One pot synthesis of highly luminescent polyethylene glycol anchored carbon dots functionalized with a nuclear localization signal peptide for cell nucleus imaging

Lei Yang,Weihua Jiang,Lipeng Qiu,Xuewei Jiang,Daiying Zuo,Dongkai Wang,Li Yang

Strong blue fluorescent polyethylene glycol (PEG) anchored carbon nitride dots (CDs@PEG) with a high quantum yield (QY) of 75.8% have been synthesized by a one step hydrothermal treatment. CDs with a diameter of ca. 6 nm are well dispersed in water and present a graphite-like structure. Photoluminescence (PL) studies reveal that CDs display excitation-dependent behavior and are stable under various test conditions. Based on the as-prepared CDs, we designed novel cell nucleus targeting imaging carbon dots functionalized with a nuclear localization signal (NLS) peptide. The favourable biocompatibilities of CDs and NLS modified CDs (NLS-CDs) are confirmed by in vitro cytotoxicity assays. Importantly, intracellular localization experiments in MCF7 and A549 cells demonstrate that NLS-CDs could be internalized in the nucleus and show blue light, which indicates that CDs may serve as cell nucleus imaging probes.

Imaging of the nuclei of living tumor cells by novel ruthenium(II) complexes coordinated with 6-chloro-5-hydroxylpyrido[3,2-a]phenazine

Yang Ding,Qiong Wu,Kangdi Zheng,Linkun An,Xiaoying Hu,Wenjie Mei

Two novel ruthenium(II) complexes coordinated with 6-chloro-5-hydroxylpyrido[3,2-a]phenazine (CQM), [Ru(L)2(CQM)]ClO4 [L = 1,10-phenanthroline, (1) and 2,2′-bipyridine, (2)], were investigated as potential fluorescent probes to track dynamic changes in the nuclei of living cells. Confocal laser technology was used to observe their co-location inside the cells. The results showed that both complexes were taken up by HepG2 cells, especially 1, which was localized in the cell nuclei, whereas 2 was distributed in the cell nuclei and mitochondria. Further studies by real-time fluorescence observations revealed that 1 rapidly entered the living cells, namely, HepG2, HeLa and MCF-7 cells, imaged the dynamic changes of the nuclei of living tumor cells, and exhibited low toxicity toward cells. The results demonstrated that 1 may be developed into a novel fluorescent probe for living cell nuclei. This study facilitates the development of fluorescent chemosensors with metal complexes.

Hydrogen-Bond-Induced Emission of Carbon Dots for Wash-Free Nucleus Imaging

Haifang Liu,Jie Yang,Zhaohui Li,Lehui Xiao,Aaron Albert Aryee,Yuanqiang Sun,Ran Yang,Hongmin Meng,Lingbo Qu,Yuehe Lin,Xiaobing Zhang

Carbon dots (CDs) are emerging as powerful tools for biosensing and bioimaging because of their intrinsic properties such as abundant precursors, facile synthesis, high biocompatibility, low cost, and particularly robust tunability and stability. In this work, a new type of CDs was prepared from m-phenylenediamine and folic acid by hydrothermal method. Interestingly, the as-prepared CDs show blue emission in non-hydrogen-bonding solution, whereas robust green emission in hydrogen-bonding solution. Based on this phenomenon, a novel fluorescence sensing mechanism named as hydrogen-bonding-induced emission (HBIE) was proposed. The HBIE-CDs have large Stokes shift (141 nm) in water, good biocompatibility, and ultrasmall size, which facilitates their translocation into living cells. Very importantly, the as-prepared HBIE-CDs show strong affinity toward nucleic acid without interference from other biological species. After binding with DNA/RNA through hydrogen bond, as high as 6-fold green fluorescence enhancement of HBIE-CDs was observed. Since the nucleus is rich in DNA/RNA, these HBIE-CDs were successfully used for rapid and, especially, wash-free subcellular in situ imaging of the nucleus in living cells in a fluorescence turn on mode, which has a great practicability to be used for nucleus imaging in bioanalytical studies and clinical applications.

