Heterologous overexpression of resveratrol synthase (PcPKS5) gene enhances antifungal and mite aversion by resveratrol accumulation

Luo Zaiqi,Guo Huili,Yang Yadong,Yang Mingfen,Ma Lanqin,Wang Younian

Huping jujube is widely cultivated in northern China. PcPKS5, a resveratrol synthase (STS) gene, contains all functionally divergent plant specific type III PKSs and is involved in resveratrol synthesis. This study aimed to assess the effects of heterologous overexpression of the resveratrol synthase (PcPKS5) gene on Huping jujube (Ziziphus jujuba Mill.) plant resistance and nutritional quality. Stems with leaves and shoot tips of Huping jujube (Ziziphus jujuba Mill) were infected with agrobacterium expressing PcPKS5 and GUS, and three positive plants were identified. Resveratrol (0.45 μg/g fresh weight) was detected by HPLC in line 2 transgenic plants. Interestingly, 100 μl extract of 1 g/ml transgenic plant crude extract showed an inhibitory effect equivalent to 1.0 μg of pure resveratrol. In addition, transgenic plants displayed increased anti-mite resistance compared with the wild-type counterparts. Importantly, higher PcPKS5 expression levels were found in young tissues compared with the stem and mature leaves. This study also provides a stable genetic transformation mechanism for developing new plant germplasms in Huping jujube.

Fructus Jujubae cooperated with water-expelling members in Shizao decoction alleviated intestinal injury and malignant ascites by modulating gut microbiota and metabolic homeostasis

Shi-Kang Zhou,Jin-Di Xu,Xiao-Qin Gao,Ren-Jie Zhang,Fang-Fang Cheng,Wei-Feng Yao,Yi Zhang,Ting Geng,Li Zhang

Background Shizao decoction (SZD) consisted of Euphorbia kansui (EK), Euphorbia pekinensis (EP), Daphne genkwa (DG), and Fructus Jujubae (FJ) is a classic Chinese herbal medicine formula for treating malignant ascites, which is closely related to the modulation of gut microbiota by our previous study. For water-expelling members (WEM) including EK, EP, and DG may have side effects on the intestine, FJ is employed for detoxification and effectivity enhancement of WEM. However, the underlying mechanism for the compatibility of WEM and FJ is still unknown. Purpose To investigate the effect of the compatibility of WEM with FJ in SZD on malignant ascites and elucidate the potential mechanism from the perspective of the modulation of gut microbiota and related metabolic function. Methods Qualitative and quantitative evaluation of main components was conducted for comprehensive characterization of SZD and WEM. The effect of WEM and SZD was compared on malignant ascites effusion (MAE) rats. The intestinal injury was evaluated by HE staining and oxidative damage. Ascites weight, urine amount, fecal water content, the expression of aquaporins, and cytokines in ascites (IL-6, VEGF, and TNF-α) were measured to estimate the water-expelling activity. The intestinal flora was detected by 16S rDNA sequencing and the content of fecal short-chain fatty acids (SCFAs) was analyzed using gas chromatography-mass spectrometry. Pseudo-germ-free (PGF) and fecal bacteria transplantation animal experiments were subsequently employed to validate this finding. The fecal metabolomics and correlation analysis were finally conducted to explore the related metabolic changes. Results 51 and 33 components were identified in SZD and WEM, respectively. Compared to WEM alone, the compatibility with FJ remarkably reduced intestinal oxidative damage in MAE rats. Ascites was also relieved by downregulating the expression of AQP3 in the colon and decreasing the levels of IL-6, TNF-α and VEGF in ascites. The diversity of gut microbiota was reversed with an increase in Lactobacillus and Clostridia _ UCG-014 while a decrease in Colidextribacter . Under the PGF condition, compatibility of WEM with FJ failed to reduce intestinal injury and alleviate MA significantly, but this effect was further enhanced after FMT. 23 potential fecal metabolites were finally identified. Correlation analysis further showed that Lactobacillus and Clostridia_UCG-014 were positively correlated with SCFAs and l-tryptophan. Colidextribacter was negatively correlated with thymidine but positively correlated with ursodeoxycholic acid and deoxycholic acid. Conclusion FJ cooperated with WEM reduced intestinal injury and alleviated malignant ascites by modulating gut microbiota, short-chain fatty and tryptophan metabolism. These findings provide a scientific basis for the clinical application of FJ from SZD and the safe usage of SZD.

