GHK-Cu/Pionin-loaded in situ electrospun PVB/PVP smart dressing promotes wound healing via anti-oxidant, anti-inflammatory, antimicrobial, and tissue regenerative effects

Yan Wang,Zhichao Zheng,Janak L. Pathak,Haoyu Cheng,Sizhen Huang,Zi Fu,Ping Li,Lihong Wu,Huade Zheng

Biocompatible, antibacterial, anti-inflammatory, and tissue regenerative smart dressings are in urgent need for infected skin wound repair in clinics. Portable in situ 3D-electrospun technology has the great advantage of direct deposition of required smart dressing on wound surfaces. Here, we fabricated a copper peptide (GHK-Cu) and quaternium-73 (Pionin) dual-drug-loaded polyvinyl butyral (PVB)/polyvinylpyrrolidone (PVP) smart nanofibers dressing using in-situ electrospinning technology for skin wound repair. The physicochemical properties and biological activities of smart dressing were extensively analyzed. In vivo biocompatibility, anti-oxidant, anti-inflammatory, and antimicrobial activities of smart dressing were analyzed during wound healing. Stable and homogeneous PVB/PVP nanofiber smart dressing with uniformly distributed GHK-Cu and Pionin and anabolic biological activities was obtained. Biocompatible smart dressing gave a sustained release of GHK-Cu and Pionin, promoted the migration and adhesion of dermal fibroblasts and keratinocytes, and showed anti-oxidant and anti-apoptotic effects in vitro and in vivo. Smart dressing exerted anti-inflammatory properties via M1 to M2 macrophage phenotype switching and effectively promoted wound healing. Pionin released from smart dressing not only killed but also inhibited the expression and delivery of virulence factor expression and bacteria adhesion. During wound healing, smart dressing promoted the proliferation of cells in the epidermal tissue, epithelial tissue remodeling, granulation tissue growth, neovascularization, and M2 macrophage polarization. Moreover, this smart dressing scavenged excess reactive oxygen species, increased SOD3 and SIRT1 expression, and repaired peroxidized tissue, consequently promoting wound healing. In summary, our results indicate that GHK-Cu/Pionin-loaded in situ electrospun PVB/PVP smart dressing promotes wound healing via anti-oxidant, anti-inflammatory, antimicrobial, and tissue regenerative effects, suggesting clinical application of this smart dressing for complicated skin wound healing.

Bioinspired shape-changing nanofiber dressings for intelligent wrapping and promoting healing of superficial wounds

Zizhao Chen,Pingping Feng,Ruqi Wang,Dongmin Chen,Chunmei Feng,Qishu Jin,Chen Yang,Botao Song

The use of dressings in clinical settings is common for the purpose of wound wrapping and creating an optimal microenvironment to enhance the healing process. Proper coverage of wounds with dressings serves as the fundamental basis for effective wound healing. Unfortunately, non-standard coverage by hands can cause pain and secondary damage to patients, while slow manual application during treatment of extensive burns may increase the risk of wound infection. Herein, drawing inspiration from the microstructure and hygroscopic deformation observed in pine cones, we propose a polyvinyl alcohol/polysulfone (PVA/PSF) smart dressing. This bioinspired smart dressing exhibits rapid bending deformation under high moisture condition, allowing easy adjustment of bending amplitude, speed, and direction. Moreover, the smart dressing is capable of rapid bending and autonomous wrapping around “artificial wounds” on a doll’s body, as well as fitting irregularly shaped “hand wounds” and extensive “arm wounds” on human subjects. By integrating two layers into one dressing design, we endow it with dual functionality: The hygroscopic PVA layer facilitates transversal liquid transport to effectively reduce exudate accumulation in the wound bed while maintaining proper moisture levels; meanwhile, the highly hydrophobic PSF layer repels various aqueous solutions to protect against external contaminants. In vivo results confirm that this multifunctional smart dressing promotes collagen synthesis and accelerates angiogenesis for accelerated wound healing. We believe that this innovative multifunctional approach to wound management will provide valuable insights into wound healing therapy.

