Naringenin Attenuates Isoprenaline-Induced Cardiac Hypertrophy by Suppressing Oxidative Stress through the AMPK/NOX2/MAPK Signaling Pathway

Yu Li,Bo He,Chao Zhang,Yanji He,Tianyang Xia,Chunyu Zeng

Cardiac hypertrophy is accompanied by increased myocardial oxidative stress, and whether naringenin, a natural antioxidant, is effective in the therapy of cardiac hypertrophy remains unknown. In the present study, different dosage regimens (25, 50, and 100 mg/kg/d for three weeks) of naringenin (NAR) were orally gavaged in an isoprenaline (ISO) (7.5mg/kg)-induced cardiac hypertrophic C57BL/6J mouse model. The administration of ISO led to significant cardiac hypertrophy, which was alleviated by pretreatment with naringenin in both in vivo and in vitro experiments. Naringenin inhibited ISO-induced oxidative stress, as demonstrated by the increased SOD activity, decreased MDA level and NOX2 expression, and inhibited MAPK signaling. Meanwhile, after the pretreatment with compound C (a selective AMPK inhibitor), the anti-hypertrophic and anti-oxidative stress effects of naringenin were blocked, suggesting the protective effect of naringenin on cardiac hypertrophy. Our present study indicated that naringenin attenuated ISO-induced cardiac hypertrophy by regulating the AMPK/NOX2/MAPK signaling pathway. Keywords:naringenin;cardiac hypertrophy;oxidative stress;AMPK;NOX2

Simultaneous enantioseparation of citrus flavonoids in chiral microemulsion electrokinetic chromatography using deep eutectic solvents

Huang-Fei Jin,Qian-Xue Shen,Xin-Ran Ma,Jun Cao

A novel, deep eutectic solvent (DES)-assisted sodium cholate (SC)-chiral microemulsion electrokinetic chromatography method is presented for separation of five flavonoid glycoside enantiomers, namely hesperidin, naringin, narirutin, eriocitrin, and neoeriocitrin. Herein, we develop a novel, green DES, whose addition considerably enhances the separation performance through multiple synergistic effects. A series of factors notably affecting chiral separation are systematically optimized: type and concentration of cyclodextrin, DES, and SC, oil phase type, and pH. The microemulsion comprises 0.5 % ( v /v) n-octanol, 0.6 % (v/v) DES prepared using betaine and citric acid, 20 mM 2-HP-β-CD, 100 mM scdium cholate hydrate, and 15 mM borax buffer under the optimized conditions. Enantioseparations of the five enantiomers are achieved within 7 min, with resolutions ranging from 1.29 to 2.88. The method developed provides an innovative and effective way for chiral separation of enantiomers in complex food matrices.

Volumetric Properties, Viscosity, and Refractive Indices of Different Naringenin Solutions at Several Temperatures

Yingying Shan,Behnaz Asadzadeh,Weidong Yan

Thermophysical properties including densities and viscosities of naringenin in aqueous solutions of ethanol/1-propanol with alcohol concentrations of (24.00 to 40.00) mol·kg–1 have been measured as a function of concentration of naringenin at temperatures from (293.15 to 323.15) K and atmospheric pressure. Also, refractive indices for studied systems have been determined at 298.15 K. The measured data were used to calculate the apparent molar volume (Vϕ), standard partial molar volume (Vϕ0), partial molar volume (V̅), viscosity B-coefficients, and molar refractions (Rm). The viscosity and refractive index data have been analyzed by Jones–Dole and Lorentz–Lorenz equations, respectively. The trends of variation of experimental and calculated parameters have been discussed according to the interactions between solvents and solute. The obtained results imply that naringenin acts as a structure maker in the studied system.

The increase in autophagy induced by naringin alleviates Schwann cell apoptosis mediated by the AMPK/mTOR signalling pathway