Rapid and sensitive quantitation of DDMP (2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one) in baked red jujubes by HS-SPME-GC-MS/MS

Yening Qiao,Jinfeng Bi,Qinqin Chen,Xinye Wu,Xinwen Jin,Min Gou,Xinrui Yang,Giorgia Purcaro

Baked red jujube is a common food ingredient used to enrich the nutritional value and enhance the aroma. DDMP (2,3-dihydro-3,5-dihydroxy-6-methyl-4H-pyran-4-one) with intense bitterness and potential toxicity may be present in several baked red jujube-based products but there are no well-defined methods to measure it quickly, sensitively, and accurately. Headspace solid-phase microextraction with gas chromatography-tandem mass spectrometry (HS-SPME-GC-MS/MS) was applied for the analysis of DDMP. The standard curve was linear between the concentrations of 0.01 and 100 μg/mL, with a limit of detection at 0.1 ng/g that below the sensory threshold of DDMP (2.06 μg/g). Five cultivars of baked red jujubes from China were evaluated with the method for DDMP content (4.55–46.50 μg/g). The results indicated that jujube cultivars with a higher molar ratio of proline to glucose, lower moisture content, and stronger ability to resist water stress should be avoided from baking to modulate the formation of DDMP.

Influence of calcium propionate on the control of post-harvest rots of jujube and the biocontrol activity of an antagonistic yeast

D. Q. Guo,W. H. Wang,Y. F. Pu,Q. Xu,L. X. Zhu,B. Q. Yang

The effect of calcium propionate (CaP) on post-harvest pathogens of winter jujube (Zizyphus jujuba Mill.), and on improving the efficacy of the biocontrol yeast, Metschnikowia pulcherrima, were evaluated. In vitro, CaP at ≥2% (w/v) had significant inhibitory effects on spore germination and germ-tube growth in Penicillium citrinum and Alternaria alternata. However, the application of 2% (w/v) CaP did not affect population growth of M. pulcherrima in nutrient yeast dextrose in vivo. With or without CaP, yeast cell numbers increased after 72 h by approx. 11.1-fold and 13.4-fold, respectively. The concentration of the antagonistic yeast suspension influenced the incidence of both pathogens and lesion development on jujube fruit, and the effect increased with an increase in yeast cell concentration. A combination of the antagonistic yeast plus 2% (w/v) CaP resulted in a lower incidence of disease and smaller lesion diameters compared to treatments with the antagonistic yeast alone. In this case, the incidence of disease was reduced by 16.8–27.3% for P. citrinum and by 25.2–31.4% for A. alternata. Lesion diameters on these fruit were 2.7–6.2 cm and 1.8–2.4 cm for P. citrinum and A. alternata, respectively. Calcium propionate had a consistent ability to enhance the biocontrol activity of M. pulcherrima against blue mould (P. citrinum) and Alternaria rot on jujube fruit. Thus, the application of 2% (w/v) CaP could be a useful approach to improve the efficacy of the antagonistic yeasts used for post-harvest disease control in jujube.

Immunomodulatory effect of a formula developed from American ginseng and Chinese jujube extracts in mice

Zhuo-ping Yu,Dong-dong Xu,Lai-feng Lu,Xiao-dong Zheng,Wei Chen

Background: American ginseng ( Panax quinquefolius L.) and Chinese jujube ( Zizyphus jujuba Mill.) are commonly used in traditional Chinese medicine to enhance immune function. Objective: The present study aimed to develop one Chinese prescription, Shenzao Cha (SZC), consisting of American ginseng and Chinese jujube, and systematically investigate its immunomodulation in healthy ICR mice. Methods: Normal ICR mice received intragastric administration of SZC (1.3, 2.6, and 5.2 g raw material/kg body weight) once daily for four weeks, while a control group received the same amount of sterile water. Results: SZC significantly increased the spleen and thymus indices and T-lymphocyte proliferation, while the T-lymphocyte proliferation in the 5.2 g/kg group was 1.4-fold higher than that in the control. Further, 1.3 g/kg SZC could markedly improve hemolytic activity by 25.2%, and 2.6 g/kg SZC increased the NK cell activity by 78.6% relative to the control. In addition, the activities of antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase), that participated in modulating oxidative stress, were significantly increased in the liver, spleen, thymus, and serum, while the contents of malondialdehyde were dramatically decreased. Conclusions: SZC exhibited potent immunomodulatory effects on innate and adaptive immunity in healthy ICR mice, as well as potential antioxidant activity for prevention of oxidative stress, which was suggested to partly contribute to the immune enhancement.

