Unlocking full potential of bamboo waster: Efficient co-production of xylooligosaccharides, lignin, and glucose through low-dosage mandelic acid hydrolysis with alkaline processing

Hong Liao,Baojun Feng,Xueping Song,Junhua Zhang,Zhanying Zhang

Mandelic acid (MA), a natural and environmentally friendly organic acid, demonstrates high selectivity and efficiency in hydrolyzing hemicellulose, making it an excellent candidate for xylooligosaccharides (XOS) production at low acid dosages. Despite its potential, the application of MA for XOS production has not been evaluated. The study first investigated the effectiveness of MA in hydrolyzing hemicellulose in bamboo into XOS. Under optimized conditions (50 mM MA, 180 °C, 45 min), a high XOS yield of 65.9 % was achieved, with a total xylobiose and xylotriose yield of 43.5 %. Subsequent alkaline pretreatment enabled 92.1 % lignin removal from MA-pretreated bamboo. The recovered lignin exhibited a high purity of 95.2 % and retained fundamental structure and functional groups of native lignin. The resulting residue displayed enhanced crystallinity and accessibility, with reduced hydrophobicity and surface area lignin compared to untreated bamboo. At high substrate concentration of 20 %, cellulase hydrolysis resulted in a glucose conversion efficiency of 83.9 %. Overall, this integrated strategy offered an efficient approach for the co-production of valuable XOS, lignin, and glucose from bamboo. The efficient energy utilization and economic viability further highlighted the potential of this method for large-scale industrial applications, making it an attractive option for biomass valorization.

Efficient production of xylooligosaccharides from Camellia oleifera shells pretreated by pyruvic acid at lower temperature

Yuejie Qiu,Leping Zhang,Fenglun Zhang,Xichuang Cheng,Li Ji,Jianxin Jiang

XOS production from lignocellulose using organic carboxylic acids and alkyd acids has been widely reported. However, it still faces harsh challenges such as high energy consumption, high cost, and low purity. Pyruvic acid (PYA), a carbonyl acid with carbonyl and carboxyl groups, was used to produce XOS due to its stronger catalytic activity. In this work, XOS was efficiently prepared from COS in an autoclave under the condition of 0.21 M PYA-121 °C-35 min. The total yield of XOS reached 68.72 % without producing any toxic by-products, including furfural (FF) and 5-hydroxymethylfurfural (5-HMF). The yield of xylobiose (X 2 ), xylotriose (X 3 ), xylotetraose (X 4 ), and xylopentaose (X 5 ) were 20.58 %, 12.47 %, 15.74 %, and 10.05 %, respectively. Meanwhile, 89.05 % of lignin was retained in the solid residue, which provides a crucial functional group for synthesizing layered carbon materials (SRG-a). It achieves excellent electromagnetic shielding (EMS) performance through graphitization, reaching −30 dB at a thickness of 2.0 mm. The use of a PYA catalyst in the production of XOS has proven to be an efficient method due to lower temperature, lower acid consumption, and straightforward operation.

Yields of three acids during simulated fermentation of inulin and xylo-oligosaccharides enhanced by six exogenous strains

Kong Lu,Zhao Xin-Huai

Three acids (acetic, propionic and butyric acids) yielded from simulated colonic fermentation of inulin and xylo-oligosaccharides (XOS) in the absence and presence of six exogenous strains were investigated in this study. Both inulin and XOS were fermented by fecal extract (FE) prepared from healthy adults and by an exogenous strain for 48 h, and three acids generated in fermentation products at five time periods were detected by gas chromatography. Inulin and XOS during fermentation were capable of producing more propionic and butyric acids, respectively, whilst inoculation using one strain culture into fermentation systems mostly increased yields of three acids. In inulin-containing fermentation systems, these strains increased yields of propionic and butyric acids by 6–34 and 206–250%, respectively. In XOS-containing fermentation systems, these strains did not enhance yield of propionic acid, but increased yield of butyric acid by 50–96%. In general, Lactobacillus brevis, Lactobacillus plantarum, Lactobacillus rhamnosus, Enterococcus faecalis and Enterococcus faecium were more efficient than Lactobacillus acidophilus to increase yields of propionic and butyric acids. It is thus concluded that the six strains in total displayed beneficial effects on simulated fermentation of inulin and XOS through enhancing yields of three acids especially propionic and butyric acids with healthcare functions.

