GhENODL6 Isoforms from the Phytocyanin Gene Family Regulated Verticillium Wilt Resistance in Cotton

Man Zhang,Xingfen Wang,Jun Yang,Zhicheng Wang,Bin Chen,Xinyu Zhang,Dongmei Zhang,Zhengwen Sun,Jinhua Wu,Huifeng Ke,Liqiang Wu,Guiyin Zhang,Yan Zhang,Zhiying Ma

Verticillium wilt (VW), a fungal disease caused byVerticillium dahliae, currently devastates cotton fiber yield and quality seriously, yet few resistance germplasm resources have been discovered inGossypium hirsutum. The cotton variety Nongda601 with suitable VW resistance and high yield was developed in our lab, which supplied elite resources for discovering resistant genes. Early nodulin-like protein (ENODL) is mainly related to nodule formation, and its role in regulating defense response has been seldom studied. Here, 41 conserved ENODLs inG. hirsutumwere identified and characterized, which could divide into four subgroups. We found thatGhENODL6was upregulated underV. dahliaestress and hormonal signal and displayed higher transcript levels in resistant cottons than the susceptible. TheGhENODL6was proved to positively regulate VW resistance via overexpression and gene silencing experiments. Overexpression ofGhENODL6significantly enhanced the expressions of salicylic acid (SA) hormone-related transcription factors and pathogenicity-related (PR) protein genes, as well as hydrogen peroxide (H2O2) and SA contents, resulting in improved VW resistance in transgenicArabidopsis. Correspondingly, in theGhENODL6silenced cotton, the expression levels of both phenylalanine ammonia lyase (PAL) and 4-coumarate-CoA ligase (4CL) genes significantly decreased, leading to the reduced SA content mediating by the phenylalanine ammonia lyase pathway. Taken together,GhENODL6played a crucial role in VW resistance by inducing SA signaling pathway and regulating the production of reactive oxygen species (ROS). These findings broaden our understanding of the biological roles ofGhENODLand the molecular mechanisms underlying cotton disease resistance. Keywords:Gossypium hirsutum;GhENODL6;Verticillium wilt resistance;SA;ROS

Degradation of α-Subunits, Doa1 and Doa4, are Critical for Growth, Development, Programmed Cell Death Events, Stress Responses, and Pathogenicity in the Watermelon Fusarium Wilt Fungus Fusarium oxysporum f. sp. niveum

Muhammad Noman,Azizullah,Temoor Ahmed,Yizhou Gao,Hui Wang,Xiaohui Xiong,Jiajing Wang,Jiajun Lou,Dayong Li,Fengming Song

The ubiquitin–proteasome system (UPS) regulates protein quality or control and plays essential roles in several biological and biochemical processes in fungi. Here, we present the characterization of two UPS components, FonDoa1 and FonDoa4, in watermelon Fusarium wilt fungus, Fusarium oxysporum f. sp. niveum (Fon), and their biological functions. FonDoa1 localizes in both the nucleus and cytoplasm, while FonDoa4 is predominantly present in the cytoplasm. Both genes show higher expression in germinating macroconidia at 12 h. Deletion of FonDoa1 or FonDoa4 affects vegetative growth, conidiation, conidial germination/morphology, apoptosis, and responses to environmental stressors. FonDoa1, but not FonDoa4, positively regulates autophagy. The targeted disruption mutants exhibit significantly attenuated pathogenicity on watermelon due to defects in the infection process and invasive fungal growth. Further results indicate that the WD40, PFU, and PUL domains are essential for the function of FonDoa1 in Fon pathogenicity and environmental stress responses. These findings demonstrate the previously uncharacterized biological functions of FonDoa1 and FonDoa4 in phytopathogenic fungi, providing potential targets for developing strategies to control watermelon Fusarium wilt.