Pharmacokinetic Comparisons of Multiple Triterpenic Acids from Jujubae Fructus Extract Following Oral Delivery in Normal and Acute Liver Injury Rats

Yao Li,Sheng Guo,Quanjin Ren,Dandan Wei,Ming Zhao,Shulan Su,Zhishu Tang,Jin-Ao Duan

Jujubae Fructus, the dried fruit ofZiziphus jujuba, has been used as Chinese medicine and food for centuries. Triterpenic acids have been found to be the major bioactive constituents inJujubae Fructusresponsible for their hepatoprotective activity in previous phytochemical and biological studies, while few pharmacokinetic studies have been conducted. To reveal the kinetics of the triterpenic acids under the pathological liver injury state, an established ultra-performance liquid chromatography coupled with a mass spectrometry method was applied for the simultaneous quantitation of seven triterpenic acids (ceanothic acid, epiceanothic acid, pomonic acid, alphitolic acid, maslinic acid, betulinic acid, and betulonic acid) in plasma samples of normal and acute liver injury rats induced by CCl4. The results showed that there were significant differences (p< 0.05) in the pharmacokinetic parameters of seven triterpenic acids between model and normal groups. The AUC0–tand AUC0–∞of epiceanothic acid (5227 ± 334 μg⋅h/L vs. 1478 ± 255 μg⋅h/L and 6127 ± 423 μg⋅h/L vs. 1482 ± 255 μg⋅h/L, respectively) and pomonic acid (4654 ± 349 μg⋅h/L vs. 1834 ± 225 μg⋅h/L and 4776 ± 322 μg⋅h/L vs. 1859 ± 230 μg⋅h/L, respectively) in model rats were significantly higher than those in normal rats, and the CLz/F of them were significantly decreased (0.28 ± 0.02 L/h/kg vs. 1.36 ± 0.18 L/h/kg and 19.96 ± 1.30 L/h/kg vs. 53.15 ± 5.60 L/h/kg, respectively). In contrast, the above parameters for alphitolic acid, betulinic acid and betulonic acid exhibited the quite different trend. This pharmacokinetic research might provide useful information for the clinical usage of triterpenic acids fromJujubae Fructus. Keywords:Ziziphus jujuba;triterpenic acids;pharmacokinetic study;acute liver injury

A novel acidic polysaccharide from blackened jujube: Structural features and antitumor activity in vitro

Zhang Guifeng,Liu Chuang,Zhang Rentang

Liver cancer is one of the most common cancers, with increasing trends in incidence and mortality. A novel acidic polysaccharide (BJP-2) obtained from blackened jujube was extracted by hot water followed by chromatographic purification employing DEAE-cellulose 52 and Sephadex G-100 column. And then BJP-2 was identified by SEC-MALLS-RI, GC-MS, methylation and NMR for the following characteristics: molecular weight of 6.42 × 104 Da, monosaccharide composition of glucuronic acid (GalA), arabinose (Ara), galactose (Gal), rhamnose (Rha), xylose (Xyl), glucuronic acid (GlcA), glucose (Glc), fucose (Fuc) and mannose (Man) with the percentage of 39.78, 31.93, 16.86, 6.43, 1.86, 1.28, 1.02, 0.61, and 0.23%, as well as the main chain of → 5)-α-L-Araf (1 → 4)-β-D-Gal(1 → , T-α-L-Araf (1 → 4)-β-D-Gal(1 → , and → 4)-α-L-6MeGalAp(1 → . The effect of BJP-2 on the apoptosis of HepG2 cells and its anti-tumor mechanism were further explored. The analysis by MTT and flow cytometry showed that BJP-2 suppressed cell proliferation by inducing apoptosis in a concentration-dependent manner. Cell scratching and Transwell revealed that BJP-2 was able to block the invasion and metastasis of tumor cells. Western blot results demonstrated that BJP-2 exhibited antitumor activity through a mitochondria-dependent pathway, as evidenced by overexpression of Bax, Cleaved Caspase-3/Caspase-3 and Cleaved Caspase-9/Caspase-9 and downregulation of Bcl-2. Therefore, BJP-2 has broad research prospects as a tumor preventive or therapeutic agent.