Lv Jianwei,Shan Duo,Jia Haobo,Wang Zengliang

Background Peripheral nerve injury (PNI) is currently an intractable disease for clinical therapeutics. Schwann cells (SCs) play pivotal roles in peripheral nerve regeneration. Naringin, a natural flavonoid, has potent beneficial effects on peripheral neuropathy. Researchers have rarely reported whether naringin attenuates SC apoptosis and the relevant mechanisms. Therefore, this study aimed to evaluate the antiapoptotic effects of naringin on SCs and probe the potential molecular mechanisms involved. Methods An in vitro SC apoptotic model was established by culturing RSC96 cells in foetal bovine serum (FBS)-free medium, namely, serum starvation. SC apoptosis was assessed by TUNEL staining and flow cytometry. The activities of the mitochondrial apoptotic pathway and autophagy in SCs were detected by western blotting and immunofluorescence staining. The autophagy inhibitor 3-methyladenine (3-MA) and the AMPK inhibitor Compound C were used to investigate whether autophagy mediated by AMPK/mTOR signalling is involved in the regulation of SC apoptosis by naringin. Results The results showed that starvation induced SC apoptosis and blocked autophagic flux. Naringin ameliorated SC apoptosis under starvation conditions by increasing the autophagy activity mediated by the AMPK/mTOR cascade, as Compound C inhibited its autophagy-activating effect, and 3-MA reversed its antiapoptotic efficacy. Conclusions Naringin mitigated starvation-induced SC apoptosis by increasing autophagy, which was attributed to the regulation of the AMPK/mTOR pathway. This study provides evidence that naringin could be an effective alternative drug for promoting peripheral nerve regeneration.

Autophagy in Cadmium-Induced Hepatic Injury of Rats: Protective Role of Naringenin

Mengmeng Gao,Hao Ling,Chengxiang Guo,Yinan Hu,Jing Zhu,Jicang Wang

Cadmium, a bluish-white metallic, possesses a prolonged half-life in organisms, causing damage to multiple tissues and organs. Naringenin, a natural flavonoid antioxidant, mitigates cadmium-induced damage in organisms. However, the mechanism by which naringenin attenuates Cd-associated hepatocellular damage has not been fully elucidated. The present investigation consequently aimed to elucidate the mechanistic involvement of autophagy in cadmium hepatotoxicity using the rat model, with parallel assessment of naringenin's hepatoprotective efficacy. In this experiment, 24 SD rats underwent randomization to four experimental groups: (1) control (intraperitoneal saline injection), (2) Cd (1 mg/kg b.w. CdCl₂ intraperitoneal injection), (3) Cd + Nar (1 mg/kg b.w. CdCl₂ injection + 50 mg/kg b.w. naringenin oral gavage), and (4) Nar (50 mg/kg b.w. naringenin oral gavage) groups. After the 14-day interventions, serum and liver tissues were collected post-euthanasia. Hepatic injury markers (AST and ALT) and antioxidant enzymes (CAT and SOD) were quantified. Histopathology utilized HE and TUNEL staining. RT-qPCR and western blot analyses determined mRNA and protein expression levels of autophagy-related factors (p62 and LC3) and apoptosis-related factor (Caspase-3). The results demonstrated that cadmium exposure significantly reduced body weight, increased relative liver weight, elevated serum AST/ALT levels, and diminished hepatic CAT/SOD activity. Cadmium significantly upregulated both mRNA and protein expression levels of p62 and Caspase-3 while suppressing LC3 expression. Naringenin co-administration attenuated cadmium-induced hepatic injury, oxidative stress, impaired autophagy, and enhanced apoptosis. These findings collectively demonstrate that cadmium exposure induces hepatic injury and oxidative stress in rats through autophagy inhibition and apoptotic activation, while naringenin exerts protective effects by modulating these pathological processes.

Optimization of Naringenin Nanoparticles to Improve the Antitussive Effects on Post-Infectious Cough

Zhengqi Dong,Xiangtao Wang,Mingyue Wang,Rui Wang,Zheng Meng,Xiaotong Wang,Bo Yu,Meihua Han,Yifei Guo

Naringenin (NRG) is a natural compound with several biological activities; however, its bioavailability is limited owing to poor aqueous solubility. In this study, NRG nanoparticles (NPs) were prepared using the wet media milling method. To obtain NRG NPs with a small particle size and high drug-loading content, the preparation conditions, including stirring time, temperature, stirring speed, and milling media amount, were optimized. The NRG (30 mg) and D-α-tocopherol polyethylene glycol succinate (10 mg) were wet-milled in deionized water (2 mL) with 10 g of zirconia beads via stirring at 50 °C for 2 h at a stirring speed of 300 rpm. As a result, the NRG NPs, with sheet-like morphology and a diameter of approximately 182.2 nm, were successfully prepared. The NRG NPs were stable in the gastrointestinal system and were released effectively after entering the blood circulation. In vivo experiments indicated that the NRG NPs have good antitussive effects. The cough inhibition rate after the administration of the NRG NPs was 66.7%, cough frequency was three times lower, and the potential period was 1.8 times longer than that in the blank model group. In addition, the enzyme biomarkers and histological analysis results revealed that the NRG NPs can effectively regulate the inflammatory and oxidative stress response. In conclusion, the NRG NPs exhibited good oral bioavailability and promoted antitussive and anti-inflammatory effects. Keywords:naringenin;nanoparticle;antitussive effect;antioxidant effect;anti-inflammatory effect

