Silver magnetic amino silicone adhesive (Fe 3 O 4 @SiO 2 @NH 2 @Ag) particles were prepared for the purification of α-linolenic acid from tree peony seed oil under applied magnetic field. First, Fe 3 O 4 @SiO 2 @NH 2 @Ag particles were prepared and physicochemically characterized, including XRD, TG, FTIR, SEM, magnetic hysteresis curves and elemental analysis. The static process for the purification of α-linolenic acid using Fe 3 O 4 @SiO 2 @NH 2 @Ag particles was investigated, including adsorption curve, desorption curve, elution solvent composition and adsorption isotherm. The result indicated that 0-1–4% acetone- n -hexane elution solvent was selected for the gradient elution process, 2 h and 60 min were the time required to reach adsorption and desorption equilibrium, 20 °C was selected as the adsorption temperature, Langmuir model was suitable to fit and explain the equilibrium data, and the adsorption process was spontaneous and exothermic. Under applied magnetic field, the dynamic process for the purification of α-linolenic acid using Fe 3 O 4 @SiO 2 @NH 2 @Ag particles was investigated, and the optimum conditions were 20:1 µL/g loading amount, 0.5 mL/min flow rate and 51.73 Oe magnetic field intensity. After purification, the purity and recovery ratio of α-linolenic acid were calculated to be 94% and 74%, respectively. Furthermore, the recycled Fe 3 O 4 @SiO 2 @NH 2 @Ag particles still achieved better purification result. Therefore, the developed method shows a good application prospect in the field of separation and purification of α-linolenic acid.

The use of a mixed-salt substrate (sodium chloride-choline bromide-1-ethyl-1-methyl-pyrrolidinium bromide; molar ratio: 1:1:1) has been proposed for hydrodistillation of essential oils. Tree peony ‘Taiping Hong’ (Paeoniaceae), a cultivated essential oil-bearing plant, was investigated. After using the salt substrate, distillation time decreased from 140 to 40 min, essential oil yield increased from 0.48 % to 0.69 % and the pseudo-first-order rate constant increased from 0.0233 to 0.123 min-1. Potential mechanisms are (1) osmosis effect and hydrogen-bond interactions, which improve cell wall rupture; (2) the increase in polarity of aqueous distillation media, which prevents emulsification. The essential oil composition was influenced by an increase in oxygenated compound content (32.9 % to 49.0 %) and a decrease in hydrocarbon content (65.3 % to 49.6 %). Furan linalool oxide (cis and trans), linalool and α-terpineol contents increased by 400 %, 250 % and 200 %, respectively. (E,E)-α-farnesene, α-muurolene and octadecane contents decreased by 50 %, 46 % and 38 %, respectively. Possible mechanisms are: (1) improved cell rupture, which significantly facilitates extraction of oxygenated compounds; (2) hydrogen bonding with oxygenated compounds, which enhances their transfer from the inside of cells to bulk solution; (3) salting-out effect, which decreases dissolution of oxygenated compounds and improves their distillation. The essential oil obtained was mainly composed of phloroglucinol trimethyl ether (21.9 %), nerol (10.2%), tricosane (9.1 %), nonadecane (9.0 %), α-muurolene (5.0 %) and furan linalool oxides (3.0 %). Target prediction analysis indicated that it has potential applications in aromatherapy. In conclusion, the use of the mixed-salt substrate combined with hydrodistillation can result in efficient isolation of essential oils.

Cross-incompatibility is the major challenge in inter-sectional distant hybridization between tree peonies ( Paeonia sect. Moutan ) and herbaceous peonies ( Paeonia sect. Paeonia ). Most of the intersectional cross combinations rarely produce hybrid progenies or even seeds. However, a combination with Paeonia × lemoinei Rehd. ( P. delavayi × P. suffruticosa , tree type) as the pollen parent and P. lactiflora (herbaceous) as the seed parent resulted in many progenies. This research focuses on cytogenetics of two highly compatible intersectional combinations, P. lactiflora ‘Martha W.’ × P. × lemoinei ‘Golden Era’ (‘MW’ × ‘GE’) and P. lactiflora ‘Fen Yun Fei He’ × P. × lemoinei ‘L’Esperance’ (‘FYFH’ × ‘LE’). Karyotype analysis combined with genomic in situ hybridization (GISH) showed that the seed parents, ‘MW’ and ‘FYFH’, are both diploids (2 n = 2 x = 10, CC), while the pollen parents, ‘GE’ and ‘LE’, are tetraploid (2 n = 4 x = 20, AABB) and diploid (2 n = 2 x = 10, AB) respectively. Four progenies from ‘MW’ × ‘GE’, and another four progenies from ‘FYFH’ × ‘LE’, are all triploids (2 n = 3 x = 15, ABC). P. × lemoinei contributes two sets of chromosomes (AB) showing distinct signals in all tested triploids. The essential of a successful intersectional hybridization is to obtain 2 x gametes from tree peony. P. × lemoinei has better compatibility in intersectional hybridization than other tree peony species, probably because of its higher frequency of 2 x gamete (unreduced in diploid, or reduced in tetraploid) generation. Tetraploid tree peonies, such as ‘GE’, might produce abundant 2 x gametes through normal meiosis, which is the possible reason for their better fertility and superior intersectional compatibility than diploid tree peonies. The mechanism of compatibility in intersectional hybridization is supposed to be a rebalance between the distant divergent parents, tree peonies and herbaceous peonies, triggered by ploidy increase of gametes from tree peonies. The discovery of tetraploid tree peony also provides valuable materials to study the mechanism of polyploidization in sect. Moutan , and to promote the distant hybridization breeding in Paeonia.

Tree peonies are well-known horticultural and medicinal plants. The tree peony seeds, as emerging woody oil crops, recently have attracted great attention for their metabolites and bioactivities. In this study, the phytochemicals isolated from tree peony seed coats were systematically investigated. Seven polyphenolics were separated and prepared, mainly belonging to resveratrol derivatives. There was a great variation in the seed coat polyphenolic content among eight Paeonia species, and the contents of the resveratrol trimers and dimers were significantly higher in the seed coats of Paeonia ostii than other species. Based on the HPLC fingerprint characteristics and chemometric analysis, a clear discrimination among Paeonia plants was found, including the composition patterns and contents of the constituents. Moreover, the characteristic phytochemicals (vateriferol and trans-ε-viniferin) could significantly reduce the starch-mediated levels of postprandial blood glucose in diabetic/normal mice. In addition, in vitro enzyme tests showed that the two compounds could effectively and competitively inhibit α-glucosidase, with the IC50 values of 3.01 and 7.75 μM, respectively, indicating that vateriferol and trans-ε-viniferin could be therapeutic potential agents for hyperglycemia and diabetes mellitus.
