Photocatalytic generation of oxidative free radicals is an effective method to eliminate viruses and bacteria in air. Nevertheless, the underlying mechanisms governing the gas-phase sterilization remain unclear. In this study, the self-doped Bi 2.15 WO 6 was synthesized to prepare the Bi 2.15 WO 6 coating. In the presence of Bi 2.15 WO 6 coating, the average logarithmic degradation efficiencies ( LDEs ) are 3.82 and 3.07 for E. coli and S. aureus , respectively under UV illumination for 12.9 s, indicating its desirable sterilization efficiencies. In addition, the sterilization rates of E. coli and S. aureus slightly decrease by 3 % and 6 % after 30-repeated cycles, indicating the high stability of this catalytic coating. The 2,7-dichlorofluorescein (DCF) fluorescence experiments and SEM analysis indicate that reactive oxygen species (ROS) produced by Bi 2.15 WO 6 have effectively destroyed cell structure to achieve the complete inactivation. The N,N -diethyl-p-phenylenediamine sulfate (DPD) and radical trapping experiments further reveal that H 2 O 2 is the primary oxidizing species, and its yields are linearly proportional to the LDE values. The produced H 2 O 2 is further decomposed to •OH under UV irradiation to kill bacteria. Raman analysis confirms the presence of the intermediate species of surface ≡Bi-OO• superoxo and/or ≡Bi-OOH peroxo groups on Bi 2.15 WO 6 , which are the precursors of H 2 O 2 . In addition, the 3-D filter facilitates sterilization by the Bi 2.15 WO 6 in that it prolongs the duration of free radical via physically intercepting bioaerosols, thus apparently improving the degradation rate. This study provides new insights on the sterilization mechanism involved in the gas-phase photocatalytic processes.

As an efficient green water disinfection technology, photothermal synergy sterilization has been paid more attention in recent years with its immeasurable advantages. The development of photocatalysts for efficient sterilization is currently an urgent need. In this manuscript, pure Ti3C2Tx is first studied as a photothermal synergy sterilization material and shows an excellent sterilization effect. The sterilization rate of Escherichia coli (E. coli) reaches 99.9% under visible infrared (vis-IR) light for 40 minutes. The peeling of the Al layer from Ti3AlC2 is the fundamental reason for improving the photothermal conversion ability, and photogenerated free radicals have also proved to be improved. The dilution spreading plate method, enzyme activity test, and leakage of cell contents confirmed the complete death of bacteria. Two aspects, photogenerated free radicals and photothermal synergy, were carried to illuminate the mechanism.

The fabrication of montmorillonite (Mt) decorated with lysozyme-modified silver nanoparticles (Ag/lyz-Mt) was reported. The lysozyme (lyz) was served as both reducing and capping reagent. Coupling the bactericidal activity of the lyz with AgNPs, along with the high porous structure and large specific surface area of the Mt , prevented aggregation of AgNPs and promoted nanomaterial-bacteria interactions, resulting in a greatly enhanced bactericidal capability against both Gram positive and Gram negative bacteria. This paper systematically elucidated the bactericidal mechanisms of Ag/lyz-Mt. Direct contact between the Ag/lyz-Mt surface and the bacterial cell was essential to the disinfection. Physical disruption of bacterial membrane was considered to be one of the bactericidal mechanisms of Ag/lyz-Mt. Results revealed that Ag + was involved in the bactericidal activity of Ag/lyz-Mt via tests conducted using Ag + scavengers. A positive ROS (reactive oxygen species) scavenging test indirectly confirmed the involvement of ROS (O 2 − , H 2 O 2 , and OH) in the bactericidal mechanism. Furthermore, the concentrations of individual ROS were quantified. Results showed that Ag/lyz-Mt nanomaterial could be a promising bactericide for water disinfection.

