Preparation of Azoxystrobin-Zinc Metal–Organic Framework/Biomass Charcoal Composite Materials and Application in the Prevention and Control of Gray Mold in Tomato

Xiao Han,Yinjie Qian,Jiapeng Li,Zhongkai Zhang,Jinbo Guo,Ning Zhang,Longyu Liu,Zhiqiang Cheng,Xiaobin Yu

In order to reduce the use of fungicide and ensure food safety, it is necessary to develop fungicide with low toxicity and high efficiency to reduce residues. Azoxystrobin (AZOX), which is derived from mushrooms, is an excellent choice. However, conventional AZOX release is difficult to regulate. In this paper, a pH-responsive fungicide delivery system for the preparation of AZOX by impregnation method was reported. The Zinc metal–organic framework/Biomass charcoal (ZIF-8/BC) support was first prepared, and subsequently, the AZOX-ZIF-8/BC nano fungicide was prepared by adsorption of AZOX onto ZIF-8/BC by dipping. Gray mold, caused byBotrytis cinerea, is one of the most important crop diseases worldwide. AZOX-ZIF-8/BC could respond to oxalic acid produced byBotrytis cinereato release loaded AZOX. When pH = 4.8, it was 48.42% faster than when pH = 8.2. The loading of AZOX on ZIF-8/BC was 19.83%. In vitro and pot experiments showed that AZOX-ZIF-8/BC had significant fungicidal activity, and 300 mg/L concentration of AZOX-ZIF-8-BC could be considered as a safe and effective control ofBotrytis cinerea. The above results indicated that the prepared AZOX-ZIF-8/BC not only exhibited good drug efficacy but also demonstrated pH-responsive fungicide release.Keywords:AZOX-ZIF-8/BC;Botrytis cinerea;fungicide;nano fungicide;pH-responsive release

Impact of Fungicide Program and Powdery Mildew Resistance in Three Varieties of Pumpkin

Timothy Coolong,Kenneth Seebold

The effects of fungicide program and varietal resistance on the development of cucurbit powdery mildew [PM (Podosphaera xanthii)] in pumpkin (Cucurbita pepo) were tested in 2009 and 2010 in Lexington, KY. Three fungicide programs consisted of a no-fungicide control, low input (chlorothalonil), and high input (chlorothalonil alternated with myclobutanil) were applied to three varieties of pumpkin with different levels of cucurbit PM resistance. Varieties tested were Howden, Aladdin, and Camaro, which had no, moderate, and high levels of cucurbit PM resistance, respectively. There were no significant variety by fungicide program interactions for yield, number fruit per acre, and average fruit weight in both years. In 2009 and 2010, variety significantly affected yield. Yield increases corresponded to the level of cucurbit PM resistance. The high-resistance variety, Camaro, had the greatest yields in both years, while Howden, with no PM resistance, had the lowest yields. In 2010, yield significantly increased with the intensity of fungicide program. With the exception of cull percentage in 2009, there were no fungicide program by variety interactions for yield parameters. In both years, PM disease severity, characterized by the area under the disease progress curve (AUDPC), was lowest on ‘Camaro’ and highest on ‘Howden’. In 2009, unsprayed ‘Camaro’ had similar AUDPC values as ‘Aladdin’ and ‘Howden’ treated with low- or high-input fungicide programs. In 2010, unsprayed ‘Camaro’ had less disease than ‘Aladdin’ or ‘Howden’ that had received the high-intensity fungicide program. These results suggest that yields can be maintained with a minimal fungicide program when using a variety with high cucurbit PM resistant in some environments. A high-input fungicide program may be necessary to maintain yields of varieties with low cucurbit PM resistance.

