Insecticidal Endostemonines A–J Produced by Endophytic Streptomyces from Stemona sessilifolia

Huimin Zhao,Aiping Yang,Nan Zhang,Shiyang Li,Tianjie Yuan,Ning Ding,Siwang Zhang,Sheng Bao,Chang Wang,Yinan Zhang,Xiachang Wang,Lihong Hu

The discovery of new, safe, and effective pesticides is one of the main means for modern crop protection and parasitic disease control. During the search for new insecticidal secondary metabolites from endophytes in Stemona sessilifolia (a traditional Chinese medicine with a long history as an insecticide), 10 new insecticidal endostemonines A–J (1–10) were identified from an endophytic Streptomyces sp. BS-1. Their structures were determined by comprehensive spectroscopic analysis. Endostemonines A–J represent the first reported naturally occurring pyrrole-2-carboxylic ester derivatives, which consisted of different fatty acid chains at the C-2 of pyrrole ring were produced by traditional Chinese medicine endophytic microbes. All new tested compounds exhibited strong lethal activity against Aphis gossypii (LC50 value range of 3.55–32.00 mg/L after 72 h). This research highlighted the discovery of pesticide natural products from insecticidal medicinal plant endophytes for the first time, paving a new pathway for the development of pest control.

Meta-Aerogel Electric Trap Enables Instant and Continuable Pathogen Killing in Face Masks

Dingming Yu,Lifang Liu,Jianyong Yu,Yang Si,Bin Ding

The tremendous menace of the COVID-19 pandemic has underscored the urgency for antipathogen masks to stop the transmission of airborne infectious diseases. Most prevailing antipathogen masks manifest a slower sterilization rate that lags behind the pathogen momentum traversing the masks, thereby engendering an elevated susceptibility to infection. Here we tailor nanofibrous meta-aerogel electric traps, 3D-assembled from self-knotted carbon nanotube networks in an all rigid nanofibrous skeleton. This superior configuration revolves around the creation of numerous “dielectrophoretic–aerodynamic grippers”, which are capable of directional manipulation of microbes toward the region of the lethal intensive electric field. Based on this, we present a disinfection unit comprising a pair of aerogel electrodes that demonstrate a rapid killing rate (>99.99% biocidal efficacy within 0.016 s) and long-term durability (12 h of continuous operation). Additionally, a microbutton lithium cell is employed as a power supply to fabricate an antipathogen face mask with this disinfection unit, which exhibits superior pathogen inactivation efficacy compared to commercial masks. This scalable biocidal protective equipment holds great potential for use in emergency medical services.

Cationic Glycopolymers with Aggregation-Induced Emission for the Killing, Imaging, and Detection of Bacteria

Die Li,Jing Chen,Mei Hong,Yan Wang,David M. Haddleton,Guang-Zhao Li,Qiang Zhang

Cationic glycopolymers with structures similar to those of typical poly(ionic liquid)s (PILs) were synthesized via the quaternization reaction of poly(4-vinyl pyridine) with halogen-functionalized d-mannose and tetraphenylethylene units. Such postpolymerization modification provided PILs with aggregation-induced emission effect as well as specific carbohydrate-protein recognition with lectins such as concanavalin A. The interactions between cationic glycopolymers and different microorganisms, including Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli, were used for the killing, imaging, and detection of bacteria. Besides, these sugar-containing PILs showed a relatively low hemolysis rate due to the presence of saccharide units, which may have potential application in the field of biomaterials.

Light-Enhanced Bacterial Killing and Less Toxic Cell Imaging: Multicationic Aggregation-Induced Emission Matters

Hengchang Ma,Yucheng Ma,Lei Lei,Weidong Yin,Yuan Yang,Tao Wang,Pei Yin,Ziqiang Lei,Manyi Yang,Yanfang Qin,Shaoxiong Zhang

The newly emerged pathogenic bacteria leads to the bactericides generations one by one, which is definitely a long-term and costly process. For the small organic molecule used as antibacterial materials, photostability, biomembrane penetrating ability, and long-term antibacterial ability still are questionable. In this work, we synthesized the multiple-cation-charged aggregation-induced emission (AIE)-active polymer 1,2-diphenyl-1,2-bis(4-(pyridin-4-yl)phenyl) ethane–1,8-dibromooctane (DBPE-DBO). Due to the incorporating the dark toxicity from positively charged polymers, and the phototoxicity from generation of singlet oxygen, as well as AIE-active fluorescence performance, DBPE-DBO could be employed as an efficient bacteria targeting, imaging, and killing materials. Interestingly, DBPE-DBO was less toxic to the normal mammalian cells, which leads us to verify that DBPE-DBO could be used as a very cell-membrane permeable nucleus imaging probes. Thus, our research results may provide a new concept for the designing advanced biomaterials with versatile antibacterial properties and cell imaging ability.

