A colorimetric and ratiometric fluorescent probe for hydrazine and its application in living cells with low dark toxicity

Zhanxian Li,Wenying Zhang,Chunxia Liu,Mingming Yu,Hongyan Zhang,Liang Guo,Liuhe Wei

It is significant to develop probes for rapid, selective, and sensitive detection of the highly toxic hydrazine in both environmental and biological science. In this work, under mild condition, a novel colorimetric and ratiometric fluorescent probe ( probe 1 ) was synthesized. Probe 1 with a chemoselective reaction mediated by hydrazine in which the diethyl malonate moiety was transformed into an aldehyde group exhibited high sensitivity and good selectivity in hydrazine recognition. Probe 1 can quantitatively detect hydrazine in concentration range from 0 to 120   μM with the LOD of 1.6   μM. Further, it displayed good selectivity and anti-interference over many species including some nucleophilic species, anions and metal ions. The ratiometric fluorescence change of this probe upon addition of hydrazine in living cells has been successfully demonstrated. Furthermore, the probe displays low dark toxicity under physiological temperature.

A mitochondrial-targeting and NO-based anticancer nanosystem with enhanced photo-controllability and low dark-toxicity

Jiangsheng Xu,Fang Zeng,Hao Wu,Shuizhu Wu

Compared to the generation of singlet oxygen in photodynamic therapy, photo-generation of nitric oxide (NO) would not be limited by the concentration of molecular oxygen. However, therapeutic applications of exogenous nitric oxide are usually limited by its short half-life and its vulnerability to many biological substances, thus straightforward and precise control over NO delivery may be critical to its therapeutic effects. Herein, we demonstrate a mitochondrial-targeting and photoactive NO-releasing system as an anticancer drug. Fabricated by covalently incorporating a photo-responsive NO-donor and a mitochondrial targeting ligand onto carbon dots, this nanosystem exhibits a multi-functional nature which combines mitochondrial-targeting, photocontrollable NO-releasing and cell imaging. Upon cellular internalization, the nanosystem could target mitochondria effectively. Furthermore, the system displays little dark toxicity under physiological temperature; but upon light irradiation, it could release NO, efficiently damage mitochondria and consequently cause prominent apoptosis of cancer cells. Moreover, evaluated by using MTT assay, this nanosystem shows high cytotoxicity towards two cancer cell lines. These observations provide new insights for exploiting NO in disease therapy.

A new near-infrared phosphorescent iridium(III) complex conjugated to a xanthene dye for mitochondria-targeted photodynamic therapy

Yongquan Wu,Jie Wu,Wai-Yeung Wong

Iridium(III) complexes are potent candidates for photodynamic therapy (PDT), but some key drawbacks still hamper clinical translation, such as poor operability in the phototherapeutic window, high dark toxicity, and low reactive oxygen species (ROS) production efficiency. In this work, a near-infrared phosphorescent Ir(III) complex conjugated to a xanthene dye, NIR-Ir-XE, is reported with highly favourable properties for mitochondria-targeted imaging and cancer phototherapy. The generation of the triplet excited state of a xanthene moiety endows the NIR-Ir-XE to form singlet oxygen (1O2) for use as a photodynamic therapy agent after irradiation with visible light. Compared with the xanthene-free Ir(III) counterpart (NIR-Ir-bpy), the xanthene-modified cyclometalated Ir(III) photosensitizer NIR-Ir-XE exhibits higher 1O2 generation efficiency, negligible dark toxicity and a better therapeutic effect. Importantly, a clear correlation between cell death and intracellular generation of 1O2 derived from NIR-Ir-XE after light irradiation was demonstrated. The corresponding in vivo photo-antitumor performance was further demonstrated to be effective in tumor-bearing mice. The observed properties of NIR-Ir-XE qualify it as a promising PDT agent.

