The quantification of mitochondrial morphology and mechanical properties is useful for the diagnosis and treatment of mitochondrial and alcoholic liver disease. In this study, the effects of ginsenoside Rg1 (G-Rg1) on the morphology and mechanical properties of mitochondria that had suffered alcohol-induced damage were investigated under near-physiological conditions. Additionally, the morphological and mechanical properties of mitochondria were quantified through atomic force microscopy . Atomic force microscopy revealed that alcohol-induced significant morphological changes in mitochondria. Compared with that of the mitochondria of normal hepatocytes, the average surface area of the damaged mitochondria was found to have increased significantly under the influence of alcohol. Furthermore, the mitochondrial area tended to be normal under the action of G-Rg1, whilst other parameters (length, width and perimeter) were significantly different from those of the mitochondria with the alcohol-induced damage. Simultaneously, alcohol significantly reduced the adhesion and elastic modulus of mitochondria, whilst the adhesion and elastic modulus of mitochondria in the G-Rg1 treatment group were closer to the values of normal mitochondria. This study overall showed that G-Rg1 could effectively alleviate the swelling and anomalous mechanical properties of mitochondria induced by alcohol.

The humidity sensors based on metal halide perovskite have achieved outstanding improvement. However, there are few reports of perovskite nanocrystals for humidity sensors. Herein, CsPdBr 3 nanocrystals with high surface-to-volume ratio are synthesized by the in situ spin-coating method. Subsequently, the humidity sensor based on CsPdBr 3 nanocrystals is prepared and the response mechanism is demonstrated to originate from the formation of the Pd––O bond on the surface by performing density functional theory calculation. The humidity sensor shows a fast response/recovery time of 0.68/2.43 s in a relative humidity range from 88.9%RH to 4.7%RH; the fast response/recovery properties are used to monitor human respiratory and noncontact switch. Moreover, the hydromechanical characteristics of unsaturated soil are obtained by noncontact testing with the humidity sensor for the first time in geological engineering, and the strong correlation between relative humidity and hydromechanical characteristics is revealed by mathematical derivation, which lays the theoretical foundation for the noncontact real-time sensing of hydromechanical characteristics of unsaturated soil. These findings not only open the door to the development of perovskite nanocrystals materials for humidity measurement, but also offer a new method for nondestructive and real-time test of the mechanical properties of soil in geological engineering.

With the continuous advancement of electronic devices, flexible thin films with both thermal management functions and excellent electromagnetic interference (EMI) shielding properties have received much attention. Hence, inspired by Janus, a CNF/MXene/ZnFe 2 O 4 @PANI composite film with an asymmetric gradient alternating structure was successfully prepared by adjusting the filler content of the conductive and magnetic layers using a vacuum-assisted filtration method. Benefiting from the magnetic resonance and hysteresis loss of ZnFe 2 O 4 @PANI, conductive loss and dipole polarization of MXene, as well as the exclusive “absorption-reflection-reabsorption” shielding feature in the alternating multilayered films, CM&CZFP-4G film has superior EMI shielding performance, with an EMI SE of up to 45.75 dB and shielding effectiveness of 99.99%. Surprisingly, the composite film maintains reliable EMI shielding properties even after prolonged erosion in harsh environments such as high/low temperatures, high humidity, acids and alkalis. Furthermore, the CM&CZFP-4G responded quickly within about 50 s and reached a maximum steady-state temperature of 235.8 °C at an applied voltage of 9.0 V, indicating the obtained film acquired outstanding and controllable Joule heating performance. This result was attributed to the homogeneous dispersion of MXene to build up a conductive network and endow the CNF/MXene with high conductivity. Meanwhile, the fire resistance of CM&CZFP-4G was significantly improved compared to pure CNF, which guaranteed fire safety during its application. Additionally, contributed by long fiber entanglement of CNF, extensive hydrogen-bonding interactions and multilayer structural design, the CM&CZFP-4G film exhibits excellent mechanical characteristics, with the tensile strength and fracture strain of 27.74 MPa and 6.21%, separately. This work offers a creative avenue to prepare multifunctional composite films with electromagnetic shielding and Joule heating for various application environments.