Bacteria killing and release of salt-responsive, regenerative, double-layered polyzwitterionic brushes

Lei Huang,Lixun Zhang,Shengwei Xiao,Yin Yang,Feng Chen,Ping Fan,Zhenping Zhao,Mingqiang Zhong,Jintao Yang

Antibacterial surfaces that not only can kill bacteria but also can release the dead bacteria have become one of most promising antibacterial strategies, but the realization of high efficiency, rapid response and long-term reusability of such surfaces remains many challenges. Herein, we reported new antibacterial surfaces with high killing and release capabilities based on a salt-responsive brush recently developed by our group. These surfaces consist of two polymer brushes with two-layer architecture, where the upper-layer bactericidal brushes (poly[(trimethylamino) ethyl methacrylate chloride], polyMETAC or poly[2-(tert-butylamino)ethyl methacrylate], polyTA) were built on background layer of salt-responsive polyzwitterionic brush (poly(3-(dimethyl(4-vinylbenzyl) ammonio) propyl sulfonate), polyDVBAPS) via sequential surface initiated atom transfer radical polymerization . By the combination in this manner, the bacteria attach/release function of polyDVBAPS and bactericidal function of polyMETAC/polyTA were successfully integrated, resulting in the smart surfaces which can reversibly kill and release bacteria in response to the switch between water and salt solution. Both two systems in this study, i.e. poly (DVBAPS-b-METAC) and poly(DVBAPS-b-TA), exhibit high bactericidal activity by killing more than 93% and highly efficient regeneration capability by rapidly releasing ∼90% of the attached bacteria. Specifically, such excellent antibacterial performance of poly(DVBAPS-b-TA) can be well retained even after four killing-release cycles, indicating the great potential in reusable biological materials and devices. Furthermore, the correlation between physicochemical properties of the two-layer brushes with their bactericidal efficiency and regenerative capability was also investigated and discussed.

Proton-mediated burst of dual-drug loaded liposomes for biofilm dispersal and bacterial killing

Da-Yuan Wang,Guang Yang,Xiao-Xiao Zhang,Henny C. van der Mei,Yijin Ren,Henk J. Busscher,Linqi Shi

Exposure of infectious biofilms to dispersants induces high bacterial concentrations in blood that may cause sepsis. Preventing sepsis requires simultaneous biofilm dispersal and bacterial killing. Here, self-targeting DCPA(2-(4-((1,5-bis(octadecenoyl)1,5-dioxopentan-2-yl)carbamoyl)pyridin-1-ium-1-yl)acetate) liposomes with complexed water were self-assembled with ciprofloxacin loaded in-membrane and PEGylated as a lipid-membrane component, together with bromelain loaded in-core. Inside biofilms, DCPA-H 2 O and PEGylated ciprofloxacin became protonated, disturbing the balance in the lipid-membrane to cause liposome-burst and simultaneous release of bromelain and ciprofloxacin. Simultaneous release of bromelain and ciprofloxacin enhanced bacterial killing in Staphylococcus aureus biofilms as compared with free bromelain and/or ciprofloxacin. After tail-vein injection in mice, liposomes accumulated inside intra-abdominal staphylococcal biofilms. Subsequent liposome-burst and simultaneous release of bromelain and ciprofloxacin yielded degradation of the biofilm matrix by bromelain and higher bacterial killing without inducing septic symptoms as obtained by injection of free bromelain and ciprofloxacin. This shows the advantage of simultaneous release from liposomes of bromelain and ciprofloxacin inside a biofilm.