Clusteroluminescence from Random Aggregation of Micrometers to Ordered Assembly of Sub-10 Nanometers by Space-Confined Template Synthesis

Riliga Wu,Hongyu Dai,Weijiang Guan,Chao Lu

Clusteroluminescence (CL) clusters in the absence of aromatic or π-conjugated structures have emerged as a new family of luminescent materials due to their abundant sources and inherent biocompatibility. However, there is an inborn challenge in controlling the size of CL clusters from random aggregation of micrometers to ordered assembly of sub-10 nanometers. Such an inherent drawback significantly restricts their progress from theoretical research to practical applications. To address this obstacle, a novel approach using space-confined templates is proposed to synthesize CL clusters with tunable sizes. Space-confined templates with sizes of ≈2–6 nm are constructed to effectively confine non-aromatic molecules, yielding size-controllable and luminescence-tunable CL clusters. The versatility of this synthetic strategy is further proved by using non-conjugated molecules, such as L-valine and L-isoleucine. Finally, the potential applications of the synthesized CL clusters have been implemented in cell nucleus imaging owing to their sub-10 nm size and efficient luminescence. The success of this work not only offers a versatile space-confined approach to synthesize size-tunable CL clusters within sub-10 nm, but also opens new avenues for the deployment of CL clusters in advanced applications.

N-doped carbon dots derived from bovine serum albumin and formic acid with one- and two-photon fluorescence for live cell nuclear imaging

Mingqian Tan,Xintong Li,Hao Wu,Beibei Wang,Jing Wu

Carbon dots with both one- and two-photon fluorescence have drawn great attention for biomedical imaging. Herein, nitrogen-doped carbon dots were facilely developed by one-pot hydrothermal method using bovine serum albumin and formic acid as carbon sources. They are highly water-soluble with strong fluorescence when excited with ultraviolet or near infrared light. The carbon dots have a diameter of ∼8.32   nm and can emit strong two-photon induced fluorescence upon excitation at 750   nm with a femtosecond laser. X-ray photoelectron spectrometer analysis revealed that the carbon dots contained three components, C, N and O, corresponding to the peak at 285, 398 and 532   eV, respectively. The Fourier-transform infrared spectroscopy analysis revealed that there are carboxyl and carboxylic groups on the surface, which allowed further linking of functional molecules. pH stability study demonstrated that the carbon dots are able to be used in a wide range of pH values. The fluorescence mechanism is also discussed in this study. Importantly, these carbon dots are biocompatible and highly photostable, which can be directly applied for both one- and two-photon living cell imaging. After proper surface functionalization with TAT peptide, they can be used as fluorescent probes for live cell nuclear-targeted imaging.

The Synthesis and Functional Study of Multicolor Nitrogen-Doped Carbon Dots for Live Cell Nuclear Imaging

Yanan Zhang,Xingwei Zhang,Yanping Shi,Chao Sun,Nan Zhou,Haixia Wen

The nitrogen-doped carbon dots (N-CQDs) were synthesized by citric acid as a raw material and propylene diamine as a passivation agent. Structure, optical properties and biocompatibility of N-CQDs were analyzed. It was found that the N-CQDs possessed concentration-dependent, multicolor photoluminescence and low toxicity. As demonstrated in the imaging of bioluminescence, by adjusting the concentration of N-CQDs, the cell imaging effect can be adjusted. The internalized N-CQDs were concentrated in the nucleus. A novel tool for studying the nuclear changes during the cell cycle was developed.Keywords:nitrogen-doped carbon dots;multicolor illumination;multicolor cell nuclear imaging;mitosis

Conjugated Oligoelectrolyte with DNA Affinity for Enhanced Nuclear Imaging and Precise DNA Quantification

Xinmeng Zhang,Cheng Zhou,Jianxun Hou,Gang Feng,Zhourui Xu,Yonghong Shao,Chengbin Yang,Gaixia Xu

Precise DNA quantification and nuclear imaging are pivotal for clinical testing, pathological diagnosis, and drug development. The detection and localization of mitochondrial DNA serve as crucial indicators of cellular health. We introduce a novel conjugated oligoelectrolyte (COE) molecule, COE-S3, featuring a planar backbone composed of three benzene rings and terminal side chains. This unique amphiphilic structure endows COE-S3 with exceptional water solubility, a high quantum yield of 0.79, and a significant fluorescence Stokes shift (λex= 366 nm, λem= 476 nm), alongside a specific fluorescence response to DNA. The fluorescence intensity correlates proportionally with DNA concentration. COE-S3 interacts with double-stranded DNA (dsDNA) through an intercalation binding mode, exhibiting a binding constant (K) of 1.32 × 106M−1. Its amphiphilic nature and strong DNA affinity facilitate its localization within mitochondria in living cells and nuclei in apoptotic cells. Remarkably, within 30 min of COE-S3 staining, cell vitality can be discerned through real-time nuclear fluorescence imaging of apoptotic cells. COE-S3’s high DNA selectivity enables quantitative intracellular DNA analysis, providing insights into cell proliferation, differentiation, and growth. Our findings underscore COE-S3, with its strategically designed, shortened planar backbone, as a promising intercalative probe for DNA quantification and nuclear imaging.