Electrochemical behavior of naringenin and its sensitive determination based on a single-walled carbon nanotube modified electrode

Wenjing Wang,Jian Gao,Lu Wang,Baoxian Ye

A stable voltammetric sensor (SWCNTs/GCE), single-walled carbon nanotubes (SWCNTs) cast on a glassy carbon electrode (GCE), was employed in the initial electrochemical determination of naringenin. The complex electrochemical behavior of naringenin on this sensor was investigated in detail, and the dynamic and thermodynamic parameters were calculated. The new electroanalytical method for naringenin was proposed with a low detection limit of 2.0 × 10−8 mol L−1 (S/N = 3) and wide linear ranges from 8 × 10−8 to 5 × 10−6 mol L−1 and 5 × 10−6 to 1.2 × 10−5 mol L−1, respectively. More importantly, this sensor exhibited good reproducibility and long-term stability. Then this sensor was successfully applied for the determination of naringenin in Fructus Aurantii Immaturus with satisfactory results.

Effects of Naringin on the Activity and mRNA Expression of CYP Isozymes in Rats

Keling Cheng,Xuan Zeng,Hao Wu,Weiwei Su,Weiyang Fan,Yang Bai,Hongliang Yao,Peibo Li

Naringin (NRG) is a common dietary flavonoid in citrus fruits and has been documented to possess multiple pharmacological activities, including anti-oxidant, anti-inflammatory, and neuroprotective effects. Naringin is frequently consumed in combination with common clinical drugs. To date, the effects of NRG on cytochrome P450 enzymes have not been fully investigated yet. In this study, the activities of hepatic CYP1A2, CYP2D2, CYP2C9, CYP2C19, and CYP2E1 in rats after the continuous oral administration of NRG (50 and 500 mg/kg) were evaluated using cocktail probe-drug method. The concentrations of 5 probe drugs (phenacetin, dextromethorphan, diclofenac sodium, omeprazole, and chlorzoxazone) in rat plasma were simultaneously determined with a validated HPLC-MS/MS (high performance liquid chromatography-tandem mass spectrometry) method and then used to calculate corresponding pharmacokinetic parameters. Compared with the control group, the AUC(0-t), AUC(0-∞), t 1/2, and C max of each probe drug in treatment groups showed no significant differences. Meanwhile, fluorescence quantitative polymerase chain reaction (FQ-PCR) analysis revealed that NRG did not significantly affect the mRNA expressions of genes CYP1a2, CYP2d2, CYP2c6, CYP2c11, and CYP2e1 in rat liver. Based on these results, it could be concluded that NRG showed no significant effects on the activities and mRNA expressions of tested CYP450 in rats.

Protective effect of naringenin on cadmium chloride-induced renal injury via alleviating oxidative stress, endoplasmic reticulum stress, and autophagy in chickens

Shi Yaning,Gao Zhixin,Xu Bing,Mao Junbing,Wang Yue,Liu Zongping,Wang Jicang

Cadmium (Cd) is a highly hazardous toxic substance that can cause serious harm to animals. Previous studies have indicated that cadmium chloride (CdCl2) can damage organs, such as the liver, ovaries, and testicles. Naringenin (Nar) represents a flavonoid with various properties that promote the alleviation of Cd-induced damage. In this experiment, 60 chickens were divided into the control group, 150 mg/kg CdCl2 treatment group, 250 mg/kg Nar treatment group, and 150 mg/kg CdCl2 + 250 mg/kg Nar co-treatment group, which were treated for 8 weeks. Kidney tissues samples were collected to investigate kidney function, including oxidative stress (OS), endoplasmic reticulum (ER) stress, and autophagy activity. Experimental results showed the decreased weight of chickens and increased relative weight of their kidneys after CdCl2 treatment. The increase in NAG, BUN, Cr, and UA activities, as well as the increase in MDA and GSH contents, and the decrease activities of T-AOC, SOD, and CAT in the kidney, manifested renal injury by OS in the chickens. TUNEL staining revealed that CdCl2 induced apoptosis in renal cells. CdCl2 upregulates the mRNA and protein expression levels of GRP78, PERK, eIF2α, ATF4, ATF6, CHOP, and LC3, and inhibited the mRNA and protein expression levels of P62 proteins, which leads to ER stress and autophagy. The CdCl2 + Nar co-treatment group exhibited alleviated CdCl2-induced kidney injury, OS, ER stress, and autophagy. Research has demonstrated that Nar reduces CdCl2-induced kidney injury through alleviation of OS, ER stress, and autophagy.