Butyrate-inserted Ni–Ti layered double hydroxide film for H2O2-mediated tumor and bacteria killing

Donghui Wang,Feng Peng,Jinhua Li,Yuqin Qiao,Qianwen Li,Xuanyong Liu

The selective tumor- and bacteria-killing property is promising for biomedical implants in directly contact with tumor tissue but still presents a considerable challenge. In this study, a butyrate-inserted Ni–Ti layered double hydroxide film (LDH/Butyrate) is prepared on the surface of nitinol alloy via a simple hydrothermal treatment. The prepared film can selectively inhibit tumor growth and metastasis and bacterial infection by taking advantage of the overproduced H 2 O 2 in tumor and infection microenvironments: the pro-tumor and pro-infection molecule H 2 O 2 can be consumed by LDH/Butyrate, and cytotoxic butyrate inserted in LDH/Butyrate is subsequently exchanged out. Such a novel endogenous stimuli-responsive platform is expected to prevent tumor overgrowth, metastasis and bacterial infection and will find a promising application in the design of localized drug-eluting systems.

Dual Electric Fields induced Synergistic Photocatalytic Inactivation: Unveiling Mechanisms and Phenomena of Enhanced Performance

Rui Chen,Na Wang,Yiwen Xie,Yayun Hu,Yuxia Song,Rong Wang

Photocatalytic inactivation, emerging as a novel bactericidal technique, is gaining increasing attention. While oxygen vacancies (OVs) and heterogeneous junction structures have been extensively investigated to enhance photocatalytic performance, the synergistic effect between the two remains largely unexplored. In this study, a Bi 2 S 3 @BiOCl-OV heterojunction structure enriched with oxygen vacancies as a promising photocatalyst was developed. Bi 2 S 3 @BiOCl-OV effectively improved photocatalytic properties due to the presence of oxygen vacancies and the unique structure of Z-type heterojunction. Further mechanism investigation shows that the unique heterojunction structure can induce the formation of internal electric field, the existence of oxygen vacancies can promote the generation of micro local electric field, and effectively promote the charge redistribution under the action of dual-electric field. Simultaneously, O 2 and H 2 O in the system are adsorbed by oxygen vacancies, and are activated by electrons to form the reactive oxygen species, thereby achieving the goal of killing microorganisms.

Regenerable bacterial killing–releasing ultrathin smart hydrogel surfaces modified with zwitterionic polymer brushes

Shengwei Xiao,Yuyu Zhao,Shuqi Jin,Zhicai He,Gaigai Duan,Haining Gu,Hongshun Xu,Xingyu Cao,Chunxin Ma,Jun Wu

Building long-lasting antimicrobial and clean surfaces is one of the most effective strategies to inhibit bacterial infection, but obtaining an ideal smart surface with highly efficient, controllable, and regenerative properties still encounters many challenges. Herein, we fabricate an ultrathin brush–hydrogel hybrid coating (PSBMA-P(HEAA- co -METAC)) by integrating antifouling polyzwitterionic (PSBMA) brushes and antimicrobial polycationic (P(HEAA- co -METAC)) hydrogels. The smart bacterial killing–releasing properties can be achieved independently by the opposite volume and conformation changes between the swelling (shrinking) of P(HEAA- co -METAC) hydrogel layer and the shrinking (swelling) of PSBMA brushes. The friction test reveals that both METAC and SBMA components support great lubrication. By tuning the initial organosilane (BrTMOS:KH570) ratios, the prepared PSBMA-P(HEAA- co -METAC) coating exhibits different antibacterial abilities from single “capturing–killing” to versatile “capturing–killing–releasing.” Most importantly, 99% of the bacterial-releasing rate can be easily achieved via 0.5 M NaCl treatment. This smart surface not only possesses long-lasting antibacterial performance, only ∼1.09 × 10 5 cell·cm −2 bacterial residue even after 72 h exposure to bacteria solutions, but also can be regenerated and triggered between water and salt solution multiple times. This work provides a new way to fabricate antibacterial smart hydrogel coatings with bacterial “killing–releasing” functions and shows great potential for biomedical applications.

Two-dimensional NbS2 nanosheets with hyperthermia for killing bacteria to promote infected wound healing

