Proposal and application of a novel extraction and purification technology for juglone from Juglans mandshurica waste branches: Reverse micellar microemulsion combined with magnetic solid phase extraction

Mengfei Tian,Chunyu Liang,Naveed Ahmad,Chunying Li,Weihao Wang,Yu Zhang,Zidan Luo,Lequn Qiu,Chunjian Zhao

A reverse micellar microemulsion combined with magnetic solid phase extraction (RMM-MSPE) technology was developed for the extraction and purification of juglone from Juglans mandshurica . Reverse micellar microemulsion (RMM) (Tween 80: n-propanol: n-hexane: 0.05 mol/L KCl aqueous solution = 27:13.5:4.5:55; w/w/w/w) was used to extract juglone based on microwave. The optimum conditions were a pH of 5.6, a liquid-solid ratio of 20:1 (mL/g), temperature of 40 °C, time of 63 s, and power of 400 W. Graphene oxide/nano diamonds/ Fe 3 O 4 (GO/ND/Fe 3 O 4 ) was prepared as adsorbent and applied to purify juglone by magnetic solid-phase extraction (MSPE). The optimum adsorption conditions were time of 14 h, solid to liquid ratio of 30 mg/mL, pH of 5.5 and shaker speed of 200 r/min. Anhydrous methanol provided the best desorption of juglone and the optimum conditions were a desorption time of 16 h, pH of 8, and the solid-liquid ratio of 40 mg/mL. The purity of juglone was 69.08% and the recovery yield was 78.67%. RMM and GO/ND/Fe 3 O 4 could be reused three times and eight times after the first use, respectively. The proposed RMM-MSPE is a green and efficient alternative method for extraction and purification of juglone.

Enrichment and purification of total flavonoids from Cortex Juglandis Mandshuricae extracts and their suppressive effect on carbon tetrachloride-induced hepatic injury in Mice

Pan Zhao,Chao Qi,Gang Wang,Xinpeng Dai,Xiaohong Hou

In the present work, a simple and efficient chromatographic separation method was developed for preparative separation and enrichment of total flavonoids (TFs) from Cortex Juglandis Mandshuricae (CJM) extracts and then the protective effect of TFs against CCl 4 -induced acute liver injury in mice was investigated. Enrichment and purification of TFs from CJM extracts were studied using six macroporous resins and HPD-750 resin was selected as the best resin according to its adsorption and desorption properties. The operating parameters of resin column chromatography were optimized. Under the optimal conditions, TFs from CJM with purity larger than 50% were produced and their antioxidant activity was further evaluated in vitro. The mice were orally administrated with the purified TFs for seven days and then given CCl 4 (0.3%, 10   mL/kg i.p.). The results showed that TFs of CJM significantly attenuated the activities of serum aspartate transaminase (AST) and alanine transaminase (ALT) compared with model group, as well as the relative liver weight. Histopathological observation also revealed that TFs reduced the incidence of liver lesions and improved hepatocyte abnormality. Moreover, oral administration of TFs significantly enhanced antioxidant enzyme activities (superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px)) and decreased the content of malondialdehyde (MDA). Histopathological and biochemical results elicited that TFs of CJM had significant hepatoprotective activity comparable to the standard silymarin. This is the first time to reveal the protective actions of the TFs from CJM against CCl 4 -induced liver damage in mice and this natural product should be developed as a new drug for treatment of live injury in future.