Healing of the cutaneous wound requires macrophage recruitment at the sites of injury, where chemotactic migration of macrophages toward the wound is regulated by local inflammation. Recent studies suggest a positive contribution of DNA methyltransferase 1 (Dnmt1) to macrophage pro-informatory responses; however, its role in regulating macrophage motility remains unknown. In this study, myeloid-specific depletion of Dnmt1 in mice promoted cutaneous wound healing and de-suppressed the lipopolysaccharides (LPS)-inhibited macrophage motility. Dnmt1 inhibition in macrophages eliminated the LPS-stimulated changes in cellular mechanical properties in terms of elasticity and viscoelasticity. LPS increased the cellular accumulation of cholesterol in a Dnmt1-depedent manner; cholesterol content determined cellular stiffness and motility. Lipidomic analysis indicated that Dnmt1 inhibition altered the cellular lipid homeostasis, probably through down-regulating the expression of cluster of differentiation 36 CD36 (facilitating lipid influx) and up-regulating the expression of ATP-binding cassette transporter ABCA1 (mediating lipid efflux) and sterol O-acyltransferase 1 SOAT1 (also named ACAT1, catalyzing the esterification of cholesterol). Our study revealed a Dnmt1-dependent epigenetic mechanism in the control of macrophage mechanical properties and the related chemotactic motility, indicating Dnmt1 as both a marker of diseases and a potential target of therapeutic intervention for wound healing.

This study aimed to evaluate the efficiency of edible films made from chlorogenic acid/chitosan (CGA/CS) nanoparticles combined with photodynamic technology (PDT). Hydroxypropyl starch (HS) and κ-carrageenan (KC) were used as the main ingredients in the preservation of Mongolian cheese under the PDT condition. The mechanical characteristics, water vapor adsorption, solubility, permeability, and release of chlorogenic acid in aqueous media were evaluated. The incorporation of CGA/CS significantly enhanced the tensile strength and barrier characteristics of the edible films. The antimicrobial efficacy of the edible film was assessed over a period of 7 days while the cheese was being stored, followed by PDT application. The use of antimicrobial PDT did not cause lipid oxidation in cheese samples. Additionally, the combination of CGA/CS@HS/KC helped to reduce fat oxidation in Mongolian cheese. Utilizing an edible film in conjunction with PDT presents a viable solution for prolonging the shelf life of Mongolian cheese while maintaining its sensory attributes and nutritional qualities.

Effective storage and utilization of limited donor corneal resources are in high demand to alleviate the shortage of donor corneal tissue. Here, we designed a static air-lifted organ culture system equipped with a protective coverage membrane, namely, an air-lifted OC-P system, to provide a biomimetic physiological environment for full-thickness corneal preservation. The air-lifted OC-P system features a unique collagen-based protective coverage membrane that can offer a moist, oxygen-rich environment for corneal epithelium, produce an appropriate intraocular pressure onto the cornea by gravity, and facilitate the maintenance of the organ culture medium level for nutrient supply during corneal preservation. Compared with conventional submerged and air-lifted corneal preservation methods, the air-lifted OC-P system remarkably improved the overall quality of the preserved corneas. These preserved corneas not only exhibited superior controllability of corneal swelling and extraordinary maintenance of the morphology and viability of all three types of corneal cells (i.e., corneal epithelium, keratocytes, and endothelium) but also demonstrated optimal optical, thermal, and mechanical properties. This air-lifted OC-P system presents a biomimetic strategy that provides a static and efficient method to replicate the corneal natural conditions for corneal preservation effectively.

The contamination of bone defects is a serious therapeutic problem. The treatment of infected bone defects involves rigorous infection control followed by bone reconstruction. Considering these two processes, the development of biomaterials possessing antibacterial and osteogenic properties offers a promising approach for the treatment of infected bone defects. In this study, a dual-functional, thermosensitive, and injectable hydrogel composed of chitosan (CS), quaternized CS (QCS), and nano-hydroxyapatite (nHA) was designed, and the ratio of CS to QCS in the hydrogel was optimized to enhance the antibacterial efficacy of CS while reducing the cytotoxicity of QCS. In vitro studies demonstrated that the hydrogel with an 85 %:15 % ratio of CS to QCS exhibited excellent biocompatibility and antibacterial properties while also possessing suitable mechanical characteristics and degradability. The incorporation of nHA into the hydrogel enhanced MC3T3-E1 proliferation and osteogenic differentiation. Moreover, this hydrogel demonstrated superior in vivo therapeutic effectiveness in a rabbit model of infected bone defect. In summary, this study provides a promising material design and a comprehensive one-step treatment strategy for infected bone defects.