Titanium dioxide (TiO 2 ) has become a prominent material in photocatalysis research, particularly for antibacterial applications in the post-pandemic era. However, conventional solid TiO 2 nanoparticles tend to agglomerate, reducing the contact area between the catalyst and bacteria, which limits their antibacterial efficiency. To overcome this, an ultra-simple hydrolysis strategy was used to create a highly dispersed TiO 2 “solution” catalyst, rich in hydroxyl groups. The resulting TiO 2 colloid exhibited excellent dispersion in water and showed the Tyndall effect. Experimental results demonstrated that the TiO 2 achieved a 100% sterilization rate within 5 min of light exposure, surpassing commercial P25. This outstanding photocatalytic antibacterial performance is due to its smaller particle size and improved dispersion, which enhance superoxide radical production and provide more active sites. Moreover, an innovative bactericidal mechanism was proposed: when hydroxyl-rich TiO 2 adsorbs onto bacterial surfaces, it dehydrates the bacteria, similar to the effect of a salt solution. This research offers a new, straightforward strategy for developing highly photocatalytically active catalysts that combine both photocatalytic and dehydration-based antibacterial mechanisms.

Multidrug resistant bacterial infection has become one of the most serious threats to human health. Antimicrobial peptides (AMPs) have been identified as potential alternatives to antibiotics owing to their excellent bactericidal activity. However, the complicated bactericidal mechanism of AMPs is still poorly understood. Fluorescence imaging has many advantages in terms of dynamic monitoring, easy operation, and high sensitivity. In this study, we developed an aggregation-induced emission (AIE)-active probe AMP-2HBT by decorating the antimicrobial peptide HHC36 (KRWWKWWRR) with an AIEgen of 2-(2-hydroxyphenyl)benzothiazole (HBT). This AIE-active probe exhibited an excellent light-up fluorescence after binding with bacteria, enabling a real-time monitoring of the binding process. Moreover, a similar time-dependent bactericidal kinetics was observed for the AIE-active probe and HHC36 peptide, which indicated that the bactericidal activity of the peptide was not compromised by decorating with the AIEgen. The bactericidal mechanism of HHC36 peptide was further investigated by super-resolution fluorescence microscopy, transmission electron microscopy (TEM), and scanning electron microscopy (SEM), which suggested that the probe tended to accumulate on the bacterial membrane and efficiently disrupt the membrane structure to kill both Gram-positive and -negative bacteria. This AIE-active probe thus provided a convenient tool to investigate the bactericidal mechanism of AMPs.

Simple SummaryThe intensification of aquaculture practices elevates risks of pathogen transmission, necessitating advanced disinfectants. Polyhexamethylene biguanide (PHMB), a promising disinfectant, exhibits unclear seawater applicability and species-specific mechanisms against marine bacteria. This study demonstrated that PHMB exerts bactericidal effects by disrupting cell membrane permeability, yet its efficacy varies with bacterial characteristics. Notably, the ionic composition of seawater was identified as a potential limiting factor, particularly through calcium-mediated interference in PHMB’s activity against marine pathogens (such asVibrio parahaemolyticusandPhotobacterium damselaesubsp.damselae). These findings provide critical insights into optimizing PHMB-based disinfection strategies in aquaculture systems.Guanidine disinfectants are cationic polymers recognized for their effective sterilization properties and their ability to prevent bacterial resistance. As a result, they are widely utilized in medical, healthcare, household, and animal husbandry settings. However, the bactericidal effects and mechanisms of guanidine in marine aquaculture systems remain unclear due to the polymeric nature of guanidine ions and the complexity of marine environments. The inhibitory effects and bactericidal mechanisms of polyhexamethylene biguanide (PHMB) on key pathogens and probiotics are examined in this study. It was shown that PHMB had inhibitory effects onVibrio parahaemolyticus(VP),Photobacterium damselaesubsp.damselae(PDD),Bacillus subtilis(BS),Escherichia coli(EPEC), andStaphylococcus aureus(SAU), with minimum inhibitory concentrations (MICs) ranging from 3.91 to 125.0 µg/mL, and minimum bactericidal concentrations (MBCs) from 15.63 to 250.0 µg/mL. A stronger bactericidal effect of PHMB on marine bacteria compared to EPEC and SAU was exhibited. It was shown in ion interference experiments that the addition of calcium ions reduced the bactericidal effectiveness of PHMB against VP and PDD by 87.73% and 53.35%, respectively. At a PHMB concentration of 62.50 µg/mL, minor changes in cell surface potential energy (CSPE) were exhibited by Gram-positive bacteria (SAU and BS), while more significant alterations were shown by Gram-negative pathogens. It was revealed by propidium iodide staining and scanning electron microscopy (SEM) analysis that the bacterial cell membrane was directly disrupted by PHMB. DNA and RNA release analysis further revealed that following PHMB treatment, changes in membrane permeability were exhibited by Gram-negative pathogens, with a significant increase in extracellular DNA content as PHMB concentration increased. No such effect was observed in Gram-positive bacteria. Additional evidence was provided by the findings that PHMB effectively inhibits bacterial pathogens in mariculture systems, with a significantly stronger inhibitory effect on Gram-negative pathogens than on Gram-positive bacteria. These results indicated that PHMB could serve as a new antimicrobial agent in mariculture.

