High efficiency biosynthesis of gardenia blue and red pigment by lactic acid bacteria: A great potential for natural color pigments

Ling Li,Ling Zhou,Gongshuai Song,Danli Wang,Gongnian Xiao,Fuping Zheng,Jinyan Gong

Current production methods of the food colorants, gardenia blue (GB) and red (GR) pigments have low efficiency. One potential approach involves using lactic acid bacteria (LAB), which produce a high level of β-glucosidase, produce the GB and GR using non-toxic and harmless process. The isolated strain Lactobacillus plantarum S3 and the reference strain Lb. plantarum KCTC3104 showed high β-glucosidase activity levels of 1.01 and 1.44 unit/mL, respectively. The 12-h bioconversion yield of geniposide to genipin using two strains were 93.4% and 100%, respectively, which are high conversion percentage. For GB, the maximal production yield obtained using Lb. plantarum S3 and Lb. plantarum KCTC3104 under optimal conditions were 2.17 and 2.18 mg/mL, respectively. For GR, glutamic acid (Glu) with Lb. plantarum S3 is the best combination. To the best of our knowledge, this is the first report of an effective alternative method for the production of natural food colorants using LAB.

Triple-induced gardenia fruit extract-enriched gelatin/polysaccharides microgels for O/W emulsions, and food 3D printing

Zhiying Ouyang,Juncheng Zhu,Yang Cheng,Liangzhe Chen,Yuxin Yang,Liang Ma,Hankun Zhu,Yong Yu,Yuhao Zhang,Hongxia Wang

Gardenia fruit extract like gardenia blue is untable under light exposure and its application need expansion. Gardenia blue was proposed to be stabilized in triple-induced microgels from cooperative effects of gelatin, β-cyclodextrin and chitooligosaccharide, and its emulsifying ability and 3D printing potential were subesequently explored. The synergetic effects on gardenia blue due to the intermolecular interactions including hydrogen bonding and electrostatic interaction, contributed to the high UV irradiation stability of complexes. The complex microgels could absorb onto oil-water interface with emulsifying property, and the obtained high internal phase emulsions (HIPEs) illustrated outstanding centrifugal stability and great storage stability with low and uniform optical microscopic droplet diameters. The obtained HIPEs possessed excellent shearing thinning properties, excellent structure reversibility, high-degree reticulation, outstanding thixotropy , great elasticity, high cohesive energy and mechanical strength. 3D printing of HIPEs onto food also demonstrated high shape fidelity with low deviation. The findings illustrate the way of stabilizing gardenia blue in microgels by triple synergies, demonstrate the potential as stabilizer for developing HIPEs, and offered reference for highly-added-value utilization in the personalized 3D food products.

Surface Modification of Silk Fabric by Polysaccharide Derivatives towards High-Quality Printing Performance Using Bio-Based Gardenia Blue Ink

