A novel dual-site recognition protocol was developed for chemiluminescent (CL) detection of Streptococcus mutans ( S. mutans ) based on a designed antibiotic-affinity strategy. Teicoplanin , a broad-spectrum antibiotic against Gram-positive bacteria, was adopted to functionalize magnetic particles and recognize S. mutans utilizing the strong affinity between this agent and D-Alanyl-D-Alanine peptide moieties in the bacterial cell wall. To achieve ideal specificity for S. mutans detection, rat immunoglobulin G2a (rat IgG2a) tagged with horseradish peroxidase (HRP) was used as the second recognition agent and signal tracer since Fab region of rat IgG2a could bind with streptococcal protein G highly expressed in the cell wall of S. mutans . Thus HRP-tagged sandwich complex of teicoplanin/ S. mutans /rat IgG2a was formed on the magnetic particles, followed by a CL quantification of S. mutans based on a HRP-catalyzed luminol-H 2 O 2 -p -iodophenol CL reaction. This dual-site recognition protocol showed a linear range of 1.0 × 10 2 –1.0 × 10 6 CFU mL −1 and a detection limit of 33 CFU mL −1 for S. mutans detection. The whole detection process could be completed within 70 min. The recovery tests for food, environmental, pharmaceutical and biological samples showed acceptable recovery values between 83.0% and 110.0%, demonstrating its application potential for detection of bacteria in various sample matrixes.

Superficially porous particles (SPPs) demonstrate superior efficiency than totally porous particles in chiral separations. In order to obtain high efficiency and fast separation, sub-2 μm SPPs with high surface area are synthesized, and with teicoplanin bonded, such materials are successfully applied into the rapid enantioseparation of native amino acids. In brief, 1.27 ± 0.06 μm nonporous silica particles are prepared by a modified seeded growth method, followed by mesoporous shell fabrication via one-pot templated dissolution and redeposition strategy, and pore size expansion via acid-refluxing. The diameter of the formed SPPs is 1.49 ± 0.04 μm, with the shell thickness as 206 nm. Nitrogen physisorption experiments show that the Brunauer–Emmett–Teller (BET) specific surface area is 213.6 m 2 /g and pore size is 9 nm. After teicoplanin derivatization with bonding capacity as 83.3 μmol/g, the prepared chiral stationary phase is packed into a stainless steel tube with the geometry of 50 mm × 2.1 mm i.d.. In less than 6.4 min, six native amino acids (norleucine, alanine, valine, methionine, leucine, norvaline) are enantioseparated with resolution factors ranging from 1.9 to 5.0. Besides, the resolution for chiral separation is improved with ethanol-water instead of methanol-water as the mobile phase. Moreover, the low temperature gives higher resolution, but longer retention time and higher backpressure. Finally, the effect of flow rate on enantiomeric separation is studied and fast chiral separation within 1 min is obtained with flow rate of 0.4 mL/min. All these results show that the synthesized teicoplanin bonded sub-2 μm SPPs have great potential to achieve the enantioseparation of native amino acids with high resolution and rapid speed.

A molecularly imprinted membrane (MIM) with teicoplanin (TE) as a template and other related material membranes (organic nylon microporous membranes, polyvinylidene fluoride (PVDF) membranes, and polypropylene membranes) as supports were synthesized for the selective absorption of TE. These membranes can be used for pretreatment of biological samples and rapid determination of TE. Our results showed that PVDF MIM exhibited the best selectivity among the other three synthesized membranes and therefore, it was used in this study. The performance of PVDF MIM was examined, including surface structure, adsorption capacity, desorption capacity, selective adsorption capacity, and recovery rate; the precision, stability, repeatability, linearity and recovery rate of this method were investigated at the same time. Clean-up and enrichment of samples by TE MIM were reliable.