Aptamer-Based Cell Nucleus Imaging via Expansion Microscopy

Yue Sun,Zhicheng Zhang,Tao Bing,Jing Liu,Weiwei Li,Xiangjun Liu,Nan Zhang,Yang Shu,Jianhua Wang,Dihua Shangguan

Expansion microscopy (ExM) is a newly developed technology in recent years that enables nanoscale imaging under conventional microscopes. Herein, we report an aptamer-based ExM imaging strategy. A nucleus-targeting aptamer Ch4–1 was chemically labeled with a dye and an acrydite at each end to perform the functions of molecular recognition, fluorescence reporting, and gel anchoring. After binding cell nucleus, the dual labeled aptamer Ac-Ch4–1-FAM directly participated in gelation and anchored in polyacrylamide gel. After expanding the gel, high-resolution imaging was achieved by confocal microscopy. Multicolor ExM imaging was also realized by combining Ac-Ch4–1-FAM, antibodies and fluorescent dyes. This aptamer-based ExM could clearly image the chromatin morphology at different mitotic stages. The expansion process is simple and the aptamer labeling is easy. The aptamer-based ExM holds great promise in super-resolution imaging of cells and tissues.

One-pot synthesis of efficient multifunctional nitrogen-doped carbon dots with efficient yellow fluorescence emission for detection of hypochlorite and thiosulfate

Yifang Gao,Yang Liu,Huilin Zhang,Wenjing Lu,Yuan Jiao,Shaomin Shuang,Chuan Dong

CD-based ratiometric fluorescence probes are of great significance for visual detection, but accomplishing this goal is still a particularly challenging task. Herein, nitrogen-doped carbon dots (NCDs) with bright yellow fluorescence were easily manufactured via a one-pot hydrothermal method for visual detection of hypochlorite (ClO−) and thiosulfate (S2O32−) under UV light irradiation. The as-prepared NCDs demonstrate favorable water solubility, excellent biocompatibility, superior optical properties and low cytotoxicity. Strikingly, the fluorescence of the NCDs could be quenched with ClO−. Based on these results, an original fluorescent nanoprobe was constructed for the highly discriminating recognition of ClO− by oxidation of the amino groups on their surface to nitro groups. The assay covered the ranges from 0.067 to 19.33 μM and 24 to 98 μM with a limit of detection (S/N = 3) of as low as 0.013 μM. Remarkably, a growing peak appears at 537 nm and the emission at 492 nm shrinks with the introduction of S2O32−, which demonstrates ratiometric fluorescence emission characteristics (F537nm/F492nm) in the range of 6.6–100 μM with a limit of detection (S/N = 3) of as low as 0.78 μM. In addition, the fluorescence color of the NCDs also changes (yellow-green-blue) after adding various ClO− concentrations. The fluorescence color of the NCDs–ClO− also changes (blue-green-yellow) after adding various S2O32− concentrations. This excellent ratiometric fluorescence probe was successfully further used for nuclear imaging. Accordingly, an easy-to-prepare paper-based sensor to identify ClO− and S2O32− was fabricated, which demonstrated their adaptability for in situ on-site testing. This research further opens up new opportunities for the development of efficient yellow fluorescent probes based on NCDs nanomaterials for visual detection, biomarking, and biomedical optical imaging.

