Osteoarthritis (OA) is a disabling disease and seriously affects the quality of life of patients. is A potential medicine for the treatment of OA is Eucommia ulmoides leaves (EULs). However, its active compounds and therapeutic mechanisms are unclear. Therefore, it is necessary to develop a method using LC–MS and network pharmacology for the detection and identification of compounds and the mechanisms of action of EULs. The compounds were detected and identified based on ultra-high-performance liquid chromatography coupled with Quadrupole Exactive-Orbitrap MS (UHPLC-Q Exactive-Orbitrap MS) and followed by the network pharmacology analysis. Seventy-three compounds, including 15 flavonoids, 8 iridoids, 10 lignans, 24 phenolic acids, and 16 additional compounds, were identified by UHPLC-Q Exactive-Orbitrap MS. The network of the pharmacological analysis revealed that 29 active compounds regulated 17 main pathways through 38 target genes, including NF-kappa B signaling pathway, PI3K-Akt signaling pathway, AMPK signaling pathway, etc. In conclusion, EULs were effective in the treatment of OA by regulating the abovementioned key pathways. This study showed that LC–MS/MS followed by network pharmacology analysis is useful to elucidate the complex mechanisms of action of Chinese herb. Practical applications This study describes a rapid method of detecting and identifying the constituents and systematic mechanism of Eucommia ulmoides based on LC–MS and network pharmacology. Our results show that 73 compounds of E. ulmoides leaves were identified and predicted that E. ulmoides leaves were effective in the treatment of OA by regulating key pathways, including NF-kappa B signaling pathway, PI3K-Akt signaling pathway, and AMPK signaling pathway by network pharmacology, which lays the foundation for subsequent research.

In order to achieve the desirable extraction yield of thermosensitive chlorogenic acid, a novel air–steam explosion with low temperature was developed to pretreat Eucommia ulmoides leaves in this study. The correlation of temperature and different partial pressure of air was investigated, and the pretreatment performance was evaluated by the extraction efficiency and antioxidant capacity of chlorogenic acid. Results showed that the pretreatment temperature obviously decreased with the increase of air partial pressure. Mixed with 0.4 MPa of air partial pressure, the temperature of gas mixture was 26.69% lower than that without air (0.8 MPa of steam). After pretreatment for 120 s, the yield, extraction rate, and OH − and O 2 − scavenging ratios of chlorogenic acid from air–steam exploded sample were 2.83%, 0.182 min −1 , 31.10% and 95.85%, respectively, which were 62.0%, 423.0%, 94.6% and 9.5% higher than those from raw sample. Additionally, the equilibrium time of chlorogenic acid extraction decreased from 120 min to 15 min. The reason for these results were that the fibrous networks and the cell walls of the leaves were destroyed after air–steam explosion pretreatment, which improved the solute–solvent accessibility and the internal mass transfer in extraction process. Therefore, air–steam explosion was an efficient pretreatment to enhance the extraction performance of chlorogenic acid from E. ulmoides leaves.

This study presented the development of a nanoscale, tunable surfactant-free microemulsion (SFME) formulated with natural deep eutectic solvents (NaDES) for the efficient extraction of phytochemicals. The NaDES-SFME, composed of octanol, thymol, ethanol and water, was characterized using a ternary phase diagram, conductivity measurements and UV absorption spectroscopy. The optimal NaDES-SFME composition (15 % v/v NADES, 45 % v/v ethanol and 40 % v/v water) demonstrated superior extraction performance compared to conventional solvents, achieving extraction yields of seven phytochemicals from Eucommia ulmoides leaves that were 1.15–3.55 times higher. FT-IR, dynamic light scattering (DLS) and fluorescence microscopy revealed a reverse W/O structure at 46.7 nm, mimicking cell membranes to penetrate plant matrices and dissolve compounds. Ultrasound-assisted extraction, optimized via response surface methodology, achieved a maximum total yield of 9.763 mg/g under the conditions of 50 °C, 41 min and a solid–liquid ratio of 31 mg/mL. The Pareto plot indicated that the extraction time was the main factor affecting the extraction efficiency and the secondary kinetic model could better fit the dissolution pattern of the phytochemicals. Density functional theory (DFT) analysis confirmed hydrogen bonding as the key mechanism for the high extraction yield. Finally, the combination of ethanol addition and rotary evaporation allowed for a high recovery of the phytochemicals, with a recovery rate of more than 96 %. This green, cost-effective and high-yield microemulsion holds promise for industrial-scale phytochemical extraction and utilization.