An affinity-based protocol is described for the detection of Staphylococcus aureus ( S. aureus ). It is utilizing teicoplanin-functionalized magnetic beads as carriers. Teicoplanin, which binds to the walls of cells of S. aureus via five hydrogen bonds, acts as the recognition agent. Captured S. aureus is magnetically separated from the sample matrix and then specifically lysed by lysostaphin which cleaves the cross-linking pentaglycine bridges of peptidoglycan in the cell wall. Lastly, S. aureus is quantified via the inhibitory effect of released intracellular catalase on a chemiluminescent (CL) system composed of peroxidase, luminol, H 2 O 2 and p -iodophenol because catalase decomposes H 2 O 2 . S. aureus can be detected with CL response in the 140 to 1.4 × 10 7 CFU·mL −1 concentration range and a detection limit as low as 47 CFU·mL −1 at a signal-to-noise ratio of 3. The method was evaluated by analyzing spiked samples including milk, human urine and saline injection solutions. The reliability was demonstrated by a recovery test and by comparison with a conventional plate counting method.
An antibiotic-affinity protocol is developed to detect Staphylococcus aureus ( S. aureus ) by utilizing teicoplanin-functionalized magnetic beads (Teic-MBs) as carriers. S. aureus can be quantified by measuring the inhibition of luminol chemiluminescence (CL) signal by intracellular catalase.

A novel teicoplanin functionalized organic-silica hybrid chiral monolithic column was prepared by a facile single-step approach. The conditions for the preparation of the organic-silica hybrid chiral monolithic column were systematically optimized, including the amount of poly (ethylene glycol) (PEG) and urea, the tetramethyl orthosilicate (TMOS)/3-(trimethoxysilyl)-propylmethacrylate (γ-MAPS) ratio (v/v), the content of teicoplanin-2-isocyanatoethyl methacrylate (Tei-ICNEML) monomer, and the reaction temperature and time. The optimum column exhibited satisfactory efficiency, permeability and stability. In order to obtain satisfactory enantioseparation performance, reversed phase (RPM) and polar organic phase (POM) modes were employed, and different experimental conditions were optimized, such as buffer concentration and pH, amount of the organic solvent, concentration and ratio of TEA/HAc, and MeOH/ACN ratio. The stability of the Tei-ICNEML organic-silica hybrid monolithic column was also investigated in both POM and RPM separation modes. Then, this novel monolithic column was successfully applied to achieve the enantioseparation of amino alcohols, N-derivatized amino acids and mandelic acids. 15 out of 20 amino alcohols were baseline separated with good column efficiency in the POM mode, and 3 out of 5 mandelic acids and 5 out of 6 N-derivatized amino acids were baseline separated in the RPM mode.

Timely and accurate anti-infective therapy significantly benefits treatment of various life-threatening bacterial infectious diseases. Unfortunately, owing to the lack of selectivity for recognizing bacterial species, most of the reported phenotypic antimicrobial susceptibility testing protocols suffer from time-consuming bacterial isolation and identification. In this work, a highly-selective molecular recognition-based sandwich fluorimetric assay was developed to rapidly detect Staphylococcus aureus ( S. aureus ), and furtherly to assess its susceptibility to antibiotics. Immobilized porcine IgG was utilized to capture S. aureus through the selective interaction between Fc fragment of porcine IgG and protein A in the cell wall of S. aureus . Fluorescein isothiocyanate-labeled teicoplanin was adopted as the signal tracer utilizing the binding behavior between teicoplanin and d -Ala- d -Ala moieties in the peptidoglycans of Gram-positive bacterial cell wall. S. aureus could be detected within a wide linear rang of 1.0 × 10 3 to 1.0 × 10 7 CFU mL −1 . Subsequently, this protocol utilizing porcine IgG and teicoplanin as the indicator molecules was utilized to assess the antimicrobial susceptibility of S. aureus after the bacteria were treated with antibiotics. The minimum inhibitory concentrations of penicillin, cefoxitin, clindamycin, trimethoprim/sulfamethoxazole and erythromycin were estimated to be >0.25,<4, <0.25, <2/38 and <0.5 μg mL −1 , respectively. The whole process for antimicrobial susceptibility testing can be completed within 4 h since it avoided extra time-consuming isolation and identification of bacterial species.