Benzoxazinoids secreted by wheat root weaken the pathogenicity of Fusarium oxysporum f. sp. fabae by inhibiting linoleic acid and nucleotide metabolisms

Ma Guanglei,Hu Bijie,Yang Siyin,Cen Zixuan,Zheng Yiran,Dong Yan

Key message The regulatory action of BXs secreted by wheat on the pathogenicity of FOF causing Fusarium wilt in faba bean were analyzed. DIMBOA and MBOA weakened the pathogenicity of FOF. Abstract A large number of pathogenic bacteria in continuous cropping soil infect faba bean plants, leading to the occurrence of wilt disease, which restricts their production. Faba bean–wheat intercropping is often used to alleviate this disease. This study investigates the effect of benzoxazinoids (BXs) secreted by wheat root on the pathogenicity of Fusarium oxysporum f. sp. Fabae (FOF) and underlying molecular mechanisms. The effects of DIMBOA(2,4-dihydroxy-7-methoxy-1,4-benzoxazine-4-one) and MBOA(6-methoxybenzoxazolin-2-one) on the activity of cell-wall-degrading enzymes in FOF(cellulase, pectinase, amylase, and protease), FOF Toxin (fusaric acid, FA) content were investigated through indoor culture experiments. The effect of BXs on the metabolic level of FOF was analyzed by metabonomics to explore the ecological function of benzoxazines intercropping control of Fusarium wilt in faba bean. The results show that the Exogenous addition of DIMBOA and MBOA decreased the activity of plant-cell-wall-degrading enzymes and fusaric acid content and significantly weakened the pathogenicity of FOF. DIMBOA and MBOA significantly inhibited the pathogenicity of FOF, and metabolome analysis showed that DIMBOA and MBOA reduced the pathogenicity of FOF by down-regulating related pathways such as nucleotide metabolism and linoleic acid metabolism, thus effectively controlling the occurrence of Fusarium wilt in faba bean.

Evaluation of the Inhibitory Efficacy of Eugenol against the Pathogen of Fusarium Wilt in Ginger Seedlings

Xian Zhou,Hui-Hui Ma,Shi-Jie Xiong,Ling-Ling Zhang,Xue-Dong Zhu,Yong-Xing Zhu,Li-Rong Zhou

Fusariumwilt, mainly caused byFusarium oxysporum, affects ginger yield and quality worldwide. To explore a safe and sustainable method of preventing and controllingFusariumwilt, the inhibitory effects of eugenol onF. oxysporum FOX-1were analysed in this study. The results showed that eugenol inhibited the reproductive and vegetative growth ofF. oxysporum FOX-1in vitro. Microscopic observations revealed that eugenol disrupted the hyphal morphology ofF. oxysporum. In addition, eugenol destroyed the cell membrane integrity of the pathogenic fungi, resulting in the leakage ofF. oxysporum FOX-1intercellular contents, including electrolytes, soluble proteins, nucleic acids, and malonydialdehyde. Results of an in vivo inoculation test showed that eugenol exerted a strong inhibitory effect onFusariumwilt in ginger seedlings. In summary, eugenol had an inhibitory effect on the growth ofF. oxysporum FOX-1and controlledFusariumwilt in ginger seedlings. These findings provide a foundation for future development of botanical antifungal agents to manageFusariumwilt. Keywords:Fusariumwilt;eugenol;essential oil;antifungal activity;Zingiber officinale

Bacterial wilt pathogen induced spatial shifts of root-associated microbiome and metabolome of potatoes

Xianjun Lai,Zhouhua He,Shuyan Wang,Feng Zhang,Haiyan Wang,Xiyao Wang,Shifeng Liu,Lang Yan