Injectable Hydrogels with Integrated Ph Probes and Ultrasound-Responsive Microcapsules as Smart Wound Dressings for Visual Monitoring and On-Demand Treatment of Chronic Wounds

Da Huang,Jiahao Du,Fang Luo,Gang He,Minglang Zou,Yongming Wang,Zhenyu Lin,Decheng Wu,Zuquan Weng

Hydrogel dressings capable of infection monitoring and precise treatment administration show promise for advanced wound care. Existing methods involve embedd ingorganic dyes or flexible electronics into preformed hydrogels, which raise safety issues and adaptability challenges. In this study, an injectable hydrogel based smart wound dressing is developed by integrating food-derived anthocyanidin as a visual pH probe for infection monitoring and poly( L -lactic acid) microcapsules as ultrasound-responsive delivery systems for antibiotics into a poly(ethylene glycol) hydrogel. This straightforwardly prepared hydrogel dressing maintains its favorable properties for wound repair, including porous morphology and excellent biocompatibility. In vitro experiments demonstrated that the hydrogel enabled visual assessment of pH within the range of 5 ∼ 9.Meanwhile, the release of antibiotics could be triggered and controlled by ultrasound. In vivo evaluations using infected wounds and diabetic wounds revealed that the wound dressing effectively detected wound infection by monitoring pH levels and achieved antibacterial effects through ultrasound-triggered drug release. This led to significantly enhanced wound healing, as validated by histological analysis and the measurement of inflammatory cytokine levels. This injectable hydrogel-based smart wound dressing holds great potential for use in clinical settings to inform timely and precise clinical intervention and in community to improve wound care management.

Wet Spinning Fabrication of Robust and Uniform Intrinsically Conductive Cellulose Nanofibril/Silk Conductive Fibers as Bifunctional Strain/Humidity Sensor in Potential Smart Dressing

Gong Ruixin,Dong Yanjuan,Ge Dan,Miao Zhouyu,Yu Hou-Yong

Silk fibroin (SF) with skin-like features and function shows great prospects in wearable electronics and smart dressing. However, the traditional method of loading conductive materials on physical interfaces can easily lead to the detachment of conductive materials, poor mechanical properties, and unstable conductivity, which hinder their practical application. Herein, simple wet spinning was utilized to fabricate multifunctional regenerated silk fibers reinforced with different contents of intrinsically conductive cellulose nanofibril (CNFene). Significant enhancements in fiber homogeneity, thermal stability, conductivity, mechanical strength, and sensing ability were achieved due to more regular orientation of silk fibroin molecules and strong intermolecular interactions with CNFene. The optimized sample (SF 1 ) with high sensitivity (100 ms), excellent washing/rubbing resistance, and superb waterproof properties (22 days) can comprehensively monitor human motion and weak signals. Surprisingly, inspired by the different humidity levels around wounds at different stages of healing, SF 1 with favorable humidity sensitivity can be developed as a smart dressing for monitoring wound healing. Therefore, this work provides a simple preparation route of smart high-performance fiber for flexible electronic devices, smart dressing, and underwater smart textiles. Graphical Abstract

Boric acid-substituted phthalocyanine/ciprofloxacin co-assembled nanoparticles for constructing bacteria-responsive intelligent wound dressings with synergistic therapy and recognition of bacterial infections