In this investigation, to maximize the desired immunoenhancement effects of PsEUL and stimulate an efficient humoral and cellular immune response against an antigen, PsEUL and the model antigen ovalbumin (OVA) were coupled using the N-(3-dimethylaminopropyl)-N′-ethylcarbodiimide hydrochloride (EDC) reaction to yield a novel delivery system (PsEUL-OVA). The physicochemical characteristics and immune regulation effects of this new system were investigated. We found the yield of this EDC method to be 46.25%. In vitro, PsEUL-OVA (200 μg mL−1) could enhance macrophage proliferation and increase their phagocytic efficiency. In vivo, PsEUL-OVA could significantly increase the levels of OVA-specific antibody (IgG, IgG1, IgG2a, and IgG2b) titers and cytokine (IL-2, IL-4, IL-6, IFN-γ) levels. Additionally, it could activate T lymphocytes and facilitate the maturation of dendritic cells (DCs). These findings collectively suggested that PsEUL-OVA induced humoral and cellular immune responses by promoting the phagocytic activity of macrophages and DCs. Taken together, these results revealed that PsEUL-OVA had the potential to improve immune responses and provide a promising theoretical basis for the design of a novel delivery system. Keywords:polysaccharides fromEucommia ulmoides;adjuvant;delivery system;macrophage;dendritic cell

ObjectivesA biolabel research based on multi-omics, informatics, molecular docking, and experimental verification was used to investigate the cardioprotective effect and pharmaceutically active compounds of Eucommiae Folium (EF).Key findingsBased on the biolabel research pattern, metabonomics, proteomics, and bioinformatics indicated that EF has a therapeutic potential for a variety of heart diseases, especially cardiomyopathy, and the most critical mechanism involved is the diabetic cardiomyopathy pathway. Bioinformatics, cheminformatics, and molecular docking showed that 24 EF compounds may play a therapeutic role in diabetic cardiomyopathy via this pathway. Among which, four compounds (kaempferol, esculetin, (+)-catechin, and astragalin) showed appropriate pharmacokinetic parameters and formed stable binding with biolabels in the pathway. In diabetic cardiomyopathy db/db mice, histopathological analysis, mitochondrial swelling and membrane potential assay, ELISA, and biochemical analysis demonstrated that EF and four active compounds had obvious hypoglycemic effects and attenuated myocardial damage and related pathological processes, such as mitochondrial dysfunction, calcium dyshomeostasis, insulin resistance, and oxidative stress.ConclusionsThis study provides new evidence and insights into the effect, mechanism, and material basis of EF in treating diabetic cardiomyopathy.

Epilepsy is a chronic brain disease and often occurs suddenly for no reason. Eucommiae folium (EF), an edible herb, can be used in the treatment of various kinds of brain diseases in clinic. From the perspective of safety and efficacy, EF is especially suitable for the treatment of chronic brain diseases. With the help of biolabels, this study was aimed to explore the value and feasibility of EF in the treatment of epilepsy. Proteomics and metabolomics were used to explore the biolabels of EF intervention in brain tissues. Bioinformatics was then applied to topologically analyze its neuroprotective effects and mechanisms and material basis based on biolabels, which were validated in an animal model. The biolabel-led research revealed that EF may exert the therapeutic potential to treat brain diseases through the interaction between multiple compounds and multiple targets, among which its therapeutic potential for epilepsy is particularly prominent. In the pentylenetetrazole-induction model, EF and four active compounds (oleamide, catechol, chlorogenic acid, and kaempferol) protected epileptic hippocampal neurons (Nissl and FJB staining) against mitochondrial dysfunction (MYH6, MYL3, and MYBPC3, etc.) and calcium overload (TNNI3, TNNC1, and TNNT2, etc.) through the hypertrophic cardiomyopathy pathway. This study provides new evidence and insights for the neuroprotective effects of EF, in which four active compounds may be potential drug candidates for the treatment of epilepsy.
