Comparisons between gelatin-tussah silk fibroin/hydroxyapatite and gelatin-Bombyx mori silk fibroin/hydroxyapatite nano-composites for bone tissue engineering

Jiabing Ran,Jingxiao Hu,Guanglin Sun,Si Chen,Li Chen,Xinyu Shen,Hua Tong

In this research, gelatin-tussah silk fibroin/hydroxyapatite (GEL-TSF/HAp), gelatin-Bombyx mori silk fibroin/hydroxyapatite (GEL-BMSF/HAp), and gelatin/hydroxyapatite (GEL/HAp) nano-composites were synthesized by a novel in situ precipitation method. Characterizations, including surface morphology, elemental composition and distribution, structure of the crystalline phase, mechanical strength, thermal stability, and in vitro cytocompatibility, were carried out. Investigations on the crystalline phase showed that rod-like HAp crystallites in the GEL-TSF/HAp composite had a higher aspect ratio than those in the GEL-BMSF/HAp composite and the GEL/HAp composite. In addition, the GEL-TSF/HAp composite also presented better thermal stability than the other two composites revealed by differential thermal analysis (DTA) and thermogravimetric analysis (TGA). Mechanical properties testing indicated that the GEL-TSF/HAp composite had a higher elastic modulus at low strain and higher compressive modulus at high strain simultaneously than the other two composites. An in vitro cell culture showed that MG63 osteoblast-like cells on the GEL-TSF/HAp membrane took on higher proliferative potential than those on the GEL-BMSF/HAp membrane. These results indicated that compared to the GEL-BMSF/HAp composite, the GEL-TSF/HAp composite was more suitable for bone tissue engineering (BTE) applications.

A novel chitosan-tussah silk fibroin/nano-hydroxyapatite composite bone scaffold platform with tunable mechanical strength in a wide range

Jiabing Ran,Jingxiao Hu,Guanglin Sun,Si Chen,Pei Jiang,Xinyu Shen,Hua Tong

Currently, great efforts have been made to enhance the mechanical strength of bone tissue engineering (BTE) scaffolds, which are composed of biopolymeric matrices and inorganic nano-fillers. But the tunability of mechanical strength in a wide range for BTE scaffolds has seldom been investigated in spite of the great importance of this performance. In this work, a chitosan- tussah silk fibroin/hydroxyapatite (CS-TSF/HAp) hydrogel was synthesized by using a novel in situ precipitation method. Through in situ inducing the conformation transition of TSF in the CS-TSF/HAp hydrogel, which could be monitored by XRD, FT-IR, TGA, and DTA, the elastic modulus and fracture strength of the final CS-TSF/HAp composite could be tailored in a wide range without changing its composition, morphology, roughness, and crystal structures. The elastic modulus of the CS-TSF/HAp composite ranged from ∼250 to ∼400   MPa while its fracture strength ranged from ∼45 to ∼100   MPa. In order to clarify the rationale behind this process, a speculative explanation was provided. In vitro cell culture indicated that MC3T3-E1 cells cultured on the CS-TSF/HAp composite had positive adhesion, proliferation, and differentiation potential. We believed that the CS-TSF/HAp composite could be used as an ideal scaffold platform for cell culture and implantation of bone reconstruction.

An Eco-Friendly Antheraea Pernyi Silk Gland Protein/Sodium Alginate Multiple Network Hydrogel as Potential Drug Release Systems

Jia Li,Bo-Xiang Wang,De-Hong Cheng,Yue Zhang,Qiang Yao,Xin-bin Ji,Jing Guo,Yan-Hua Lu