Ziqian Xu,Haochen Li,Shangmei Zhou,Haocong Sun,Peng Zhang,Ke Zheng,Caifeng Ding

In contemporary times, the waning effectiveness of antibiotics against bacterial infections is progressively giving rise to significant concerns in public health. Although photodynamic technology possesses a potent ability to deactivate bacteria, its non-selective attack on normal cells poses potential side effects. Hence, in this study, a boric acid-substituted phthalocyanine photosensitizer (BAPc) was synthesized, exhibiting remarkable bacterial targeting capability. BAPc was assembled with antibiotic ciprofloxacin (CIP) followed by the integration into polylysine-modified alginate dressing to obtain the product BAPc-CIP@AD-PL. Interestingly, the release of antibacterial agents and generation of reactive oxygen species (ROS) from BAPc-CIP@AD-PL can be triggered by bacteria, thereby eliciting synergistic bactericidal effect and effectively promoting wound healing in bacterial infections. Additionally, the fluorescence of BAPc-CIP@AD-PL can be activated by bacteria, facilitating the recognition of bacterial infections. The construction strategy and application principle of this multifunctional antibacterial dressing provide a novel perspective for the development of intelligent antibacterial therapy.

Fabrication of PANI-modified PVDF nanofibrous yarn for pH sensor

Hongmei Zhao,Zhang Dai,Tian He,Shufang Zhu,Xu Yan,Jianjun Yang

In recent years, with the rise of an intelligent concept, oral and maxillofacial surgery smart dressing had also attracted the interest of researchers, especially for the pH sensor with flexible medium. In this study, polyvinylidene fluoride (PVDF) nanofibrous yarn was fabricated by a conjugate electrospinning process and modified with in situ polymerization of polyaniline (PANI) forming a PANI/PVDF yarn. By a weaving process, these yarns could be weaved into a fabric. It was found that both the PANI/PVDF yarn and the fabric showed a sensitivity to pH, about −48.53 mV  per pH for yarn and −38.4 mVper pH for fabric, respectively, in the pH range of 4.0–8.0. These results indicated that the prepared PANI-modified PVDF yarn and fabric might have a potential application in intelligent oral and maxillofacial surgery dressings for monitoring wound healing.

Smart and bioactive electrospun dressing for accelerating wound healing

Di Xu,Yuhang Feng,Minjie Song,Xinyu Zhong,Jiansheng Li,Zhigao Zhu,Junsong Wang

The biosafe plant extract, rich in bioactive compounds with anti-inflammatory and antioxidant properties, significantly contributes to wound healing by promoting tissue repair and regeneration. However, the powdered forms of these extracts face several limitations including poor water solubility, limited release in vitro, and compromised waterproofing and breathability. This study successfully developed a smart and bioactive wound dressing through co-electrospinning hydrophobic polylactic acid- H. rhamnoides L. petroleum ether extract with hydrophilic polyvinyl alcohol-Ag (PLA PEE /PVA Ag ). This innovative dressing exhibits robust properties such as anti-inflammatory and antibacterial activities primarily derived from bioactive compounds in PEE and Ag nanoparticles decorated within PVA nanofibers respectively. Additionally, the dressing’s intelligent waterproofing, breathability, and interstitial fluid absorption create a moist environment that accelerates wound healing . Metabolomics analysis highlights the significance of glycolysis, the TCA cycle, arginine metabolism, linoleic acid/arachidonic acid metabolism, and glutaminolysis in wound healing. These enriched pathways not only stimulate extracellular matrix synthesis specifically collagen but also demonstrate anti-inflammatory properties while promoting angiogenesis thereby enhancing the wound healing process. As an innovative medical dressing, this smart bioactive solution holds immense potential for accelerating wound healing and tissue regeneration while paving a promising path for future advancements in personalized therapeutic strategies for enhanced wound care management.