Modulatory effects of black jujube melanoidins on gut microbiota and metabolic pathways in high-fat diet-induced obesity

Xi Che,Yifei Zhao,Yuxiao Wang,Xin Sun,Hongxun Tao,Rentang Zhang

Introduction: Obesity, a growing public health issue, is closely associated with excessive energy intake and gut microbiota imbalances. Despite the increasing attention given to black jujube as a blackened fermented food in recent years, the role of its melanoidins (MLD) in obesity-related biological mechanisms has yet to be thoroughly investigated. Methods: This study investigated the effects of black jujube MLD on obesity induced by a high-fat diet in C57BL/6J mice. We hypothesize that MLD exerts an anti-obesity effect, potentially mediated by alterations in gut microbiota composition and the modulation of metabolic responses. Results: The results demonstrate that MLD administration mitigated HFD-induced weight gain, improved insulin resistance, and enhanced glucose metabolism by reducing blood glucose and insulin levels (p < 0.05). MLD also protected the liver, as evidenced by a 16.4% decrease in alanine aminotransferase (ALT) levels and a 29.6% decrease in aspartate aminotransferase (AST) levels (p < 0.05). Furthermore, MLD treatment restored the compromised intestinal microbiota to a healthier state at the genus level by lowering the Firmicutes/Bacteroidetes ratio (38.9%), suppressing the abundance of pathogenic bacteria, such as Erysipelatoclostridium and Bacteroides descendants, and promoting the growth of beneficial bacteria, including Bifidobacterium and norank_f_Muribaculaceae. Metabolomic profiling revealed that MLD can alleviate metabolic disorders by downregulating arginine metabolism and sphingolipid metabolism. Its metabolites are significantly correlated with several bacterial genera, including Bacteroides, Roseburia, Erysipelatoclostridium, Bacteroides, norank_f_Muribaculaceae, and Lachnospiraceae NK4A136 group.Discussion: Based on these findings, MLD may mitigate obesity and other associated metabolic disorders by modulating the gut microbiome–metabolism axis.

Hydrophilic interaction ultra-high performance liquid chromatography coupled with triple quadrupole mass spectrometry for determination of nucleotides, nucleosides and nucleobases in Ziziphus plants

Sheng Guo,Jin-ao Duan,Dawei Qian,Hanqing Wang,Yuping Tang,Yefei Qian,Dawei Wu,Shulan Su,Erxin Shang

In this study, a rapid and sensitive analytical method was developed for the determination of 20 nucleobases, nucleosides and nucleotides in Ziziphus plants at trace levels by using hydrophilic interaction ultra-high performance liquid chromatography coupled with triple-quadrupole tandem mass spectrometry (HILIC–UHPLC-TQ-MS/MS) in multiple-reaction monitoring (MRM) mode. Under the optimized chromatographic conditions, good separation for 20 target compounds were obtained on a UHPLC Amide column with sub-2   μm particles within 10   min. The overall LODs and LOQs were between 0.11–3.12   ng   mL −1 and 0.29–12.48   ng   mL −1 for the 20 analytes, respectively. It is the first report about simultaneous analysis of nucleobases, nucleosides and nucleotides in medicinal plants using HILIC–UHPLC-TQ-MS/MS method, which affords good linearity, precision, repeatability and accuracy. The developed method was successfully applied to Ziziphus plant ( Z. jujuba , Z. jujuba var. spinosa and Z. mauritiana ) samples. The analysis showed that the fruits and leaves of Ziziphus plants are rich in nucleosides and nucleobases as well as nucleotides, and could be selected as the healthy food resources. Our results in present study suggest that HILIC–UHPLC-TQ-MS/MS method could be employed as a useful tool for quality assessment of the samples from the Ziziphus plants as well as other medicinal plants or food samples using nucleotides, nucleosides and nucleobases as markers.

Hepatoprotective effects of polysaccharides extracted from Zizyphus jujube cv. Huanghetanzao

Guangpu Liu,Xinquan Liu,Yongchun Zhang,Fang Zhang,Ting Wei,Min Yang,Kaiming Wang,Yongjie Wang,Na Liu,Huiling Cheng,Zhongxi Zhao

Jujube polysaccharides have been proved to have various bioactivities. This study was designed to evaluate the chemical composition and hepatoprotective function of the polysaccharides extracted from Zizyphus jujube cv. Huanghetanzao (HJP). The composition of HJP was determined as heteropolysaccharides with galactose and arabinose being the main components. The pretreatment of mice with HJP significantly ( p   <   0.01) reduced the activities of serum hepatic AST, ALT, and LDH induced by CCl 4 or acetaminophen (APAP) while the commercial liver-injury treatment drug silybin did not show any prevention effects. Mechanistic study results indicate that the administration of the CCl 4 - or APAP-injured mice with HJP enhanced SOD and GSH-Px and decreased MDA, indicating that anti-oxidation and detoxification could be the pathways for the liver protection observed. In addition, the liver prevention and treatment effects of HJP on the liver damage induced by CCl 4 or APAP obtained from the liver enzyme analyses were confirmed by the hepatic histopathology studies in mice. Therefore, HJP could be used as a prevention and treatment agent for liver injury induced by liver toxic chemicals and drugs.

