β-fructofuranosidase (β-FFase) belongs to the glycosyl-hydrolase family 32 (GH32), which can catalyze both the release of β-fructose from β-d-fructofuranoside substrates to hydrolyze sucrose and the synthesis of short-chain fructooligosaccharide (FOS).BmSuc1has been cloned and identified from the silkwormBombyx morias a first animal type of β-FFase encoding gene. It was hypothesized that BmSUC1 plays an important role in the silkworm-mulberry adaptation system. However, there is little information about the enzymatic core sites of BmSUC1. In this study, we mutated three amino acid residues (D63, D181, and E234) that represent important conserved motifs for β-FFase activity in GH32 to alanine respectively by using site-directed mutagenesis. Recombinant proteins of three mutants and wild type BmSUC1 were obtained by using a Bac-to-Bac/BmNPV expression system and BmN cells. Enzymatic activity, kinetic properties, and substrate specificity of the four proteins were analyzed. High Performance Liquid Chromatography (HPLC) was used to compare the hydrolyzing and transfructosylating activities between D181A and wtBmSUC1. Our results revealed that the D63A and E234A mutations lost activity, suggesting that D63 and E234 are key amino acid residues for BmSUC1 to function as an enzyme. The D181A mutation significantly enhanced both hydrolyzing and transfructosylating activities of BmSUC1, indicating that D181 may not be directly involved in catalyzation. The results provide insight into the chemical catalyzation mechanism of BmSUC1 inB. mori. Up-regulated transfructosylating activity of BmSUC1 could provide new ideas for usingB. moriβ-FFase to produce functional FOS.Keywords:Bombyx mori;β-fructofuranosidase;site-directed mutagenesis;hydrolysis;transfructosylation

Selectively converting fructose into furfural by H-beta catalyst is a very interesting catalytic process. However, it is a challenging task to clearly elucidate the roles of framework Al for selective scission of C C and C O bond. Herein, selective elimination of framework Al atoms is realized. Several detailed investigations showed that the framework Al species located in different sites play important role in furfural formation. The framework Al atoms of pristine H-beta mainly occupied at T1, T2, T5, T6 and T7 sites. These Al species at pore mouth can be removed by citric acid, while the treatment of nitric acid mainly eliminated the framework Al atoms from T2 and T7 sites. As for oxalic acid, the Al atoms occupied at T2, T5, T6 and T7 was primarily leached. Upon citric acid and nitric acid treatment afforded the similar yield of furfural, indicating that the formation of furfural could not be governed by these Al species at pore mouth, T2 and T7 sites. However, in the case of oxalic acid treatment, complete removal of the Al species on T2, T5, T6 and T7 sites decreased the yield of furfural. Thus, it is reasonably inferred that these Al atoms populated at T1 sites and intersecting channels is more encouraging for the splitting of C C bond, while the Al species occupied at T5, T6 sites and the larger straight channels is probably more favorable to scission of C O bond.

Honey crystallization is a natural phenomenon commonly found in honey products. To better understand the characteristics of honey crystallization, the concentrations of the four major compositions (namely glucose, fructose, sucrose, and maltose) were determined in the liquid, crystal, and mixed forms of honey. Our study revealed the contents of glucose and fructose in three forms varied significantly (p < 0.05). Molecular dynamics simulation was performed to investigate honey crystallization process. All the simulation systems had a tendency to form a cubic crystal. When the ratio of glucose/fructose is 2.5:1, which is the same as the crystal found in honey, a shorter time and lower root-mean-square deviation were found compared to the ratios at 2:1, 1:1, and pure glucose. It infers that glucose/fructose at 2.5:1 is the most stable honey crystallization, relatively. The morphology of crystals found in honey, which was observed under an environment scanning electron microscope, looked like sticky balls with indistinct edges, different from that plates or granule of glucose crystal with flattened edges. These results indicated that the relative content of glucose and fructose had a significant effect on the crystallization of honey. The glucose/fructose at 2.5:1 probably is the critical ratio of honey crystallization, at which honey can form stable crystalline deposit speedily.

Fructose is generated from glucose isomerization in water over attapulgite without any addition of metal compounds. The formation of fructose was related closely to the calcination processing of attapulgite. The highest fructose yield of 20.6% was obtained with the selectivity of 77.9% over attapulgite calcined at 500 °C. Moreover, attapulgite was characterized by BET, XRD, IR and CO 2 -TPD techniques. The results indicated that proper distribution and amount of basic sites were crucial for the selectivity of fructose as well as the structural properties of attapulgite.

