There is uncertainty in quantifying the toxic effects of total chromium (Cr) in the environment by modeling the toxicity of individual Cr(III) or Cr(VI). In the present study, the effects of Cr(III) and Cr(VI) on barley root elongation were investigated in a hydroponic system where Cr(III) and Cr(VI) combination dose-response experiments under monotoxicity concentration, single-dose addition, and fixed concentration ratios were designed to identify and quantify their combined phytotoxicity of one element with various valences. The results show that the calculated mixed toxicity unit values for 50% inhibition (TU mix50 ) ranged from 1.06 to 1.45, indicating the weak antagonism effects of Cr(III) and Cr(VI) on barley root toxicity. Also, the single-dose group experiment has proved that the EC 50 of Cr(VI) was increased from 71.2 μM to 119.9 μM with Cr(III) addition, which suggested that Cr(III) has antagonism on the toxicity of Cr(VI). While EC 50 of Cr(III) was not affected by Cr(VI) addition. After introducing the expansion coefficient of Cr(III) on Cr(VI) toxicity, both the extended concentration addition model (e-CA) based on the log-logistic and Weibull equations and the extended independent action model (e-IA) could more accurately predict the barley root elongation under Cr(III) and Cr(VI) interaction. The e-CA model based on the Weibull equation had almost the best correlation coefficient (R 2 ) and lowest root mean square error (RMSE) between the measured and predicted values. Finally, the combined toxicity and antagonism of the same element with co-existing different valences simultaneously were successfully and firstly identified and quantified in the present study.

Toxicity of engineered nanoparticles on organisms is of concern worldwide due to their extensive use and unique properties. The impacts of ZnO nanoparticles (ZnO NPs) on seed germination and root elongation of corn ( Zea mays L.) and cucumber ( Cucumis sativus L.) were investigated in this study. The role of seed coats of corn in the mitigation toxicity of nanoparticles was also evaluated. ZnO NPs (1,000 mg L −1 ) reduced root length of corn and cucumber by 17 % ( p < 0.05) and 51 % ( p < 0.05), respectively, but exhibited no effects on germination. In comparison with Zn 2+ , toxicity of ZnO NPs on the root elongation of corn could be attributed to the nanoparticulate ZnO, while released Zn ion from ZnO could solely contribute to the inhibition of root elongation of cucumber. Zn uptake in corn exposed to ZnO NPs during germination was much higher than that in corn exposed to Zn 2+ , whereas Zn uptake in cucumber was significantly correlated with soluble Zn in suspension. It could be inferred that Zn was taken up by corn and cucumber mainly in the form of ZnO NPs and soluble Zn, respectively. Transmission electron microscope confirmed the uptake of ZnO NPs into root of corn. Although isolation of the seed coats might not be the principal factor that achieved avoidance from toxicity on germination, seed coats of corn were found to mitigate the toxicity of ZnO NPs on root elongation and prevent approximately half of the Zn from entering into root and endosperm.

CrRLK1L receptor kinases, particularly FERONIA ( FER ), play critical roles in plant stress adaptation and hormonal signaling, yet their functional characterization in economically vital crops like Brassica napus remains limited. Here, we identified 108 CrRLK1L family members through genome wide screening in B. napus and focused on BnFERONIA , a homolog evolutionarily conserved with FERONIA orthologs in Arabidopsis and related crucifers. CRISPR/Cas9 generated BnFERONIA knockout mutants ( cas9-fer ) in Arabidopsis exhibited contrasting abiotic stress responses: hypersensitivity to salt stress (100 mM NaCl), with root elongation was significantly inhibited compared to wild-type, and enhanced tolerance to osmotic stress (300 mM mannitol), displaying 20% longer roots. Furthermore, cas9-fer mutants showed pronounced ABA hypersensitivity, with germination rates significantly suppressed under 0.25 μM ABA and vegetative growth stunted, indicating the role of BnFERONIA in attenuating ABA signaling. Strikingly, osmotic-induced germination remained unaffected in mutants, suggesting there was a compensatory mechanisms during the early stress perception. This research provides the first functional evidence of BnFERONIA gene during stress adaptation, offering a molecular framework for creating stress-resistant oilseed crops by specifically manipulating the CrRLK1L pathways.

As representative compounds spanning three generations of diamide insecticides, flubendiamide, chlorantraniliprole, and tetraniliprole have been extensively employed in agricultural practices. However, their widespread application induces oxidative stress in plants and underlying toxic mechanisms remain poorly understood. Given the important roles in oxidative stress, glutathione reductase 2 from Arabidopsis thaliana ( At GR2) was applied as a biomarker to evaluate the toxicological effects of diamide insecticides. Firstly, the root elongation in Arabidopsis seedlings was significantly inhibited and the At GR2 gene expression level was up-regulated under exposure of the diamide insecticides. Then the glutathione reductase activity of At GR2 was decreased with the increase concentration of these three insecticides, especially for flubendiamide with similar detrimental effects at much lower concentrations. Moreover, the binding affinity between flubendiamide and At GR2 was also stronger than that of chlorantraniliprole and tetraniliprole, which was conformed with molecular docking results. Additionally, much more evident effects on the structure of At GR2 under exposure of flubendiamide was detected through multispectral techniques compared to the other two insecticides. Hence this study systematically explored the binding mechanism of these three-generations of diamide insecticides with At GR2 at the molecular level, and further provided critical assessments of their phytotoxic effects on plant development and agricultural systems.