A hybrid hydrogel constructed using drug loaded mesoporous silica and multiple response copolymer as an intelligent dressing for wound healing of diabetic foot ulcers

Wenrui Zhang,Zun Yang,Mingzu Zhang,Jinlin He,Shenzhi Li,Xingwei Sun,Peihong Ni

Traditional wound dressings have poor mechanical properties and a single function, which cannot achieve rapid healing of diabetic wounds in a unique physiological microenvironment. In order to develop multifunctional hydrogel dressings with appropriate biological activity to accelerate wound healing and obtain better clinical therapeutic effects, herein we report a hybrid system based on drug loaded mesoporous silica and injectable polymer hydrogels mixed with hypoglycemic drug metformin (Met) as a dressing for diabetic wounds. Firstly, a copolymer with the phenylboronic acid group in the side group, poly(acrylamide-co-dimethylaminopropylacrylamide-co-methacrylamidophenylboronic acid) (abbreviated as PB), was prepared. PB was mixed with polyvinyl alcohol (PVA) to obtain an injectable hydrogel (named PP) with pH/glucose dual responsiveness, which was formed through the combination of the phenylborate group of PB and o-diol of PVA. In another reaction, polydopamine-modified mesoporous silica nanoparticles (MSN@PDA) were prepared and used to adsorb antibiotic tetracycline hydrochloride (TH) to obtain drug-loaded MSN@PDA-TH nanoparticles. Subsequently, the hybrid hydrogel dressing (abbreviated as PP/MSN@PDA-TH/Met) was obtained by mixing PB, PVA, Met and MSN@PDA-TH. The self-healing, rheological and adhesive properties of the hybrid hydrogel were characterized. The results show that the hydrogel dressing has good physical properties. Met and TH were released in vitro in different pH media and glucose environments. The results show that the hydrogel dressing has dual responsiveness towards pH and glucose, and can continuously release metformin and tetracycline, which is conducive to accelerating wound healing. The antimicrobial activity, ROS clearance ability and biocompatibility of the hydrogel dressing were evaluated. The results indicate that the hydrogel dressing was multifunctional. Finally, a full-thickness wound repair model of diabetic mice induced by streptozotocin (STZ) was established. The hybrid hydrogel dressing was applied to the wound surface of mice. The wound healing testing on diabetic mice confirmed that the wound covered with the hybrid hydrogel dressing was completely healed with the formation of the new skin and hair within 9 days to 12 days. Histological analysis indicates that, compared to the PBS control, the hydrogel dressing did not cause significant inflammation in the wound, and a large number of blood vessels, glands and hair follicles appeared. This study provides a good strategy for multi-drug synergistic treatment of diabetic foot ulcers.

A Versatile Approach for Preparing Shape-Morphing Bilayer Films by Simply Adhering Two SEBS/Paraffin Films with Different Deformability and Melting Temperatures

Sijia Ren,Jiachun Feng

Shape-morphing materials that can exhibit various shape deformations are highly desirable. In this work, shape-morphing materials with diverse deformation modes are prepared via a facile approach of constructing bilayer structures of SEBS/paraffin blends with different melting temperatures. The bilayer films are prepared using a simple solvent-coating adhesion method, either by adhering two single-layers together and stretching or by adhering two pre-stretched single-layers. These bilayer films show strong interfacial adhesion due to the similar chemical composition of the two layers. By adjusting the programming method, stretching strain, and thickness ratio, the shape deformation behaviors of the resultant bilayer films can be flexibly tuned. To demonstrate the wide applicability of this approach, several 2D sheets with hinge structures as well as a smart dressing are prepared, in which the former can be transformed into 3D configurations and the latter can automatically fit to the human body and peel off. Owing to the advantages of low cost, easy and large-scale preparation, recyclability, and high designability, the approach for preparing shape-morphing bilayer films proposed by this work is versatile and the obtained bilayer films have the potential to satisfy the demands of divergent application fields.