Proton-Mediated Dynamic Nestling of DNA Payloads Within Size-Matched MOFs Nanochannels for Smart Intracellular Delivery

Xiaowen Wang,Hong Zhang,Ping-Ping He,Xiaoxue Du,Yuxin Shen,Wensheng Cai,Weiwei Guo

With sequence-programmable biological functions and excellent biocompatibility, synthetic functional DNA holds great promise for various biological applications. However, it remains a challenge to simultaneously retain their biological functions while protecting these fragile oligonucleotides from the degradation by nucleases abundant in biological circumstances. Herein, a smart delivery system for functional DNA payloads is developed based on proton-mediated dynamic nestling of cytosine-rich DNA moieties within the precisely size-matched nanochannels of highly crystalline metal–organic frameworks (MOFs): At neutral pH, cytosine-rich DNA strands exhibit a flexible single-stranded state and can be accommodated by MOFs nanochannels with a size of ca. 2.0 nm; while at acidic conditions, the protonation of cytosine-rich strands weakens their interaction with the nanochannels, and the tendency to form four-stranded structures drives these DNA strands out of the nanochannels. Results confirm the successful protection of DNA payloads from enzymatic hydrolysis by the MOFs nanochannels, and the delicate coupling of the endocytosis processes and the proton-responsive Cytosine-rich DNA/MOFs systems realized the efficient intracellular delivery of DNA payloads. Furthermore, with a complementary sequence to the telomere overhangs, direct imaging of telomeres and the nucleus is successfully achieved with the proton-mediated DNA/MOFs system.

Multimodal Imaging-Guided Strategy for Developing 177Lu-Labeled Metal–Organic Framework Nanomedicine with Potential in Cancer Therapy

Ranxi Liang,Feize Li,Xijian Chen,Fuyuan Tan,Tu Lan,Jijun Yang,Jiali Liao,Yuanyou Yang,Ning Liu

Nano-metal–organic frameworks (nano-MOFs) labeled with radionuclides have shown great potential in the anticancer field. In this work, we proposed to combine fluorescence imaging (FI) with nuclear imaging to systematically evaluate the tumor inhibition of new nanomedicines from living cancer cells to the whole body, guiding the design and application of a high-performance anticancer radiopharmaceutical to glioma. An Fe-based nano-MOF vector, MIL-101(Fe)/PEG-FA, was decorated with fluorescent sulfo-cyanine7 (Cy7) to investigate the binding affinity of the targeting nanocarriers toward glioma cells in vitro, as well as possible administration modes for in vivo cancer therapy. Then, lutetium-177 (177Lu)-labeled MIL-101(Fe)/PEG-FA was prepared for high-sensitive imaging and targeted radiotherapy of glioma in vivo. It has been demonstrated that the obtained 177Lu-labeled MIL-101(Fe)/PEG-FA can work as a complementary probe to rectify the cancer binding affinity of the prepared nanocarrier given by fluorescence imaging, providing more precise biodistribution information. Besides, 177Lu-labeled MIL-101(Fe)/PEG-FA has excellent antitumor effect, leading to cell proliferation inhibition, upregulation of intracellular reactive oxygen species, tumor growth suppression, and immune response-related protein and cytokine upregulation. This work reveals that optical imaging and nuclear imaging can work complementarily as multimodal imaging in the design and evaluation of anticancer nanomedicine, offering a MIL-101(Fe)/PEG-FA-based pharmaceutical with potential in tumor endoradiotherapy.

A transparent hybrid metal halide glassy scintillation screen for high-resolution fast neutron radiography

Zhou Zi’an,Zheng Jinxiao,Ruan Shihao,Yue Guichu,Feng Tiao,An Yini,Wu Meimei,Wang Nü,Zhou Shuyun,He Linfeng,Sun Chenghua

Fast neutron radiography offers exceptional penetration for high-density and bulky objects, yet its resolution is hindered by light scattering in conventional scintillators and screen fabrication techniques. To address this, here, we develop a transparent glassy Mn-based hybrid metal halide scintillation screen, (BTPP) 1.8 (HTPP) 0.2 MnBr 4 (BTPP + = butyltriphenylphosphonium, HTPP + = heptyltriphenylphosphonium), leveraging temperature-dependent ordered-disordered transitions. The large-area screen boasts >70% visible light transmittance (500–800 nm), a high photoluminescence quantum yield (~85.54%), and threefold higher light output than commercial ZnS (Ag): PP screens. With a spatial resolution of 5 lp mm −1 , it surpasses existing scintillators. This hybrid material enables imaging of heavy objects with clear hierarchical details, providing accurate data for non-destructive detection while offering an alternative approach to scintillator design, advancing the potential of fast neutron radiography.