Multifunctional AIE iridium (III) photosensitizer nanoparticles for two-photon-activated imaging and mitochondria targeting photodynamic therapy

Cai Xuzi,Wang Kang-Nan,Ma Wen,Yang Yuanyuan,Chen Gui,Fu Huijiao,Cui Chunhui,Yu Zhiqiang,Wang Xuefeng

Developing novel photosensitizers for deep tissue imaging and efficient photodynamic therapy (PDT) remains a challenge because of the poor water solubility, low reactive oxygen species (ROS) generation efficiency, serve dark cytotoxicity, and weak absorption in the NIR region of conventional photosensitizers. Herein, cyclometalated iridium (III) complexes (Ir) with aggregation-induced emission (AIE) feature, high photoinduced ROS generation efficiency, two-photon excitation, and mitochondria-targeting capability were designed and further encapsulated into biocompatible nanoparticles (NPs). The Ir-NPs can be used to disturb redox homeostasis in vitro, result in mitochondrial dysfunction and cell apoptosis. Importantly, in vivo experiments demonstrated that the Ir-NPs presented obviously tumor-targeting ability, excellent antitumor effect, and low systematic dark-toxicity. Moreover, the Ir-NPs could serve as a two-photon imaging agent for deep tissue bioimaging with a penetration depth of up to 300 μm. This work presents a promising strategy for designing a clinical application of multifunctional Ir-NPs toward bioimaging and PDT.

AIE-active Ir(III) complexes functionalised with a cationic Schiff base ligand: synthesis, photophysical properties and applications in photodynamic therapy

Shengnan Liu,Jiahong Han,Weijin Wang,Yulei Chang,Runlin Wang,Ziwei Wang,Guangzhe Li,Dongxia Zhu,Martin R. Bryce

Photodynamic therapy (PDT) is a promising cancer treatment method. Traditional small-molecule photosensitizers (PSs) suffer from low intersystem crossing (ISC) ability and aggregation-caused quenching (ACQ), which adversely affects the luminous efficiency and singlet oxygen (1O2) yield of PSs in the aggregated state. Ir(III) complexes are promising PSs with long excited-state lifetime, good photophysical and photochemical properties and large Stokes shifts. Aggregation-induced emission (AIE) characteristics could reduce the nonradiative recombination and improve the ISC ability of excited states through the restriction of the intramolecular motions in aggregated states. Accordingly, two AIE-active Ir(III) complexes Ir-1-N+ and Ir-2-N+ were successfully designed and obtained based on Schiff base ligands. Experimental results showed that Ir-1-N+ and Ir-2-N+ have good photophysical properties and the corresponding nanoparticles (NPs) have good water solubility and 1O2 generation ability. Notably, Ir-2-N+ NPs can be efficiently taken up by mouse breast cancer cells (4T1 cells) with good biocompatibility, low dark toxicity and excellent phototoxicity. This work demonstrates a versatile strategy for exploiting efficient transition metal PSs with a cationic ligand in PDT.

Competitive coordination assembly of light-degradable gold nanoclusters-intercalated metal organic frameworks for photoresponsive drug release

Ke Wang,Sicheng Zhai,Yuanyuan Qin,Mengke Hao,Siqi Su,Shuming Li,Xuexue Tang

Controlled drug release with on demand holds great promise for cancer therapy. Light-degradable nanocarriers have gained increasing attention for designing controllable drug delivery systems due to their spatiotemporally controllable properties. Herein, a high luminescent and light-degradable nanocarrier is constructed by intercalating glutathione-capped gold nanoclusters (AuNCs) into zeolitic imidazolate framework-8 (ZIF-8) via competitive coordination assembly, named as AuNC@ZIF-8, for light-triggered drug release. Glutathione-capped AuNCs and 2-methylimidazole (MIm) competitively coordinated with Zn2+ to form AuNC@ZIF-8 via one step process in aqueous solution. Specifically, the obtained AuNC@ZIF-8 has a high quantum yield of 52.96 % and displays a distinctive property of photolysis. The competitive coordination interactions within AuNC@ZIF-8 were evidenced by X-ray diffraction and X-ray photoelectron spectroscopy, in which Zn2+ strongly coordinated with the N of MIm and weakly coordinated with the carboxyl/amino groups in glutathione of AuNCs. Under light irradiation, the Au-S bond on AuNCs breaks, enhancing the coordination ability between carboxyl/amino groups and Zn2+ enhance. This leads to the collapse of the crystal structure of AuNC@ZIF-8 and subsequent fluorescence quenching. Additionally, AuNC@ZIF-8 is successfully employed as luminescent nanocarriers of anticancer drugs to form drug-AuNC@ZIF-8, in which three typical anticancer drugs are selected due to different coordination interaction. The obtained smart drug-AuNC@ZIF-8 can be effectively internalized into HeLa cells and degraded in response to blue light, with negligible dark cytotoxicity and high light cytotoxicity. This study highlights the crucial role of competitive coordination interactions in synthesizing functional materials with fluorescence efficiency and photolytic properties.
