The hydrogen bonds formed within the network endowed the gels with low-hysteresis under deformation. The unique semi-interpenetrating network structure provided the gels with a tissue-like low modulus. Additionally, the resulting hydrogels exhibited a high conductivity of 2.39 S/m and excellent anti-freezing properties, making them suitable for flexible strain sensors. These sensors demonstrated high sensitivity over a broad strain window of 0.1–1500 %, enabling the detection of various human motions and the recognition of different languages. These findings emphasize the potential of the composite hydrogels as wearable strain sensors for flexible devices.

Improving cyclodextrin yield via retarding retrogradation of debranched starches during the preparation of cyclodextrin with cyclodextrin glycosyltransferase and pullulanase

Yue Dai,Xiaoxiao Li,Zhengyu Jin,Yuxiang Bai

Improving substrate utilization is crucial for industrial conversion of starch. The introduction of pullulanase in cyclodextrin (CD) production has improved substrate utilization. However, complete utilization of debranched starches has not been achieved, mainly due to the retrogradation of debranched starches and the inhibitory effect of CDs on pullulanase during the catalytic reaction. This study aimed to establish an efficient CD preparation system by regulating the substrate concentration and reaction temperature. A Bacillus thermoleovorans pullulanase (BtPul) was selected for investigation of its interaction with CDs through inhibition pattern assays, fluorescence spectroscopy, and molecular docking. Results clarified that CDs inhibited BtPul, with α-CD showing the highest inhibition, followed by β-CD and γ-CD. γ-CD inhibited BtPul through a mixed mode of competitive and noncompetitive inhibition, while the other CDs inhibited BtPul competitively. When pullulan was used as the substrate, increasing the substrate concentration weakened the inhibitory effect of CDs to some degree, while changes in reaction temperature only had a minor contribution. When BtPul and β-CGTase collaboratively converted 30% (w/v) starch substrate, an increase in reaction temperature improved the conversion rate of β-CD from 67.6% to 86.8%. Optimization of substrate concentration showed that high concentration starch substrate had an adverse effect on CDs yield due to retrogradation. The optimal yields of α-CD and β-CD produced by BtPul and α-/β-CGTases were 55.7% and 90.3%, respectively. This study provides a reliable method and reference for the efficient conversion of starch into CDs.