Study on the structure and preservation mechanism of 3D-Printed surimi with Ca2+ and Xylo-oligosaccharides

Jun-qi Zhan,Yue-wen Chen,Gao-shang Li,Ya-qin Hu

The viscosity and plasticity of surimi is remarkable, which is a suitable 3D printing material. But low printing accuracy limit its wide application. Adding xylo-oligosaccharide (XOS) can enhance printing accuracy of surimi printing ink and improve the self-supporting ability and anti-deformation ability of the printed object. At the same time, XOS can impart antioxidant properties to 3D printed surimi. Additionally, adding 20 mM kg −1 of Ca 2+ to the surimi can significantly improve the printing accuracy. With the addition of Ca 2+ , the gel strength and texture of surimi are enhanced, as well as the surimi protein became denser. Moreover, the Ca 2+ in the “ink” can activate the activity of digestive enzymes which increase the contact of enzymes and surimi. The process of enzymatic hydrolysis is promoted, which improving the release rate and antioxidant properties of XOS. Functional surimi with Ca 2+ can be used to deliver functional components and enhance the printing effects.

Preparation of xylooligosaccharides from rice husks and their structural characterization, antioxidant activity, and probiotic properties

Shuangqi Tian,Ziyi Yang,Feng Yan,Xing'ao Xue,Jing Lu

Rice husks are rich in xylan , which can be hydrolyzed by xylanase to form xylooligosaccharides (XOS). XOS are a functional oligosaccharide such as improving gut microbiota and antioxidant properties. In this study, the structure and functional characteristics of XOS were studied. The optimal xylanase hydrolysis conditions through response surface methodology (RSM) were: xylanase dosage of 3000 U/g, hydrolysis time of 3 h, hydrolysis temperature of 50 °C. Under this condition, the yield of XOS was 150.9 mg/g. The TG-DTG curve showed that XOS began to decompose at around 200 °C. When the concentration of XOS reached 1.0 g/L, the clearance rate of DPPH reached 65.76 %, and the scavenging rate of OH reached 62.10 %, while the clearance rate of ABTS free radicals reached 97.70 %, which was equivalent to the clearance rate of V C . XOS had a proliferative effect on four probiotics : Lactobacillus plantarum , Lactobacillus brucelli , Lactobacillus acidophilus , and Lactobacillus rhamnosus . However, the further experiments are needed to explore the improvement effect of XOS on human gut microbiota, laying a foundation for the effective utilization of XOS. XOS have a wide range of sources, low price, and broad development prospects. The reasonable utilization of XOS can bring greater economic benefits.

Purification of xylooligosaccharides from bamboo with non-organic solvent to prepare food grade functional sugars

Yetao Jiang,Xiaoyu Wang,Zhen Wu,Jiaxing Xu,Lei Hu,Lu Lin

Food grade xylooligosaccharides (XOS) have a high application value, but most of the reported purification methods of XOS use organic solvents, which has significant problems such as large amount of solvent, residual solvent, and difficulty in solvent recovery. In addition, although there are many reported methods of separation and purification, quantitative analysis is lacking for each specific treatment step, such as sugar loss rate, decolorization rate, protein removal rate, and desalination rate. Therefore, purification of XOS from bamboo with non-organic solvent to prepare food grade functional sugars was presented by this study. What’s more, quantitative analysis and optimization was made for each step of separation, which was of great significance for guiding the actual production. Overall, through activated carbon adsorption, ion exchange and membrane filtration comprehensive treatments, food grade XOS were obtained from bamboo with 7.5% yield, a purity of 92.3%, and a total sugar proportion as high as 98.9%.