We have developed a nonenzymatic electrochemical sensor based on a carbon paste electrode modified with electrospun LaMnO 3 fibers for fructose determination. LaMnO 3 fibers, a kind of perovskite-type oxide, were prepared by electrospinning and subsequent calcination process. The morphology and structure of LaMnO 3 fibers were characterized by scanning electron microscope, X-ray diffraction and Fourier Transform Infrared spectrum. The electrochemical response of the proposed sensor was evaluated by cyclic voltammetry and amperometry. Under optimal conditions, the linear response for fructose determination was obtained in the range of 0.4–100 μM, with a low detection limit of 63 nM ( S / N = 3).

The mechanism for the formation of furfural by dehydration of d -fructose via selective carbon-carbon (C C) bond cleavage was investigated over beta zeolite (Hβ). Several different pore size zeolites were employed to determine the shape selectivity of fructose isomers. Zeolitic pore sizes that smaller than the kinetic diameter of cyclic fructose also produced yield furfural (∼22.5%), while the high yield (66.0%) of hydroxymethylfurfural (HMF) can be achieved over large pore zeolite (USY zeolite), which could accommodate the cyclic fructose. These results indicated that the furfural formation likely began with acyclic fructose. In situ 13 C NMR and GC MS studies, using labeled 13 C-1-fructose as substrate, suggested that the conversion of fructose to furfural involved with splitting of the C5-C6 bond. Furthermore, the C1 compound from the cleavage of C C bond was identified as formaldehyde, inferring that the selective scission of C C bond was ascribe to the retro-aldol reaction. Interestingly, in situ NMR studies implied that the acyclic fructose mainly derived from pyranose forms. In addition, compared with glucose, fructose directly converted to furfural at the higher yield and reaction rate.

Alkaline earth metal salts are environmentally friendly and economical. They have attracted much attention in the process of biomass conversion, especially for isomerization and dehydration. However, the lack of understanding of the alkaline earth metal-mediated catalytic mechanism prevents the achievement of high HMF yields from glucose or fructose. Herein, we performed experimental and theoretical studies on CaCl2-catalyzed glucose-to-fructose isomerization and fructose-to-HMF dehydration. In CaCl2 solution, the highest fructose yield (33.6%) from glucose can be achieved at 80 °C. Meanwhile, the presence of CaCl2 in fructose-to-HMF dehydration notably improved fructose conversion. A “one-pot” isomerization–dehydration of glucose to prepare HMF was designed and 100% conversion and 52.1% HMF yield were obtained. The CaCl2 solution can be reused for five runs while maintaining catalytic activity without regeneration. Through 1H NMR, 13C NMR, DOSY-NMR and DFT calculations, we concluded that the coordination between the C1/C2 sites of the β-glucopyranose tautomer and CaCl2 promoted the 1,2-intramolecular hydride shift of glucose, thus rapidly converting glucose into fructose. Similarly, Ca2+ can complex with the C2/C6-OH of β-D-fructofuranose and stabilize this tautomer, which easily forms HMF. This is the main reason that CaCl2 can promote the preparation of HMF from fructose.

L-Proline derived ionic liquids (ILs) used as both solvent and catalyst were efficient for transformation of fructose and sucrose to 5-hydroxymethylfurfural (HMF) in the presence of water. Response surface methodology (RSM) was employed to optimize fructose dehydration process, and a maximum HMF yield of 73.6% could be obtained at 90°C after 50 min. The recycling of the IL exhibited an almost constant activity during five successive trials, and a possible reaction mechanism for the dehydration of fructose to HMF was proposed.