To improve the versatility of the sodium alginate-loaded bio-hydrogels, Antheraea pernyi silk gland protein/sodium alginate drug-loaded hydrogels were prepared by using an eco-friendly multiple network cross-link technology. Fourier transform infrared (FT-IR) spectroscopy and UV-Vis spectrophotometer were used separately to evaluate the chemical structure and drug release behavior of drug-loaded hydrogels. The antibacterial drug carrier gels were evaluated by using inhibition zone test against theS. aureusandE. coli. The CCK-8 assay was used to assess the biocompatibility of drug loaded hydrogels. The FT-IR results showed that there was a strong interaction within the drug loaded hydrogels, and the ASGP was beneficial to enhance the interaction within the drug loaded hydrogels. UV-Vis spectrophotometer results indicated the cumulative release reached 80% within 400 min. Antibacterial bio-hydrogels had a good antibacterial property, especially the antibacterial bio-hydrogels with bacitracin exhibits superior to other antibacterial agents. The drug-loaded bio-hydrogels exhibited the adhesion and proliferation of RSC96 cells and perfected biocompatibility. This provides a new idea for further research and development of tissue-friendly drug-loaded biomaterials. Keywords:antheraea pernyi silk gland protein;sodium alginate;multiple network hydrogel;tissue engineering;drug release systems

Combined application of plant extracts (tannic acid and baicalin) with acid dyes in the simultaneous dyeing and finishing of tussah silk for enhanced antioxidant, antibacterial and UV protective properties

Ying-Jie Yu,Chen-Yu Li,Ren-Cheng Tang

The one-step dyeing and finishing of tussah fiber have advantages of short process, high efficiency, and energy and water saving. Additionally, to upgrade the functionalities of tussah silk fiber using natural chemicals with environmental benefits can increase its applicability. In this work, the combined application of plant extracts and acid dyes was presented for simultaneously imparting colorful shades, antioxidant activity , antibacterial activity, and UV protection capability to tussah silk. The stability of tannic acid , baicalin , and their mixtures with acid dyes was first discussed, and the colors and functionalities of dyed and finished tussah silk were evaluated. Tannic acid had less effect on the colors of dyed tussah silk. C.I. Acid Blue 113 was not stable in hot baicalin solution, and its pre-heated mixture with baicalin dyed tussah silk to dull red hue rather than blue hue. The bright blue and green colors of tussah silk were obtained by dyeing with anthraquinone and triarylmethane dyes, but their hues were affected by baicalin due to the light yellow color of baicalin and the matching of colors. Tannic acid exhibited better antibacterial property than baicalin on tussah silk, while baicalin had better antioxidant and UV protection capability than tannic acid . This work demonstrates that the one-step dyeing and finishing of tussah silk with acid dyes and plant extracts are feasible. However, special attention should be paid to the color changes of individual dyes for pale dyeing.

Waterproof strain sensor based on superhydrophobic, conductive carbon-coated tussah silk for human motion detection under hydrated environment

Yinghai Shao,Yuhan Zhang,Haoqian Luo,Lei Wang,Yuguo Peng,Jipeng Cao,Lanjie Xu,Yue Zhang,Junchi Ma

Conductive silk-based composites, prepared via surface modification techniques, have found extensive applications in wearable sensor technologies due to their excellent bio-compatibility and mechanical toughness. However, there is a critical limitation of traditional silk-based strain sensors that failing under hydrated conditions. Herein, we developed a waterproof strain sensor based on superhydrophobic and conductive tussah silk (SCTS) via a facile coating strategy that synergistically combined the carbon composite (one-dimensional hydrophobic MWCNTs, zero-dimensional hydrophobic CB, and TPU) with silk skeleton. The coating exhibited unprecedented mechanical robustness, the superhydrophobicity maintained stable even after stretching cycles (50 % strain), impacts, hand-kneading, and sandpaper abrasion, attributed to elasticity and strong adhesion of the TPU. Additionally, the SCTS sensor demonstrated exceptional strain sensing capabilities, featuring a wide sensing range (up to 120 % strain), high sensitivity (GF of 7.52), and superior cyclic stability (exceeding 500 cycles at 20 % strain). Owing to these merits, this wearable sensor was successfully applied for monitoring human motions under hydrated environment. Most important, it could even operate in water that traditional silk-based sensors cannot realize. The proposed tussah silk based sensor, integrated bio-inspired superhydrophobicity with strain response functionality, offered transformative potential for aqueous-environment applications ranging from marine exploration to medical diagnostics.