Insights into enhanced toxic effects by the binary mixture of carbendazim and procymidone on hepatic lipid metabolism in mice

Zhiwei Bao,Dou Wang,Yao Zhao,Ting Luo,Guilin Yang,Yuanxiang Jin

Carbendazim (CBZ) and procymidone (PRO) are two widely used fungicides in agriculture. However, there are still gaps in knowledge regarding about the potential hazards of joint exposure to CBZ and PRO in animals. Here, 6-week-old ICR mice were exposed to CBZ, PRO and CBZ + PRO for 30 days, and metabolomics were performed to discover the mechanism by which the mixture enhanced the effects on lipid metabolism. Co-exposure to CBZ + PRO elevated the body weights, relative liver weights and relative epididymis fat weights, but not in the single exposure groups. Molecular docking analysis suggested that CBZ and PRO combined with peroxisome proliferator-activated receptor (PPARγ) at the same amino acid site as the agonist rosiglitazone. The RT-qPCR and WB results demonstrated that the levels of PPARγ were higher in the co-exposure group than in the single exposure groups. In addition, hundreds of differential metabolites were discovered by metabolomics and enriched in different pathways, such as pentose phosphate pathway and purine metabolism. A unique effect, a decrease in glucose-6-phosphate (G6P) that promoted more NADPH production, was observed in the CBZ + PRO group. These results demonstrated that exposure to CBZ + PRO caused more serious lipid metabolism disorder in the liver than exposure to a single fungicide, which could provide some new insight for the toxic effects after fungicides joint exposure.

Activity of a SDHI fungicide penflufen and the characterization of natural-resistance in Fusarium fujikuroi

Yanan Sun,Haiping Shi,Chenxin Mao,Jianyan Wu,Chuanqing Zhang

The occurrence of bakanae disease of rice caused by the fungus Fusarium fujikuroi in Zhejiang Province has become increasingly aggravated in recent years, concomitant with the development of resistance to the widely applied fungicides , prochloraz and phenamacril. In this study, the activity of a novel succinate dehydrogenase inhibitor (SDHI) fungicide, penflufen, against different fungi was evaluated in addition to the potential of penflufen in controlling F. fujikuroi infections. Penflufen exhibited good bioactivity against F. fujikuroi , but weak activity against Fusarium spp. and other investigated plant-pathogenic fungi including Colletotrichum spp. In addition to inhibiting mycelial growth , penflufen effectively inhibited F. fujikuroi conidium production. For germination, penflufen could effectively inhibit that of small conidia, but only delay the germination of large conidia. In addition, the sensitivity to penflufen among 100  F. fujikuroi isolates that were collected in areas that were never exposed to SDHIs was determined based on mycelium growth. Sensitivities surprisingly exhibited bimodal distributions, indicating the presence of natural resistance. Cross-resistance was not observed between penflufen in F. fujikuroi and two fungicides that have been extensively applied in field including prochloraz (a DMI) and phenamacril (a 2-cyanoacrylate fungicide), nor with the three SDHIs, fluopyram, benzovindiflupyr, and pydiflumetofen. Additional analysis identified five different point mutations in SDH-A (i.e., at residues 46, 225, 283, 430, and 586) of naturally resistant isolates. These results inform the potential application of the new SDHI fungicide penflufen for managing crop diseases and understanding possible resistance mechanisms among pathogens .

First report of Bipolaris bicolor causing a leaf spot disease on rubber tree

Xiaoyu Liang,Yun Peng,Yanchao Liu,Meng Wang,Ye Yang,Yu Zhang

The rubber tree ( Hevea brasiliensis ) is the only resource for commercial natural rubber production and thus has economic importance in Southeast Asia. A spot disease on the leaves of a rubber tree was first discovered in 2017 in Hainan, China. In this study, the fungal isolate MA1 from the infected tissues was determined to be a pathogen of the spot disease by satisfying Koch's postulates. The isolate MA1 was identified as Bipolaris bicolor based on the morphological characteristics and multigene phylogenetic analysis. Among fungicides, prochloraz, iprodione and pyraclostrobin significantly inhibited hyphal growth of B. bicolor under in vitro conditions. This study constitutes the first report on the association of B. bicolor with leaf spot disease of rubber trees worldwide.