Yan Liang,Ni Wang,Qing Li,Huiyu Jiang

Ink-jet-printed silk, a premium textile material, was achieved by utilizing a bio-based gardenia blue dye. However, the sharpness of the printing pattern is difficult to control due to the limited water-retention capacity of silk. To address this issue, three polysaccharide derivatives, namely, sodium alginate (SA), low-viscosity hydroxypropyl methyl cellulose (HPMC-I), and high-viscosity hydroxypropyl methyl cellulose (HPMC-II), were employed as thickeners to modify the silk by the dipping–padding method. Firstly, the preparation of the gardenia blue ink and the rheology assessment of the thickener solution were conducted. Furthermore, the impacts of different thickeners on the micro-morphology, element composition, and hydrophilicity of the silk, along with the wetting behavior of the ink on the silk, were analyzed comparatively in order to identify an appropriate thickener for preserving pattern outlines. Lastly, the color features, color fastness, and wearing characteristics of the printed silk were discussed to evaluate the overall printing quality. Research results showed that the optimized ink formulation, comprising 12% gardenia blue, 21% alcohols, and 5.5% surfactant, met the requirements for ink-jet printing (with a viscosity of 4.48 mPa·s, a surface tension of 34.12 mN/m, and a particle size of 153 nm). The HPMC-II solution exhibited prominent shear-thinning behavior, high elasticity, and thixotropy, facilitating the achievement of an even modification effect. The treatment of the silk with HPMC-II resulted in the most notable decrease in hydrophilicity. This can be attributed to the presence of filled gaps and a dense film on the fibers’ surface after the HPMC-II treatment, as observed by scanning electron microscopy. Additionally, X-ray photoelectron spectroscopy analysis confirmed that the HPMC-II treatment introduced the highest content of hydrophobic groups on the fiber surface. The reduced hydrophilicity inhibited the excessive diffusion and penetration of gardenia blue ink, contributing to a distinct printing image and enhanced apparent color depth. Moreover, the printed silk demonstrated qualified color fastness to rubbing and soaping (exceeding grade four), a soft handle feeling, an ignorable strength loss (below 5%), and a favorable air/moisture penetrability. In general, the surface modification with the HPMC-II treatment has been proven as an effective strategy for upgrading the image quality of bio-based dye-printed silk.

Elucidating the formation mechanism of gardenia blue pigment from amino acid and genipin

Xiaoyu Zhang,Ruihu Zhang,Rui Li,Jiaxing Zhang,Yuefei Wang,Xin Chai,Yuefei Wang

Gardenia blue (GB), a natural active blue pigment, can be synthesized from iridoid glycosides and primary amino-containing compounds in vivo and in vitro . However, limited studies have reported GB synthetic intermediates, leading to unawareness of the formation mechanism of GB. Here, we reported that GB could be detected in rats′ feces, whose formation was simulated by in vitro reaction of amino acid-rich feces extract from rats and genipin (GP) metabolized from geniposide (GE) through β -glucosidase. Firstly, we proved that 14 amino acids (AAs) detected in rats′ feces containing primary amino groups can react with GP to produce GB. Then, taking histidine (His) as example, we characterized the key intermediates during the reaction process, including basic units genihistidine A (GH-A) and dimers. As an active basic unit, GH-A underwent covalent polymerization to form dimer, which then self-assembled into supramolecular spherical nanoparticles through multiple noncovalent interactions, including hydrogen bonds, π-π interactions, and van der Waals interactions. Our study revealed the formation mechanism of GB and provided insights into the bioavailable form of GE from traditional Chinese medicine in vivo .

Fibrous guided tissue regeneration membrane loaded with anti-inflammatory agent prepared by coaxial electrospinning for the purpose of controlled release

Min He,Jiajia Xue,Huan Geng,Hao Gu,Dafu Chen,Rui Shi,Liqun Zhang

Here, with the aim of inhibiting inflammation during guided tissue regeneration membrane (GTRM) implant surgery, coaxial electrospinning was used to fabricate drug-loaded core/sheath nanofiber GTRMs capable of controlled drug release. Various amounts of the anti-inflammatory agent metronidazole (MNA) were encapsulated into the core/sheath nanofibers (where PCL was the core, gelatin the sheath, and the gelatin shell was crosslinked with genipin) in order to establish the minimal drug content necessary to achieve the appropriate anti-inflammatory effect. By using TEM and SEM, the core/sheath structure was confirmed. In vitro drug disolution results showed that the core/sheath nanofibers exhibited sustained release profiles that were superior to those nanofibers produced by blending electrospinning. Additionally, the membrane significantly inhibited the colonization of anaerobic bacteria. Furthermore, with gelatin as a shell, the core/shell nanofiber membranes showed improved hydrophilicity, which resulted in better cell adhesion and proliferation without cytotoxicity. Therefore, in this study, a simple and effective coaxial electrospinning approach was demonstrated for the fabrication of anti-inflammatory GTRMs capable of providing controlled drug release.