Introduction: Plant root-associated microbiomes play an important role in plant health, yet their responses to bacterial wilt remain unclear poorly understood. Methods: This study investigated spatial variations in microbiome and metabolome composition across three root-associated niches—root-surrounding soil, rhizosphere, and endosphere—of healthy and Ralstonia solanacearum-infected potato plants. A total of 36 samples were analyzed, with microbial diversity assessed by full-length 16S rRNA and ITS sequencing, and metabolic profiles characterized using LC-QTOF-MS.Results: Alpha diversity analysis revealed that bacterial diversity in healthy plants was consistently higher than in diseased plants, progressively increasing from the root-surrounding soil to the rhizosphere, and most notably in the endosphere, where the Shannon index declined from 5.3 (healthy) to 1.2 (diseased). In contrast, fungal diversity was lower in diseased plants in the root-surrounding soil and rhizosphere, but significantly elevated in the endosphere, suggesting niche-specific microbial responses to pathogen stress. Beta diversity confirmed significant microbiome restructuring under pathogen stress (R² > 0.5, p = 0.001). Taxonomic analysis showed over 98% dominance of Proteobacteria in the diseased endosphere, where Burkholderia, Pseudomonas, and Massilia enriched in healthy plants were significantly reduced. R. solanacearum infection promotes the enrichment of Fusarium species in both the rhizosphere and endosphere. Metabolomic analysis revealed extensive pathogen-induced metabolic reprogramming, with 299 upregulated and 483 downregulated metabolites in the diseased endosphere, including antimicrobial metabolites such as verruculogen and aurachin A. Network analysis identified XTP as a central metabolite regulating microbial interactions, whereas antimicrobial metabolites exhibited targeted pathogen suppression. O2PLS analysis revealed that pathogen-induced antimicrobial metabolites (e.g., Gentamicin X2, Glutathionylspermine) were associated with Clostridia and Ketobacter in diseased plants, while nucleotide-related compounds (e.g., XTP) correlated with Rhodomicrobium and others, indicating infection-driven microbial adaptation and metabolic restructuring.Discussion: These findings provide insights into pathogen-driven disruptions in root microbiomes and suggest potential microbiome engineering strategies for bacterial wilt management.

Effective assessment of mixed strains against Fusarium oxysporum f. sp. lycopersici causing tomato wilt

Zhe Han,Shengyi Wang,Ao Gao,Meiqi Dong,Yufeng Xiao,Hongyu Pan,Lining Zheng,Hao Zhang

Tomato wilt disease, caused by Fusarium oxysporum f. sp. lycopersici , is a major soil-borne disease affecting tomato cultivation. In this study, two strains exhibiting significant control effects on F. oxysporum f. sp. lycopersici and tomato growth-promoting effects were isolated from the rhizosphere. Through physiological and biochemical identification, as well as 16S rRNA identification, these strains were identified as Enterobacter quasihormaechei L33 and Klebsiella oxytoca TL2, respectively. And the two strains of bacteria were confirmed to have no antagonistic effect through antagonistic compatibility tests. Notably, when these strains were mixed in a ratio of 5:1, they demonstrated remarkable antifungal activity. The mixed strains induced mycelium swelling, necrosis, and rupture in F. oxysporum f. sp. lycopersici , resulting in a 56.65 % inhibition of mycelial fresh weight. Additionally, they inhibited spore production with an inhibition rate of 787.5 %, and the inhibition rate of the lipopeptide produced reached 93.80 % at 50 mg/mL. Furthermore, in potting experiments, the mixed strains reduced the disease incidence by 36.25 % after 8 weeks. Additionally, compared to the control group, the mixed bacterial strain group enhanced fresh weight, plant height, chlorophyll a and b levels, and root activities by 96.31 %, 18.51 %, 184.48 %, 53.03 %, and 44.31 %, respectively. In addition, mixed strains can inhibit the cell wall degrading enzymes produced by Fusarium oxysporum f sp., induce plant defense enzymes, including increased activity of peroxidase, catalase, and superoxide dismutase, and reduce the decrease in malondialdehyde levels. The mixed strains could colonize tomato plant roots. Overall, these findings suggest a potential biocontrol agent deriving from the mixed strains against tomato wilt disease.

Evaluating the Potential Value of Natural Product Cuminic Acid against Plant Pathogenic Fungi in Cucumber