Bioinspired smart catalytic microneedle patch with structural color for infected wound management

Yuting Huang,Yuqi Wu,Peipei Xi,Nengjie Yang,Yongxin Xu,Hao Zhang,Yunan Wang,Xiaodong Chen,Yujuan Zhu

Infected wounds remain a major clinical challenge due to persistent inflammation, excessive reactive oxygen species (ROS), and bacterial colonization. Here, we present a bioinspired catalytic microneedle (MN) patch that integrates antibacterial, anti-inflammatory, and real-time monitoring functions into a single platform. The patch possesses a hierarchical structure with spatially distributed nanocomponents: gold-modified barium titanate (AuB) in the MN tips enables ultrasound-triggered piezoelectric antibacterial activity, while cerium dioxide nanozyme (CeNP) in the backing layer efficiently scavenges ROS and mitigates inflammation. Of note, a structural color hydrogel incorporated in the backing layer provides visual, colorimetric indication of wound exudate, allowing timely patch replacement. Based on these features, it is demonstrated that the hierarchically structured MN patch exhibits robust antibacterial efficacy as well as anti-inflammatory treatment. In vivo evaluations further demonstrate its enhanced healing outcomes, which are characterized by improved tissue regeneration and accelerated resolution of inflammation. Collectively, these results highlight the potential of this smart hierarchical MN platform as an intelligent and multifunctional therapeutic strategy for efficient management of infected wounds.

A continuous wound monitoring patch based on stretchable organic electrochemical transistors

Binbin Cui,Ivo Pang,Shilei Dai,Xiaonan Chen,Dingyao Liu,Xinyu Tian,Wensheng Lin,Jing Bai,Xinran Zhang,Shiming Zhang

Chronic wounds, especially those linked to diabetes, require continuous monitoring and timely interventions to prevent severe complications. The emergence of smart bandages equipped with multifunctional sensors offers a promising avenue to alleviate the burden on healthcare systems while minimizing delays in medical care. However, conventional electrochemical sensors often face limitations, including inadequate detection sensitivity and poor mechanical compatibility with biological tissue, thereby restricting their utility in smart bandages for long-term wound care applications. In this study, we present a stretchable organic electrochemical transistor (OECT)-based glucose sensing platform designed for a smart patch in chronic wound care. By leveraging the amplification ability and the excellent mechanical properties of stretchable OECTs, this platform achieves both high sensitivity and exceptional tissue compatibility. Specifically, we demonstrate an all-polymer stretchable OECT-based glucose sensor, which exhibits high glucose sensitivity (>20 μA/mM), robust mechanical stability under substantial deformation (up to 50% strain), and extended storage stability (up to 15 days). For practical use, the sensors are integrated into a compact, coin-sized wearable readout system, enabling continuous and comfortable wound monitoring.

Closed-Loop Responsive Smart Bandages for Dynamic Monitoring and Self-Regulated Antibacterial Therapy in Wound Healing

Zhaochuan Yu,Chao Deng,Liucheng Meng,YiJin Wang,Chengcheng Li,Chao Liu,Yuqian Liu,Farzad Seidi,Xing Zhang,Qiang Yong,Hao Wu,Huining Xiao

Currently, smart wound management systems have garnered increasing attention. However, most existing strategies still rely on predefined stimuli or threshold-based mechanisms to trigger drug release, which are insufficient to feedback to the dynamic changes in infection status, and their independently designed sensing and therapeutic modules further limit functional integration and coordinated intervention. To address these challenges, this study presents a smart bandage system that integrates a closed-loop mechanism encompassing sensing, therapy intervention, and feedback. The system is built by embedding carbon dots (CDs) into metal–organic frameworks (MOFs) formed via Fe 3+ -carbenicillin (CARB) coordination, then immobilized on a cellulose nonwoven (CNW) to create a composite smart bandage. In the infection-induced acidic microenvironment, the bandage enables concurrent fluorescence recovery and CARB release, enabling real-time infection monitoring and potent antibacterial activity (>99.99%). As infection resolves, diminished bacterial activity raises local pH, thereby reducing CARB release through pH-responsive negative feedback. Mechanistic studies indicate that Fe 3+ competitively coordinates between bacterial ligands and CDs, enabling coupled sensing and therapy. In a murine wound model, the bandage significantly accelerated wound healing, suppressed bacterial growth, and allowed visual tracking of infection severity. This work provides a novel strategy for an intelligent wound bandage with autonomous sensing and therapeutic functions.