Sub-Chronic Difenoconazole Exposure Induced Gut Microbiota Dysbiosis in Mice

Zhiwei Bao,Weitao Wang,Xiaofang Wang,Mingrong Qian,Yuanxiang Jin

Difenoconazole (DIF) is a widely separated triazole fungicide in many countries. The excessive usage of DIF increases the high volume of residues in agriculture production and water bodies. Some previous studies demonstrated the toxic effects of DIF on non-target animals, however, there were still some gaps in the knowledge of the potential hazards of DIF to mammals and human health. Herein, 7-week-old male mice were exposed to 30 and 100 mg/kg/day DIF for 14 and 56 days. We observed that 56 days of DIF exposure decreased the colonic mucus expression of alcin blue-periodic acid-schiff (AB-PAS) stain and the immunochemical stain ofmuc2protein. The transcript levels of mucin protein (muc1,muc2andmuc3) decreased significantly in the gut of mice followed 56 days of 100 mg/kg/day DIF exposure. In addition, the gut microbiota composition was also affected after 14 or 56 days of DIF exposure. Although the mucus expression after 14 days of DIF exposure only decreased slightly, the gut microbiota composition compared with the control group was changed significantly. Moreover, the DIF-30 and DIF-100 caused respectively different changes on the gut microbiota. The relative abundance of Bacteroidetes decreased significantly after 14 days and 56 days of DIF exposure. After 14 days of DIF exposure, there were 35 and 18 differential genera in the DIF-30 and DIF-100 group, respectively. There were 25 and 32 differential genera in the DIF-30 and DIF-100 group after 56 days of exposure, respectively. Meanwhile, the alpha diversity indexes, including observed species, Shannon, Simpson, Chao1 and ACE, in gut microbiota decreased significantly after 56 days of DIF exposure. Interestingly, the relative abundance ofAkkermansiaincreased significantly after 56 days of 100 mg/kg/d DIF exposure. AlthoughAkkermansiawas considered as one probiotic, the phenomenon of dramaticAkkermansiaincrease with the decrease in gut microbiota diversity needed further discussion. These results provided some new insights on how DIF exposure impacts the mucus barrier and induces gut microbiota dysbiosis. Keywords:fungicides;Difenoconazole;gut microbiota;mice

A beta-cyclodextrin-functionalized magnetic metal organic framework for efficient extraction and determination of prochloraz and triazole fungicides in vegetables samples

Guangyang Liu,Lingyun Li,Yuhang Gao,Mingkun Gao,Xiaodong Huang,Jun Lv,Donghui Xu

A β-cyclodextrin-functionalized magnetic zinc-metal organic framework (M-MOF/β-CD) was synthesized via a facile one-pot reaction. M-MOF/β-CD was used as a magnetic porous absorbent for the extraction and determination of prochloraz and three triazole fungicides in vegetable samples. M-MOF/β-CD was prepared by creating MOF layers on the surface of a Fe 3 O 4 –graphene oxide (GO) nanocomposite and bonding them with β-CD molecules. Characterization suggested that a 3D porous structure was formed, with M-MOF/β-CD exhibiting high superparamagnetism and a large surface area. As a new strategy, integrating MOFs with Fe 3 O 4 –GO could improve their water-resistance and mechanical strength by providing a rigid nanosupport interface. Combining M-MOF and β-CD resulted in excellent selective adsorption capacities for prochloraz and three triazole fungicides. The static adsorption process was evaluated and the results were in good agreement with the Freundlich model. Subsequently, M-MOF/β-CD was applied to extracting prochloraz and triazole fungicides from tomato and lettuce vegetables, followed by HPLC-MS/MS determination. The limits of detection for the above fungicides were found to be 0.25–1.0 μg/L at a signal-to-noise ratio of 3, with spiked recoveries of 74.13%–119.83%, indicating that M-MOF/β-CD was promising for application to the extraction and determination of fungicides in complex matrices.

