Alginate-containing 3D-printed hydrogel scaffolds incorporated with strontium promotes vascularization and bone regeneration

Afeng Miao,Qingsong Li,Genling Tang,Qifeng Lu

Bone defects persist as a significant challenge in the field of clinical orthopedics. This study focuses on the fabrication and characterization of 3D-printed composite hydrogel scaffolds composed of sodium alginate, gelatin, and α-tricalcium phosphate (α-TCP) with varying ratios of Strontium ions (Sr 2+ ). These scaffolds aim to address the clinical challenges associated with bone defect repair by providing mechanical support and promoting bone formation and vascularization. The degradation, swelling, mechanical properties, and release profiles of Sr 2+ from the hydrogel scaffolds were comprehensively characterized. In vitro tests were conducted to assess cell viability and proliferation, as well as osteogenic and angiogenic gene expression, to investigate the osteogenic and pro-angiogenic potential of the composite hydrogel scaffolds. Furthermore, skull defect simulations were performed, and composite scaffolds with varying Sr 2+ ratios were implanted to evaluate their effectiveness in bone repair. This research establishes a foundation for advancing bone tissue engineering through composite scaffolds containing biological macromolecules and strontium, with alginate serving as a key element in enhancing performance and expanding clinical applicability.

Biomimetic construction of a lubricious hydrogel with robust mechanics via polymer chains interpenetration and entanglement for TMJ disc replacement

Yi Hou,Mengmeng Jin,Yao Liu,Nan Jiang,Li Zhang,Songsong Zhu

Developing a temporomandibular joint (TMJ) disc substitute with reliable long-term functional support has faced a huge challenge in replicating the unique microarchitecture, biomechanics and lubrication of the natural tissue. Herein, a novel biomimetic hydrogel consisting of graphene oxide nanosheets modified anionic polyurethane (nGO-APU) and polyvinyl alcohol (PVA) was successfully fabricated through combined freeze-thawing and annealing treatment. Of which, the annealed PVA (a-PVA) with entanglement network acted as the main frame to provide mechanical support, while the interpenetrated nGO-APU chains endowed the function for water retention and lubrication, thus resisting the compressive loads and fulfilling a durable energy absorbing and cushion. The resultant nGO-APU/a-PVA hydrogel achieved better lubrication effect through the synergistic effect of fluid film and boundary lubrication, exhibiting a low friction coefficient of about 0.06, close to the natural TMJ disc. After implantation into rabbit TMJ for up to 24 weeks, the nGO-APU/a-PVA hydrogel kept the structural stability to offer effective protection against wearing of the cartilage, meanwhile ameliorating the worsening of osteoarthritis. The finite element analysis (FEA) further proved that the resultant hydrogel could effectively disperse the applied stress and dissipate energy under practical loading conditions. This strategy based on a biomimetic hydrogel with enhanced mechanical stability and lubrication performance shows clinical promise for TMJ disc replacement.

Ion-induced electrospinning of hierarchical spiderweb-like bioscaffolds

Yayun Wang,Zhigang Chen,Jurui Liu,Zhongqing Wu,Xiao Wang,Junfeng Chen,Hongjing Zhang,Mingming Wu,Kang Yang,Changshun Ruan,Bin Wang

Tissue engineered scaffolds need to possess various functionalities, including biocompatibility, mechanical support, bioactivity , and vascularization. The design and fabrication of bioscaffolds to attain mutual coordination among these functionalities with minimal processing complexity are a highly challenging but rewarding task. In this study, a simple, effective one-step electrospinning method was developed to fabricate hierarchical spiderweb-like bioscaffolds that achieve both superior biological and mechanical functionalities. By incorporating ionic drugs of deferoxamine mesylate and lithium chloride, the spiderweb-inspired structures with adjustable coverages (up to 100 %) were successfully created, imparting the fibrous bioscaffolds with remarkable tensile strengths (∼88.28 MPa). The strengthening mechanisms endowed by the spiderweb structure in optimizing stress distribution to delay damage and enhance load-bearing ability were elucidated through finite element simulations . Furthermore, this hierarchical spiderweb-like bioscaffold demonstrated favorable biological characteristics, including biocompatibility, osteogenesis, angiogenesis, and hemostasis. The presence of the nano-spiderweb structures significantly improved cell adhesion and differentiation on the scaffold and increased the spreading area of cells by 2–3 times. The dual-drug loaded bioscaffolds with full coverages of the spiderweb structure exhibited the least amount of bleeding (45.33 ± 27.47 mg) and the fastest hemostasis speed (82 ± 8.19 s) in the hemostasis test, compared to the control group. Overall, the outstanding performance makes the developed bioscaffolds a promising alternative for tissue repair and regeneration in the field of tissue engineering .

3D Printing Cervical Implant Scaffolds Incorporated with Drug-Loaded Carboxylated Chitosan Microspheres for Long-Term Anti-HPV Protein Delivery

Jingyuan Ji,Chenjia Zhao,Chen Hua,Lu Lu,Yuan Pang,Wei Sun

As attempting personalized medicine, 3D-printed tissue engineering scaffolds are employed to combine with therapeutic proteins/peptides especially in the clinical treatment of infectious diseases, genetic diseases, and cancers. However, current drug-loading methods, such as immersion and encapsulation, usually lead to the burst release of the drugs. To address these issues, we proposed an integrated strategy toward the long-term controlled release of protein. In this study, patient-customized 3D scaffolds incorporated with drug-loaded microspheres were printed to realize the effective delivery of the anti-human papillomavirus (anti-HPV) protein after cervical conization in the treatment of cervical cancer. The 3D-printed scaffold could provide mechanical support to the defect site and ensure local release of the drug to avoid systemic administration. Meanwhile, microspheres serve as functional components to prevent the inactivation of proteins, as well as regulate their release period to meet the time requirement of different treatment courses. The research also utilized bovine serum albumin as a model protein to validate the feasibility of these scaffolds as a generic technology platform. The bioactivity of the released anti-HPV protein was validated using a pseudovirus model, which demonstrated that the microsphere encapsulation would not cause protein denaturation during the scaffold fabrication process. Besides, 3D-printed scaffolds incorporated with carboxylated chitosan microspheres were biocompatible and beneficial for cell attachment, which have been demonstrated by favorable cell viability and better coverage results for HeLa and HFF-1. This study highlights the great potential of scaffolds incorporated with microspheres to serve as tissue engineering candidate products with the function of effective protein delivery in a long-term controlled manner and personalized shapes for clinical trials.