Pretreatment of moso bamboo with p-toluenesulfonic acid for the recovery and depolymerization of hemicellulose

Meixin Wang,Yawei Zhan,Jiayue Zhao,Zhiqiang Li

Bamboo and its mechanical processing residues have broad prospects for high value-added utilization. In this research, p-toluenesulfonic acid was used for the pretreatment of bamboo to investigate the effects of extraction and depolymerization of hemicellulose. The response and behavior of changes of cell-wall chemical components were investigated after different solvent concentration, time, and temperature pretreatment. Results indicated that the maximum extraction yield of hemicellulose was 95.16 % with 5 % p-toluenesulfonic acid at 140 °C for 30 min. The depolymerized components of hemicellulose in the filtrate were mainly xylose and xylooligosaccharide, with xylobiose accounting for 30.77 %. The extraction of xylose from the filtrate reached a maximum of 90.16 % with 5 % p-toluenesulfonic acid at 150 °C for 30 min pretreatment. This research provided a potential strategy for the industrial production of xylose and xylooligosaccharide from bamboo and for the future conversion and utilization.

Efficient production of xylobiose and xylotriose from xylan in moso bamboo by the combination of pH-controlled lactic acid and xylanase hydrolysis

Baojun Feng,Hong Liao,Wenjun Ying,Junhua Zhang,Zhangjing Chen

Short-chain xylo-oligosaccharides (XOS) such as xylobiose (X2) and xylotriose (X3) have higher biological activities. Therefore, it is interesting to produce highly active XOS enriched with X2 and X3. In this work, pH-controlled lactic acid (LA) hydrolysis was used to produce XOS from xylan in moso bamboo and xylanase was used to convert high DP XOS into low DP XOS to increase the percentage of X2 + X3 in XOS. A 33.1 % XOS yield was obtained from 2 % LA hydrolysis (pH = 3.2). After xylanase hydrolysis of the LA hydrolysate, the total XOS yield reached 64.1 %, with X2 + X3 yield reaching 58.4 %. The percentage of X2 + X3 increased from 52.3 % to a high level of 91.0 %. The deep eutectic solvent pretreatment removed 87.2 % lignin from the residue and the glucose yield of the delignified residue hydrolyzed by cellulase was 96.9 %. The results suggested that the integrated process of LA and xylanase hydrolysis could effectively produce X2 + X3 from xylan in moso bamboo.

Production of Xylooligosaccharide, Nanolignin, and Nanocellulose through a Fractionation Strategy of Corncob for Biomass Valorization

Qixuan Lin,Yuhuan Yan,Xinxin Liu,Bei He,Xiaohui Wang,Xiaoying Wang,Chuanfu Liu,Junli Ren

The three-step fractionation strategy of corncob was performed for the efficient production of xylooligosaccharide, nanolignin, and nanocellulose. High yield of xylooligosaccharide (XOS) was produced from corncob through organic acid–NaOH pretreatment. The influences of the type and concentration of organic acids and the amount and time of NaOH added on the XOS yield were investigated. For resource efficiency, the pretreated corncob residue was further soaked in p-toluenesulfonic acid (p-TsOH) at 80 °C to extract lignin, and then the residue after secondary treatment was catalyzed by TEMPO to produce cellulose nanofibers. Results showed that 38.3% of XOS was achieved by formic acid–NaOH pretreatment, and the addition of NaOH at the later stage of the pretreatment process was proved to prevent the excessive hydrolysis of xylan, resulting in the selective depolylmerization of xylan during pretreatment. In addition, the atomic force microscopy (AFM) analysis showed that nanoscale lignin and cellulose were obtained by the subsequent treatment. TEMPO-oxidized cellulose remarkably swelled in water, and the nanocellulose paper with high transparency was successfully prepared.