Photolysis of Fungicides on Simulated Leaf Surfaces vs. Aqueous Solutions: Pathways, Kinetics, and Environmental Detoxification Insights

Chenyang Zhang,Xuewei Zhang,Xuerui Yang,Guangli Xiu,JM CHOVELON,Lei Zhou

The photochemical behaviors of fungicides under irradiation differ significantly between leaf surfaces and aqueous solutions. However, few studies have compared the photolysis of fungicides between these two environmental media. In this study, three typical fungicides—kresoxim-methyl (KM), pyraclostrobin (PAS), and cyprodinil (CRL)—were examined using carnauba wax to simulate leaf surfaces The photolysis kinetics, products, and pathways were systematically compared between aqueous solutions and wax films. On wax film, the photolysis rate constants of selected fungicides were observed to be significantly faster than in aqueous solution, with increases of 44, 5 and 36-fold for KM, PAS and CRL, respectively. The primary photolysis pathways on wax films involved photoisomerization and ether cleavage; while the hydroxylation process, commonly observed in aqueous systems, was absent. Toxicity assessments demonstrated that photolysis effectively reduced their ecotoxicities in both systems, with the photolysis on wax film documented as a more profound detoxification process. Moreover, wax film thickness and pesticide additives significantly influenced photodegradation kinetics of all the three fungicides. The present study highlighted the significant influence of environmental media on the photodegradation pathways of pesticides.

Dissipation Behavior and Residue Distribution of Famoxadone and Cymoxanil in Cucumber and Soil Ecosystem Under Open-Field Conditions

Rao Jifa,Chen Ling,Xiang Yujia,Li Jiayu,Chen Xiao,Zhou Yaoyu

In this study, a practical and excellent method was used to determine the famoxadone and cymoxanil via high-performance liquid chromatography equipped utilizing ultraviolet detector lamp (HPLC-UV) for investigating the dissipation behavior and residue distribution of famoxadone and cymoxanil in cucumber and soil ecosystem. The limit of quantification (LOQ S ) of famoxadone and cymoxanil in cucumber were 0.50, 1.00, 2.00 and 0.05, 0.50, 1.00 mg kg −1 in soil, respectively. The limit of detection (LOD S ) of both famoxadone and cymoxanil were 8.0 ng. The average recoveries ( n  = 5) of the fungicide ranged from 84.10 to 108.02% with the relative standard deviations (RSDs) typically < 9.23%. The fungicide was applied to cucumber and soil at the range of doses (275.6–413.4 g a.i.ha −1 ) three or four times. The half-lives of famoxadone and cymoxanil in cucumber and soil were 1.34–16.12 days, which followed the first-order chemical reaction kinetics equation C t  =  C 0  ×  e -kt . The residues of famoxadone and cymoxanil in cucumber at the pre-harvest interval (PHI, 3 days) were below 8.0 × 10 −8  g and 8.0 × 10 −9  g, respectively. Overall, this study evaluated the food safety and the environmental fate of famoxadone and cymoxanil in cucumber and soil ecosystem. In addition, this study would promote the series of work on the pesticide exposure assessment of these fungicides as well.

Dual-function multinuclear microcapsules with long-term scale inhibition and antibacterial properties

Xiaoyu Chen,Jianwei Zhang,Hengtong Xia,Zhihao Chen,Xiaoshuang Yin,Ying Liu,Wenzhong Yang

Scale and biofouling are two critical problems in industrial circulating cooling water systems, urging the development of innovative solutions to address both issues simultaneously. The direct usage of liquid scale inhibitors and biocides often fails to provide long-term effective control and results in significant wastage of agents. In this work, the melamine formaldehyde resin/clove oil/1‑hydroxy ethylidene-1,1-diphosphonic acid (MCH) microcapsules were successfully synthesized by double emulsion water-in-oil-in-water (W/O/W) in-situ polymerization method. The 1‑hydroxy ethylidene-1, 1-diphosphonic acid (HEDP) acted as a scale inhibitor and clove oil as a natural biocide, which offered a comprehensive solution to both scaling and biofouling issues. The as-prepared MCH microcapsule exhibited a smooth multinuclear spherical structure, a uniform size. UV–vis results confirmed the successful encapsulation of HEDP and clove oil within the MF shell, with encapsulation amounts of approximately 7.47 % and 23.67 %, respectively. Remarkably, the release of the HEDP and clove oil could extend up to 60 h and 160 h, respectively, which was consistent with the First-order model, displaying an excellent sustained-release performance. Static scale inhibition experiments indicated that the microcapsules possess sustained scale inhibition ability. The scale inhibition efficiency of the dynamic release solution of MCH microcapsules remained above 90 % for the initial 14 h, with the highest scale inhibition rate exceeding 98 %. Furthermore, these microcapsules displayed remarkable antibacterial activity against Escherichia coli and Bacillus subtilis, with both antibacterial rates exceeding 99.9 %. Overall, the sustained release of the scale inhibitors and biocides ensures long-term control and reduction in agent wastage, which presents a promising application in addressing the scaling and biofouling issues in industrial circulating water systems.