A novel chiral stationary phase (CSP), based on a monolithic organic polymer chemically modified with teicoplanin, was fabricated within a 100 μm I.D. fused silica capillary. The teicoplanin was firstly derivatized with 2-isocyanatoethyl methacrylate (ICNEML) and then thermally co-polymerized with the crosslinker ethylene dimethacrylate (EDMA) in presence of porogens (methanol and dimethylsulfoxide). The optimal experimental conditions (e.g., concentration and ratio of the reagents), considering enantioresolution and permeability, were systematically investigated. The prepared monolith was evaluated using scanning electron microscopy, and the column exhibited quite good morphology. In order to further evaluate the enantioresolving power of the poly(ICNEML-teicoplanin- co -EDMA) monolith, a series of basic and acidic chiral compounds were analyzed using an isocratic mode of polar organic solvents (methanol and acetonitrile) or the same solvents in combination with water (reversed-phase) by nano-liquid chromatography. Five mandelic acids and six derivatized amino acids were enantioresolved under reversed-phase mode ( R s = 1.22–3.47 and α = 1.43–6.33). This monolithic teicoplanin-CSP was also effective in the enantioseparations of 17 amino alcohol drugs employing polar-organic phase mode (MeOH/ACN/TEA/HOAc (80/20/0.03/0.055, v/v/v/v)). Ten of them were baseline enantioresolved (alprenolol, betaxolol, clenbuterol, isoproterenol, metoprolol, pindolol, propranolol, salbutamol, sotalol, tertatolol) ( R s = 1.55–2.48 and α = 1.21–1.55), while the others were partially enantioseparated ( R s = 1.14–1.48).

A simple, sensitive, and simultaneous method was established and validated for the active clinical components of teicoplanin and ramoplanin in environmental water by LC-MS/MS coupled with cascade elution. Moreover, a cascade elution method, which was rapid, solvent-less, and high-extraction efficient was successfully proposed to realize the extraction and purification of seven targets in one step. Under optimized conditions, the method showed excellent linearity with the correlation coefficient (R2) ≥0.998 in the range of 1.0–100.0 ng L−1. Low matrix effects and good recoveries which ranged from 86 to 114% were reached with RSDs lower than 3.0% for most targets. The limits of detection and limit of quantification were 0.1–1.3 and 0.3–4.0 ng L−1, respectively. This method was successfully applied for the determination of teicoplanin and ramoplanin in water samples from the Pearl River and the South China Sea. TA2-2,3 was quantified in only one sample with the concentration of 8.0 ng L−1.

Bacterial infection is a very troublesome issue in wound treatment, which stimulates exudate formation and severely delays the healing process. Herein, a thermogelling dressing system composed of two triblock copolymers of poly(D,L-lactic acid- co -glycolic acid)- b -poly(ethylene glycol)- b -poly(D,L-lactic acid- co -glycolic acid) (PLGA-PEG-PLGA) with different block lengths was developed to deliver teicoplanin (TPN), a glycopeptide antibiotic, for cutaneous wound repair. The TPN-loaded thermogel was a free-flowing sol at room temperature and formed a semi-solid gel at physiological temperature. In vitro studies demonstrated that the TPN-loaded thermogel system exhibited desired tissue adhesiveness and realized the sustained release of TPN in a fast-followed-slow manner for over three weeks. Furthermore, a full-thickness excision wound model in Sprague-Dawley (SD) rats was constructed to assess the efficacy of TPN-loaded thermogel formulation. Gross and histopathologic observations implied that treatment with the thermogel formulation reduced inflammation response, promoted disposition of collagen, enhanced angiogenesis, and accelerated wound closure and maturity of SD rats. The combination of the bioactivity of TPN and the acidic nature of the thermogel matrix was responsible for such an enhanced wound healing process. Consequently, the TPN-loaded PLGA-PEG-PLGA thermogel is a good candidate of wound dressing for full-thickness excision wound healing.