Dongqian Xu,Xiaojun He,Enoch Obeng,Zhangyan Ye,Jianliang Shen,Xi Ding

Antibiotic resistance and bacteria-related infections pose a threat to lives around the globe, which calls for urgent development of advanced therapy to curb this menace. Although photothermal therapy (PTT) is promising, most photothermal agents lack good photothermal conversion performance which restrains their application. Transition metal dichalcogenides (TMDs) nanosheets can exhibit many advantages in antibacterial applications due to their unique two-dimensional structure with larger specific surface area and higher near-infrared (NIR) absorption. Here, we prepared two-dimensional NbS 2 nanosheets (NbS 2 NSs) with good biological stability and high biocompatibility through a high-temperature reduction reaction. NbS 2 NSs exhibited excellent and stable photothermal properties, which were capable of killing Pseudomonas aeruginosa ( P. aeruginosa ) and Staphylococcus aureus ( S. aureus ) under 5 min of 808 nm laser irradiation. In addition, the morphological characterization of bacteria showed bacterial cell membrane destruction. Furthermore, NbS 2 NSs under NIR irradiation inhibited the formation of biofilms and quickly eradicated S. aureus from an infected wound to promote wound healing through a hyperthermia effect. Overall, this work provides an attractive strategy for synthesizing and applying two-dimensional NbS 2 NSs with significant implications for bacterial eradication and future biomedical applications.

Host–Guest Self-Assembly Toward Reversible Thermoresponsive Switching for Bacteria Killing and Detachment

Zhen-Qiang Shi,Yu-Ting Cai,Jie Deng,Wei-Feng Zhao,Chang-Sheng Zhao

A facile method to construct reversible thermoresponsive switching for bacteria killing and detachment was currently developed by host–guest self-assembly of β-cyclodextrin (β-CD) and adamantane (Ad). Ad-terminated poly(N-isopropylacrylamide) (Ad-PNIPAM) and Ad-terminated poly[2-(methacryloyloxy)ethyl]trimethylammonium chloride (Ad-PMT) were synthesized via atom transfer radical polymerization, and then assembled onto the surface of β-CD grafted silicon wafer (SW-CD) by simply immersing SW-CD into a mixed solution of Ad-PNIPAM and Ad-PMT, thus forming a thermoresponsive surface (SW-PNIPAM/PMT). Atomic force microscopy (AFM), X-ray photoelectron spectrometry (XPS), and water contact angle (WCA) analysis were used to characterize the surface of SW-PNIPAM/PMT. The thermoresponsive bacteria killing and detachment switch of the SW-PNIPAM/PMT was investigated against Staphyloccocus aureus. The microbiological experiments confirmed the efficient bacteria killing and detachment switch across the lower critical solution temperature (LCST) of PNIPAM. Above the LCST, the Ad-PNIPAM chains on the SW-PNIPAM/PMT surface were collapsed to expose Ad-PMT chains, and then the exposed Ad-PMT would kill the attached bacteria. While below the LCST, the previously collapsed Ad-PNIPAM chains became more hydrophilic and swelled to cover the Ad-PMT chains, leading to the detachment of bacterial debris. Besides, the proposed method to fabricate stimuli-responsive surfaces with reversible switches for bacteria killing and detachment is facile and efficient, which creates a new route to extend the application of such smart surfaces in the fields requiring long-term antimicrobial treatment.

A modified hyaluronic acid hydrogel with strong bacterial capture and killing capabilities for drug-resistant bacteria-infected diabetic wound healing

Yulong Lan,Yao Wang,Xiaoliang Qi,Erya Cai,Yajing Xiang,XinXin Ge,Hangbin Xu,Xiaojing Chen,Ying Li,Yizuo Shi,Jianliang Shen,Zhiyong Liao

Management of diabetic wounds becomes increasingly challenging as bacterial infections intensify the inflammation. Employing polysaccharide hydrogels with inherent antibacterial qualities can significantly reduce the need for antibiotics to manage infections in diabetic wounds. The typical approach to achieving antibacterial outcomes with hydrogels relies on the penetration of bacteria into their porous architecture. Such penetration not only takes time but can also prolong inflammation, thus impeding the healing of wounds. Hence, the quick capture and eradication of bacteria are essential for optimizing the hydrogel's antibacterial performance. Herein, we introduce a multifunctional polysaccharide hydrogel dressing—designated as HAQ—created for managing bacterial infections in diabetic wounds. This dressing is based on hyaluronic acid, which is modified with methacrylic anhydride, and special functional groups are added to the modified hyaluronic acid matrix: phenylboronic acid for capturing bacteria and quaternary ammonium chitosan for bacterial destruction. As expected, the HAQ system exhibits robust antibacterial effectiveness against both methicillin-resistant Staphylococcus aureus and multidrug-resistant Pseudomonas aeruginosa in vitro and in vivo. Consequently, HAQ stands as a promising hydrogel dressing with intrinsic antibacterial capabilities and offers significant potential for managing diabetic wounds infected by drug-resistant bacteria.