Co-pollution of antibiotics and heavy metal copper (Cu) is common in freshwater environments because of their wide use as antimicrobial agents, especially in aquaculture. However, the toxic effects of coexisting antibiotics and heavy metals on aquatic plants remain unclear. This study investigated the effect of four antibiotics (i.e., enrofloxacin, ENR; tetracycline, TC; sulfamethoxazole, SMX; erythromycin, ERY), Cu, and their mixture on the growth and physiological responses of Hydrilla verticillata (L.f.) Royle. Results showed that the four antibiotics exhibited toxic effects on the growth and physiological indicators of H. verticillata , and root elongation was the most sensitive endpoint of the phytotoxicity test. The median effect concentration (EC 50 ) of root elongation indicated that TC (EC 50 = 10.05 mg/L) has the highest level of growth toxicity, and the toxicity of ENR to aquatic plants was close to TC (EC 50 = 10.44 mg/L), followed by SMX (EC 50 = 20.08 mg/L). However, there was no significant toxic effect of 20 mg/L ERY on the root elongation. Hydrophobicity may be a key factor affecting the phytotoxicity of antibiotics. Moreover, antagonistic toxic effects were observed under ENR + Cu, TC + Cu, SMX + Cu, and ERY + Cu co-exposures at all the experimental concentrations (0.01–20 mg/L). Due to the concentrations of antibiotics in natural waters usually with ng/L levels, our results suggested that environmental antibiotic concentrations probably pose low ecological risk to aquatic plants and indicated the H. verticillata could be used as phytoremediation candidate to remove antibiotic or antibiotic–Cu pollutions in general nature water.

Ammonium toxicity affecting plant metabolism and development is a worldwide problem impeding crop production. Remarkably, rice ( Oryza sativa L.) favours ammonium as its major nitrogen source in paddy fields. We set up a forward-genetic screen to decipher the molecular mechanisms conferring rice ammonium tolerance and identified rohan showing root hypersensitivity to ammonium due to a missense mutation in an argininosuccinate lyase (ASL)-encoding gene. ASL localizes to plastids and its expression is induced by ammonium. ASL alleviates ammonium-inhibited root elongation by converting the excessive glutamine to arginine. Consequently, arginine leads to auxin accumulation in the root meristem, thereby stimulating root elongation under high ammonium. Furthermore, we identified natural variation in the ASL allele between japonica and indica subspecies explaining their different root sensitivity towards ammonium. Finally, we show that ASL expression positively correlates with root ammonium tolerance and that nitrogen use efficiency and yield can be improved through a gain-of-function approach.

Structural analogs of enzyme substrates involved in plant hormone biosynthesis can either inhibit or enhance hormone-related responses depending on their specific chemical characteristics. Ethylene is an important gaseous phytohormone regulating a myriad of physiological and developmental processes. The substrate of the ethylene biosynthesis key enzyme ACC oxidase (ACO), 1-aminocyclopropane-1-carboxylate acid (ACC), is the direct precursor of ethylene. Previous studies have identified a series of ACC analogs as ethylene antagonists competitively binding with ACO. Here, this study identified methyl 1-aminocyclopropanecarboxylate (methyl-ACC), a structural analog of ACC, as an agonist of ethylene response in plants. Methyl-ACC triggered enhanced ethylene-related responses in plants similar to the effects of ACC, such as restrained root elongation, increased root hair number, promoted dark-induced leaf senescence, and accelerated ripening of postharvest tomato fruit. A higher amount of ethylene release was detected from the tomato leaves treated with methyl-ACC than from the mock control. These bioactivities of methyl-ACC render it a potential plant growth regulator (PGR) for the agricultural and postharvest industries. RT-qPCR revealed a generally upregulated expression of the ethylene biosynthesis genes in detached tomato leaves under methyl-ACC treatment. In vivo and in vitro enzymatic activity analyses showed that methyl-ACC had no obvious effect on the total ACO activity in detached tomato leaves nor on the activity of recombinant SlACO1, the predominantly expressed ACO family member in this tissue. This study offers a promising new PGR and adds to the current knowledge on the structure–function-regulation relationship of compounds associated with ethylene biosynthesis. The mechanism by which methyl-ACC exerts its function is discussed.

Soil pollution caused by complex organochloride mixtures has been increasing in many parts of the world in recent years; as a result, countless numbers of people are exposed to dangerous pollutions; hence, the treatment of organochlorides-polluted soils is gaining considerable attention. In this study, the potential of unactivated peroxymonosulfate (KHSO 5 ) in remediating soil co-contaminated with trichlorophenol, para-dichlorobenzene, and para-chloro-meta-cresol was investigated. In addition, the treatment's collateral effect on critical soil properties was explored. The result revealed that treating 10 g of soil with 20 mL of 5 mM KHSO 5 for 60 min could oxidize 70.49% of the total pollutants. The pH of the soil was decreased following the treatment. The significant decrease, ( p < 0.05), in the soil organic matter following the remediation has affected cation exchange capacity, and available nitrogen. It was also observed that the treatment reduced the β-glucosidase, urease, invertase, and cellulase activities significantly, ( p < 0.05). The treatment, on the other hand, brought negligible effects on available phosphorus, available potassium, and particle size distribution . The phytotoxicity tests, which included seed germination and root elongation and soil respiration tests revealed that the treatment did not leach toxins into the treated soil. The treatment method was found to be relatively ecofriendly and cost effective.