Yong Wang,Jie Zhang,Yang Sun,Juntao Feng,Xing Zhang

Fusarium wilt and anthracnose are two major diseases which limit the yield and quality of cucumber worldwide. Cuminic acid was extracted from the seed ofCuminum cyminumL. The mean EC50values of cuminic acid for inhibiting mycelial growth and zoospore germination of fiveFusarium oxysporumf. sp.cucumerinumstrains were 25.66 ± 3.02 μg/mL and 15.99 ± 2.19 μg/mL, and of fourColletotrichum lagenarium(Pass.) Ellis and Halsted strains were 29.53 ± 3.18 μg/mL and 18.41 ± 2.78 μg/mL, respectively. In greenhouse experiments, cuminic acid at 2000 μg/mL exhibited 70.77% protective and 62.63% curative efficacies againstF. oxysporum, and 65.43% protective and 55.46% curative efficacies againstC. lagenarium. Moreover, the translocation behavior of cuminic acid, determined by high performance liquid chromatography (HPLC), showed that it could be readily absorbed and transported upward and downward in cucumber. Importantly, superoxide dismutase (SOD) and pyphenol oxidase (PPO) activities of cucumber leaves treated with cuminic acid increased significantly. All results indicated that cuminic acid showed antifungal activity, and could be used as a botanical fungicide in disease management. This study encourages further investigation on the mechanism of action of cuminic acid and the development of alternative antifungal drugs. Keywords:botanical fungicide;cuminic acid;cucumber;F. oxysporum;C. lagenarium;control efficacy

Coating Seeds with Paenibacillus polymyxa ZF129 Microcapsule Suspension Enhanced Control Effect on Fusarium Root Rot and Promoted Seedling Growth in Cucumber

Jiayi Ma,Jialin Liu,Yanxia Shi,Xuewen Xie,Ali Chai,Sheng Xiang,Xianhua Sun,Lei Li,Baoju Li,Tengfei Fan

In this study, a Paenibacillus polymyxa ZF129 microcapsule suspension (CS-ZF129) was prepared. P. polymyxa ZF129 was microencapsulated by gum arabic via the spray-drying method. Coating seeds with the CS-ZF129 not only increased the control efficacy against cucumber root rot to 46.30 ± 0.02%, but also promoted the growth of the cucumber roots and other aspects, effectively improving the colonization ability of polymyxa ZF129 in the cucumber root system, which was analyzed to clarify the biological control mechanism of the cucumber Fusarium root rot disease through the enzyme activity. This study provides an efficient means based on seed coating for the biological control of cucumber Fusarium root rot. Fusarium root rot, a destructive soil-borne fungal disease, necessitates eco-friendly biocontrol strategies. This study developed a microbial seed-coating approach using the antagonistic strain Paenibacillus polymyxa ZF129, formulated into a microencapsulated powder (10 8 CFU/g) and a suspension (CS-ZF129). CS-ZF129 application enhanced cucumber resistance, achieving 46.30 ± 0.02% disease suppression while promoting root growth. The maximum increase in the fresh weight of the root in the promotion of rectangular growth was 47.16%. The colonization dynamics of ZF129 in the rhizosphere were systematically tracked, revealing its antagonistic correlation with Fusarium proliferation. An enzymatic activity analysis further uncovered the underlying regulatory mechanisms, demonstrating induced defense responses through pathogenesis-related protein activation. These findings highlight ZF129’s dual functionality as a biocontrol agent and a plant growth promoter, offering a sustainable strategy against soil-borne pathogens.

Fluorine-containing amphiphilic quaternary ammonium salts for the suppression of Banana fusarium wilt

Yaling Lin,Deqiang Zhang,Shijie Shan,Wei Zhang,Rui Li,Anqiang Zhang

As the pathogen of banana Fusarium wilt, Fusarium oxysporum f . sp. cubense race 4 (Foc4) has the strongest toxicity and pathogenicity due to its long-term viability in soil, but there is still no effective way to control it at present. Based on the activity parameters of polymeric quaternary ammonium salt in our previous work, this paper hopes to enhance the amphiphilicity of polymeric quaternary ammonium salt by introducing a fluoroalkyl group to improve the antifungal property of polymeric quaternary ammonium salt and achieve stable adsorption and long-term inhibition of saprophytic Foc4 conidia in soil. Here, we prepared a series of fluorine-containing ammonium salts with random and block structures by atom transfer radical polymerization (ATRP), and we systematically studied the effects of the introduction of fluoroalkyl groups, the proportion of hydrophobic segments and polymer structure on micelle properties, antifungal properties, adsorption, migration and long-term antifungal properties against Foc4 in soil. Finally, the preferred polymeric quaternary ammonium salts were selected for pot experiments in the prevention and control of banana Fusarium wilt caused by Foc4, and positive experimental results were achieved. In conclusion, we made an exploratory attempt at the practical application of polymeric amphiphilic quaternary ammonium salt in plant disease control.