Molecular Identification and Engineering a Salt-Tolerant GH11 Xylanase for Efficient Xylooligosaccharides Production

Jiao Ma,Zhongke Sun,Zifu Ni,Yanli Qi,Qianhui Sun,Yuansen Hu,Chengwei Li

This study identified a salt-tolerant GH11 xylanase, Xyn st , which was isolated from a soil bacterium Bacillus sp. SC1 and can resist as high as 4 M NaCl. After rational design and high-throughput screening of site-directed mutant libraries, a double mutant W6F/Q7H with a 244% increase in catalytic activity and a 10 °C increment in optimal temperature was obtained. Both Xyn st and W6F/Q7H xylanases were stimulated by high concentrations of salts. In particular, the activity of W6F/Q7H was more than eight times that of Xyn st in the presence of 2 M NaCl at 65 °C. Kinetic parameters indicated they have the highest affinity for beechwood xylan ( K m = 0.30 mg mL −1 for Xyn st and 0.18 mg mL −1 for W6F/Q7H), and W6F/Q7H has very high catalytic efficiency ( K cat / K m = 15483.33 mL mg −1 s −1 ). Molecular dynamic simulation suggested that W6F/Q7H has a more compact overall structure, improved rigidity of the active pocket edge, and a flexible upper-end alpha helix. Hydrolysis of different xylans by W6F/Q7H released more xylooligosaccharides and yielded higher proportions of xylobiose and xylotriose than Xyn st did. The conversion efficiencies of Xyn st and W6F/Q7H on all tested xylans exceeded 20%, suggesting potential applications in the agricultural and food industries.

Co-production of xylooligosaccharides and activated carbons from Camellia oleifera shell treated by the catalysis and activation of zinc chloride

Yanzhi You,Xiankun Zhang,Pengfei Li,Fuhou Lei,Jianxin Jiang

Camellia oleifera shell (COS) is a worthy byproduct in woody edible oil production enriched in hemicellulose and lignin. This paper aims to explore the high-value transformation of COS for the production of xylooligosaccharides (XOS) with main degree of polymerization (DP) of 2–5 by the catalysis of ZnCl 2 . The effect of pretreatment temperature, reaction time and ZnCl 2 concentration on the contents and DP distributions of XOS were analyzed. Moderate reaction conditions tended to achieve high content XOS, and the maximum value 61.38% and 14.39 g/L of XOS yield and concentration, respectively, peaked at 170 °C for 30 min using 0.5% (w/w) ZnCl 2 . The first time the solid residues derived from the production process of XOS were used as the precursor for the co-production of activated carbons (AC). The maximum iodine values and BET surface area were 5623.94 mg/g and 1244.46 m 2 /g, respectively, using 2.20 M ZnCl 2 as the activating agent .

Insights into the mechanisms underlying the degradation of xylooligosaccharides in UV/H2O2 system

Biyang Zhu,Dongxiao Sun-Waterhouse,Lijun You

UV/H 2 O 2 process is increasingly used to degrade carbohydrates, though the underlying mechanisms remain unclear. This study aimed to fill this knowledge gap, focusing on mechanisms and energy consumption involved in hydroxyl radical (•OH)-mediated degradation of xylooligosaccharides (XOSs) in UV/H 2 O 2 system. Results showed that UV photolysis of H 2 O 2 generated large amounts of •OH radicals, and degradation kinetics of XOSs fitted with a pseudo-first-order model. Xylobiose (X 2 ) and xylotriose (X 3 ), main oligomers in XOSs, were attacked easier by •OH radicals. Their hydroxyl groups were largely converted to carbonyl groups and then carboxy groups . The cleavage rate of glucosidic bonds was slightly higher than that of pyranose ring, and exo -site glucosidic bonds were more easily cleaved than endo -site bonds. The terminal hydroxyl groups of xylitol were more efficiently oxidized than other hydroxyl groups of it, causing an initial accumulation of xylose . Oxidation products from xylitol and xylose included ketoses , aldoses , hydroxy acids and aldonic acids , indicating the complexity of •OH radical-induced XOSs degradation. Quantum chemistry calculations revealed 18 energetically viable reaction mechanisms, with the conversion of hydroxy-alkoxyl radicals to hydroxy acids being the most energetically favorable (energy barriers <0.90 kcal/mol). This study will provide more understanding of •OH radicals-mediated degradation of carbohydrates.