Mechanisms Underlying the Action of Ziziphi Spinosae Semen in the Treatment of Insomnia: A Study Involving Network Pharmacology and Experimental Validation

Bian Zhenhua,Zhang Wenming,Tang Jingyue,Fei Qianqian,Hu Minmin,Chen Xiaowei,Su Lianlin,Fei Chenghao,Ji De,Mao Chunqin,Tong Huangjin,Yuan Xiaohang,Lu Tulin

Purpose: This study aimed to investigate the potential mechanisms and related bioactive components of ZSS for the treatment of insomnia.Method: The insomnia model of rat induced by PCPA was established. After oral administration of ZSS extract, the general morphological observation, pentobarbital sodium-induced sleep test and histopathological evaluation were carried out. Network pharmacology, assisted by UHPLC-Q-Exactive-MS/MS analysis, was developed to identify the targets of ZSS in the treatment of insomnia, as well as the corresponding signaling pathways. In addition, we validated the identified targets and pathways by RT-qPCR and immunohistochemical analysis. Results: The pentobarbital sodium-induced sleep test, determination of 5-HT and GABA levles in hypothalamic tissues and HE staining showed that ZSS extract was an effective treatment for insomnia. Network pharmacology analysis identified a total of 19 candidate bioactive ingredients in ZSS extract, along with 433 potentially related targets. Next, we performed protein-protein interaction (PPI), MCODE clustering analysis, GO functional enrichment analysis, KEGG pathway enrichment analysis, and ingredient-target-pathway (I-T-P) sub-networks analysis. These methods allowed us to investigate the synergistic therapeutic effects of crucial pathways, including the serotonergic and GABAergic synapse pathways. Our analyses revealed that palmitic acid, coclaurine, jujuboside A, N-nornuciferine, caaverine, magnoflorine, jujuboside B, and betulinic acid, all played key roles in the regulation of these crucial pathways. Finally, we used the PCPA-induced insomnia in rats to validate the data generated by network pharmacology; these in vivo experiments clearly showed that pathways associated with the serotonergic and GABAergic system were activated in the rats model. Furthermore, ZSS treatment significantly suppressed high levels of HTR1A, GABRA1, and GABRG2 expression in the hypothalamus and reduced the expression levels of HTR2A.Conclusion: Based on the combination of comprehensive network pharmacology and in vivo experiments, we successfully identified the potential pharmacological mechanisms underlying the action of ZSS in the treatment of insomnia. The results provide a theoretical basis for further development and utilization of ZSS, and also provide support for the development of innovative drugs for the treatment of insomnia.

Fungicidal efficiency of DBD cold plasma against Aspergillus niger on dried jujube

Qiaoyun Wei,Yuan Yuan,Jianhao Zhang,Jin Wang

This study investigated the fungicidal efficiency and mechanism of action of dielectric barrier discharge cold atmosphere plasma (DBD-CAP) in inactivating Aspergillus niger ( A. niger ) spores. The disinfection efficacy and quality of dried jujube used as the processing application object were also studied. The results indicated that the Weibull +  Tail model performed better for spore inactivation curves at different voltages among various treatment times, and the spore cells were reduced by 4.05 log (cfu/mL) in spores suspension at 70 kV after 15 min of treatment. This disinfection impact was further supported by scanning electron microscope (SEM) and transmission electron microscopy (TEM) images, which showed that the integrity of the cell membrane was damaged, and the intracellular content leaked out after DBD-CAP treatment. Elevated levels of reactive oxygen species (ROS) during the treatment increased the relative conductivity of cells, and leakage of nucleic acids and proteins further supported the disinfection impact. Additionally, the growth and toxicity of surviving A. niger spores after treatment were also greatly reduced. When DBD-CAP was applied to disinfecting dried jujube, the spore number exhibited a 2.67 log cfu/g reduction after treatment without significant damage observed onto the quality ( P  >  0.05 ).
