Conductive hydrogels require tunable mechanical properties, high conductivity and complicated 3D structures for advanced functionality in (bio)applications. Here, we report a straightforward strategy to construct 3D conductive hydrogels by programable printing of aqueous inks rich in poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) inside of oil. In this liquid-in-liquid printing method, assemblies of PEDOT:PSS colloidal particles originating from the aqueous phase and polydimethylsiloxane surfactants from the other form an elastic film at the liquid-liquid interface, allowing trapping of the hydrogel precursor inks in the designed 3D nonequilibrium shapes for subsequent gelation and/or chemical cross-linking. Conductivities up to 301 S m −1 are achieved for a low PEDOT:PSS content of 9 mg mL −1 in two interpenetrating hydrogel networks. The effortless printability enables us to tune the hydrogels’ components and mechanical properties, thus facilitating the use of these conductive hydrogels as electromicrofluidic devices and to customize near-field communication (NFC) implantable biochips in the future.

3D printing hydrogels with a good resolution, diversified geometries, and a tunable mechanical performance is still a big challenge, which is mainly due to the low photo-reactivity of the high-water containing precursor and the low modulus of the photo-cured hydrogel. In the present work, an acryloylmorpholine - acrylic acid - PEG diacrylate drygel was firstly printed with a good fidelity and subsequently post- treated with an aqueous solution containing specific ions, during which hydration of the hydrophilic network and coordination between the carboxyl and ions occurred simultaneously. The obtained dual-crosslinked hydrogel combined a complex geometry and a good stretchability. This approach decoupled the printability and mechanical properties of the hydrogels. To furtherly show this advantage over conventional direct hydrogel 3D printing, ultrasoft hydrogel lattice structures which cannot be self-supported and purely ionic crosslinked hydrogels with a good self-weldability were demonstrated.

Liver cancer organoids replicate the pathophysiology of primary tumors, making them ideal for drug screening and efficacy evaluation. However, their growth in complex, variable, animal-derived matrices hinders practical application. Here, we designed an easily accessible, chemically defined, biocompatible double-network hydrogel (HADR) using methacrylated hyaluronic acid (HAMA), sodium alginate (SA), methacrylamide dopamine (DMA), and c(RGDFC) for liver cancer organoid culture. By optimizing critical extracellular matrix (ECM) parameters, the HADR hydrogel achieves compatibility with the physiological mechanics of the human liver and fosters the adhesion and proliferation of multiple cell types. In vitro drug efficacy tests showed that HepG 2 cell line-derived liver cancer organoids exhibited higher IC50 values than 2D cultures, indicating greater drug resistance. Subcutaneous tumor models in nude mice revealed that HADR hydrogels created a microenvironment for HepG 2 cells mirroring the natural tumor ECM, leading to increased tumor volume, denser cell arrangement, and concurrent microvascular development. In vivo drug efficacy evaluations indicated that DOX treatment downregulated Ki-67 and MMP-9 expression, inhibiting HepG 2 cell proliferation, invasion, and metastasis. These findings demonstrate the potential of HADR hydrogels for liver cancer organoid culture, offering new strategies for personalized drug screening and efficacy evaluation.

Heart valve replacement have numerous problems, such as calcification, inflammation, and limited endothelialization. Herein, we used bio-degradable arginine-modified polylysine and oxidized hyaluronic acid as the substrate, expanding the advantages of rapid Schiff-base reaction of aldehydes and amino groups under physiological conditions to form dynamic acyl-hydrazone bonds. These were used to design dynamic adaptive hydrogels, and a polysaccharide/protein dual-network system that mimicked the extracellular matrix, realizing the excellent mechanical tunability and shear thinning properties of its hydrogel. The dual antibacterial mechanism effectively avoided the occurrence of infection-like inflammation after valve implantation. The hydrogel has great ability in promoting cellular proliferation, and can also promote cell migration through a potential cell recruitment mechanism. The hydrogel dynamically triggers and sustains NO release in vivo , removing highly expressed ROS in inflammatory site as needed. It further promotes angiogenesis, and provides targeted treatment for calcified aortic valve disease caused by oxidative stress signals. Furthermore, it exhibits exceptional anti-calcification properties and can enhance the upregulation of CD31 while suppressing the expression of IL-10 and TNF-α, thereby significantly promoting angiogenesis and reducing inflammation. Additionally, 3D printing technology enables the individualized customization of valves with diverse heterogeneous structures to achieve a high degree of biomimetic tissue.