The present study evaluated inactivation efficiency of a sonophotocatalytic process using ZnO nanofluids including ultrasonic parameters such as power density, frequency and time. The result showed that inactivation efficiency was increased by 20% when ultrasonic irradiation was combined with photocatalytic process in the presence of natural light. Comparison of inactivation efficiency in photocatalytic, ultrasonic and sonocatalytic processes using Escherichia coli as a model bacteria identified that inactivation efficiencies are shown in the following order: ultrasonic irradiation < sonocatalysis < photocatalysis < sonophotocatalysis. Furthermore, inactivation mechanism of sonophotocatalysis was proposed. Studies of reactive oxygen species (ROS) and zinc ions (Zn 2+ ) release evaluation revealed that ROS play a key role in bacterial inactivation rather than Zn 2+ . Permeability of outer membrane (OM) and inner membrane (IM) of E. coli bacterial cells were studied and exhibited that sonophotocatalysis increased the permeability of OM and IM significantly. The enhanced bacterial inactivation effect in sonophotocatalytic process contributed to acoustic cavitation, sonocatalysis of ZnO and sonoporation phenomenon.

The emergence of drug-resistant bacteria poses a serious threat to public health. The traditional antibiotics have specific intracellular targets and disinfect via chemical ways, which easily lead to the development of drug resistance, therefore, cationic peptides as promising antibiotic agents have attracted extensive attention due to their unique properties. Herein, we report a class of amphiphilic peptide-based pectinate polymers with primary amino groups. The polymers spontaneously self-assembled into the positively charged nanoparticles, which were evaluated and confirmed by scanning electron microscopy (SEM) and dynamic light scattering (DLS). Biological assays revealed that the nanoparticles showed broad-spectrum antibacterial efficacy against both Gram-positive and Gram-negative bacteria, exhibiting a MIC of 16 μg mL−1 against six clinical bacteria, namely, E. faecalis, S. aureus, MRSA, VRE, P. aeruginosa, and K. pneumonia, and three bacterial strains E. coli and E. coli producing NDM-1 and ImiS, and showed a sterilization rate of 95.6% and 94.7% on S. aureus and E. coli, respectively. Importantly, the nanoparticles did not result in drug-resistance for both the normal and drug-resistant bacteria tested after 14 passages and showed low toxicity on the mouse fibroblast cells (L929). The fluorescence staining, electrical conductivity, SEM, and surface plasmon resonance (SPR) characterization suggested that the nanoparticles initially bound to the surface of the bacteria, then pierced into the membranes of the bacteria with their phenyl groups, and finally disrupted the membranes, resulting in ions leaking out and thus exhibiting broad-spectrum antibacterial efficacy. This bactericidal mechanism that the nanoparticles employed does not lead the bacteria susceptible to developing drug resistance. This study provides a promising pathway for the development of the efficient antibacterial materials.

The traditional antibiotics have specific intracellular targets and disinfect in chemical ways, and the drug-resistance induced by the antibiotics has grown into an emerging threat. It is urgent to call for novel strategies and antibacterial materials to control this situation. Herein, we report a class of silver-decorated nanocomposite AgNPs@PCL-b-AMPs as potent nanoantibiotic, constructed by ring-opening polymerization of the monomers ε-caprolactone, Z-Lys-N-carboxyanhydrides (NCAs), and Phe-NCAs, then decorated with AgNPs, and characterized by SEM, TEM, and DLS. The biological assays revealed that the nanocomposite possessed strong antibacterial efficacy against both Gram-positive and Gram-negative bacteria including clinical isolated bacteria MRSA, VRE, P. aeruginosa, and K. pneumonia, exhibiting a MIC value range in 2–8 μg/mL. Importantly, the S. aureus and P. aeruginosa treated with the nanocomposite did not show drug-resistance even after 21 passages. Also, in vivo anti-infective assays showed that the nanocomposite was able to effectively kill bacteria in the infected viscera of mice. The study of the sterilization mechanism showed that the nanocomposite exhibited a multimodal antimicrobial mechanism, including irreversibly damaging the membrane structure, making the leakage of intracellular ions and subsequently inducing generation of the reactive oxygen species (ROS), ultimately sterilizing the bacteria. The nanocomposite exhibits effective broad-spectrum antibacterial properties and shows low toxicity to the mammalian cells/animal. Overall, the AgNPs@PCL-b-AMPs gained in this work show great potential as a highly promising antibacterial material for biomedical applications including drug-resistant bacterial infection.