Driven by sustainable development goals, the wood-based panel industry increasingly favors renewable biomass-based and formaldehyde-free adhesives. Saccharides, owing to their renewable environmental friendliness and high reactivity, have garnered attention for green adhesive development. However, existing saccharide-based adhesives face challenges due to demanding curing conditions (hot-pressing temperature ≥160 °C), limiting their industrial application. In this study, a fructose and toluene-4-sulfonic acid adhesive (FT adhesive) was prepared by a simple one-pot process. FT adhesive can be hot-pressed at 110 °C to prepare plywood in line with the China National Standard GB/T 9846–2015 (wet strength ≥0.7 MPa). Compared with other saccharide-based adhesives, FT adhesives show superior curing efficiency while maintaining high bonding strength. During the synthesis phase, fructose reacts with toluene-4-sulfonic acid at lower temperatures to produce various active compounds, including 5-HMF and its derivatives, formic acid, acetic acid, and furan compounds. These compounds undergo condensation reactions during high-temperature curing, forming polyfurans, humin polymers, and other insoluble polymers predominantly bonded by C–O–C, C–O, and C═O linkages. Additionally, toluene-4-sulfonic acid acts as a catalyst and an enhancer in both synthesis and curing phases, facilitating the conversion and cross-linking of 5-HMF and its derivatives. This process results in a robust and dense network structure, significantly enhancing the adhesive performance of FT adhesives at lower curing temperatures (110–130 °C). This research offers insights into the development of efficient and environmentally friendly saccharide-adhesives for wood-based panels.

Efficient and selective isomerization of glucose into fructose is the most crucial process of biorefining. Despite its rapid isomerization rate under low-temperature conditions, alkali-catalyzed glucose isomerization is less attractive due to the occurrence of extremely complex side reactions. Herein, we explored an approach for promoting the efficiency of alkali-catalyzed glucose isomerization into fructose by adding alkaline earth metal salts. Kinetic analysis showed that the addition of alkaline earth metal salts increased the Ea of fructose degradation from 35.99 to 74.04 kJ mol−1 and decreased the Ea of glucose conversion from 40.35 to 24.62 kJ mol−1. Adding alkali earth metal salts simultaneously stabilized fructose and promoted glucose isomerization. 70.3% fructose yield with 99% fructose selectivity can be achieved in 0.011 g mL−1 Ca(OH)2/0.5 g mL−1 CaCl2 solution at 50 °C for 25 min. From the DFT calculation and DOSY NMR characterization, we inferred that the interaction of CaCl2 with glucose can reduce the energy barrier of glucose ring opening, and the complexation between CaCl2 and fructose maintains the stability of fructose. Meanwhile, 1H NMR titration revealed that the addition of CaCl2 dramatically promoted the conversion of α-glucopyranose to β-glucopyranose. This alkali-salt-catalyzed glucose isomerization is simple and efficient, providing a new strategy for preparing platform chemicals from glucose.

Sulfonated carbon dots (SCDs) were synthesized from plant leaves via continuously hydrothermal treatment by hydrogen peroxide and sulfuric acid, used as catalyst for converting fructose to 5-hydroxymethylfurfural (HMF). Owing to nanosize effect and moderate acidic intensity, SCDs could thoroughly distribute in the solvent with an improved interfacial compatibility and selectively convert fructose to HMF. Under the optimal condition, the yield of HMF was 92.6% along with a fructose conversion of 100%, benefiting from a low activation energy of 52.9 kJ/mol when dimethylsulfoxide was used as solvent. The SCDs catalyst can be recovered, after six recycles, the fructose conversion and HMF yield were remained 66.1% and 56.2% under condition with incompletely conversion of fructose, respectively. This work provides a sustainable route to prepare carbon dots with a superior catalytic performance for converting biomass to important biobased platform chemicals.

The selective synthesis of furfural or hydroxymethylfurfural (HMF) from fructose on single catalyst (Beta zeolite, Hβ) is challenging task. However, in this study, selectivity of Hβ zeolite was discovered easily to tune by solvent effects. Strong solvent effects on the selectivity of fructose conversion were observed in different manners depending on the solvent used. It was shown that the coordinated state of framework aluminum, induced by solvent effects, has a major impact on the selectivity. The solvents with amide group were discovered to induce the reversible tetrahedral-octahedral framework aluminum transformation, but the configuration of aluminum was no influenced by other solvents such as γ-butyrolactone (GBL). Compared with other solvents, the GBL was not able to enhance turnover frequency (TOF) value of reaction but also suppress the degradation of furfural. Interestingly, a considerable yield of furfural (50.25%) was obtained combined with tetrahedral and octahedral framework aluminum active sites in GBL, while high selectivity of HMF (83.3%) was achieved in presence of single tetrahedral framework aluminum over Hβ in NMP.