Effect of organic fertilizers prepared from organic waste materials on the production of antibacterial volatile organic compounds by two biocontrol Bacillus amyloliquefaciens strains

Waseem Raza,Zhong Wei,Ning Ling,Qiwei Huang,Qirong Shen

Three organic fertilizers made of different animal and plant waste materials (BOFs) were evaluated for their effects on the production of antibacterial volatile organic compounds (VOCs) by two Bacillus amyloliquefaciens strains SQR-9 and T-5 against the tomato wilt pathogen Ralstonia solanacearum (RS). Both strains could produce VOCs that inhibited the growth and virulence traits of RS; however, in the presence of BOFs, the production of antibacterial VOCs was significantly increased. The maximum inhibition of growth and virulence traits of RS by VOCs of T-5 and SQR-9 was determined at 1.5% BOF2 and 2% BOF3, respectively. In case of strain T-5, 2-nonanone, nonanal, xylene, benzothiazole, and butylated hydroxy toluene and in case of strain SQR-9, 2-nonanone, nonanal, xylene and 2-undecanone were the main antibacterial VOCs whose production was increased in the presence of BOFs. The results of this study reveal another significance of using organic fertilizers to improve the antagonistic activity of biocontrol agents against phytopathogens.

Phenazine-1-carboxylic Acid Has a Broad-Spectrum Antifungal Effect by Targeting Isocitrate Lyase

Jing Huang,Siyi Chen,Weipeng He,Yinan Xiao,Nana Wang,Lili Huang

Phenazine-1-carboxylic acid (PCA), as a broad-spectrum antifungal agent, showed significant resistance against the Apple Valsa canker caused by Valsa mali (V. mali). Therefore, it is of great importance to reveal the functions and mechanisms of PCA in plant pathogens, which would provide potential targets for crop disease control. In this study, we identified the major contribution of PCA in the resistance of Pseudomonas chlororaphis subsp. aureofaciens W9-1 to V. mali by gene knockout and high-performance liquid chromatography. Subsequently, we demonstrated that PCA could target isocitrate lyase (ICL) to exert a broad-spectrum antifungal effect by molecular docking, microscale thermophoresis, and isothermal titration calorimetry. Furthermore, lipidomics analysis shows that PCA alters the lipid balance at the cell membrane of V. mali. In summary, this study demonstrates that PCA has a broad-spectrum antifungal effect by targeting the ICL, which may be a potential target for the development and utilization of fungicides.

Antipathogenic Activities of Volatile Organic Compounds Produced by Bacillus velezensis LT1 against Sclerotium rolfsii LC1, the Pathogen of Southern Blight in Coptis chinensis

Tao Tang,Fanfan Wang,Houyun Huang,Nengneng Xie,Jie Guo,Xiaoliang Guo,Yuanyuan Duan,Xiaoyue Wang,Qingfang Wang,Jingmao You

This study explores the antipathogenic properties of volatile organic compounds (VOCs) produced by Bacillus velezensis LT1, isolated from the rhizosphere soil of Coptis chinensis. The impact of these VOCs on the mycelial growth of Sclerotium rolfsii LC1, the causative agent of southern blight in C. chinensis, was evaluated using a double Petri-dish assay. The biocontrol efficacy of these VOCs was further assessed through leaf inoculation and pot experiments. Antifungal VOCs were collected using headspace solid-phase microextraction (SPME), and their components were identified via gas chromatography–mass spectrometry (GC–MS). The results revealed that the VOCs significantly inhibited the mycelial growth and sclerotia germination of S. rolfsii LC1 and disrupted the morphological integrity of fungal mycelia. Under the influence of these VOCs, genes associated with chitin synthesis were upregulated, while those related to cell wall degrading enzymes were downregulated. Notably, 2-dodecanone and 2-undecanone exhibited inhibition rates of 81.67% and 80.08%, respectively. This research provides a novel approach for the prevention and management of southern blight in C. chinensis, highlighting the potential of microbial VOCs in biocontrol strategies.
