Dark fermentation is an effective means to convert organic waste to hydrogen for bioenergy utilization but is limited by non-hydrogen-producing pathways. Emerging electro-fermentation technology promises to improve it by modulating dominant metabolic pathways. This study investigated the impact of electro-fermentation performance with voltages and substrate concentrations. The results revealed that the specific hydrogen yields at 0.3–0.6 V voltage were increased by 31.5%–91.3% over the control, and substrate degradation efficiency were also improved by 13.3%–20.7%. When the voltage was applied, the abundance of the hydrogen-producing bacteria Clostridium species increased 2.6–8.1 times more than the conventional. Meanwhile, an increase of 74.6%–311.5% in electron transfer system activity was quantified with nearly doubling the thickness of the biofilm. High soluble metabolite acetate/butyrate expression provided evidence that electro-fermentation promotes both favorable hydrogen production pathways. This study provides new insights into the application of electro-fermentation as a strategy for improving sustainable hydrogen production.

Objectives To investigate the effect of zero-valent iron and copper (Fe/Cu) micro-electrolysis on dark fermentative hydrogen production from glucose by a mixed bacterial consortium and the possible mechanisms of increasing hydrogen yield. Results Compared to zero-valent iron and activated carbon (Fe/C) micro-electrolysis, Fe/Cu micro-electrolysis could increase hydrogen yield by 32.2%, hydrogen production potential by 27.1%, and the maximum hydrogen production rate by 62.0%. Meanwhile, the number of ferrous ions released into the liquid phase with Fe/Cu micro-electrolysis was about 27.0% greater than that released by Fe/C micro-electrolysis, because the dispersion of copper on the surface of iron could markedly improve electrochemical corrosion activity. Metabolic analysis revealed that Fe/C micro-electrolysis promoted acetate formation, which may have been responsible for the observed improvement in fermentative hydrogen production. Further investigation indicated that Fe/Cu micro-electrolysis increased the activity of hydrogenases and stimulated the expression of the [FeFe]-hydrogenase gene. Conclusion Fe/Cu micro-electrolysis is better than Fe/C micro-electrolysis or Fe corrosion alone for dark fermentative hydrogen production.

Chitosan is widely used as a dewatering flocculant , but whether it affects hydrogen production from sludge anaerobic fermentation is unclear. This study aimed to elucidate the role of chitosan in the dark fermentation of waste activated sludge for hydrogen production. The results showed that chitosan had a negative effect on hydrogen production from sludge. Chitosan at 30 g/kg total suspended solids reduced hydrogen accumulation by 56.70 ± 1.22 % from 3.94 ± 0.12 to 1.71 ± 0.10 mL/g volatile suspended solids. Chitosan hindered the solubilization of sludge by flocculation , which reduced the available substrate for anaerobic fermentation. In addition, chitosan interfered with the electron transport system by reducing cytochrome C and caused lipid peroxidation by inducing reactive oxygen species , thereby inhibiting the activity of enzymes involved in anaerobic fermentation. Hydrogen production was reduced because hydrogen-producing processes (i.e., hydrolysis, acidification, and acetification) were inhibited more strongly than hydrogen-consuming processes (i.e., methanogenesis , sulfate reduction, and homoacetogenesis). Furthermore, chitosan enriched the abundance of Spirochaetaceae sp. and Holophagaceae sp., which occupied the survival space of hydrogen-producing microorganisms . This study reveals the potential impact of chitosan on hydrogen production in dark fermentation of sludge and provide direct evidence that chitosan triggers oxidative stress in anaerobic fermentation.

The efficiency of microbial electron transfer is fundamental for determining the performance of fermentative hydrogen/methane production. To facilitate microbial electron transfer, conductive magnetite nanoparticles (MNPs) were added into a cascading dark fermentation and anaerobic digestion system that was inoculated with Enterobacter aerogenes ZJU1 and methanogenic activated sludge (MAS), respectively. During the hydrogen-producing stage, the ratio of NADH/NAD + and the activities of hydrogenase and electron transport system (ETS) of E. aerogenes ZJU1 were all increased by dosing 200 mg/L MNPs, which was conducive to hydrogen production through the NADH-dependent pathway. In the presence of 200 mg/L MNPs, hydrogen production increased by 21.1%, while subsequent methane production improved by 22.9%. Electrochemical analysis demonstrated the improvement in extracellular electron transfer capacity of MAS after adding MNPs, which can be ascribed to the contribution of MNPs and electrochemically active extracellular polymeric substances (EPS) induced by MNPs, such as humic acid-like and fulvic acid-like substances. Bacteria Syntrophomonas and Archaea Methanosarcina were the dominating enriched syntrophic partners, and the expression of functional genes involved in CO 2 reduction to methane pathway was found to increase. Therefore, a more efficient fermentative hydrogen and methane co-production system was established by improving microbial electron transfer with the addition of MNPs.