Bioassay-guided isolation of antifungal amides against Sclerotinia sclerotiorum from the seeds of Clausena lansium

He Yan,Zi Xiong,Na Xie,Shanzhi Liu,Lianlian Zhang,Fang Xu,Wenhui Guo,Juntao Feng

Clausena lansium ( Lour.) Skeels is an attractive shrub or small tree, a member of the Rutaceae family. The seeds, leaves and roots of this plant have been used as herbal medicines for a long time. In this study, seven amides were isolated from the seeds of Clausena lansium . The antifungal assay indicated that lansiumamide B ( 4 ) and lansiumamide C ( 2 ) displayed the highest antifungal activity against Sclerotinia sclerotiorum with EC 50 values of 4.95 and 13.24 μg/mL, which were both lower than that of carbendazim (EC 50  = 0.64 μg/mL). The structure-activity relationships of these amides were investigated using assays to measure their in vitro antifungal activities against Sclerotinia sclerotiorum . The effect of the most potent compound lansiumamide B on morphology and physiology of S. sclerotiorum were also evaluated. Lansiumamide B caused cell rupture and mycelial abnormalities of S. sclerotiorum , and its curative efficacy (75.17%) against S. sclerotiorum infection was better than that of carbendazim (56.57%). This study demonstrated that amides isolated from C. lansium possessed the potential to be exploited as botanical fungicides for commercial application or can be used as templates for designing new fungicides with novel action modes.

Roles of ompA of Citrobacter werkmanii in bacterial growth, biocide resistance, biofilm formation and swimming motility

Zhou Gang,Wang Ying-si,Peng Hong,Li Su-juan,Sun Ting-li,Shen Peng-fei,Xie Xiao-bao,Shi Qing-shan

Abstract The genus Citrobacter is commonly found in environmental and industrial settings, some members of which have been used for bioremediation of heavy metals owing to the absorption ability of their biofilms. Although our previous studies have found that the outer membrane protein A (OmpA) contributes to the process of Citrobacter werkmanii biofilm formation, the underlying mechanisms remain elusive. Therefore, we deleted ompA from the genome of C. werkmanii and investigated its phenotypes in comparison to the wild type strain (WT) and the complementary strain using biochemical and molecular techniques including RNA-Seq. Our results demonstrated that the deletion of ompA led to an increase in biofilm formation on both polystyrene and glass surfaces due to upregulation of some biofilm formation related genes. Meanwhile, swimming ability, which is mediated by activation of flagellar assembly genes, was increased on semi-solid plates in the ∆ompA strain when compared with WT. Additionally, inactivation of ompA also caused increased 1,2-benzisothiazolin-3-one (BIT) resistance, differential responses to Ca 2+ stress, curli protein expression and cellulose production. Finally, ∆ompA caused differential expression of a total of 1470 genes when compared with WT, of which 146 were upregulated and 1324 were downregulated. These genes were classified into different Gene Ontology (GO) and KEGG pathways. In summary, ompA in C. werkmanii contributes to a variety of biological functions and may act as a target site to modulate biofilm formation. Key points • ompA is a negative regulator for biofilm formation by C. werkmanii. • ompA inhibits swimming motility of C. werkmanii. • ompA deletion causes different expression profiles in C. werkmanii.
