A novel quinoline/benzothiazole pH probe (E)-2-(2-(quinolin-4-yl)vinyl)benzothiazole (BTVQ) was facilely synthesized. BTVQ exhibits remarkable emission response to pH and can image pH changes within the extreme acidity range of 3.0–3.8 with a pKa of 3.52. Moreover, BTVQ displays a notably large Stokes shift of 110 nm, which can reduce the excitation interference. BTVQ was successfully applied in HeLa cells and E. coli cells, suggesting that BTVQ can be employed as one of the few lysosome specific probes for imaging acidic intracellular pH changes, especially under extreme-acidity conditions in bacteria.

Carbon dots are popular luminescent materials because of their excellent fluorescence properties, but the low quantum yield limits their application. Heteroatom doping is a more convenient and popular approach to increase the quantum yield of carbon dots. Here, novel N,S,P heteroatom co-doped carbon dots (N,S,P-CDs) were synthesized by a simple one-step hydrothermal method using m-phenylenediamine, L-cysteine and phosphoric acid as raw materials. The as-prepared N,S,P-CDs showed excellent photoluminescence properties with a fluorescence quantum yield of up to 41%, which greatly encourages their application in fluorescence sensing. The N,S,P-CDs exhibited good fluorescence stability under salt solution, xenon lamp irradiation and ultraviolet lamp irradiation except for a high sensitivity to extreme acidity. The fluorescence intensity of the N,S,P-CDs can be decreased by as much as 85% when the pH of the solution changes from 2.50 to 4.75, that is, a small fluctuation in pH can cause an intense response of the fluorescence of the N,S,P-CDs. Therefore, an excellent fluorescence sensing platform for accurately monitoring the pH of extreme acidity has been constructed. In addition, the N,S,P-CDs can be applied for quantitative detection of folic acid based on the strong quenching effect of folic acid on the fluorescence of the N,S,P-CDs. Good linearity was obtained in the concentration range of 4.85–82.45 μM, with a detection limit of 0.148 μM. The constructed sensing platform was used for the determination of folic acid in actual samples of orange juice, oatmeal and tablets with satisfactory results.

Adhesive hydrogels have become an ideal alternative to sutures and staples for sealing tissue defects and anchoring devices. However, most existing adhesive hydrogels are designed for two-sided adhesion and suffer from irreversible swelling in the in vivo environment. In this work, we report a nanoscale phase separation induced self-reinforced PABA-EGCG-ALC Janus hydrogel as a sutureless therapeutic solution for gastrointestinal (GI) defect sealing and gastric perforation repair. The Janus hydrogel is prepared by a one-step method and exhibits excellent mechanical compliance, one-sided adhesion behavior, and ROS scavenging ability. Notably, the nanoscale phase separation structures formed by polymer chain entanglements effectively inhibit the swelling behavior of the hydrogel (60.2 % at equilibrium) and induce self-reinforced mechanical strength and interfacial robustness under extreme acidity. After soaking the hydrogel in simulated gastric fluid (SGF, pH 2.0) for 72 h, its tensile stress, elongation at break, toughness, Young's modulus, and burst pressure are increased by 2.9, 1.7, 6.4, 2.4, and 2.1 folds, respectively. Subsequent in vivo animal experiments demonstrate that the PABA-EGCG-ALC Janus hydrogel provides a convenient, fluid-tight, gastric fluid-resistant, robust and suture-free sealing for GI defects and effectively preventing postoperative adhesions while addressing the key limitations of commercially available tissue adhesives and Janus hydrogels.