The inhibitory effect of citrus flavonoids naringenin and hesperetin against purine nucleoside phosphorylase: Spectroscopic, atomic force microscopy and molecular modeling studies

Xingang Lv,Qilei Wang,Lang-Hong Wang,Er-Fang Ren,Deming Gong

In this work, the inhibitory effect of two citrus flavonoids naringenin and hesperetin on human purine nucleoside phosphorylase (hPNP) and their binding mechanism were evaluated. Results from enzymatic kinetics revealed that naringenin and hesperetin reversibly inhibited hPNP via a mixed-type manner with IC 50 values of 4.83 × 10 −4  M and 5.32 × 10 −4  M, respectively. Analysis of molecular modeling revealed that both naringenin and hesperetin bound directly into the active site by generating multiple forces including hydrogen bonding , π–π and π-Alkyl interactions with His64, Glu201, Ser220, His257, Phe200 and Val217 residues of hPNP, which caused the inhibition of hPNP activity. Moreover, conformational analysis by three-dimension fluorescence, circular dichroism and atomic force microscopy revealed that the binding of naringenin and hesperetin to hPNP induced changes in the microenvironment, secondary structure and morphology of hPNP. These results suggested that occupying the active site and enzymatic conformational perturbation induced by naringenin and hesperetin are the main reasons for reducing the inhibition of hPNP activity, which would be helpful in understanding the inhibitory mechanism of naringenin and hesperetin against hPNP.

Preparation and characterization of three-dimensional porous bioelectrets and study on bone repair in osteoporosis

Xinran Wang,Junfei Li,Jian Sha,Feilong Zhao,Liyang Zhang,Jianming Zhao,Abudureheman Bahatibieke,Biao Zhang,Yudong Zheng

Bone tissue possesses inherent electroactivity that is physiologically associated with osseous repair processes. Electret materials, which are capable of maintaining stable surface charges without external power, offer novel opportunities for in situ electrical stimulation therapies. This study fabricated a three-dimensional porous microcarrier bioelectret that combines polarized electret effects with naringin (NG) delivery to enhance bone regeneration in osteoporotic zebrafish. The microcarrier was prepared through in situ dopamine polymerization on nano-hydroxyapatite (nHAP@PDA), followed by integration with chitosan (CS) matrix, and was finalized with charge implantation via high-voltage polarization. The optimized 1 mg nHAP@PDA/CS (1nHAP@PDA/CS) formulation exhibited interconnected pores with high porosity. The surface charge density reached 105.2 nC/m 2 post-polarization and retained 12.4 nC/m 2 after 14 days. In rat bone mesenchymal stem cells (rBMSCs) culture, NG-loaded electrets (NG/nHAP@PDA/CS) increased ALP expression compared to nHAP@PDA/CS microcarriers. In the osteoporotic zebrafish model, NG/nHAP@PDA/CS electrets demonstrated therapeutic effects: The spinal calcein staining area showed enhancement at 8 days post-treatment, and skull mineralization, as quantified by alizarin red S staining, expanded. Treated zebrafish exhibited improved locomotion capacity, with increased swimming velocity and extended travel distance. These findings demonstrate the potential of NG/nHAP@PDA/CS electrets for bone regeneration applications.

Surface Deposition of Juglone/FeIII on Microporous Membranes for Oil/Water Separation and Dye Adsorption

Hui Yu,Qi-Zhi Zhong,Tian-Geng Liu,Wen-Ze Qiu,Bai-Heng Wu,Zhi-Kang Xu,Ling-Shu Wan

Deposition of dopamine and tannic acid has received great attention in the fields of surface and interface science and technology. The deposition behaviors of various metal–phenolic systems have been investigated, and it is generally accepted that at least one catechol group is essential to the formation of the coatings. Herein, we report a novel and effective surface-coating system based on the coordination complexes of FeIII ions with a natural product juglone that contains only one phenolic hydroxyl. We investigated the deposition behaviors of this novel system on various substrates. Microporous polypropylene membrane modified with juglone/FeIII coatings is superhydrophilic and underwater superoleophobic, showing high separation efficiency and good reusability for various oil/water emulsions. In addition, the modified membrane can adsorb anionic dyes and selectively remove them from dye mixtures with high efficiency. We further demonstrated that the coating is a result of the synergetic effect of juglone/FeIII coordination and FeIII hydrolysis. This work not only provides new insights into surface deposition systems but also expands the polyphenol family for surface coatings of multifunctional materials.