Based on the specific affinity behavior of teicoplanin towards Gram-positive (G + ) bacteria, teicoplanin-coated magnetic particles were employed to separate G + bacteria. Moreover, intracellular catalase released by catalase-positive bacteria led to catalyzed hydrolysis of H 2 O 2 and inhibition of chemiluminescent signal of luminol-H 2 O 2 system. By combining the separation capability of teicoplanin-coated magnetic particles and the inhibition of luminol chemiluminescence by catalase, a facile method was proposed for quantifying catalase-positive G + bacteria. In this proof-of-principle work, Staphylococcus aureus , Bacillus subtilis and Micrococcus luteus were adopted as model bacteria to evaluate its quantification performance. The three bacteria were quantified within the dynamic ranges of 5.0 × 10 2 –5.0 × 10 7 CFU mL −1 , 4.0 × 10 2 –4.0 × 10 7 CFU mL −1 and 2.0 × 10 2 –2.0 × 10 7 CFU mL −1 , with detection limits of 156 CFU mL −1 , 86 CFU mL −1 and 44 CFU mL −1 , respectively. Gram-negative bacteria and catalase-negative G + bacteria both displayed minor interference. The results for quantifying bacteria in milk, human urine and physiological saline samples demonstrated its reliability for real application. It provided a promising technique tool for food safety, medical diagnosis as well as drug quality control.

Injudicious or inappropriate use of antibiotics has led to the prevalence of drug-resistant bacteria, posing a huge menace to global health. Here, a self-assembled aggregation-induced emission (AIE) nanosphere (AIE-PEG1000 NPs) that simultaneously possesses near-infrared region II (NIR-II) fluorescence emissive, photothermal, and photodynamic properties is prepared using a multifunctional AIE luminogen (AIE-4COOH). The AIE-PEG1000 NPs were encapsulated with teicoplanin (Tei) and ammonium bicarbonate (AB) into lipid nanovesicles to form a laser-activated “nanobomb” (AIE-Tei@AB NVs) for the multimodal theranostics of drug-resistant bacterial infections. In vivo experiments validate that the “nanobomb” enables high-performance NIR-II fluorescence, infrared thermal, and ultrasound (AB decomposition during the photothermal process to produce numerous CO2/NH3 bubbles, which is an efficient ultrasound contrast agent) imaging of multidrug-resistant bacteria-infected foci after intravenous administration of AIE-Tei@AB NVs followed by 660 nm laser stimulation. The highly efficient photothermal and photodynamic features of AIE-Tei@AB NVs, combined with the excellent pharmacological property of rapidly released Tei during bubble generation and NV disintegration, collectively promote broad-spectrum eradication of three clinically isolated multidrug-resistant bacteria strains and rapid healing of infected wounds. This multimodal imaging-guided synergistic therapeutic strategy can be extended for the theranostics of superbugs.

Bacterial analysis and antibiotic resistance testing (ART) are of great significance in clinical diagnosis and therapy of bacterial infectious diseases. In this work, a portable antibiotic-affinity chromatographic test strip has been developed for rapid analysis of Staphylococcus aureus ( S. aureus ) and further applied for ART of this pathogen. Porcine IgG was immobilized on a nitrocellulose membrane for capturing S. aureus based on the selective binding capability of the Fc fragment of IgG toward protein A on the surface of the target bacteria. Fluorescent microspheres modified with teicoplanin (TEI) were applied as signal substances to trace S. aureus utilizing the hydrogen bond conjugation between this antibiotic and Gram-positive bacteria. S. aureus can be analyzed within the concentration range from 1.4 × 10 3 CFU mL −1 to 1.4 × 10 7 CFU mL −1 . The recovery values for spiked samples were 93.3–110.0%. The obtained results of ART for penicillin, daptomycin , gentamicin, cefoxitin and clindamycin against S. aureus showed agreement with those of traditional broth dilution method. The procedures for bacterial analysis and ART can be accomplished within 20 and 110 min, respectively. The antibiotic-affinity chromatographic test strip showed great promise in point-of-care testing because of its ideal portability and rapidity.