AimsThis study focuses on the bactericidal mechanism of the new combination of disinfectant glutaraldehyde‐didecyldimethylammonium bromide (GA‐DDAB) against Escherichia coli. Methods and ResultsEscherichia coli were exposed to GA‐DDAB, and assays for cell morphology, K+ and Ca2+ leakage, H+‐ATPase activity and DNA degradation were performed. GA‐DDAB damaged the cell wall and disrupted cell‐membrane integrity. Leakage of K+ and Ca2+ increased, resulting in significantly lower intracellular concentrations within 60 min of treatment. In addition, H+‐ATPase was inactivated and DNA was degraded.ConclusionsLeakage of intracellular components indicated that GA‐DDAB damaged the cell membrane of E. coli. This may have caused the observed disruption in equilibria of metal ions, inactivation of H+‐ATPase, and DNA damage.Significance and Impact of the StudyUsing a low concentration of GA and DDAB, a new combination disinfectant was developed. GA‐DDAB displayed higher antimicrobial activity than treatment with GA or DDAB alone. Therefore, GA‐DDAB may be a more cost‐effective and efficient antimicrobial agent than others in use today. Furthermore, this study provided a paradigm for developing high‐efficiency disinfectant to help address the growing problem of bacterial resistance.

With the gradual development of carbon dots (CDs) in the antimicrobic application, efforts have been still devoted to overcome the dilemma that more effective antibacterial treatment would lead to inevitable higher cytotoxicity. In addition, their bactericidal mechanisms are still in urgent need to be clarified. Therefore, silver (copper)-containing carbon dots have been prepared via one microwave-irradiation strategy using glucose as precursors and metal salts as passivators. It was discovered that the as-prepared metal-containing CDs have the comparable or better bactericidal effect while lower cytotoxicity towards mammalian cells compared with most of corresponding commercial metal-containing antibacterial agents and their free metal ions, because of the good permeability towards bacterial cells originating from the excellent metal dispersion and glucose disguiser in/on the CDs. The minimum bactericidal concentration of silver(copper)-containing carbon dots against 1.5 (0.5) × 10 7 CFU/mL Escherichia coli was less than 25 (75) μg/mL. Different bactericidal mechanisms between the two metal-containing CDs and the free copper/silver ions were revealed in details. It was found that the metal ions could directly inhibit the function of ribosomes in bacterial cells, while the metal-containing CDs showed negligible influence towards ribosome directly, instead, they inhibited bacterial metabolism through more complex pathways such as metabolic disorders, transporter inactivation or inhibition of the nucleic acid synthesis. Since the mammalians cells can switch to other pathways more flexibly than bacterial cells under the disturbance of the same metabolism pathway inhibition, the metal-containing CDs provides excellent biocompatibility.

The objective of this study was to investigate the antibacterial activity and potential mechanism of alkyl gallates against Escherichia coli and Staphylococcus aureus . Results show that the length of the alkyl chain plays a pivotal role in eliciting the activity and octyl gallate (OG) exerted excellent bactericidal activity through a multiple bactericidal mechanism. OG functions against both bacteria through damaging bacterial cell wall integrity, permeating into cells and then interacting with DNA, as well as disturbing the activity of the respiratory electron transport chain to induce a high-level toxic ROS (hydroxyl radicals) generation and up-regulation of the ROS genes. Also, electrospun nanofibers with OG have unique superiorities for maintaining the freshness of the icefish (4 °C). This research not only provides a more in-depth understanding of the interaction between OG and microorganisms but also highlights the great promise of using OG as a safe multi-functionalized food additive for food preservations.
